A morphological reexamination on the genus Adonis L. sensu lato (Ranunculaceae) in Korea (한국산 복수초속(미나리아재비과)의 형태분류학적 재검토)

  • The morphology of plants and scanning electro-microscopical features of fruits were examined to evaluate the taxonomic entities of Korean Adonis L. in sensu lato (Ranunculaceae). The species of Korean Adonis were readily diagnosed by the branching pattern of stem, the number of flowers, the bifurcation of leaf main axis, the number of tepals, the flowering periods in relation to leaf sprouting, and the number of stomata and the texture of fruit surface. This study demonstrates that there are three species, A. amurensis, A. pseudoamurensis, and A. multiflora., found in Korea. However, A. ramosa, which has been often claimed to be distributed in Korea, is not present in Korea.

Changes in resource allocation among vegetative organs during the clonal growth of Polygonatum humile (Liliaceae) grown in a temperate forest gap

  • Background: The aim of this study was to examine the resource allocation among the organs and the leaf morphology of Polygonatum humile. The data were collected from June 2014 to May 2015 in a natural P. humile stand of a temperate forest gap. Results: The dry weight of new rhizome ($R_0$) sharply decreased to the time of new shoot sprouting in next year and was constant for 2 ~ 3 years. However, dry weight of root on $R_0$ increased to the end of growth season and, thereafter, decreased slowly along the time elapsed. The correlation coefficients between the rhizome and the leaf sizes were 0.5 for the last year's rhizome ($R_1$) and 0.6 for $R_0$ and were significant at 0.1% level for $R_0$. The increase of one leaf a plant led to increase of the total leaf area, total leaf weight, and stem length. Moreover, the organ sizes' differences between two plants of the one leaf difference were almost significant at 1% level. In 11-leaf plant, the leaf length/width ratio and specific lea area increased to the second, relative leaf area to the fourth and relative leaf dry weight to the fifth, and thereafter, decreased to the last leaf. The differences in the values of these four parameters between two leaves on a stem were almost significant at 5% level. Conclusion: P. humile examined showed the close relationships between the last year's rhizome or this year's one and the shoot system (leaf and stem) sizes. The leaf number a shoot greatly influenced specific leaf area. P. humile's leaf might showed diverse morphology with leaf rank on a stem in a forest gap environment.

A Study on Schütte-Lihotzky's Concepts of Residential Planning in the Red Wien Era ('레드 빈' 시대 쉬테-리호츠키의 주거 계획 개념에 대한 연구)

  • In addition to the well-known 'Frankfurt Kitchen', Austrian female architect Margarette $Sch{\ddot{u}}tte-Lihotzky^{\prime}s$ architectural achievements include a study on the housing for working class, contribution to the resident participation movement, the planning of multi-family housing complexes, rational floor plan design, and the design and study of interior spaces. The purpose of this study is to investigate how she, who played intermediate roles between an architect and a social reformer, reflected social issues and the demands of living on the housing plans in the Red Wien era and to discuss diverse issues of modern residential planning furthermore. In this study, first, the housing situation in Vienna immediately after World War I was grasped and the architectural and housing discourses at that time were examined. Thereafter, cases of $Sch{\ddot{u}}tte-Lihotzky^{\prime}s$ works were investigated and analyzed. In the period of transition to modern times, the concepts of residential planning of $Sch{\ddot{u}}tte-Lihotzky$, who began as a social democratic architect, often showed transitional tendencies in private and public functions of housing, spatial distribution for the daily life, and functions of modern family and home. However, thereafter, the paradigm of the rationalism-functionalism was already sprouting from her architectural and residential plans. In conclusion, it can be said that the works of $Sch{\ddot{u}}tte-Lihotzky$ has realized the social responsibility of residential planning and become a cornerstone of the later modern housing.

A new functional mushroom cultivated variety Jangsaeng of Fomitella fraxinea (기능성버섯 장수버섯의 신품종 『장생』)

  • A new functional mushroom cultivated variety "Jangsaeng" of Fomitella fraxinea was developed by crossing monokaryons derived from collected strains for contribution to mushroom farmers. The major characteristics of this mushroom were compared to Yeongji 2ho of Ganoderma lucidum because of their morphological similarities. The optimal temperature of mycelial growth and fruitbody sprouting and growth were 30 and 25~30, respectively. The surface color of fruitbody pilus was reddish brown and the fruitbody was flat kidney-shaped like Ganoderma lucidum except that the stipe was absent. Its texture was so tough and rigid that the yield per 1,000 ml bottle might be as high as 136 g. The hot water extract of this medicinal mushroom is fit to drink just like Ganoderma lucidum but its taste is not bitter.

Developing a mass propagation technique for Aralia elata via somatic embryogenesis

  • Aralia elata is found in mountain areas all over Korean peninsula. Aralia elata is the scientific name for Japanese angelica tree. The tree belongs to the family Araliaceae, commonly known as ginseng family. Bud sprouts from apical shoot tip of the plants are rich in flavor and thus mainly used for both folk medicine and vegetable. The stalks with apical buds are gathered in the early spring and planted in sandy soil or water in the greenhouse. The sprouting buds are then collected and sold as fresh vegetable. Although the plants have been used for food, they have been cultivated in a very small scale. In spring, local farmers just go around mountain areas to search the trees and gather the stalks as much as they get and sell them to the market. No conservation efforts have been made to stop the exploitation or to save the dwindling population. We tried to provide local farmers with the plants that may be used as an alternative to stalks from wild populations. This will hel! p conserve the wild populations. However, it is hard to propagate them either by conventional cuttings or by seed germination in a short period of time. Mass propagation using tissue culture systems have shown a great promise with several woody plants. Recently we developed a mass propagation technique via somatic embryogenesis system using mature and/ or juvenile explants for Aralia elata. Several factors affecting somatic embryogenesis system including SE(somatic embryo) induction, embryogenic callus proliferation, SE germination, plant regeneration and transplanting to field will be presented. And some problems arising for the somatic embryogenesis system will be also discussed.lso discussed.

Controlled Release of Nerve Growth Factor from Sandwiched Poly(L-lactide-co-glycolide) Films for the Application in Neural Tissue Engineering

  • In order to fabricate new sustained delivery device of nerve growth factor (NGF), we developed NGF-loaded biodegradable poly(L-lactide-co-glycolide) (PLGA, the mole ratio of lactide to glycolide 75:25, molecular weight: 83,000 and 43,000 g/mole, respectively) film by novel and simple sandwich solvent casting method for the possibility of the application of neural tissue engineering. PLGA was copolymerized by direct condensation reaction and the molecular weight was controlled by reaction time. Released behavior of NGF from NGF-loaded films was characterized by enzyme linked immunosorbent assay (ELISA) and degradation characteristics were observed by scanning electron microscopy (SEM) and gel permeation chromatography (GPC). The bioactivity of released NGF was identified using a rat pheochromocytoma (PC-12) cell based bioassay. The release of NGF from the NGF-loaded PLGA films was prolonged over 35 days with zero-order rate of 0.5-0.8 ng NGF/day without initial burst and could be controlled by the variations of molecular weight and NGF loading amount. After 7 days NGF released in phosphate buffered saline and PC-12 cell cultured on the NGF-loaded PLGA film for 3 days. The released NGF stimulated neurite sprouting in cultured PC-12 cells, that is to say, the remained NGF in the NGF/PLGA film at 37 $^{\circ}C$ for 7 days was still bioactive. This study suggested that NGF-loaded PLGA sandwich film is released the desired period in delivery system and useful neuronal growth culture as nerve contact guidance tube for the application of neural tissue engineering.

Content of Rutin and Monacolin K in the Red Buckwheat Fermented with Monascus ruber (홍국균 발효 메밀에서의 rutin과 monacolin K의 함량 변화)

  • Buckwheat contains dietary rutin, a flavonol glycoside, which is able to antagonize hypertension in human and has antioxidant activity. Monacolin K, a secondary metabolite produced by fungi Monascus species, is an effective specific inhibitor of cholesterol biosynthesis. To develop functional health food, dehulled buckwheat was fermented with Manascus ruber to produce Anka, and contents of rutin and monacolin K and Anka were determined. The rutin content in dehulled buckwheat decreased slightly after germination, but increased with the growth of sprout. Growth of fungi had no influence on rutin content in dehulled buckwheat Anka, Monacolin K content in fermented dehulled buckwheat malt was lower than that in dehulled buckwheat Anka. Monacolin K content in dehulled buckwheat Anka decreased with increasing length of sprout in fermented dehulled buckwheat malt, probably due to consumption of nutrients, such as polysaccharides as carbon sources, during sprouting for growth of M. ruber.

Effect of Freezing on Hydration Rate and Sprouting Characteristics of Soybeans (대두의 수화속도와 콩나물의 발아특성에 대한 동결처리 효과)

  • Effect of freezing on the hydration rate and growth characteristics of bean sprout of two domestic cultivars of soybean, Eunha beans and Taegwang beans, was investigated. Freezing of the soybeans at $-80^{\circ}C$ affected characteristics of the hydration and growth of soybean sprout of the beans. Hydration rate of the frozen beans depended on the hydration temperature. It decreased up to 50% at lower than $60^{\circ}C$ of hydration temperature, but it did not decrease at higher than $60^{\circ}C$. Activation energies of frozen Eunha and Taegwang beans were 39.79 and 39.25 kJ/mol, respectively. Kinetic compensation effect between activation energy values and the pre-exponential factor for the hydration of soybeans with or without freezing was observed. Germination rate and thickness of the bean sprout increased by freezing, however, yield and weight were not affected by freezing.

Studies on the Optimal Conditions for the Storage of Fresh Garlic Bulbs (생체(生體)마늘의 적정(適定) 저장조건(貯藏條件) 설정연구(設定硏究))

  • To investigate the optimal conditions for the storage of fresh garlic bulbs, garlic was stored at the different temperature, relative humidity and moisture content. From the standpoint of weight loss and sprouting ratio of garlic bulbs, the optimal temperature for storage was $-4^{\circ}C$. No significant changes in quality of garlic bulbs occurred even after 10 months of storage, when preserving the completely predried sample at $-4^{\circ}C$. However, freezing injury was observed in sample with incomplete drying or without predrying. From these results, The optimal condition for long term storage of fresh garlic was concluded to preserve at $-4^{\circ}C$ after complete predrying.

Effects of Sprout Length, Amounts and Times of Nitrogen Application, Transplanting Rate and Depth of Wheat Straw Mulch on the Fall Crop Production of Potato (정식시 아장, 질소의 시용량과 시용시기, 재식거리 및 피복정도가 감자추작에 미치는 영향)

  • In order to find out the effect of five kinds of cultural treatments in fall crop production of potatoes the experiments were carried out with Simabara cultivar at Chonan in 1976. Sprouting of dormant seed pieces were induced by soaking in 2ppm GA and 250ppm Ethrel mixed solution for 60 minutes. Seed pieces with longer sprouts at transplanting, and increasing of urea(nitrogen) application from 10 to 30 Kg per 10a resulted better growth, yields, and quality. Application of total nitrogen at one time on 24th July(transplanting date) resulted lower emergence, yields, growth, and quality as compared with divided application on 24th July and 15th August and 5th September. Higher transplanting than 60$\times$20cm or 70$\times$17cm resulted slender growth and lower yields. Deeper mulch with wheat straw (from 4 to 12cm) induced lower maximal earth temperature in summer and higher minimal earth temperature in autumn.
