Comparison of Antioxidant Activities of Radish Bud (Raphanus sativus L.) According to Extraction Solvents and Sprouting Period (추출용매 및 발아시기에 따른 무순 추출물의 항산화 활성 비교)

  • The purpose of this study is to investigate the total polyphenol and antioxidant activities of radish buds (Raphanus sativus L.) based on sprouting periods and extraction solvents in order to present basic data that are needed for using the radish buds as functional food material. The antioxidant activities were assessed by using various antioxidant models (DPPH, TBARS, Rancimat method, POV). The total polyphenol contents according to the extraction solvents were 84.11 and 296.51 mg/g, and the ethanol extract on day 4 of showed the highest value as 296.51 mg/g. As for DPPH radical-scavenging activity, on the day 4 of sprouting, water extracts indicated the highest scavenging activity by 86.67%, and the acetone extracts indicated a rather low scavenging activity as 77.23%. As for TBARS measurement of the radish bud extracts on day 4 of sprouting the extract of 70% ethanol was highest (71.48%). On day 8 of sprouting the TBARS value was increased and the methanol extract was highest (78.99%). As for the oxidative induction period on day 4 of sprouting in Rancimat measurement, the methanol extract was highest (6.07 hours) on day 4 and the antioxidant index was 1.16. On day 12 of sprouting, the general oxidative induction period tended to be reduced to 5.25 to 5.91 hours. In the peroxide value measurement on day 4 of sprouting and beginning of the storage, the extracts showed no difference between 3.02 meq/kg oil and 4.12 meq/kg oil, and on the day 60 of storage, the water extract (43.83 meq/kg oil) and the methanol extract (45.42 meq/kg oil) were lowest with higher antioxidant effect. In conclusion, the radish bud extract with higher total polyphenol contents and antioxidant activities may serve as functional material for food additives, such as natural antioxidants and food preserving agents.

Studies on the Induction of Sprouting of Dormant Seed Potato in Fall Crop Production (추작감자의 최아법에 관한 연구)

  • To find out the most effective method of treatments for the induction of sprouting of dormant seed potato pieces for the fall crop production of Irish cobbler, this experiment was carried out with sprout inducing bed and field performance trial. In GA treatments, about 10 days were required to sprout and resulted uniform and thin 3-4 sprouts per tuber piece, but sprouts were slender and rooting was not observed. In Ethrel treatments, 20-25 days were required, and sprouts were inferior than that of GA treatment in uniformity of sprouting, and percentage of rotten pieces and of healthy sprouted tuber pieces, but number of sprouts per tuber pieces was low, being 1-2, and sprouts were short, thick, and healthy, and showed good rooting. In GA and Ethrel mixture treatments, 1-2 more days were required to sprout than GA treatments, but sprouts were relatively healthy, and other sprouting pattern were like that of GA treatments. In Ethylene chlorohydrin and 6-Benzyl-adenine treatments, sprouting was like that of Ethrel treatments, but much more days were required than Ethrel treatments and tendency of severe rotting was observed. Optimum treating methods of promising chemicals found to be 1-2 and 2-5 ppm GA solution, 500 and 1000-2000 ppm Ethrel solution, and 1-2+250-500 and 5+250-500 ppm GA and Ethrel mixture solution for 60 min. treatment of tuber piece and whole-tuber, respectively. Induction of sprouting in dry and hot time resulted severer rotting of tuber pieces during the induction of sprouting and with the advancement of dormancy, being delayed in date of treatment, tendency of promotion of sprouting and rooting was observed. When sprouted tuber piece was transplanted at the same date, yields were in order of Ethrel, GA and Ethrel mixture, and GA treatment, indicating the correlation between yield and healthiness of sprout and rooting status of sprouted tuber piece. In all treatments, earlier transplanting resulted higher yields.

Effect of Storage Temperature and Humidity on the Quality Stability of Potatoes (감자의 품질안정성에 대한 저장 온.습도의 영향)

  • Potatoes, Irish cobbler, were subject to quality evaluation from the physiological and physicochemical points of view during storage under different temperatures and relative humidities for 11 months. Quality criteria were sprouting, rotting, weight changes, moisture, total sugar, reducing sugar and vitamin C. Low temperature condition(LT, 2∼4$^{\circ}C$, 80% RH) was enough to preserve Potatoes for 11 months, but it was indicated to be limitations that the increase in reducig sugar as well as sprouting by about 3.3 to 6.7% occurred from the 3rd month of storage. Meanwhile, pit temperature(PT, 3∼15$^{\circ}C$, 75∼85% RH), room temperature(RT, 7∼17$^{\circ}C$, 80∼95% RH) and ambient temperature(AT, 2∼25$^{\circ}C$, 62∼72 RH) were shown infeasible conditions for the storage of potatoes mainly due to the increase In sprouting-rate and subsequent quality-loss after 3 months of storage. Based on the results, it proposed that more economical conditions, ranging 10 to 15$^{\circ}C$ and about 80% RH following sprout inhibition treatment should be used for the long-term storage of potatoes.

LDB2 regulates the expression of DLL4 through the formation of oligomeric complexes in endothelial cells

  • Delta-like ligand 4 (DLL4) expression in endothelial cells is intimately associated with angiogenic sprouting and vascular remodeling, but the precise mechanism of transcriptional regulation of DLL4 remains incompletely understood. Here, we showed that LIM-domain binding protein 2 (LDB2) plays an important role in regulating basal DLL4 and VEGF-induced DLL4 expression. Knockdown of LDB2 using siRNA enhanced endothelial sprouting and tubular network formation in vitro. Injection of ldb2-morpholino resulted in defective development of intersegmental vessels in zebrafish. Reduction or over-expression of LDB2 in endothelial cells decreased or increased DLL4 expression. LDB2 regulated DLL4 promoter activity by binding to its promoter region and the same promoter region was occupied and regulated by the LMO2/TAL1/GATA2 complex. Interestingly, LDB2 also mediated VEGF-induced DLL4 expression in endothelial cells. The regulation of DLL4 by the LDB2 complex provides a novel mechanism of DLL4 transcriptional control that may be exploited to develop therapeutics for aberrant vascular remodeling.

Changes in Physiologcal Properties of Garlic (Allium sativum L.) by Storage and Marketing Condition after Storage (저장 및 저장후 유통조건에 따른 마늘(Allium sativum L.)의 생리적 특성 변화)

  • To find out suitable storage and marketing condition, northern type 'seosan' garlic was stored for 3 months at controlled atmosphere(CA), modified atmosphere(MA), low(0~1$^{\circ}C$) and room temperature(20$\pm$5$^{\circ}C$), and then marketed at low(2~3$^{\circ}C$) and room temperature (20$\pm$5$^{\circ}C$)after storage. The physiological properties of garlic were investigated among treatments. The garlic respiration increased after 2 months at room temperature storage, which seem to be dormancy break time. During storage of garlic by CA, MA condition, sprouting and weight loss were prevented effectively by suppress of respiration, and hardness was maintained higher then those of the others. Decay rate occurred 8.1% at room temperature storage, 2% at MA storage but didn't occurred at CA and low temperature storages. When marketed at room and low temperature after storage, increment of respiration and sprouting were delayed, and changes of weight and hardness were low in garlics stored at CA and MA. Decay rate occurred high in garlics stored at MA and room temperature.

Effects of Storage Humidity on the Sprout Growth of Mulberry Cut Twigs (뽕나무 절지의 저장 습도가 새싹의 생장에 미치는 영향)

  • This study was conducted to develop the industrial producing technology of sprout vegetables using the cut twigs of woody plants. We have cut the twigs of wild mulberry tree on 13 April,2006 to examine the sprouting rate as affected by the storage conditions on relative humidity of $40{\sim}50%$ and $80{\sim}90%$ under $4{\sim}6^{\circ}C$ temperature. And we have also investigated the sprouting growth following those storage period of twigs cut with $1{\sim}3$ buds. The longer storage periods, the less survival rate of wild mulberry twigs cut. The sprouting rate of cut twigs gathered on the day which had not stored at low temperature was 97.4%. And The sprouting rate at the relative humidity of $40{\sim}50%$ after storing at low temperature for 122 days were 34.2%, and that of at $80{\sim}90%$ relative humidity was 85.7%. The longer storage periods, the more the number, length and width of sprouts. Therefore, the twigs of a wild mulberry trees cut with $1{\sim}3$ buds were able to produce the sprout vegetables under the condition of temperature $4{\sim}6^{\circ}C$ and relative humidity $80{\sim}90%$.

Effect of Depth of Tuber Burial, Soil Temperature, and Soil Moisture on Tuber Sprouting of Eleocharis Kuroguwai Ohwi (올방개 괴경(塊莖)의 맹아(萌芽)에 미치는 매몰심(埋沒深), 토양온도(土壤溫度) 및 수분조건(水分條件)의 영향(影響))

  • A study was conducted to determine the effect of depth of burial, soil temperature and/or moisture on tuber sprouting of E, kuroguwai. Tubers were evenly distributed in the upper 30cm of soil. Tuber weight increased as depth of tuber formed increased. No dormancy in newly formed tubers was found, whereas mature tubers were dormant. When new tubers were subjected both to continuous $5^{\circ}C$ and to gradual decreasing temperatures regimes ranged from 20 to $1^{\circ}C$ for 30 days, the tubers remained non-dormant. Viability of tubers was reduced when buried at 5cm depth in dry and moist soil conditions, but no reduction was obtained when buried at 25cm depth, regardless of soil moisture conditions employed. Percent sprouting of tubers buried at 25cm depth increased with increasing duration of burial in three soil moisture conditions studied, wherease in dry and moist conditions percent sprouting of tubers buried at 25cm depth increased by 60-day burial and thereafter decreased. In submerged condition, tuber sprouting was greater when buried at 5cm depth than when buried at 25cm depth, and increased as duration of burial increased at the both depths.

Effects of Bulb Size, Type of Media, Depth of Planting, and Nutrient Compositions on the Growth of Tissue Cultured Garlic Microbulbs in Hydroponic Culture (조직배양 마늘의 양액재배시 종구크기, 상토, 재식깊이 및 양액조성이 맹아 및 생육에 미치는 영향)

  • This study was carried out to investigate the effects of bulb size, type of media, depth of planting, and nutrient compositions on sprouting and growth of tissue cultured garlic microbulbs in hydroponic culture. Early and increased sprouting were observed when the microbulbs were transplanted into soil planted in shallow (1 cm in depth), while bulb size and fresh weight of the whole plant increased in deep planting (3 cm in depth). Bulb size have greatly influenced on not only sprouting rate but also plant growth after planting. Large bulbs resulted in high growth rate such as number of leaves, stem width, plant height, and increase in bulb size after planting. It was shown that Oriental nutrient solution (N=0.17, P=0.45, K=1.29, Ca=2.44, and Mg=0.98 me/L) was more effective in sprouting and further growth of microbulbs as compared to Yamazaki solution (N=0.27, P=0.16, K=1.50, Ca=1.36 and Mg=0.78 me/L). Microbulbs grown in mixture of leaf mould + bark + sand or Baroko showed the most vigrous growth.

Freezing Hardiness of Several Pear Cultivars According to Degree and Duration of Low Temperatures (저온 처리 온도 및 지속 시간에 따른 배의 품종별 내동성 비교)

  • Freezing hardiness of winter bud and branch of several pear (Pyrus pyrifolia) cultivars according to degree and duration of low temperatures was investigated by sprouting, electrolyte leaching rate and triphenyltetrazolium chloride (TTC). Sprouting rate as infected by degree and duration of low temperature were different between cultivars. The lower temperature, the longer duration, sprouting rate was decreased. Electrolyte leaching rate was showed above 30% at below $-30^{\circ}C$ treatment regardless of cultivars and duration. The lower temperature and the longer duration, Electrolyte leaching rate was increased. Electrolyte leaching rates of Manpungbae, Niitaka and Chuwhangbae at $-30^{\circ}C$ for 9 hours treatment which were observed high sprouting rate, were lower than those of other varieties. Absorbance rates by TTC test at $-21^{\circ}C$ treatment were 66.0 to 96.5% for 6 hours, 49.4 to 91.9% for 9 hours, and 37.3 to 89.4% for 12 hours. Freezing hardiness of pear cultivars at ecodormancy was different according to degree and duration of low temperature treatments.

Development of the Automatic Watering System for the Soybean Sprouting Equipment by Siphon and Well Bucket (싸이펀과 두레박을 이용한 콩나물 재배기용 자동 수주 시스템 개발)

  • The new watering system for the cabinet type of soybean sprouting equipment was developed. It was made of one siphon $({\Phi}35 mm)$, one well bucket $(13{\times}10{\times}10\;cm)$ that its weight-center is changed by water level, and bottle-neck water tank (bottom: $18.5^W{\times}32^L{\times}29^H\;cm$, top: $14^W{\times}14^L{\times}25^H\;cm)$. The watering system can be automatically watered more than 750 mL/sec without electric supply. The soybean sprouting equipment is consisted of the watering system and nine sprouting buckets. In addition siphon phenomena brought about easily by narrowing same as bottle-neck shape top portion of water tank that inserted a siphon. The equipment with the watering system may be produced more than three box a day in the condition of $25{\pm}1^{\circ}C$, and $6{\sim}7$ watering times a day.
