Effect of Fluorescent Light Treatment during Imbibition and Culture on Growth of Soybean Sprout

  • The lateral root formation in soybean sprout culture declines its quality. This study was done to measure the effect of fluorescent light treatment during 24 hour imbibition and 6-day culture on seed germination and growth of soybean sprout. After 6 day culture, the sprouts were sorted as normal (>4cm), abnormal (<4cm) and non-germination by their hypocotyl lengths, and lateral roots, fresh and dry weights were measured. Lateral roots were less formed in the fluorescent light treatment lasted during the whole period of the imbibition than in the treatment for 50 minutes a day during the culture. The fluorescent light treatment during the imbibition mainly affected the germination and growth compared to the treatment done during the culture. Compared to the dark imbibition, the light treatment during the imbibition resulted in more normal sprouts, thicker diameters of hypocotyl and hook, and more fresh weights in cotyledon, hypocotyl, whole sprout, and economic yield. However, these results were reverse in lengths of hypocotyl and root, and fresh and dry weights of roots. It is concluded that the fluorescent lamp mainly irradiating red and blue lights can be used for the sprout production as an alternative light replacing blue and red lights treated during the imbibition because it blocked the lateral root appearance and stimulated growth of the sprout.

Physicochemical Components and Antioxidative Activity of Wheat Sprout Powder Prepared by the Enzyme and the Lactic Acid Bacteria (효소와 유산균 전처리 밀싹분말의 이화학적 성분 및 항산화 활성)

  • The purpose of this study was to evaluate the physicochemical components and antioxidant activities of wheat sprout powder prepared by the enzymatic hydrolysis and lactic acid bacteria treatment. The four kinds of pre-treatment were: no treatment (WP), treated with enzyme (WPE), treated with lactic acid bacteria (WPL), and treated with enzyme and lactic acid bacteria (WPEL) were applied to the wheat sprout powder. The WPEL had higher total free amino acid and essential amino acid content than the other samples. As for the volatile aroma of the wheat sprout powder, 29 types of compounds were identified in the WP and WPL, 28 types in the WPE, and 27 types in the WPEL, respectively. The total polyphenols and flavonoids contents, in the wheat sprout powder was enhanced with the enzyme and the lactic acid bacteria pre-treatment. The WPEL had highest DPPH radical scavenging activities. The overall acceptability was the highest at 6.24 points in the WPEL. Based on these observations, it was confirmed that the enzyme and lactic acid bacteria pre-treatment could improve the antioxidant activities and active component of the wheat sprout powder.

Elucidation of Cause of Cotyledon Black-Decay of Soybean Sprout by Bean Bug, Riptortus clavatus

  • Cotyledon Black Decay (CBD) on soybean sprout mimics the black spot due to microbial infection. CBD, not visible or predictable at seedlot state, for some reason, shows up exclusively on cotyledon of soybean sprout during sprouting process. Such an incidence rate fluctuated from 0.8 to 19.5% over three years from 2004. We suspected some pod-infecting anthracnose fungi and/or pod-blight pathogen, or pod-sucking bean bug, one of the major pests of soybean, might have involved, of which we ruled out fungal pathogen because it was preventable through heat treatment, a proven method for seedlot disinfestation. The healthy seeds artificially fed by bean bug for one to seven days were sprouted, and 6 to 41% of the soybean sprout revealed the CBD mimic to those occurred in soybean sprout from previous commercial seedlot screening experiments. This finding is the first report to confirm that bean bug damage to pod at $R_8$ stage is directly responsible for the CBD, which did not concur with any other deleterious effects on sprouting such as reduction in hypocotyls elongation and rooting except unsightly sprout quality. However, earlier feeding either at green pod or greenish yellow pod stage ($R_6$ -early $R_7$ stage) resulted in rather severe damages, which strikingly reduced hypocotyls growth to about one forth to about two third, as well as the reduction in rates of seed germination.

Quality Characteristics of String Cheese Prepared with Barley Sprouts (새싹보리를 첨가한 스트링치즈의 품질 특성)

  • The objective of this study was to evaluate the quality characteristics of string cheese with different concentrations (0, 1, and 2%) of barley sprout (Hordeum vulgare L.) powder. The pH of barley sprout string cheese (6.26) was significantly lower compared to that of the control (6.46). Chromaticity tended to decrease with increasing amounts of barley sprout powder in terms of L* (lightness) and a* (redness), whereas b* (yellowness) tended to increase. In the texture profile analysis, string cheese prepared with barley sprout powder showed higher values of hardness, gumminess, and chewiness but lower value of cohesiveness. The string cheese prepared with barley sprout powder also showed higher values for total amino acids and stretchability. Total polyphenol contents of string cheese showed the highest value when barley sprout powder was 2%. Therefore, addition of barley sprout powder to string cheese can be valuable for improving quality of string cheese.

Study on the whitening effect and deterrent effect on gene expression of MMP-1 in wheat sprout extracts (밀싹 추출물이 MMP-1의 유전자 발현 억제효과 및 미백효과에 관한 연구)

  • This study set its purpose on to specify whether it is possible to use wheat sprout extracts for source of cosmetics, and its biological activity. Cellular experiment was conducted to apprehend toxicity of wheat sprout extracts and through study on the whitening and aging activity of skin cell. As a result, it is appeared that wheat sprout extracts have weak toxicity for HDF, B16F10 cells. Also, it is appeared that wheat sprout extracts have weak deterrent effect on melanin biosynthesis, but since the extracts impede expression of MMP-1, which is induced UV ray, the extracts hold down the aging effect by UV rays. Hence the extracts are effective on preventing wrinkles caused by UV ray and aging. Therefore, this study is expected to be utilized usefully, if the wheat sprout extracts are used as source of functional cosmetics from the perspective of preventing skin-aging in the future.

Preparation and Quality Characteristics of Soymilk added with Buckwheat Sprout (메밀 싹 첨가 두유의 제조와 품질 특성)

  • This study investigated the physicochemical properties and antioxidative activity of soymilk added with buckwheat sprout (SBS), and sensory properties of SBS were analyzed with control soymilk (CS). Moisture content decreased while protein, ash, and lipid contents increased according to content of buckwheat sprout. Solids content and viscosity tended to increase with addition of buckwheat sprout from 0% (CS) to 2.5% (2.5SBS), 3.5% (3.5SBS), and 4.5% (4.5SBS). SBS was found the significant pH drop from 7.08 to 6.43, 6.34, and 6.21. Suspension stability of soymilk slightly decreased with addition of buckwheat sprout. Hunter's color value L of SBS decreased while a and b values increased in comparison with CS. Rutin content was measured 23.78 mg/100 g (2.5SBS), 39.68 mg/100 g (3.5SBS), and 44.80mg/100 g (4.5SBS). Vitamin C content in SBS was higher than CS. Daidzin content increased as buckwheat sprout was added. Total phenolic content increased from 100.95 mg/100 g (CS) up to 315.71 mg/100 g (4.5SBS). For free radical scavenging activity, SBS was significantly higher than CS. In conclusion, SBS can be used as a functional food with higher amounts of rutin, vitamin C, total phenolics, and daidzin. 2.5SBS and 3.5SBS also showed better overall quality characteristics.

Resveratrol and Aspartic acid Contents and Antiadipogenic Effect of Peanut and Peanut Sprout Extracts (땅콩과 땅콩새싹 추출물의 resveratrol과 aspartic acid 함량분석 및 지방세포분화 억제효능)

  • Interest in peanuts driven by their various biological activities and abundant nutrition engenders research interest in peanut sprouts for determining their biological activities and nutrition. Several research groups have recently studied the peanut sprout ingredients, but these studies focused on the peanut sprout extracted by methanol, an inedible extraction solvent. Thus, the present study provides the contents of two biologically active compounds, resveratrol and aspartic acid, in peanut and peanut sprout extracted by hot water and fermented ethanol, two edible extraction solvents, using UPLC-MS/MS technique. The UPLC-MS/MS results show that the peanut sprout extracted by fermented ethanol has the highest resveratrol and aspartic acid contents. This extract also exhibited the highest inhibitory effect on adipocyte differentiation, which the resveratrol may induce. Consequently, peanut sprout extracted by fermented ethanol might be helpful for the development of functional food plant materials.

Anti-oxidative effects of broccoli (Brassica oleracea var. italica) sprout extract in RAW 264.7 cell and cisplatin-induced testicular damage

  • Background: Brassica oleracea var. italica (broccoli), a rich source of antioxidants, can prevent various diseases and improve human health. In this study, we investigated the antioxidative effects of broccoli sprout extract on oxidative stress induced by lipopolysaccharide and cisplatin in cell and organ tissue models. Methods: Antioxidative effect of BSE was evaluated using DPPH and ABTS in RAW 364.7 cells, and effects of BSE on testes were investigated using Cisplatin-induced testicular damage model with an in vitro organ culture system. Results: The DPPH assay showed that the antioxidant activity of the alcoholic broccoli sprout extract was higher than that of the water extract. Additionally, the expression levels of antioxidation-related genes, Nrf2, Gsr, HO-1, and catalase, were significantly increased in broccoli sprout extract-treated RAW 264.7 cells, and the extract suppressed lipopolysaccharide-induced mitochondrial dysfunction. Based on the results in the RAW 264.7 cell culture, the antioxidative effects of the extracts were investigated in a mouse testis fragment culture. The expression of Nrf2, HO-1, and Ddx4 was clearly decreased in cisplatin-treated mouse testis fragments and not in both broccoli sprout extract- and cisplatin-treated mouse testis fragments. In addition, the oxidative marker O-HdG was strongly detected in cisplatin-treated mouse testis fragments, and these signals were reduced by broccoli sprout extract treatment. Conclusions: The results of this study show that broccoli sprout extracts could serve as potential nutraceutical agents as they possess antioxidant effects in the testes.

Physiochemical property of edible tissues (sprout and root) of steam-treated reed (증자처리에 의한 갈대 식용가능 부위(어린순과 뿌리)의 이화학적 특성)

  • This study was performed for the comparison of the general components, minerals, amino acids, organic acids, free sugars, ascorbic acid, polyphenol content and DPPH free radical scavenging activity of steamed-treated reed (sprout and root) to those of raw reed. Moisture content of sprout and root of reed after steaming treatment decreased from 81.28% and 81.64% to 70.18% and 65.50%, respectively. Crude ash content was the highest in raw sprout and steam-treated root. Crude lipid content of raw sprout and root were almost similar. In addition, crude lipid content of steam-treated sprout was greater than that of root. Nitrogen free extract content of root was 2 times as high as that of sprout. Total free sugar contents of raw sprout and root increased from 1,311.39 mg% and 4,130.98 mg% to 1,157.79 mg% and 3,750.90 mg%, respectively, after steam treatment. Furthermore, the organic acid contents of sprout and root of reed after steam treatment were less than those of raw sprout and root. Calcium and potassium contents were the highest among others in both steam-treated and raw reed. Amino acid content of sprout was higher than that of root in both before and after steam treatment. Among the amino acids, serine content was the best presented in both before and after steam treated reed. Vitamin C content of steam-treated sprout and root of reed decreased from 61.74 mg% and 6.57 mg% to 4.54 mg% and 80.79 mg%, respectively. Total polyphenol content of sprout was greater than that of root in raw and steam-treated reed. DPPH free radical scavenging activity of ethanol extraction of root was greater than those of other extracts of root.

Analysis of Chemical Factors Determining Taste of Soybean Sprouts (콩나물 식미 결정 성분 요인 분석)

  • Soybean sprout is a year-round traditional vegetable that is easily produced and relatively inexpensive in Korea. In addition, the sprout is known as a good source of protein, vitamins and minerals. The quality of the soybean sprout has been mainly evaluated only by its appearance like length, width, color, and the others without considering any odor or taste attributes. We studied the chemical factors affecting taste of soy sprouts cultivated with 5 recommended soybean cultivars through evaluation of chemical constituents in relation to their sensory characteristics. Correlation coefficient among the chemical constituents and sensory characteristics of soybean sprout showed that the linolenic acid and Ca contents were positively correlated with total acceptability of soybean sprout and histidine, aspartic acid, and serine showed a negative association with beany odor of soybean sprout. Multiple regression analysis was done to formulate selection criteria for good taste of soy sprout. The estimation of step-wise regression analysis conducted by 47 chemical components for major quality-related characteristics showed that linolenic acid and mineral contents were the main components increasing the acceptability of soybean sprout.
