Objectives : This study was designed to investigate effects of Lumbricus pharmacopuncture (LP) on the lipid lowering, anti-oxidative activity and concentration of proinflammatory cytokines in rat fed high fat diet. Methods : Sprague-Dawley male rats were fed high fat diet for 8 weeks and experimental groups were divided into 4 groups as follows : Control, Lumbricus Jungwan ($CV_{12}$) pharmacopuncture (T I), Lumbricus Joksamni ($ST_{36}$) pharmacopuncture (T II), Lumbricus Jungwan ($CV_{12}$) and Joksamni ($ST_{36}$) pharmacopuncture (T III). Results : The levels of plasma FFA, TG, total cholesterol, LDL-C, TBARS, IL-$1{\beta}$, IL-6, TNF-$\alpha$ and liver total cholesterol, TG, TBARS, SOD, catalase, IL-6 in more than one LP group were significantly lower than those of Control groups. The level of IL-10 in one of LP groups was significantly higher than that of Control group. Conclusions : As the results were shown, LP treatment have considerable effects on lipid lowering, anti-oxidative activity and concentration of proinflammatory cytokines in rat fed high fat diet.
The aim of this study was to investigate the effect of n-3 polyunsaturated fatty acids (PUFAs) on eruptive movement during tooth development. Sprague-Dawley (SD) rat pups were randomly divided into two groups; control group and experimental group. The experimental group was administered daily with n-3 PUFA by intraperitoneal (IP) injection. After 10 days postpartum, rat pups were sacrificed to evaluate the effect of n-3 PUFA on eruptive tooth movement. Histological analyses were by hematoxylin-eosin (H&E) staining. Tartrate-resistant acid phosphatase (TRAP) assay was performed to compare the osteoclast distribution in the bone matrix above the developing molar teeth. Incisor teeth eruptions were noticeably observed in IP group, as compared to control group. Rat pups in IP group showed faster tooth eruption on day 8 after birth. Through histological analyses, IP group showed thinner bone matrix and more osteoclasts above the $1^{st}$ molar teeth, as compared to control group. TRAP assay showed significantly stronger stained pattern that the osteoclast above the $1^{st}$ molar teeth in IP group, as compared to control group. The results suggested that n-3 PUFA could affect osteoclastic activity involved in bony remodeling during eruptive tooth movement.
The purpose of this study was to investigate the irradiation effects on the secretory granules of the rat submandibular gland acinar cell. For this study, Sprague-Dawley strain rats were irradiated to their neck region with the dose of 5Gy and l0Gy by 6MV X-radiation, and sacrificed on the experimental periods after irradiation. The authors observed the ultrastructural changes of the secretory granules of the rat submandibular gland acinar cell under a trasmission electron microscope. The results were as follows: In the transmission electron microscopic examination, secretory granules were blurred in limiting membrane on the 3hours groups after irradiation. And they showed decrease in number, irregularities in shape and distributional pattern, and inhomogeneous internal electron density on the Iday and 3days groups. After then, these changes were recovered in shape and distributional pattern on the 14days groups, and changes of internal electron density and limiting membrane were recovered on the 28days groups after irradiation. Among the intracellular organelles, rough endoplasmic reticulum was scattered, but golgi complex was not changed. And such pathologic changes were earlier and more prominent in 10Gy irradiated groups than in 5Gy irradiated groups.
The present study investigated the protective effects of green tea against acute ethanol-induced lipid peroxidation and the change of antioxidative enzyme activities in various regions of rat brain : cortex, cerebellum, striatum and hippocampus. The following parameters were examined : malondialdehyde(MDA) levels and activities of superoxide dismutase(SOD), catalase and glutathione peroxidase(GSH-Px). Male Sprague-Dawley rats were given the experimental containing 1% green tea powder or control diet for 4 weeks, and at the end of feeding diet group received acute ethanol(5g/kg body weight) or equicaloric sucrose solution intragastrically. Green tea powder significantly decreased MDA levels in the striatum compared to control-non alcohol treated group to 1% green tea-non alcohol treated group without altering the antioxidative enzyme activities. Green tea resulted in a significant increase in GSH-Px activities in the hippocampus compared to either control-non alcohol treated group(0.043units/mg protein) or 1% green tea-non alcohol treated group(0.071units/mg protein). In conclusion, these results suggest that moderate consumption of green tea leaves can exert protective effects against ethanol-induced oxidative stress in brain regions, by reducing MDA concentrations in the striatum and enhancing GSH-Px activities in the hippocampus. Changes in the expression level of splenocyte $IFN-{\gamma}$mRNA of Sprague-Dawley (SD) rats infected with Paragonimus westermani were analyzed by competitive reverse transcription-polymerase chain reaction (RT-PCR) followed by southern blot. The template RNA was extracted from the splenocytes of rats infected with 20 metacercariae of P. westermani. The products of competitive RT-PCR were subjected to southern blot and enhanced chemiluminescence (ECL), and analyzed with a densitometer. In comparison with that of uninfected control rat splenocytes (value of 1), the levels of mRNA expression of $IFN-{\gamma}$had changed to 0.747 at 1 week post infection (PI), 0.00175 at 2 week PI, 0.0217 at 3 week PI, 0.194 at 4 week PI and then to 0.537 at 5 week PI. The level at 7 week PI had returned to 1.25, comparable with that of uninfected rats. These results show that, when infected with p. westermani, the levels of $IFN-{\gamma}$ mRNA of SD rat splenocytes were remarkably reduced by more than 500 times at 2 week PI and restored to normal level at 7 week PI.
This experiment was carried out to investigate effects of DMSO or ethylene glycol as a cryopotectant and of freezing methods on survival rate of forozen-thawed rat 2-cell embryos by morphological observation. 2-cell embryos were recovered from oviducts of Sprague Dawley females mated with males of same strain on day 2 of pregnancy after inducing superovulation by intrapertioneal injection of PMSG and HCG. In slow freezing and thawing groups, embryos were frozen to -79$^{\circ}C$ or -196$^{\circ}C$ in a glass test tube containing 0.2ml PBI with 1.5M DMSO or 1.2M ethylene glycol at a rate of 0.3-1.0C/min. and thawed slowly. When samples were frozen to -79$^{\circ}C$, higher survival rate was obtained in the medium containing DMSO (43.9%) than ethylene glycol (41%). And similar result was obtained (32.5% in DMSO vs. 31.4% in ethylene glycol) when samples were frozen. In rapid freezing and thawing groups, embryos were frozen to -79 or -196$^{\circ}C$ in a glass test tube containing 0.2ml of PBI with 1.5M DMSO or 1.2M ethylene glycol by rapid cooling, and thawed rapidly. When samples were frozen to -79$^{\circ}C$, 1.5M DMSO (13.2%) was more effective than 1.2M ethylene glycol (6.1%). When the storage temperature was -196$^{\circ}C$, survival rates were 9.8% in 1.5M and 5.4% in 1.2M ethylene glycol.
The purpose of this study were to 1) determine the analgesic effect of 632.8 nm of helium-neon (He-Ne) laser stimulation on acupuncture point in rat and 2) determine the reversal of analgesic effect by naloxone injection. Eighteen Sprague-Dawley rats were devided into three groups : control (n=6) : laser (n=6), laser stimulation at $3.58\;J/cm^2$ ; and naloxone (n=6), 1 mg/kg of naloxone chloride inject into peritoneum before laser stimulation at $3.63J/cm^2$. Tail-flick latency were measured pretreat and posttreat with hot plate $(55^{\circ}C)$. Data were analyzed using one-way ANOVA and paired t-teat for tail-flick latency. No significant change was noted in the tail-flick latency in either control or naloxone groups. But significant increased in tail-flick latency in taller group. The results suggest that He-Ne laser induced analgesic effect, and endogenous opioids may be involved in He-Ne laser induced analgesia.
The aim of present study was to investigate the possible influence and related mechanism of resveratrol on U-46619 (high concentration)-induced vasoconstriction. Agonist-induced vascular smooth muscle contractions involve the activation of thick or thin filament pathway. However, there are no reports addressing the question whether this pathway is involved in resveratrol-induced relaxation in rat aortae contracted with high U-46619. We hypothesized that MEK or Rho-kinase inhibition plays a role in vascular relaxation evoked by resveratrol in rat aortae. Endothelium-denuded arterial rings from male Sprague-Dawley rats were used and isometric contractions were recorded using a computerized data acquisition system. Resveratrol fully inhibited U-46619 in low concentration-induced contraction regardless of endothelial function. However, resveratrol partially decreased U-46619 in high concentration-induced contraction regardless of endothelial function. Interestingly, only in U-46619 (high concentration)-induced contraction, no significant decrease was observed in phospho-ERK1/2 levels and slight decrease in phospho-MYPT1 levels suggesting that additional pathways different from them or endothelial nitric oxide synthesis might be involved in the vasorelaxation. In conclusion, in high U-46619-contracted rat aortae, resveratrol showed relaxation response regardless of endothelial function significantly but slightly decreasing MYPT1 phosphorylation rather than ERK1/2 phosphorylation.
It is important to study using experimental animals for research about stroke. Magnetic Resonance Imaging(MRI) is avaluable diagnosticmethods for stroke diagnosis. The purpose of this research is to know the Magnetic Resonance Imaging(MRI) and histopathological characteristics findings after induction of photothrombotic cerebral infarction in rat brain. Male Sprague-Dawley rats were anesthetized, Rose Bengal dye(20 mg/kg) was intravenously injected. The right sensonrimotor cortex of rat brain was exposed to cold light of 7 mm diameter at a position of 1 mm anterior and 3.5 mm lateral to bregma for 20 min. The post-infarction effects were monitored by T1 weighted and T2 weighted images of brain MRI. Histopathological changes were observed after Hematoxylin & Eosin staining. The lesion appeared clearly high signal intensity area on T2 weighted images(the major axis $7.04{\pm}0.11$ mm, the minor axis $3.08{\pm}0.04$ mm) and also H&E staining was same result. In conclusion, MRI was avaluable diagnostic methods for diagnosis and serial changes of stroke.
The purpose of this study was to find an effect of honey on the lipid metabolism of Sprague Dawley rats. All experimental rats were fed ad libitum, for seven weeks, 68% saccharide diet and 10% or 20% honey from acacia, sumac and miscelllaneous flower honey, respectively, and sucrose. The food efficiency ratio of rat taken diet with honey and high fructose of control group was increased in comparison with the control group. The concentration of cholesterol in serum of rats take총 diet with and high fructose of control group was more increased in comparison with the control and normal group. The concentration of H DL-cholesterol in serum of rats taken sumac honey was increased 57.0% in comparison with the control group, but the concentration of VLDL, LDL-cholesterol in serum of rats taken diet 10PA sumac honey was decreased 48.36% in comparison with the control group. The concentration of phospholipid in serum of rats taken diet with 20% acacia or 10% miscellaneous honey was increased 24.7, 16.25%, respectively, in comparison with the control group. The concentration of free fatty acid in serum of rats taken Inlet with sumac or miscellaneous honey and high fructose was increased in comparison with the comparison with the control group. The concentration supplemented diet with acasia honey was increase in comparison with the control group. The concentration of triglyceride in serum of rat was increased by feeding of honey. The concentration of triglyceride in liver was increased, but the level of phospholipid was decreased by feeding of honey.
