In vitro를 통한 혼합시료의 물 추출물과 에탄올 추출물을 시료로 사용하였고, 마우스 비장세포 증식능 및 활성 복강 대식세포에서 분비하는 사이토카인(IL-1 ${\beta}$, IL-6, TNF-${\alpha}$)의 분비능을 측정하였다. 그 결과 혼합시료의 물 추출물과 에탄올 추출물 모두 5${\sim}$250 ${\mu}$g/ml의 농도로 첨가했을 때 세포증식을 촉진하는 효과가 있는 것으로 나타났고, 물 추출물인 경우 고농도인 1,000 ${\mu}$g/ml에서, 에탄올 추출물인 경우는 500${\sim}$1,000 ${\mu}$g/ml 이상의 고농도 첨가시 세포증식이 억제되는 효과를 보였다. 복강 대식세포의 사이토카인 분비량을 측정한 결과, 혼합 물 추출물과 에탄올 추출물 모두에서 대조군보다 높은 분비량을 보였다. IL-1${\beta}$ 생성량 검색 결과, 물 추출물 투여군 10 ${\mu}$g/ml 농도 첨가에서 유의적(p<0.05)으로 높은 분비능을 보여주었고, 에탄올 추출물을 10 ${\mu}$g/ml와 100 ${\mu}$g/ml 첨가시에도 대조군보다 높은 분비량을 보였다. IL-6의 결과에서도, 물 추출물 10 ${\mu}$g/ml을 첨가했을 때 유의적(p<0.05)으로 높은 분비량을 보였고, TNF-${\alpha}$의 경우, 물 추출물의 10 ${\mu}$g/ml와 100 ${\mu}$g/ml의 농도에서 대조군에 비해 높은 TNF-${\alpha}$ 분비능을 보였다. 이상의 결과에 따르면, 혼합 물 추출물이 비장세포 증식능과 복강 대식세포에 의한 사이토카인 분비능을 상승시킴으로서 면역 기관의 주요 기능을 증진시키는 것으로 사료된다.
Streptozotocin으로 당뇨를 유발시킨 생쥐에서 fructan(치커리이뉼린과 치커리이뉼린올리고당, 프럭토올리고당)이 혈당, 인슐린과 소장 및 신장의 이당류분해효소 활성 및 면역능에 미 치는 영향을 조사한 결과는 다음과 같이 요약할 수 있다. 혈당은 당뇨군들이 정상군보다 유의하게 높았으며, 치커리이뉼린군의 혈당이 당뇨대조군에 비하여 유의하게 낮았다. Fructan은 혈장 인슐린 농도에는 영향을 미치지 않았다. 소장점막의 maltase와 sucrase활성은 당뇨군들이 정상군에 비하여 유의적으로 높았으나, 당뇨군 중에서 치커리이뉼린군이 가장 낮은 경향을 보였다. 신장 maltase 활성은 정상군과 당뇨군이 유사한 수준이었으나, 신장 sucrase와 lactase 활성은 정상군에 비하여 당뇨군에서 유의하게 높은 수준을 보였으며, 치커리이뉼린군이 당뇨대조군에 비하여 유의하게 낮은 수준을 보였다. LPS에 의한 비장세포증식에서는 당뇨대조군에 비하여 치커리이뉼린올리고당군이 유의 적으로 증가하여 정상군과 비슷한 수준을 보였다. ConA에의한 비장세포증식은 군간 유의적인 차이를 보이지 않았으나 LPS에 의한 증식 결과와 비슷한 경향을 보였다. 비장세포로부터의 IL-2의 분비는 당뇨군들이 정상군에 비하여 낮았으나, 당뇨군간에는 유의한 차이가 없었다. 결론적으로 당뇨 생쥐에서 프럭토올리고당, 치커리이뉼린올리고당, 치커리이뉼린을 비교한 결과, 혈당 강하에 미치는 효과가 치커리이뉼린에서 유의하게 나타났으며, 비장세포 증식능은 정상군에 비하여 당뇨군에서 감소하는 경향을 보였으나, 치커리이뉼린올리고당 섭취군이 당뇨대조군에 비해 비장세포 증식능이 유의하게 증가하여 면역능이 개선되는 효과를 보였다.
These experiments were conducted to investigate the effects of glycyrrhizin(GL) and glycyrrhetinic acid(GA) on histamine synthesis, lymphocyte blastogenesis in C57BL/6J mice splenocytes, IL-1 production, $Ca^{2+}$ uptake by macrophage-like P388D$_{1}$ cells and plaque forming cell assay against SRBC. Histamine contents, lymphocyte blastogenesis, IL-1 activity, $Ca^{2+}$ uptake and plaque forming cell were determined by enzyme isotope method, [sup 3/H]-thymidine incorporation, C3H/HeJ mouse thymocytes proliferation, the addition of 5 $\mu$Ci/ml $^{45}$Ca$^{2+}$ to P388D$_{1}$, cell suspension and assay to sheep red blood cell, respectively. Cytotoxicity, which was expressed as 50% mortality, was occurred by the addition of GL(10$^{-3}$M) and GA(10$^{-4}$M). Histamine production in mouse spleen cell culture was significantly increased by the addition of 0.25 $\mu\textrm{g}$/ml of Con A, after 48 hour incubation. Con A dependent T-lymphocyte proliferation was also enhanced by the addition of 0.25 .mu.g/ml of Con A. The effects of GL on histamine contents and T-lymphocyte proliferation were significantly decreased at high dose (10$^{-5}$M), while IL-1 activity was remarkably suppressed by 10$^{-8}$~10$^{-4}$M of GL. $Ca^{2+}$ uptake was not changed, but antibody production was increased by GL(10 mg/kg). GA inhibited histamine contents at 10$^{-9}$~10$^{-7}$ and depressed Con A (0.25 $\mu\textrm{g}$/ml) dependent T-lymphocyte proliferation at 10$^{-7}$~10$^{-5}$M of GA, but increased suboptimal dose (Con A 0.1 $\mu\textrm{g}$/ml) at 10$^{-9}$~10$^{-7}$M of GA. IL-1 activity was suppressed by 10$^{-8}$~10$^{-4}$M of GA and $Ca^{2+}$ uptake was enhanced by 10$^{-9}$~10$^{-6}$ of GA, but antibody production was not changed by GA. From the above results, it is suggested that GL and GA have immuno-regulatory action. GL decreased cell-mediated immune response, and increased humoral immune response at high dose. On the other hand, low dose of GA enhanced cell-mediated immune response, while high doses of GA decreased humoral immune reaction.
Objective : Sinapis alba L. (SA) is a korean traditional herbal medicine that is usually used to prevent or treat inflammatory diseases, such as respiratory infection and rheumatoid arthritis. However, the effects of SA supplementation in vitro on serum antibody levels, splenocyte and peritoneal macrophage immune responses have not yet been determined. In this study, we examined the effect of SA on the production of Th1/Th2 cytokines. Methods : Splenocytes were isolated from naive C57BL/6 mice. Cells were enriched for CD4+ cell populations by first staining the cells with anti-CD4 (BD PharMingen, Calif, USA). CD4+ T cells were selected on a (CS) column, and the flow-through was collected as CD4+ T cells. Isolated cells were activated by overnight incubation on 24-well plates coated with $1{\mu}g/mL$ anti-CD3, $1{\mu}g/mL$ anti-CD28 and with SA ($100{\mu}g/mL$). Primary macrophages were collected from the peritoneal cavities of mice (8-week-old female C57BL/6). The peritoneal macrophages were washed and plated with RPMI-1640 overnight for the experiments. After 48-hours cultures, samples were centrifuged at 2000 rpm for 10 minutes, and the supernatants were stored at $-80^{\circ}C$. Mouse IL-4, IFN-$\gamma$ and TNF-$\alpha$ were quantified using ELISA kits (BioSource International, Camarillo, Calif, USA) according to the manufacturer's protocols. Results : SA at 100ug/ml decreased the generation of Th1 cytokine (IFN-$\gamma$) by 0.5-fold. However, SA has no effect on Th2 (IL-4) production. Conclusions : These results suggest that SA may play an important role in the control of T-cell-mediated autoimmunity by down-regulation of Th1 cytokine (especially IFN-$\gamma$, TNF-$\alpha$). These data may contribute to the design of new immunomodulating treatments for a group of autoimmune diseases.

SPDJTK medicines controlled $CD4^+/IFN-\gamma$, and $CD4^+/CD25^+/foxp3^+$ revelation that an experiment that motive allergy immune reponse because an in vitro experiment stimulates T cells of a NC/Nga mouse same time by anti-CD40/rmIL-4, and interleukin-$1{\beta}$, IL-6, TNF-$\alpha$, and TGF-$\beta$ mRNA outturn that bear in T and B cells decreased remarkably by SPDJTK medicines. Intracellular staining of splenocytes anti-CD40/rmIL-4 plus rmIL-4 stimulated as described in a, assessed after 24 h, SPDJTK exerts a mainly immunosuppressive effect that acts at least partially through suppression of the transcription factor GATA3 expression in $CD4^+$ T cells. Atopic dermatitis (AD) usually develops in patients with an individual or family history of allergic diseases, and is characterized by chronic relapsing inflammation seen specially in childhood, association with IgE hyperproduction and precipitation by environmental factors. However, the exact etiology of AD has been unclear. To further explore the pathogenesis and treatment of AD, a suitable animal model is required. We found that skin lesions, which were clinically and histologically very similar to human AD, mite antigen-induced dermatitis on the face, neck, ears and dorsal skin of inbred NC/Nga mice. Result that Th1 cell and Th2 cell observe to be shifted by cytokine expression with $CD4^+/CD25^+/foxp3^+$ Treg cells induction by SPDJTK medicines could know that SPDJTK medicines can use usefully in allergy autoimmnune diease.
Objectives: Maier symptoms of allergic rhinitis are nasal obstructions, sneezing and watery rhinorrhea. When exposed to certain allergens, the IgE covered mast cells degranulate and release inflammatory mediators and cytokines which result in a local inflammatory reaction. In many recent studies, molecular biological methods have been used to investigate the role of cytokines in pathogenesis and new therapeutic targets of allergic rhinitis. This experimental study was done to reveal the effects of the Bojungikgi-tang on the allergic rhinitis. We have studied effect of mice on OVA-induced production of IL-4, IL-5, $INF-{\gamma}$ by murine splenocytes and effect of OVA-induced Total IgE and OVA-specific IgE Meterial and Metheds: 21 BALB/c rats were divided into three groups: normal group, control group, experimental group. To induce the allergic rhinitis in control group and experimental group, rats were sentitized intraperitioneally with 0.1% ovalbumin solution 3 times at intervals of 2 weeks. Then intranasal sensitization was performed by diffusing 0.1% ovalbumin solution 3 times at intervals of 2 days for a week. After that time, rats in experimental group were oral administration treated by the Bojuogikgi-tang fer 28 days. We observed changes of IL-4, IL-5, $INF-{\gamma}$, Total IgE and OVA-specific IgE. We used independent-test statistically. Results: 1. In IL-4 study, Bojungikgi-tang treated group didn't show significant differences. 2. In IL-5 study, Bojungikgi-tang treated group shows significant differences. 3. In $INF-{\gamma}$ study, Bojungikgi-tang treated group shows significant differences. 4. In Total IgE, Bojungikgi-tang treated group shows significant differences. 5. In OVA-specific IgE, Bojungikgi-tang treated group didn't show significant differences. According to this result, Bojungikgi-tang was concluded to be effective on lowering the total IgE. Through this. Bojungikgi tang seems to reduce the symptoms of allergic rhinitis. More studies are required to know exact mechanism of Bojungikgi tang to show the anti allergenic effect.
Objectives The purpose of this study is to investigate the effect of KKHS on atopic dermatitis in an in-vivo experiment using an NC/Nga atopic dermatitis mouse, which has histological and clinical similarities to this condition in humans. Methods To investigate the effect of KKHS on atopic dermatitis (AD), we evaluated atopic dermatitis-like skin lesions by clinical skin index and analyzed immunological parameters in peripheral blood mononuclear cells(PBMCs), splenocytes, draining lymph node(DLN) and performed skin histology in ears and dorsal skin of atopic dermatitis of NC/Nga mouse in vivo. Results In vivo, clinical skin severity score was significantly lower in the KKHS group than in the control group. IgE, IL-6, TNF-${\alpha}$, IgM, IgG2a and IgG2b levels in serum decreased remarkably in the KKHS group than in the control group, and the level of IFN-${\gamma}$ production which is secreted from Th1 cell was increased by KKHS. After this experiment we analyzed immunological cells ($CD3^+$, $CD19^+$, $CD4^+$, $CD8^+$, $CD3^+CD69^+$, $CD4^+CD25^+$ and $CD49b^+$) by flow cytometry. It results that the total absolute number of $CD3^+$, $CD19^+$, $CD4^+$ and $CD8^+$ cells were recovered as much as normal state, and the level of $CD3^+CD69^+$ in isolated DLN and PBMCs were significantly decreased, and total absolute number of $Gr-1^+$, $CD11b^+$ and $CD3^+$ in dorsal skin of NC/Nga mouse were decreased by KKHS. We analyzed ear, DLN, and neck-back skin after biopsy and dyeing by hematoxyline/eosin(H&E), toluidine staining (mast cells marker). KKHS were very effective to the histological symptoms which are in dermal and epidermal thickening, hyperkeratosis and inflammatory cell infiltration. Ear thickness was significantly decreased compared with the control group and the size of inflammatory lymphocytes cells (ILC) and plasma cells (PC) in DLN were also decreased. Conclusions KKHS on atopic dermatitis in an in-vivo experiment using an NC/Nga atopic dermatitis mouse was very effectiveness to the atopy dermatitis treatment.
In the recent, increased concern has been focused on the pharmacology and clinical utility of herbal extracts and derivatives as a drug or adjunct to chemotherapy and immunotherapy. Here we investigated the role of the extract of Anethi Fructus in the expression of inflammatory mediators, surface molecule, and related receptors in vitro. In murine macrophage RAW 264.7 cells and peritoneal macrophages of C57BL/6N mice, water extract of Anethi Fructus increased the production of secretary tumor necrosis factor (TNF)-a and Nitric oxide (NO), and the expression level of CD14, LPS co-receptor and CD86, co-stimulatory molecule compared to negative natural extract ex vivo. The water extract of Anethi Fructus increased the production of interferon (IFN)-g from splenocytes. Also, water extract of Anethi Fructus increased ConA-induced cell proliferation. These results suggest that water extract of Anethi Fructus may enhance the immune response through immune modulation of macrophage and lymphocytes.
원발성 아메바성 수막 뇌염을 일으키는 병원성이 강한 자유 생활 아메바인 Naegleria fowleri이 영양형을 마우스 비강에 접종시킨 후 12시간, 1일, 2일, 3일 및 5일째에 비장 세포의 자연살세포 독성을 측정하였다. 51Cr 방출 경사법과 단세포 독성 검사에 의하여 표적 세포 결합능, 활성 자연살세포, Vmaz 및 재순환능을 측정하였다. 감염 마우스의 사망률은 접종 후 20일까지 76.7%이였으며 사망한 마우스의 평균 생존 기간은 12.87±0.71일이었다. 자연살세포의 독성은 아메바 접종후 12시간째에 대조군에 비하여 유의하게 증가하기 시작하여 접종후 1일째 가장 증가하였다. 표적 세로 결합능간에는 경과 기간별로 유의한 차이가 없었으며 환성 자연살세포와 재순환능은 자연살세포의 독성이 증가함에 따라 증가하였다. N. fowleri가 감염되었을 때 자연살세포의 활성에 미치는 영향을 보면 자연살세포의 독성이 증가함에 따라 환성 자연살세포 및 재순환능이 증가하였고, 표적 세포 결합능에는 영향을 미치지 않는 것을 알 수 있었다.
한국 전통발효음료 중에 존재하는 특이다당류의 화학적 특성 및 생물활성을 규명할 목적으로, 농가에서 직접 발효한 감식초로부터 다당류를 분리하고 이들의 각종 면역증강활성에 대해 검토하였다. 감식초에 80% ethanol 침전을 행하여 얻어진 조다당 획분 PV-0을 이용, 연속적인 3회의 column chromatography를 행하여, 감식초의 주요 다당인 PV-1b-I으로 정제할 수 있었다. PV-1b-I획분은 HPLC상에서 대칭을 유지하는 단일 peak로 검출되었으며, 분자량은 약 110 kDa으로 평가되었다. 정제다당인 PV-1b-I의구성당 조성을 확인한 결과, mannose(46.8%), galactose(28.5%) 및 arabinose(19.1%)를 높은 비율로 함유하고 있었다. 또한 본 정제다당은 ${\beta}$-glucosyl Yariv reagent와 강하게 반응하는 특성을 보여주었으며 이러한 사실로부터 PV-1b-I이 arabino-3,6-galactan 부위를 소유함을 확인할 수 있었다. 한편 감식초에서 정제한 다당 PV-1b-I은 in vitro상에서 macrophage의 활성화를 높은 비율로 유도하였으며, 비장세포에 대해 높은 증식능을 보였다. 또한 PV-1b-I을 정맥 투여한 경우, NK cell을 활성화하여 YAC-1 종양세포에 대한 세포독성을 증가시킴이 관찰되었다. 한편 고분자 정제 다당인 PV-1b-I는 비특이적 면역계에 있어 중요 역할을 담당하고 있는 보체계에 대하여 농도 의존적인 활성화 경향을 보였다. 이들은 $Ca^{2+}$ 이온이 제거된 상태에서의 항보체 활성과 anti-human C3를 이용한 2차원 면역전기영동에 의하여 C3 산물을 동정한 결과로부터 보체계의 classical pathway와 alternative pathway 양 경로를 모두 경유하여 활성을 나타냄을 확인할 수 있었다.

④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
