Effects of Dietary Folate Content on Folate Concentration and DNA Strand Breaks after Alkaline Treatment in Immune Cells (식이엽산함량이 흰쥐의 면역세포의 엽산농도와 알칼리 처리후의 DNA 이중 나산선 잔존율에 미치는 영향)

  • Folate, a precursor of the coenzyme tetrahydrofolate, plays an important role in DNA replication and cell proliferation, and thus could influence rapidly proliferating immune cells such as leukocytes and splenocytes. The effects of dietary folate on folate concentrations of plasma, thymus, spleen and leukocytes were investigated in rats. The animals were raised for 6 weeks on semipurifed experimental diets containing 0mg, 2mg, 4mg, 8mg folate/kg diet. Folate concentrations were determined microbiologically using Lactobacillus casei(ATCC 7469), and DNA strand breaks produced by alkaline treatment were analyzed fluorometrically. When compared to folate adequate diet, the folate deficient diet(0mg folate/kg diet) resulted in lowest folate levels in plasma, thymus, spleen and leukocytes, and the highest DNA strand breaks in spleen cells and leukocytes. Dietary folate levels significantly increased folate concentrations of immune tissues, leukocytes, and the plasma in a dose dependent manner, folate concentrations being highest with a diet providing 8mg folate/kg diet. The percentages of the double strand DNA remaining in the splenocytes and leukocytes after alkaline treatment were significantly increased with higher amounts of dietary folate in a dose dependent manner. Folate intakes of 8mg than 4mg/kg diet was found to be more effective in the prevention of DNA strand breaks. The results of this study suggest that increased folate above the requirement level could improve DNA stabilities in immune cells.

Anti-metastatic and Immunomodulating Activity of Water-Soluble Components from Five Mushroom Extracts

  • Objectives : This experiment was conducted to evaluate the inhibitory effects against lung metastasis and promotion of splenocytes by water-soluble components from five mushrooms extracts (WEFM): Garnoderma frondosa, Corious versicolor, Codyceps militaris, Hericium erinaceus and Lentinula edodes. Methods : Colon 26-L5 carcinoma cells were injected through the tail vein to induce lung metastatic cancer. Changes in weight of lung were observed and cytokine level was analyzed to evaluate immunological changes. Results : Oral administration of WEFM resulted in a significant inhibition of lung metastasis after intravenous injection of colon 26-L5 cells in a dose-dependent manner. There was also a significant increase in T cell and B cell mitogenic stimuli and production of IFN-g by splenocytes stimulated with Con A compared to untreated controls. Conclusion : WEFM may have anti-tumor activities via Th1-type dominant immune responses.

Bamboo Culm Extract Downregulated Activation of NKT- and B- cells and Production of IL-6 in Pristane-Induced Lupus Mice

  • Lupus is characterized by immunoregulatory abnormalities between T- and B-cells leading to autoantibody production and multiorgan injuries. We investigated whether bamboo culm extract (BC) ameliorates aberrant activation of T cells and B cells and attenuate production of IL-6 in pristane-induced lupus mice. Lupus was induced by i.p. a single injection of 0.5 ml of pristane in female BALB/c mice, which, later about 4 months, were used as a lupus model. The pristane-induced lupus mice and healthy mice were injected i.p. with BC 5 ${\mu}l$/kg or PBS once a day for 3 weeks. These results demonstrated that BC significantly decreased levels of serum and BAL IL-6 and production of IL-6 by macrophages with/without LPS, and downregulated expression of NKT cell and CD86+ CD45R/B220+, but not CD80+CD45R/B220+ and CD69+CD4+ in the splenocytes in pristaneinduced lupus mice. Moreover, BC greatly increased Con A-stimulated production of IL-6, IL-10 and IFN-${\gamma}$ by splenocytes obtained from pristane-induced lupus mice. Therefore, our findings suggest that BC may ameliorate lupus pathogenesis in pristane-induced lupus mice via downregulation of aberrant activation of NKT cells and B cells and inhibition of production of IL-6.

Effects of Neasookseul-Tang on Anti-Cancer Action in Mice (내소옥설탕(內消沃雪湯)의 항암효과(抗癌效果))

  • Objective: This Study was to investigate effects of Neasookseul-Tang on the anti-cancer, proliferation of immunocytes and nitric oxide(NO) production of peritoneal macrophages. Methods: We used Neasookseul-Tang extract(NOT) with freeze-dried, 8wks-old male mice(balb/c, ICR) and cancer cell lines(L1210, sarcoma-180) for this Study. The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). Results: 1. NOT was significantly showed cytotoxicity on the L1210 and Sarcoma-180 cell lines. 2. NOT was significantly increased proliferation of thymocytes and splenocytes in vitro. 3. NOT was significantly decreased proliferation of L1210 cells in L1210 cells transplanted mice. 4. NOT was significantly increased proliferation of thymocytes and splenocytes in L1210 cells transplanted mice. 5. NOT was significantly increased NO production from peritoneal macrophages in L1210 cells transplanted mice. 6. NOT was significantly inhibited body weight and tumor weight in Sarcoma-180 cells transplanted mice. 7. NOT was significantly increased in the mean survival days in Sarcoma-180 cells transplanted mice. Conclusions: The present author thought that NOT had action of anti-cancer by accelerating immuno-function.

Inhibitory Effects of Soamsan1 on Lung Metastasis of B16 Melanoma Cells (B16 흑색종 세포의 폐전이에 대한 소암산1의 억제효과)

  • We have examined whether Soamsan 1 (SA 1) augment the inhibitory effect of oral administration of Soamsan (SA) on lung metastasis of mouse 816 melanoma cells. The inhibitory effect was slightly enhanced by increase in administration dosage of SA 1. SA 1 as well as SA inhibited effectively the lung metastasis regardless of the pretreatment with anti-mouseNK monoclonal antibody. However, in the case of 2-chloroadenosine-pretreated mice, the inhibitory effects of SA and SA 1 were decreased by 18 and 23%, respectively. In vitro stimulation of the mouse splenocytes with mitogens showed that SA or SA 1 significantly augmented the proliferation of mouse splenocytes. Especially, the activity was more prominent in the presence of a B cell mitogen. LPS than a T cell mitogen, Con A. These results suggest that oral administration of SA 1 or SA inhibited lung metastasis of B16 melanoma cells, possibly through a mechanism mediated by the activation of macrophages and B lymphocytes in the host immune system. However, SA 1 did not showed more significant augment of the activation of immune system than SA.

Effects of Methidathion on Humoral Immune Response in Mice (Methidathion이 체액성 면역 반응에 미치는 영향)

  • The effects of methidathion on humoral immune response were studied in BALB/c mice. 0.5 or 5.0 mg/kg/day methidathion were administered orally for 14 days. The parameters examined to assess apparent toxicity of methidathion included changes of body weight, relative weight of spleen, thymus, sidney and liver, and viable spllenic cell numbers. To evaluate the humoral immune response, thymus, kidney and liver, and viable splenic cell numbers. To evaluate the humoral immune resopnse, the plaque forming cell(PFC) responses sheep the red blood cells (SRBC) and the lovels of serum IgG to hen egg lysozyme (HEL) were determined. No alterations were observed in changes of body weights, relative organ weights and the numbers of viable splenocytes by exposure to any dose of methidathion. At the dose of 0.5mg/kg only PFC response was decreased, whereas both PFC response and the level of serum IgG were decreased significantly at the dose of 5.0 mg/kg. These results indicate that exposure to methidathion may cause sup[pression of humoral immune reponse in mice without overt changed in lymphoid organ weight or viability of splenocytes.

Anti Inflammatory Effect of Low Level Laser Irradiation on the LPS-stimulated Murine Immunocytes

  • Pro-inflammatory cytokines, such as tumor necrosis factor $(TNF)-{\alpha}$, interleukin-12 (IL-12) and interleukin $(IL-1)-{\beta}$, play a key role in causing inflammatory diseases, which are rheumatoid arthritis, Crohn's disease and sepsis. Accumulating evidences suggest that low level laser irradiation (LLLI) may have an anti-inflammatory action. However, there are few data regarding down regulation of Th1 immune response by using the diod typed laser emitting device for human patients. As a fundamental step in order to address this issue, we investigated immunological impact of the low level laser irradiation (10 mw laser diode with a wavelength of 630 nm) on expression of pro-inflammatory cytokines in murine immunocytes (splenocytes and peritoneal macrophages) in vitro. The LLLI on lipopolysaccharide (LPS 100 ng/ml)-stimulated murine splenocytes and macrophages, clearly down regulated mRNA expression of $TNF-{\alpha}$ and IL-12 in dose-dependent manner. In addition, LLLI significantly inhibits the NO production in the LPS-stimulated murine macrophages. This data suggests that LLLI (wavelength of 630 nm) may exert an anti-inflammatory action via modulation of pro-inflammatory cytokine and NO production pathway.

Immunomodulatory effect of Tinospora cordifolia in tumor-bearing host

  • The present investigation was undertaken to study whether tumor-associated macrophages of Daltons lymphoma (DL), a spontaneous transplantable T cell lymphoma can be activated to tumoricidal state by alcoholic extract of Tinospora cordifolia (ALTC). In vivo administration of ALTC (200 mg/kg body weight) in DL-bearing mice resulted in an enhanced RNI production and an augmented cytotoxic response of tumor-associated macrophages. Earlier we had reported that DL-bearing mice show a regression of thymus and an enlargement of spleen. In vivo administration of ALTC to DL-bearing hosts resulted in a decrease in the weight of spleen and counts of splenocytes along with an increase in the weight of thymus as compared to control DL-bearing mice. In vivo administration of ALTC in DL-bearing mice also resulted in an increase in the proliferation of splenocytes/thymocytes and BMC. The results of this study indicate that the ALTC upon in vivo administration in DL-bearing shows immuno-modulatory effects and thus may have clinical significance.

Effect of the Seeds of Coix Lachryma-Jobi L. var. Ma-yuen Stapf. on the Proliferation of L1210 cells, a Leukemic cell-lines (율무가 백혈병세포주인 L1210 세포의 증식에 미치는 영향)

  • The purpose of this research was to investigate the effects of Coix Lachryma-Jobi L. var. Ma-yuen Stapf. on the proliferation of L1210 cells and immune cells. The hexane fraction of Coix Lachryma-Jobi decreased the proliferation of L1210 cells and induced DNA fragmentation of L1210 cells. Also, the fraction decreased the cell viability of murine thymocytes and splenocytes, but increased the phagocytic activity of murine peritoneal macrophages. In addition, fatty acids and fatty acid methyl esters decreased the proliferation of L1210 cells and induced DNA fragmentation of L1210 cells. The main components of the fraction are fatty acid and their derivatives. These results indicate that Coix Lachryma-Jobi has a cytotoxicity on tumor cells and immune cells.

Role of Metabolism by Intestinal Bacteria in Arbutin-Induced Suppression of Lymphoproliferative Response in vitro

  • Role of metabolism by intestinal bacteria in arbutin-induced immunotoxicity was investigated in splenocyte cultures. Following an incubation of arbutin with 5 different intestinal bacteria for 24 hr, its aglycone hydroquinone could be produced and detected in the bacterial culture media with different amounts. Toxic effects of activated arbutin by intestinal bacteria on lymphoproliferative response were tested in splenocyte cultures from normal mice. Lipopolysaccharide and concanavalin A were used as mitogens for B- and T-cells, respectively. When bacteria cultured medium with arbutin was treated into the splenocytes for 3 days, the medium cultured with bacteria producing large amounts of hydroquinone induced suppression of lymphoproliferative responses, indicating that metabolic activation by intestinal bacteria might be required in arbutin-induced toxicity. The results indicated that the present testing system might be applied for determining the possible role of metabolism by intestinal bacteria in certain chemical-induced immunotoxicity in animal cell cultures.
