The 2'-fucosyllactose (2'-FL) is the richest components in a human milk oligosaccharide. Several studies have reported that 2'-FL has beneficial effects in infants. However, there are few studies on its immune-enhancing effects. This research aimed to examine the immune-enhancing effect of 2'-FL on immunosuppression by cyclophosphamide (CCP) in ICR mice. Mice were orally administered distilled water or 0.5 mg/kg B.W. 2'-FL for 14 days. An immunocompromised mouse model was induced using CCP 80 mg/kg B.W. at 12-14 days. Using the CCP had effects on reducing their body weight, organ weight, spleen index, natural killer (NK) cell activity, and cytokines concentration and expression. This study also used concanavalin A-mediated T-cell proliferation to verify the immune-enhancing effects in the sample. Body weight, spleen index, organ weight, and cytokine levels were measured to estimate the immune-enhancing effects. The body weight at 14 days tended to increase, and the spleen weight and index significantly increased in the 2'-FL group compared to the CCP group. The NK cell activity increased in the 2'-FL group compared to the CCP group, but there was no significant difference. The concentration of interleukin (IL)-2 tended to recover in the 2'-FL group compared to the CCP group. The 2'-FL group showed a significant increase of IL-10 and IFN-gamma concentration compared to the CCP group. In addition, there was a trend of increased IL-10 mRNA expression compared to the CCP group. These results revealed that 2'-FL improved CCP-induced immunosuppression, suggesting that 2'-FL may have the potential to enhance the immune system.
Effects of royal jelly(RJ) on the immune system in normal and cyclophosphamide(CY)-treated mice were investigated. The results were as following: 1. Body weight, spleen weight, thymus weight, WBC, cell-mediated immunity (CMI, contact hypersensitivity to DNFB), humoral immunity (HI, Hemagglutinin-, Hemolysin-titer) were increased or decreased dependent on the day of administration of RJ in normal mice. But it showed no effect on liver weight and RBC. 2. Combined treatment with RJ in CY-treated mice on the day which RJ showed the increasing activities in normal mice inhibited the decrease of survival rate, body weight, spleen weight, WBC and CMI caused by CY, but no effect on the decrease of thymus weight and HI induced by CY.
To investigate the possibility of Panax ginseng as a therapeutic agent for the immune suppression, ginseng total saponin (GTS) extracted from korean red ginseng was tested on immune functions from morphine-induced immune suppressed mice. To study how immune functions are affected by morphine and also to test whether GTS can be an useful therapeutic agent for morphine toxicity, several parameters were employed, body weight, immune organ weight, B cell functions, and T cell function. Morphine impaired the development of body weight and immune organ weight such as spleen and thymus. Morphine also depressed a B-cell function, antibody production. T-cell functions studied by type IV hypersensitivity test were most markedly affected by morphine treatment. GTS restored most of morphine-induced immune suppression. GTS restored the morphine-induced decrease in spleen weight to body weight ratio in a dose dependent manner, but not the body weight decrease. Also all of the morphine-induced impairments of B cell functions and cellmediated immunity were fully recovered by GTS. These results suggest that ginseng product could be very helpful for the treatment of immune suppression occurring in morphine abusers.
Changes in gastrointestinal tissue blood volume induced by variations of venous pressure between 6 and 40 mmHg were studied in 32 rabbits. Venous pressure lowering was produced by withdrawal of appropriate volume of blood and venous pressure elevation was obtained by partial occlusion of intra-thoracic vena cava inferior. Estimation of regional tissue blood volume was performed by means of regional distribution of injected $Cr^{51}-labeled$ red blood cells. The following results were obtained. 1. At the normal control venous pressure value of 18 mmHg, spleen showed the highest value of tissue blood volume expressed on weight basis, namely, $111{\mu}l/gm$, Liver tissue blood volume was $95\;{\mu}l/gm$, small intestine 24 and stomach $21\;{\mu}l/gm$, respectively. 2. Linear relationships were observed between venous pressure change and gastrointestinal tissue blood volume. The coefficients of correlation were: in spleen r=0.723; in liver r=0.791; in stomach r=0.704, respectively. In small intestine the relationship was less clear and r=0.358. Tissue blood volume of extrabdominal tissue, such as M. gastrocnemius was not influenced by venous pressure change. 3. The highest change in tissue blood volume expressed on weight basis was observed in spleen. The liver tissue showed the next highest change. Change in total tissue blood volume, however, was greatest in liver and next greatest in small intestine. This was interpreted by the fact that total weight of these two organs was much greater than that of spleen. 4. The mechanism that the change in tissue blood volume lies in the venous system which has a great compliance was discussed.
Experiments were performed on rats to investigate the effect of doses of capsaicin on the immune response. Olive oil and the 0.3 mg, 1.0 mg and 3.0 mg/kg administration of capsaicin in olive oil were injected intraperitoneally every day for 4 weeks. Rats were sensitized and challenged with sheep red blood cells (S-RBC). Immune responses were evaluated by organ weight, HA and HY titer, Arthus reaction, delayed type hypersensitivity (DTH) and Rosette froming cell. Following results have observed. 1) The weight of spleen and thymus were increased by doses of compared with control group, but the body weight of rats was decreased. 2) HA titer, Arthus reaction and DTH were significntly decreased by doses of capsaicin as compared with control group. 3) Rosette forming cell in spleen cells was decreased according to the increase of capsaicin doses. These results suggested that high dose of capsaicin decrease humoral and cellular immune response in rats.
We purified a protein carboxyl O-methyltransferase (protein methylase II) from porcine spleen to homogeneity. The molecular weight of the porcine spleen protein methylase II (ps-PM II) was estimated to be 27,500 daltons on SDS-PAGE. Amino acid sequence of N-terminal 28 residues for ps-PM II was identified. Amino-terminal three amino acid residues of ps-PM II were deleted when compared to those of other protein carboxyl methytransferase. S-Adenosyl-L-homocysteine competitively inhibits ps-PM II with a K, value of $1.63{\times}10^{-7}M$. Myelin basic protein exhibited the highest methyl-accepting capacity among the proteins tested.

The co-administration of BPA and BBP induced slow weight gain compared with single administration in dams. Also, such mixture induced low neonatal body weights in next generation. The dams treated with BPA and BBP showed significant organ weight changes in liver, spleen exposed during lactational periods. But the dams exposed during lactational periods showed significant organ weight changes not only in liver, spleen but also in kidney, uterus and ovary. The F1 female rats exposed during lactation periods showed significant organ weight changes in liver, spleen, ovary. The F1 male rats showed significant organ weight changes in liver, kidney, epididymis, vesicular glands, prostate. However no clear synergistic effects of BPA and BBP could be found. Estrogen receptor ${\alpha}$ expression by BPA and BBP in the uterus(dam, F1 female) and testis(F1 male) were studied. There was no significant different $ER{\alpha}$ expression pattern between control and treated groups. But $ER{\alpha}$ expression were increased in F1 male testis and female uterus. F1 male showed distinct $ER{\alpha}$ expression, especially in the group of lactational combined exposure. Synergistic $ER{\alpha}$ expression was found by combined treatment of BPA and BBP.
This study was carried out to investigate the effect of addition of cellulose in the diet on the metabolism in rat fed high and low level of zinc. The experimental animals were consisted of 24 male weaning rats of Sprague-Dawley strain(mean weight 72.3g), and they were devided into 4 groups of 6 rats and fed experimental diets for four weeks. Dietary zinc levels used were 10 ppm, and 300ppm and cellulose levels were 2.5% and 10% of diet by weight. Throughout the experimental period, feed consumption and body weight gain were measured and feed efficiency ratio was calculated. The weight of live, kidney and spleen were measured, and the contents of zinc in feces, urine, liver, kidney, spleen and serum were determined. The results obtained are summarized as following ; 1. Body weight gain in high zinc-adequate cellulose group was significantly higher than the other groups. Feed consumptions were significantly higher in high zinc groups and no significant difference was found with dietary cellulose levels. 2. Fecal zinc excretions of four groups were not different at the first week, but at the end of fourth week, high zinc groups experince significantly more zinc excretion than low zinc groups, and also high cellulose groups had higher zinc contents in the feces than the adequate ones within the same zinc levels(p<0.05). There was no significant difference in the urinary zinc excretion. 3. The weights of liver, kidney and spleen were heavier in the high zinc groups than the lower ones, and higher in the high cellulose groups(p<0.05). The liver zinc contents were significantly lower in the low zinc and high cellulose groups. However zinc contents in the kidney and serum were not influenced by dietary zinc level but by cellulouse level. High cellulose diet lowered serum and kidney zinc concentrations(p<0.05).
This experiment was carried out to study the toxic effect of solublized methylcellulose (MC). Sprague-Dawley rats were dosed with 1%(w/v) MC in 0.9% saline by gavage at a dose of 10ml/kg b.w/day or by intravenous injection at a dose of 5ml/kg b.w/day for 28 days. Clinical signs were observed once a day. Body weights, water and food consumptions were measured and urinalysis was performed several times during the experiment. Rats were sacrificed on days 3, 7, 15 and 28 for hematology, blood chemistry, organ weights and histopathology. The relative weight of the spleen and foamy cells of the spleen were increased in the gavage group. Body weight gain, food consumptions, the values of RBC, Hb, MCH, Hct, serum proteins, glucose, bilirubin, AST, and ALP were decreased in I.V. treatment group. On the other hand, water consumptions, the values of serum cholesterol, creatinine, and BUN were increased. Microscopic findings were granulomas, distended sinusoids, and hypertrophy of Kupffer cells with vacuoles in the liver. Spleen exhibited granuloma, increased extramedullary hematopoiesis, and congestion. Kidney exhibited foamy cells in the glomeruli, distension of the tubules. The findings appeared more severe when the treatment was extended. In conclusion, MC solution is not a safe vehicle for intravenous administration because of the toxic effects on the liver, kidney and spleen. In addition, a long-term and large dosage of oral administration of MC appears to be unsafe also and needs to be investigated further.
Bisphenol A (BPA) is a monomer widely used in the manufacturing polycarbonate plastics or epoxy resin, and xenobiotics recently known as endocrine disrupting chemical. In this paper, to assess the effects of bisphenol A on immunopathological parameters (body weight, organ weight, hematological parameters, cellularity and surface marker) in mice, ICR female mice were orally exposed to BPA dissolved in olive oil as concentrations of 100, 500, 1000 ㎎/㎏/day b.w. 5 days a week for 30 days (subacute exposure). Liver - and kidney weight was significantly increased as dose-dependent manner, but body- , spleen- and thymus- weight didn't changed. In hematological parameters, WBC and MCHC were lowered but HCT and MCV were siginificantly enhanced. There was no significant differences in peritoneal macrophages number of the mice exposed to BPA. However, number of splenocytes of spleen, CD3/sup +/ and CD4/sup +/ cell in splenocytes, CD4/sup +/ and CD8/sup +/ cell in thymocytes were decreased at the mice subacutely exposed to BPA. In addition, BPA decreased expression of B7-1 and B7-2 on macrophages. Therefore, these results showed BPA may affect hematological parameters, cellularity and surface marker of immunocytes.
