Human Lung Cancer Cell Xenografts Implanted under the Capsule of Kidney, Spleen and Liver (폐암 세포주를 사용한 신, 비장 및 간 피막하 분식법의 비교)

  • Bakground : Complete resection by the surgery has been selected as the treatment of choice in lung cancer patients, but in cases of recurrence after excision or inoperable cases, the importance of anticancer chemotherapy has been emphasized. If one can select a set of the sensitive chemotherapeutic agents before anticancer chemotherapy, it will give more favourable results. Subrenal capsular assay has been recognized as a useful in-vivo chemosensitivity test of thoracic and abdominal tumors and it can be done in a short time for a rapid interpretation of tumor responsiveness to anticancer chemotherapeutic drugs. It has been reported that various kinds of cancer cells can be implantable to the kidney, but so far there is no comparative study of xenogeneic cell implantation on liver, spleen and kidney. The author implanted the human lung cancer cells under the capsule of S.D rat's liver, spleen and kidney respectively and compared the pattern of growth and histology. Material and Method: After incubation of human lung cancer cell line (SW-900 G IV) in RPMI 1640 (Leibovitz L-15 medium) culture media, 3${\times}$3${\times}$3 mm size fibrin clots which contain 108 cancer cells were made. Thereafter the fibrin clots were implanted at subcapsule area of liver, spleen and kidney of S.D. female rat. For immune suppression, cyclosporin-A (80 mg/Kg) was injected subcutaneously daily from post-implantation first day to sixth day. The body weight was measured at pre and post implantation periods. The growth pattern and the size of tumor mass were observed and the pathologic examination and serum tumor marker tests were performed. Result: Body weight increased in both of control and experimental groups. Serum Cyfra 21-1 was not detected. Serum levels of CEA and NSE revealed no significant change. The SCC-Ag increased significantly in implanted group. The growth rate of human lung cancer cells which was implanted on spleen was higher than on liver or kidney. The surface area, thickness, and volume of tumor mass were predominant at spleen. The success rates of implantation were 80% on kidney, 76.7% on spleen and 43.3% on liver. Pathologic examination of implanted tumors showed characteristic findings according to different organs. Tumors that were implanted on kidney grew in a round shape, small and regular pattern. In the spleen, tumors grew well and microscopic neovascularization and tumor thrombi were also found, but the growth pattern was irregular representing frequent daughter mass. Human lung cancer cells that were implanted in the liver, invaded to the liver parenchyme, and had low success rate of implantation. Microscopically, coagulation necrosis and myxoid fibrous lesion were observed. Conclusion: The success rate of implantation was highest in the kidney. And the mass revealed regular growth that could be measured easily. The SCC-Ag was presented earlier than CEA or Cyfra21-1. The Cyfra21-1 was not detected at early time after implantation. The best model for tumor implantation experiment for chemosensitivity test was subrenal capsular analysis than liver and spleen and the useful serum tumor marker in early period of implantation was the SCC-Ag.

Effects of Ethanol and Saccharin on the Immune Response in Rats (랏트에 있어서 에탄올과 삭카린이 면역반응에 미치는 영향)

  • Experiments were performed to investigate effects of ethanol and saccharin on the immune system in rats. 4% ethanol and 0.02, 0.20, 2.00% saccharin solution in 4% ethanol were provided ad libitum by tap water for 4 weeks. Rats were sensitized and challenged with sheep red blood cells (S-RBC). Immune responses were evaluated by relative immuno organ weight, antibody production, Arthus reaction, delayed type hypersensitivity, and rosette forming cell. Ethanol exposure decreased thymus weight and delayed type hypersensitivity. A combined solution of ethanol and saccharin decreased water intake, growth rate, spleen weight, thymus weight, humoral and cellular immune response. Especially, a 2% saccharin solution in 4% ethanol very significantly suppressed cellular immunity.

Oligosaccharides Affect Performance and Gut Development of Broiler Chickens

  • The effects of oligosaccharide supplementation on the growth performance, flock uniformity and GIT development of broiler chickens were investigated. Four diets, one negative control, one positive control supplemented with zinc-bacitracin, and two test diets supplemented with mannoligosaccharide (MOS) and fructooligosaccharide (FOS), were used for the experiment. Birds given MOS or FOS had improved body weight (BW) and feed efficiency (FCR), compared to those fed the negative control diet during the 35-d trial period. The effect on FCR became less apparent when the birds got older. FOS and MOS supplementation reduced the pancreas weight as a percentage of BW, with an effect similar to that of the antibiotic, at 35 d of age. Birds given MOS tended to have a heavier bursa (p = 0.164) and lower spleen/bursa weight ratio (p = 0.102) at 35 d of age. MOS and Zn-bacitracin showed a clear improvement on flock uniformity, compared to FOS. The mortality rate was not affected by FOS or MOS.

  • The effects of Chelidonii Herba on the cell mediate and humoral immune responses were investigated in ICR mice. ICR mice received intraperitoneal injection of methanol extracts (ether fraction, buthanol fraction, water fraction) for 5 days before sensitization. The change of body weight (%), the organ weight ratio (%), the numbers of RBC and WBC, the ear swelling by dinitrofluorobenzene, the titers of hemagglutination (HA) and hemolysin (HY) to SRBC were determined. The results were as follows; 1. The change of body weight (%) showed a tendency of increasing generally. 2. The weight of spleen (%) revealed decreasing but that of thymus revealed increasing. 3. Ear swelling was maxium at 48 hours after challenge and was significantly decreased in the groups treated with Ether, Buthanol and Water. 4. The numbers of RBC and WBC were decreased generally. 5. HA and HY titers were decreased significantly.

A Study of Elucidation of Protein Quality of Raw and Heated Legumes Fed by Three Different Dietary Levels on Rats (팥과 녹두단백질의 첨가수준과 가열처리가 흰쥐의 성장에 미치는 영향)

  • Various kinds of legumes have included as essential foods in Korean diet. However, a little attention have been paid on variety of those beans other than soybean. Main purposes of this study is to evaluate the protein quality of raw and cooked, red mung bean. Eighty male, sprague-Dawely rats weighing 50 gram were devided into 16 groups, five rats each. Casein protein from red and mung bean were used was included to after true digestibility of the protein of legemes. After 4 weeks feeding period animals were sacrificed and following data: all determined food intake, body weight gain, F.E.R., pp.E.R. and weights of liver, kidney heart, spleen, testis, pancreas, skeletal muscles. Food intake, body weight gain, F.E.R., pp.E.R. of casein group were higher than those of experimental groups (red bean, mung bean) body weight gain was increased with protein increment in the diet in all groups. The weight of liver showed significant difference between standard group and experimental group. (red bean, mung bean) The nitro gen content of liver and muscles were increased with level of protein in the diets.

Effect of Banhabakchulchunma-tang on the Hepatic, Splenic and Cardiac Toxicity induced by Doxorubicin (반하백출천마탕(半夏白朮天麻湯)이 Doxorubicin에 의해 유발(誘發)된 독성(毒性)에 미치는 영향(影響))

  • Object : The effect of Banhabakchulchunma-tang extracts on the hepatic, splenic and cardiac toxicity induced by Doxorubicin administration(Three injection protocol) were monitored using male ICR mice. Methods : The changes of body weight, clinical signs, necropsy findings and organ weights of liver, spleen and heart were observed with blood GOT and GPT levels. Results : 1. Decrease of body weight and The degrees of anorexia, ataxia and dehydration after Doxorubicin treatment were dose-dependently inhibited by Banhabakchulchunma-tang extracts. 2. Increase of absolute and relative liver and heart weight observed in Doxorubicin treatment group were dose-dependently inhibited by Banhabakchulchunma-tang extracts. In addition, the degrees of liver congestion necrotic spot and the degrees of heart congestion enlargement were dose-dependently decreased after Banhabakchulchunma-tang extracts dosing groups compared to that of doxorubicin treatment group. It is also demonstrated that elevated serum GOT and GPT levels in doxorubicin treatment group were significantly decreased in Banhabakchulchunma-tang extracts dosing groups. 3. Decrease of absolute and relative spleen weight observed in doxorubicin treatment group were dose-dependently inhibited by Banhabakchulchunma-tang extracts. In addition, the degrees of splenic atrophy were significantly and dose-dependently decreased after Banhabakchulchunma-tang extracts dosing groups compared to that of doxorubicin treatment group. Conclusion : the toxicity of doxorubicin treatment(decrease of body weights, clinical signs such as anorexia, ataxia and dehydration, changes of organ weights of liver, spleen and heart, elevation of serum GOT and GPT levels) was inhibited and/or prevented by Banhabakchulchunma-tang extracts. According to these results, it is considered that Banhabakchulchunma-tang has some preventive effect against to doxorubicin induced toxicity.

Effects of Curcuma longa Rhizoma on Asthma induced intra-nasal instillation of Ovalbumin in Mice (강황이 난황의 비강내 점적을 통하여 유발된 생쥐의 천식에 미치는 영향)

  • Objective : This study was designed to investigate the effects of Curcuma longa Rhizoma(CLR) on asthma. Methods : Detecting splenocyte proliferation rates, cytokines and antigen specific antibody isotypes in bronchoalveolar lavage fluid (BALF). In addition, the present author I also investigated changes in spleen and histopathological changes of lung tissues. Results : Oral administration of CLR lowered spleen weight and splenocyte proliferation rates. In addition, levels of IL-4, IL-17A and Granulocyte/Macrophage Colony-Stimulating Factor(GM-CSF), Th2 driven cytokines, were lowered respectively and IFN-g, and Th1 driven cytokine, were elevated by CLR. Levels of Ovalbumin(OVA) specific IgE and IgGl in BALF were also lowered by oral administration of CLR too. Conclusion : CLR is useful to treat patients with asthma and the mechanisms are related to the in suppression of Th2 skewing reactions.

  • DA-125, a new anthracycline antitumor antibiotic, was administered to Sprague-Dawley rats intravenously for 4 weeks to investigate the repeated dose toxicity Focal alopecia was noted in three female rats receiving 1.0mg/kg/day. In rats receiving 1.0 mg/kg/day, weight gain decreased in both sexes after first or second week. Hematological examination revealed lower counts of total leukocyte and increased numbers of platelet after second week. At terminal necropsy, atrophy of thymus and spleen was observed. Lymphocytic depletion of thymus and atrophy of white pulp in spleen were observed microscopically. A decrease in the number of hematopoietic cells in the bone marrow and degeneration of germinal epithelia in testes were also observed. These treatment-related effects were mainly confined to rats receiving 1.0 mg/kg/day. And toxic effects with microscopic changes were not observed in rats receiving 0.2 mg/kg/day or 0.04 mg/kg/day.

  • Ginseng components were examined for their ability to modify immunotoxicity of cyclophosphamide. Ginseng polysaccharide fraction (FO, 300 mg/kg) inhibited decreases in the ratio of spleen to body weight, white blood cells (WBC) count and the number of plaque forming cells (PFC) induced by cyclophosphamide (50 mg/kg, i.p.), and increased these variables in normal mice. Ginseng saponin fraction (50 mg/kg) showed to increase hemoglobin content as well as the number of PFC/spleen in normal mice. on the other hand, Panaxytriol (20 mg/kg) Prevented decrease in mc count by cyclophos phamide. Neither saponin fraction nor panaxytriol had any significant effect on the number of PFC and antibody titers in cyclophosphamide-treated mice. These results suggest that ginseng polysaccharine fraction may reduce the immunotoxicity of cyclophosphamide and may be effective in stimulating immune function in normal mice.
