To evaluate the effect of PAHs on fertilization and larval development of the Pacific oyster, Crassostrea gigas via in vivo exposure for 30 days. Conditioned C. gigas adult brood stock was taken from oyster farm in May 2003 and raised in flow through tank at 20-22$^{\circ}C$ with feeding equivalently mixed marine microalgae: Chaetoceros simplex, C. gracilis, Isochrysis galbana and Tetrasemis tetrathele. The oysters were exposed to PAHs cocktail at 200ppb (10 species) until they are fully riped with an untreated group. Motility of sperm, Fertilization and The percent of D-shaped larvae was measured under microscope (Olympus BX-50). Fertilization capability using dry sperm and eggs from both non- and PAHs treated brood stocks was significantly reduced at the treated group (P<0.01). In addition to the origin effect, it was obvious that fertilization of eggs from both group were adversely suppressed in consistent with PAHs level (P<0.01). The percent of D-shaped larvae in treated group was significantly reduced comparing to non-treated. The percent was adversely correlated with PAHs level, which was more significantly reduced in treated group. Therefore, it was obvious that PAHs exposure on the Pacific oyster has adverse affects on the sperm as well as larvae in both maternal and acute source.
Objective s: To assess the fertilizing capacity using sperm penetration assay (SPA) to predict the outcome of the in vitro fertilization-embryo transfer (IVF-ET) outcome. Materials and Methods: Semen samples were provided by 129 patients undergoing IVF. We attempted to correlate the extent of sperm penetration under enhanced SPA protocol with the results of fertilization, cleavage, preimplantation embryo development, and pregnancy. Results: Univariate analysis demonstrated a statistically significant correlation between fertilizing capacity and motility, kinetics, fertilization, cleavage and embryo development, and pregnancy rate. By logistic regression analysis, fertilizing capacity was found to be the only variable that was statistically significant with respect to pregnancy rate. Fertilizing capacity, cleavage rate and pregnant rate were significantly higher in pregnant group. However, the fertilization rates was comparable with both group. Conclusions: Lower fertilizing capacity could denote a poorer prognosis for establishing a pregnancy, even after satisfactory fertilization rate is achieved.
Artificial insemination technique has been contributed immensely for production of livestock worldwide as a critical assisted reproductive technique to preserve and propagate excellent genes in domestic animal industry. In the past decade, methods for semen preservation have been improved mostly in liquid preservation method for boar semen and freezing method for bull semen. Among many factors affecting semen quality during preservation, reactive oxygen species, produced by aerobic respiration in sperm for survival and motility, are unfavorable to sperm physiology. In mammalian cell as well as in the sperm, antioxidant system plays a role in degradation of reactive oxygen species. Magnetized water forms smaller stabilizing water clusters, resulting in high absorption and permeability of the cell for water, implicating its application for semen preservation. Therefore, this review focuses on preservation methods of boar and bull semen with respect to improvement of extender and reduction of reactive oxygen species by using magnetized water and supplementation of antioxidants.
Tributyltin chloride(TBTCl) was administered through discrete immersion(2 hr each) from the $18^{th}-25^{th}$ day after hatching(dph). At the doses of 1, 2, 4 and 8 ${\mu}g/L$, the immersion at 2 ${\mu}g/L$ ensured 93% masculinization and the highest survival of 75% after the treatment. TBTCl acted as a growth suppressant and the magnitude of its suppression was stronger in females. During the 300 day experiment, it postponed sexual maturity of females from 120$^{th}$ dph in the control to 240$^{th}$ dph in the females treated at 8 ${\mu}g/L$. It reduced spawning frequency(22-3 times) and cumulative fecundity(1,632-19 eggs) by reducing the number of vitellogenic eggs. In the treated males too, it reduced sperm motility(100-68 sec); consequently, fertilizability of the sperm cells drawn from these males was also reduced from 88 to 43%. Progeny testing showed that the cross between males treated at>2 ${\mu}g/L$ and normal females generated the presumed 'homogametic' males. Both the treated 'homogametic' and 'heterogametic' males could induce the females to spawn fewer eggs than that of the normal males. A normal female somehow deducted the differences between the control, treated and sex reversed males; it preferred a normal male over a treated one, and a treated one over the sex reversed male.

But, the role of the proteins is not limited to particular function in the biological systems. Some of the proteins act as ion channels such as catsper and proteins like Nanos acts as a translational repressor in germ cells and expressed in prenatal period whose role in male fertility is uncertain. Rbm5 is a pre mRNA splicing factor necessary for sperm differentiation whose loss of function results deficit in sperm production. DEFB114 is a beta defensin family protein necessary for sperm motility in LPS challenged mice where as TEX 101 is a plasma membrane specific germ cell protein whose function is not clearly known u to now. Gpr56 is another adhesion protein whose null mutation leads to arrest of production of pups in rats. Amyloid precursor protein role in Alzheimer's disease is already known but it plays an important role in male fertility also but its function is uncertain and has to be considered while targeting APP during the treatment of Alzheimer's disease. The study on amyloid precursor protein in male fertility is a novel thing but requires further study in correlation to alzheimer's disease.
본 연구는 종모우의 정자수정능력 평가방법의 개발과 정자 기능 및 성상 변화에 영향을 미치는 요인을 알아보기 위하여 시행하였다. 동결-융해된 종모우 정액을 대상으로 정자의 운동성과 정자의 형태를 분석하였고, 정자의 기능 검 사 항목으로서 체외수정(IVF), HOST, Ca-ionophore에 의한 첨체반응율, 정자의 ROS 측정을 위한 luminol, lucigenin-dependent chemiluminescence, LPO 분석을 위한 malondialdehyde의 측정 및 TUNEL (terminal deoxynucleotidyl transferase(TdT) dUTP nick end labelling) 기법을 이용한 정자의 DNA fragmentation를 측정하였으며 이들 각각의 조사 항목들의 분석치들과 체외수정율 및 배발생율과의 상관관계를 조사하여 다음과 같은 결과를 얻었다. 1. 고수정군과 저수정군의 체외수정율과 배반포 발생율의 평균은 각각 64.4%와 34.3%, 18.50%와 6.2%였으며 두 군간에 통계학적으로 유의한 차이를 보였다(P<0.05). 고수정군과 저수정군의 정자운동성과 첨체반응률은 각각 평균79.0 %와 66.2%, 40.7%와 22.9%로 두 군간에 통계학적으로 유의한 차이를 보였으나(P<0.05), 정상형태 정자의 비율과 HOST는 각각 평균 94.6%와 92.7%, 69.4%와 59.8%로 두 군간 유의한 차이를 보이지 않았다. 2. Luminol dependent chemiluminescence, LPO 및 DNA fragmentation의 평균은 고수정군과 저수정군에 있어서 각각 6.4와 6.5, 2.Onmol와 3.Inmol 및 2.6%와 7.4%로 두 군간 통계학적으로 유의한 차이를 보였으나(P<0.05), lucigenin dependent chemiluminescence는 4.7와 4.6로 두 군간 유의한 차이를 보이지 않았다. 3. 체외 수정율은 정자의 운동성 및 첨체반응율과 통계학적으로 유의한 정(positive)의 상관관계(r=0.87, p<0.01; r=0.81, p<0.05)를 나타내었으며, luminol dependent chemiluminescence, lipid peroxldation 및 DNA fragmentation과는 통계학적으로 유의한 부(negative)의 상관관계 (r= -0.81, p<0.05; r: -0.74, p<0.05; r : 0.81, p<0.05)를 나타내었다. 그러나 체외수정율은 정상형태 정자의 비율, HOST 및 lucigenin dependent chemiluminescence와는 유의한 상관 관계를 나타내지 않았다. 4. 배반포 발생율은 첨체반응율과 통계학적으로 유의한 정의 상관관계(r=0.71, p<0.05)를 나타내었으며, luminol dependent chemiluminescence, lipid peroxidation 및 DNA fragmentation과는 통계학적으로 유의한 부의 상관관계(r= -0.71, p<0.05; r= -0.89, p<0.01; r= -0.71, P<0.05)를 나타내었다. 배반포 발생율은 정자의 운동성, 정상형태 정자의 비율 및 HOST, lucigenin dependent chemilumihescence와는 유의한 상관관계를 나타내지 않았다. 이상의 결과를 종합해 보면 정액질의 저하에 ROS의 영향이 밀접히 연관되어 있음을 알 수 있으며, 또한 본 연구에서 적용된 기법들은 정액질의 평가 및 정자 수정능력 향상을 위한 기술개발에 있어서 유용한 평가 방법으로 이용될 수 있을 것으로 사료된다.

Objective: Sperm morphology plays an important role in infertility, especially in cases of defects in the heads of spermatozoa. Tapered-head or elongated-head spermatozoa are examples of morphological abnormalities. The aim of this study was to compare the semen parameters, levels of protamine deficiency, and frequency of apoptosis between patients with normozoospermia and those with teratozoospermia with tapered-head spermatozoa. Methods: Fifty-two semen samples (27 patients with tapered-head sperm and 25 fertile men) were collected and semen analysis was performed according to the World Health Organization criteria for each sample. Protamine deficiency and the percentage of apoptotic spermatozoa were evaluated using chromomycin A3 (CMA3) staining and terminal deoxynucleotidyl transferase dUTP nick-end labelling (TUNEL) assays, respectively. Results: Sperm concentration, motility, and normal morphology in the tapered-head spermatozoa (cases) were significantly lower than in the normozoospermic samples (controls). CMA3-reactive spermatozoa (CMA3+) in the case group were more common than in the controls. Apoptotic spermatozoa (TUNEL-positive) were significantly more common in the cases than in the controls. Conclusion: This analysis showed that tapered-head spermatozoa contained abnormal chromatin packaging and exhibited a high rate of apoptosis, which can be considered to be an important reason for the impaired fertility potential in teratozoospermic patients with tapered-head spermatozoa.
BPA, a diphenyl compound containing groups, that make it structurally similar to synthetic estrogen and is considered as one of the major endocrine disruptors. Silymarin has extensively been used to prevent and/or alleviate some human disease, especially for the treatment of adverse liver conditions. It has an antioxidative efficacy and cancer preventive efficacy. Therefore, we examined the hypothesis that silymarin can inhibit BPA-induced toxicity in boar sperm duing in vitro storage. Sperm characteristics (motility, viability, membrane integrity and mitochondrion activity) in semen exposed to BPA (10~200 uM) were sharply lowered, while it increase in a dose and time dependent manner due to silymarin addition (50~200 uM) into semen extender in the presence of BPA (100 uM). All of the evaluated characteristics were gradually improved in the groups that were treated with silymarin (50~200 uM) in the presence of BPA (100 uM) in comparison to BPA 100 uM alone group, irrespective of incubation periods (3 and 6 h). These results demonstrate that silymarin can ameliorate the toxicity of BPA on boar sperm characteristics during in vitro storage, suggesting that silymarin indirectly act as an antioxidant.
Oxidative stress is one of the major causes of failure of in vitro storage of boar semen. Reactive oxygen species (ROS) are one of the important mediators of oxidative stress during in vitro storage of boar semen. Our study examined the effects of taurine on sperm characteristic and on in vitro developmental embryos during in vitro storage of boar semen for 7 days. Semen was randomly aliquoted into 3 centrifuge tubes and treated with different concentrations of taurine (25-100 mM). The characteristics of boar sperm were analyzed for motility by light microscopy, viability by using a Makler counting chamber and membrane integrity by a hypoosmotic swelling test (HOST). The percentages of motile spermatozoa in taurine groups after 5 days were significantly higher compared to the control. Sperm viability in the control was lower than in taurine groups after 7 days irrespective of different taurine concentration. In the hyoosmotic swelling test (HOST), significantly higher results were obtained in taurine groups after 3 days. Also, the developmental rates of IVM/IVF porcine embryos from semen treated with pyruvate and taurine were significantly increased when compared with the control (p<0.05). These results indicate that supplementation of taurine as an antioxidant in boar semen extender can improve the semen quality.
