Cryopreservation of Filefish (Thamnaconus septentrionalis) Sperm

  • The present study examined the possibility of long term storage, by cryopreservation in liquid nitrogen, of the sperm of Filefish (Thamnaconus septentrionalis), and the changes in motility, survival rate and ultrastructure of the sperm after freezing and thawing. The sperm was collected by stripping and stored on ice until experiments. For selection of the immobilizing solution, diluted artificial seawater (ASW) of 20, 30 and 40% were tested. The sperm motility was significantly inhibited in 30% ASW, and restored entirely after 100% ASW was added again. Two cryoprotectants, dimethyl sulfoxide ($Me_2$SO) and glycerol, were added to 30% ASW to formulate the extenders at the concentrations between 5 to 20% by volume for freezing. The sperm was diluted at the ratio of 1 :6 with the extenders, inserted into 0.5ml plastic straws and frozen at a freezing rate of $50^{\circ}C$/min to $-100^{\circ}C$ after equilibration for 10 min at room temperature, followed by plunging into liquid nitrogen. The straws were thawed in a $30^{\circ}C$ water bath for 15 sec. The highest post-thawed sperm motility and survival rate were obtained with 5% glycerol Afterward, the effect of different freezing rates was examined using 5% glycerol as a cryoprotectant, and the rate of $20^{circ}C/min to $-80^{\circ}C$ showed the best result Some ultrastructural changes of sperm, such as the detachment of plasmatic and nuclear membranes, destruction of mitochondria, were observed after cryopreservation. Morphological normality of the sperm in 5% glycerol frozen at the ratio of 1$0^{\circ}C$/min to $-80^{\circ}C$ was better than that of others.

The Usefulness of Selected Physicochemical Indices, Cell Membrane Integrity and Sperm Chromatin Structure in Assessments of Boar Semen Sensitivity

  • The present work describes experiments undertaken to evaluate the usefulness of selected physicochemical indices of semen, cell membrane integrity and sperm chromatin structure for the assessment of boar semen sensitivity to processes connected with pre-insemination procedures. The experiments were carried out on 30 boars: including 15 regarded as providers of sensitive semen and 15 regarded as providers of semen that is little sensitive to laboratory processing. The selection of boars for both groups was based on sperm morphology analyses, assuming secondary morphological change incidence in spermatozoa as the criterion. Two ejaculates were manually collected from each boar at an interval of 3 to 4 months. The following analyses were carried out for each ejaculate: sperm motility assessment, sperm pH measurement, sperm morphology assessment, sperm chromatin structure evaluation and cell membrane integrity assessment. The analyses were performed three times. Semen storage did not cause an increase in the incidence of secondary morphological changes in the group of boars considered to provide sperm of low sensitivity. On the other hand, with continued storage there was a marked increase in the incidence of spermatozoa with secondary morphological changes in the group of boars regarded as producing more sensitive semen. Ejaculates of group I boars evaluated directly after collection had an approximately 6% smaller share of spermatozoa with undamaged cell membranes than the ejaculates of boars in group II ($p{\leq}0.05$). In the process of time the percentage of spermatozoa with undamaged cell membranes decreased. The sperm of group I boars was characterised with a lower sperm motility than the semen of group II boars. After 1 hour of storing diluted semen, the sperm motility of boars producing highly sensitive semen was already 4% lower ($p{\leq}0.05$), and after 24 hours of storage it was 6.33% lower than that of the boars that produced semen with a low sensitivity. Factors that confirm the accuracy of insemination male selection can include a low rate of sperm motility decrease during the storage of diluted semen, low and contained incidence of secondary morphological changes in spermatozoa during semen storage and a high frequency of spermatozoa with undamaged cell membranes.

  • 본 실험은 PF과 FPP가 소와 사람 동결 정자에 첨가되었을 때 응해 후 정자의 체외 생존성, 운동성 그리고 intact acrosome 향상에 기여할 수 있는지의 여부를 조사하고자 실시하였다. 사람의 정액은 TYB 동결 배양액을 사용하여 초 급속 동결하였다 PF과 FPP 첨가 효과는 각각의 시약이 동결-응해된 소나 사람 정자의 현미경적 조사에 의한 운동성에 미치는 영향과 Coomassie brilliant blue 염색방법에 의한 intact acrosome의 비율에 미치는 영향으로 조사하였다. Bovine 동결 응해 정자에 PF을 첨가하여 운동성을 조사하였던 바, 5 mM 처리군 (50.0%)이 대조군 (34.0%) 보다 유의하게 높은 운동성을 보여주었다 (F<0.05). 동결 응해된 소 정자에 FPP를 농도에 따라 처리하여 intact acrosome을 조사하였던 결과, 50 nM 치리춘 (49%)이 대조군과 25 nH 처리군 (30.0, 38.0%) 보다 유의하게 많은 intact한 acrosome을 유지하였다 (P<0.01). 사람 정자에서 동결에 앞서 PF을 농도에 따라 첨가하여 동결 응해 후 운동성을 조사한 결과, 5.0 mM 처리군 (51.0%)이 대조군과 2.5 mM (39.0, 40.0%) 처리군의 운동성보다 높았다 (P<0.01). 사람 정액의 모든 동결 처리과정 (동결전, 동결, 응해후)에서 50 nM (75.5%) FPP 첨가가 intact acrosome percentage 유지하는데 유의한 효과가 있었다 (대조군: 45.0; 25 nM: 53.0; 100 nM: 68.0%) (P<0.01). PF와 FPP 첨가하여 사람 정자의 동결융해 후 운동성과 intact acrosome에 미치는 영향을 동시에 비교해본 결과, 운동성에서는 PF 처리군이 약간 높지만 intact acrosome rate는 FPP 처리군의 결과 (65.0%)가 PF 처리군 (43.0%)보다 유의하게 높았다 (P<0.05). 따라서 본 실험은 동결-융해된 소 정자에 PF이나 FPP 첨가는 정자의 운동성이나 intact acrosome 비율을 좀더 개선시킬 수 있고, 특히 사람 정자는 동결 전 과정에 FPP를 첨가하는 것이 정자의 체외 생존성, 운동성 그리고 intact acrosome을 유의하게 향상시킬 수 있다는 것을 시사한다.

An Antioxidant Davallialactone from Phellinus baumii Enhances Sperm Penetration on In Vitro Fertilization of Pigs

  • Davallialactone (DAVA) is a hispidin analogue derived from the medicinal fungus Phellinus baumii. We examined the effect of DAVA on in vitro fertilization (IVF) of pigs. Boar spermatozoa were incubated in fertilization medium with varying concentrations of DAVA, then sperm motility and reactive oxygen species (ROS) level were evaluated. Higher sperm motility was found following the addition of 0.5 or $1{\mu}M$ DAVA after incubation than addition of other concentrations or controls. ROS level decreased significantly with the addition of DAVA. The rate of normal fertilization was higher in the presence of $1{\mu}M$ DAVA (65.1%) than were those of other concentrations or controls (45.4~59.4%), and the highest total fertilization rate (mono- and polyspermic oocytes) was observed at $1{\mu}M$ DAVA (83%). In conclusion, addition of DAVA to fertilization medium improved sperm motility, and reduced ROS level so as to potentially improve sperm-oocyte binding in IVF, suggesting the potential of a compound isolated from mushrooms in assisted reproductive technology for humans and animals.

The antioxidant roles of L-carnitine and N-acetyl cysteine against oxidative stress on human sperm functional parameters during vitrification

  • Objective: Amino acids can protect sperm structure in cryopreservation due to their antioxidant properties. Therefore, the present study aimed to investigate the protective effect of L-carnitine (LC) and N-acetyl cysteine (NAC) on motility parameters, plasma membrane integrity (PMI), mitochondrial membrane potential (MMP), DNA damage, and human sperm intracellular reactive oxygen species (ROS) during vitrification. Methods: Twenty normal human sperm samples were examined. Each sample was divided into six equal groups: LC (1 and 10 mM), NAC (5 and 10 mM), and cryopreserved and fresh control groups. Results: The groups treated with LC and NAC showed favorable findings in terms of motility parameters, DNA damage, and MMP. Significantly higher levels of intracellular ROS were observed in all cryopreserved groups than in the fresh group (p≤0.05). The presence of LC and NAC at both concentrations caused an increase in PMI, MMP, and progressive motility parameters, as well as a significant reduction in intracellular ROS compared to the control group (p≤0.05). The concentrations of the amino acids did not show any significant effect. Conclusion: LC and NAC are promising as potential additives in sperm cryopreservation.

Comparison of Spermatozoa Recovery Methods on Cauda Epididymal Sperm of Hanwoo Bulls

  • In this study, two epididymal spermatozoa recovery methods in relation to sperm number, motility, viability and acrosome reaction were examined. Seven bulls were castrated and 7 testicles with epididymides were transferred to the laboratoy. Epididymis in each bull was randomly used for flushing and mincing methods with semen extender (Optixcell, IMV, France). The recovered spermatozoa with adjusted sperm concentration to $40{\times}10^6cells/mL$ was diluted with optixcell and cryopreserved. In experiment 1, the difference in the total number of spermatozoa using flushing and mincing methods was insignificant (2570.0 and $2505.2{\times}10^6cells/mL$, respectively). For experiment 2, the percentage of motile spermatozoa and motility parameters between flushing and mincing methods were studied through the use of sperm class analyzer after frozen-thawing. The percentage of total motile sperm between flushing and mincing methods was almost the same with $89.5{\pm}12.8$ and $91.4{\pm}7.9%$, respectively. The same is the case with experiment 3 wherein the viability and acrosomal integrity of frozen-thawed epididymal spermatozoa by flushing and mincing was insignificantly different. The results from the study showed that both flushing and mincing methods can be used for epididymal spermatozoa recovery in bull.

Melatonin and selenium supplementation in extenders improves the post-thaw quality parameters of rat sperm

  • Objective: The aim of this study was to determine the effects of melatonin and selenium in freezing extenders on frozen-thawed rat sperm. Methods: Semen samples were collected from 20 adult male Wistar albino rats. Following dilution, the samples were divided into six groups: four cryopreserved groups with 1 mM and 0.5 mM melatonin and selenium supplements, and two fresh and cryopreserved control groups. The rapid freezing technique was used to freeze the samples. Flow cytometry was used to assess plasma membrane integrity, mitochondrial membrane potential, and DNA damage, while computer-assisted sperm analysis was used to assess motility. Results: Total motility was higher in the 1 mM melatonin supplementation group than in the cryopreserved control group (mean±standard error of the mean, 69.89±3.05 vs. 59.21±1.31; p≤0.05). The group with 1 mM selenium had the highest plasma membrane integrity (42.35%±1.01%). The cryopreserved group with 0.5 mM selenium had the highest mitochondrial membrane potential, whereas the cryopreserved control group had the lowest (45.92%±4.53% and 39.45%±3.52%, respectively). Conclusion: Cryopreservation of rat semen supplemented with 1 mM melatonin increased sperm motility after freeze-thawing, while supplementation with 0.5 mM selenium increased mitochondrial activity.

Comparison various level ascorbic acid and lycopene additions in semen diluent enhanced sperm quality of Sapudi ram

  • The primary cause of sperm quality decline during the freeze-thaw pathway is the peroxidation hazard caused by reactive oxygen species produced by the biological molecules of sperm. Ascorbic acid (Vitamin C) and lycopene are two potent antioxidants that operate to prevent oxidation processes. This study aimed to analyse the effects of ascorbic acid and lycopene on the motility, viability, abnormality and plasma membrane integrity of post-thawed Sapudi rams. Sperm samples were obtained and pooled from six sexually mature Sapudi rams, separated into ten equal proportions and diluted with Tris-egg yolk-glycerol (TEY) extender. Semen was supplemented with 0 (C0; L0), 1 (C1; L1), 2 (C2; L2), 3 (C3; L3) and 4 (C4; L4) mg/100 mL (1%-4%) diluent each of ascorbic acid and lycopene, respectively. Total sperm motility, viability, abnormalities and semen membrane plasma (%) were analysed after thawing. C3 and L3 extenders resulted in higher total motility (p < 0.05) compared to the other extenders, with all treatments higher than that of the control. The extender C3 (p < 0.05) exhibited the highest semen quality. Finally, the current findings show that C3 and L3 can increase the quality of post-thawed Sapudi ram spermatozoa.

Motility, Fertilizability and Subsequent Embryonic Development of Frozen-thawed Spermatozoa derived from Epididymis in Hanwoo

  • The aim of the study was to investigate the ability of sperm derived from the epididymis in regard to sperm motility, sperm penetration to oocyte and subsequent development of the embryo. Frozen-thawed sperm from epididymis showed similar percentage of motile sperm (VSL ${\geq}25{\mu}m/sec$) as compared to that of commercial sperm (control). Sperm penetration of frozen-thawed epididymal and commercial sperm was not significantly different. Moreover, cleavage and blastocyst rates were similar in both epididymal and control. Sperm derived from the epididymis also showed fertilizability and subsequent embryonic development.

동결보존액의 종류와 동결방법에 따른 해동후 인간정자 운동성의 비교분석 (Effect of Cryopreservation Medium and Freezing Method on Post-thaw Motility of Human Sperm : Comparison of Different Type of CASA)

  • 본 연구에서는 보다 효율적인 동결 보존법을 수립하기 위하여 현재 사용되는 동결 보존액과 동결방법을 정자의 운동성 측면에서 비교해 보았다. 즉, 세 종류의 조성이 다른 동결보존액인 TYB, dithiothreitol을 첨가한 TYB+DTT, KS II 등이 동결보존 전후에 있어 운동성에 미치는 영향을 조사하였으며, 또한 vapor freezing 방법과 computerized freezer를 사용한 동결방법이 정자 운동성에 미치는 영향을 알아보았다. 정자의 분석은 현미경적 방법과 두 종류의 컴퓨터 정자 자동측정기인 SAIS와 Hamilton Thorn을 사용하여 동결 전 후의 정자 운동성과 VCL, VSL, VAP, ALH, LIN 등의 sub-motility 패턴을 측정하였다. 정액성상이 정상인 군에서 동결보존액을 비교한 실험결과는 TYB군과 TYB+DTT군, 그리고 KS II군의 융해 후 운동성이 각각 28.3%, 23.0%, 34.8%로 KS II군이 우수하였고, 동결방법을 비교한 실험에서는 vapor freezing 군과 computerized freezing 군의 융해 후 정자 운동성이 각각 27.8%, 33.2%로 유의차는 없었다. 또한 무력정자증을 보인 정액군에서는 TYB군과 TYB+DTT군, 그리고 KS II군에서 융해후 정자 운동성이 각각 13.6%, 10.0%, 18.5%로 여기 KS II군이 우수하였으며, vapor freezing군과 computerized freezing군의 융해 후 정자 운동성은 12.8%, 12.9%로 유의차가 없었다. 이상의 결과로 보아 운동성이 정상인 정액군과 무력정자증을 보이는 정액군에서 KS II를 사용해 동결하는 것이 TYB나 TYB+DTT를 사용하는 것보다 운동성 있는 정자를 회수하는데 더 효율적이며, 동결방법 측면에서는 vapor freezing 방법과 computerized freezing방법간에 큰 차이가 없음을 볼 수 있었다.
