A Study on the Ultrastructure of Reproductive Organ of Korean Planaria (Dugesia japonica) (한국산(韓國産) 플라나리아(Dugesia japonica Ichikawa et Kawakatsu)생식기관(生殖器官)의 미세구조(微細構造)에 관(關)한 연구(硏究))

  • The morphological study on different types of cells of reproductive organ including spermatogenesis in the adult planaria was performed to observe their cytochemical and ultrastructural characteristics. 1. Spermatogenesis The circular luminated material appears immediately inside the nuclear envelope of early spermatid and is found also in the nucleus of sperm, but typical acrosomal structures cannot be observed. Approximately ten of small-sized mitochondria occur around the nucleus in the transitional phase from primary spermatocyte to secondary spermatocyte, but in sperm a long mitochondrion is closely associated with nucleus, parellel to long axis of it. The sperm has a relatively long head connected with two tails via hollow neck. 2. Reproductive organ The penis bulb and the bursa stalk were observed. (1) Penis bulb The cells constituted penis bulb are classified into six types on the basis of ultrastructure of the cells and cytochemistry of the cytoplasmic granules. 1) A-type cells: These cells exhibiting low electron density are mainly occupied by large nucleus. These cells possess two different types of granules: highly electron-dense round granules with an average size of $0.9{\mu}m$, and electron-dense granules exhibit PAS-positive reaction. 2) B-type cells contain PAS-positive granules with the size of about $0.4{\mu}m$. They are rich in free ribosomes and mitochondria. 3) C-type cells are found to be dark cells due to high electron-density. These cells are largely occupied by large nucleus. 4) D-type cells: These cells are seen as light cells which have poorly developed cell organelles. 5) E-type tells: These cells contain a large number of glycogen granules which occupy most of cell. 6) F-type cells: These arc parietal epidermal cells surrounding the genital antrum. These cells are characterized by their finger-like shapes and the presence of a number of electron-dense, irregularly-shaped structures inside cells. The relatively large electron-lucent granules can be also found. The F-type cells possess numerous microvilli on their free surfaces. (2) Bursa stalk The cells constituted bursa stalk are classified into 3 types on the basis of cell shapes and presences of electron-dense or electron-lucent granules. 7) G-type cells with a long cytoplasmic process. They have large nuclei and poorly developed cell organelles. 8) H-type cells: These cells are characterized by the presence of a long cytoplasmic process and relatively highly electron-dense cytoplasmic profile. They have poorly developed cell organelles. 9) I-type cells contain large electron-lucent granules which exhibit negative reactions with three kinds of cytochemical staining methods used in this experiment. The fine electron-dense structures can be found inside these granules.

Suppression of ASKβ(AtSK32), a Clade III Arabidopsis GSK3, Leads to the Pollen Defect during Late Pollen Development

  • Arabidopsis Shaggy-like protein kinases (ASKs) are Arabidopsis thaliana homologs of glycogen synthase kinase 3/SHAGGY-like kinases (GSK3/SGG), which are comprised of 10 genes with diverse functions. To dissect the function of $ASK{\beta}$ (AtSK32), $ASK{\beta}$ antisense transgenic plants were generated, revealing the effects of $ASK{\beta}$ down-regulation in Arabidopsis. Suppression of $ASK{\beta}$ expression specifically interfered with pollen development and fertility without altering the plants' vegetative phenotypes, which differed from the phenotypes reported for Arabidopsis plants defective in other ASK members. The strength of these phenotypes showed an inverse correlation with the expression levels of $ASK{\beta}$ and its co-expressed genes. In the aborted pollen of $ASK{\beta}$ antisense plants, loss of nuclei and shrunken cytoplasm began to appear at the bicellular stage of microgametogenesis. The in silico analysis of promoter and the expression characteristics implicate $ASK{\beta}$ is associated with the expression of genes known to be involved in sperm cell differentiation. We speculate that $ASK{\beta}$ indirectly affects the transcription of its co-expressed genes through the phosphorylation of its target proteins during late pollen development.

A Study on Abdominal Temperature of Male Infertility Patients (남성 불임 환자의 하복부 온도에 관한 임상적 고찰)

  • Purpose : To examine the relation between male infertility and cold hypersensitivity on lower abdomen. Methods : From 2004, 4 to 2005, 10, a total 60(infertile male 30, normal male on semen analysis 30) patients who visited the Oriental gynecological department of Conmaul Oriental Medicine hospital was selected, and their medical records were reviewed retrospectively. We compared the temperature of 3 points(CV17, CV12, CV4) of both group by DITI(Dorex, DITI-16UTI and DITI-Spectrum 9000 MB). Results and Conclusion : The general characteristics such as age, weight, height, BMI, amount and frequency of drinking and smoking of both group were not different statistically. The mean ${\Delta}T1$ between upper and lower abdomen in infertile group was higer than in normal group but they were not different statistically. The mean ${\Delta}T2$ between chest and lower abdomen was $0.46{\pm}0.41$ in infertile group, and $0.18{\pm}0.56$ in normal group, and it shows significant difference statistically. There was no significant relationship between sperm motility and ${\Delta}T1$, ${\Delta}T2$ in infertile group. In this study, we suggest that an inclination of cold hypersensitivity of lower abdomen of infertile male, but it is expressed when it is contrasted to chest of the body rather than upper abdomen.

Establishment of Cryopreservation of Leopard Cat Semen Collected by Electro-ejaculation Method

  • The aim of this study was to evaluate the post-thawed characteristics of leopard cat semen. In this experiment, semen was collected from two leopard cats (A and B) at wild animal center in Seoul Grand Park in Korea. After collection, the sperms were washed with D-PBS and diluted by the freezing medium (Irvine science, USA) and stored in liquid nitrogen. The post-thawed concentration was $357{\times}10^6sperms/ml$ for A and $97{\times}10^6sperms/ml$ for B. The viability of post-thawed sperm from A and B individual was 24.0% and 19.0%, respectively. Pre-freezing motility of A and B individual semen was 68.54% and 56.65. Leopard cat A had more normal sperm than that of B (69.5% vs. 54.5%). Acrosome integrity analysis detected live (14.5% vs. 9.0%), damage (39.0% vs. 44.0%) and dead (46.0% vs. 47.0%) in leopard cat A and B, respectively. The present results concluded that leopard cat semen can be collected successfully by electro-ejaculation method and cryopreserved successfullyfor future use in different assisted reproductive technologies. The cryopreservation protocol needs to be modified for increasing post-thawed viability of leopard cat spermatozoa.

Association of Polymorphisms in Epidermal Growth Factor, Prostaglandin-endoperoxide Synthase 2 and Prolactin Receptor Genes with Semen Quality in Duroc Boars

  • The quality characteristics of semen are important indicators of the fertility of a boar. Development of genetic markers for the semen quality in boars will be beneficial to the improvement of porcine fertility. We investigated the relationship between the polymorphisms of epidermal growth factor (EGF), prostaglandin-endoperoxide synthase 2 (PTGS2) and prolactin receptor (PRLR) genes, and semen quality traits in boars. The genomic DNA of 233 boars (157 Duroc and 86 Landrace) from a central testing station was subjected to genotyping for surveying gene frequency. The EGF, PTGS2 and PRLR genotypes were determined using the restriction fragment length polymorphism method. Thirty-seven normal, mature Duroc boars from an AI center were also genotyped and their semen quality traits were collected. The effect of genotype on semen quality traits was analyzed by the least-squares means method using data corrected for season. The frequencies of the AA genotype of EGF, PTGS2 and PRLR in Duroc boars were 0.14, 0.01 and 0.66, respectively. In Landrace, the frequencies of the AA genotype were 0.03, 0.09 and 0.62, respectively. Boars with the BB genotype in EGF, with the AB genotype in PTGS2 and with the AA genotype in PRLR had significantly better semen quality with a higher percentage of normal sperm and a lower percentage of immature sperm than those with other genotypes. These findings imply that polymorphisms of EGF, PTGS2 and PRLR genes might be used as markers for improving the semen quality of boars.

  • To investigate the spermiogenesis of the Saghalien Pygmy shrew (Sorex minutus gracillimus), the testis obtained from mature male shrew was studied by electron microscopy, and the following results obtained based on the morphological characteristics of cell differentiation of the seminiferous epithelium in the testis. According to the fine structural differentiation, spermiogenesis of S. minutus gracillimks was divided into Golgi, cap, acrosome, maturation and spermiation phases. Beside, the Golgi and cap phases were subdivided into three steps of early, middle and late phase respectively, and acrosome phase into two steps of early and late phase , and maturation and spermiation phases has only one step respectively. Thus, the spermiogenesis of S. minutus gracillimus was divided into a total of ten steps. The chromatin granules begin to be condensed in the acrosome phase, and a perfect nucleus of sperm was formed at the spermiation phase. Mancette were appeared from the late acrosome phase to the maturation phase. The formation of sperm tail began to develop in the late Golgi phase, and completed at the spermiation phase. Multivesicular bodies were appeared from the Golgi phase to the maturation phase, recognized with pale, pale and moderate, and dense at Golgi, cap and acrosomal and matulation phases respectively.

  • Objectives : There has been little known about the Tibetan medicine in the society for Korean medicine. The aim of this study is to review the system of Tibetan medicine and compare with Korean medicine. Methods : The authors investigated several literatures that mentioned Tibetan medicine and organized in physiology, pathology, diagnostics, and treatment. And then, we interpreted the characteristics of Tibetan medicine as well as compared Tibetan medicine with Korean medicine. After that, we analyzed the commons and the differences, and also found out the meaning of Tibetan medicine. Results : The theory of Tibetan medicine is basically constituted of three elements of Lung, Tripa, and Peken in every parts of physiology, pathology, diagnostics, and treatment. Many organs of human body are compared to the fabrications of building. There is a detail explanation about the process of the development of fetus in Tibetan medicine. Tibetan medicine uses taking pulse in wrist, analysis of urine, watching of tongue, sperm, and menstruation, and etc. for diagnosis. In Tibetan medicine, regimen is prior to other treatments such as surgical treatment and medications. Conclusions : There is the oriental thought of '3' in Tibetan medicine, and esoteric buddhism is solved in Tibetan medicine. The anatomy and the diagnostics using urine, sperm, and menstruation have especially been developed in Tibetan medicine. Tibetan medicine emphasizes the feature of preventive medicine.

Ultrastructure of Germ Cell during Spermatogenesis and the Reproductive Cycle of the Hanging Cultured Male Scallop Patinopecten yessoensis (Pelecypoda:Pectinidae) on the East Coast of Korea

  • Ultrastructure of germ cell differentiation during supermatogenesis and the reproductive cycle in male Patinopecten yessoensis was studied by histological and cytological observations. The gonadosomatic index (GSI) in males rapidly increased and reached a maximum in April when seawater temperature gradually increased. Then the GSI gradually decreased from May through July when spawning occurred. Accordingly, monthly changes in the GSI in males coincided with testicular maturation and spawning periods. The sperm morphology of P. yessoensis belongs to the primitive type and showed general characteristics of external fertilization species. The head of the spermatozoon is approximately $3.50{\mu}m$ in length: the sperm nucleus and acrosome are approximately $2.90{\mu}m\;and\;0.60{\mu}m$ in length, respectively. The nuclear type of the spermatozoon is vase in shape, and the acrosome is cone type. The axoneme of the tail flagellum consists of nine pairs of microtubules at the periphery and a pair of central microtubules in the center The satellite body (which is formed by the centriole) and four mitochondria appear in the middle piece of the spermatozoon. The spawning period was from April through July and the main spawning occurred from May to June when seawater temperatures gradually increased. The reproductive cycle of this species can be classified into five successive stages; early active stage (September to November), late active stage (October to March), ripe stage (February to August), spawning stage (April to July), and spent/inactive stage (July to November).

Effect of Optixcell and Triladyl extenders on frozen-thawed sperm motilities and calving rates following artificial insemination in Hanwoo

  • In this study, we examined the effect of a liposome-based extender (Optixcell) and a tris-citric egg-yolk extender (Triladyl) on the frozen-thawed spermatozoa characteristics and the calving rate. The percentages for the total motility of the frozen-thawed spermatozoa were similar in the Optixcell and Triladyl groups. However, among the motile spermatozoa with a straight line velocity (VSL) ${\geq}25{\mu}m/sec$, the curvilinear velocity (VCL, ${\mu}m/sec$), VSL (${\mu}m/sec$), average path velocity (VAP, ${\mu}m/sec$), amplitude of lateral head displacement (ALH, ${\mu}m$), beat cross frequency (BCF, Hz), and plasma membrane integrity of the frozen-thawed spermatozoa for the Optixcell group were significantly higher than those for the Triladyl group. Furthermore, the calving rate in the Optixcell group (79.0%) was higher than that of the Triladyl group (62.8%). However, the acrosomal membrane integrity of the frozen-thawed spermatozoa in the Optixcell and Triladyl groups was not significantly different. These results indicate that semen freezing with Optixcell improved the motility and plasma membrane integrity of frozen-thawed spermatozoa and the calving rate of Hanwoo cows (native Korean cattle). In conclusion, our results suggest that semen freezing with the liposome-based extender Optixcell is more efficient than with the tris-citric egg-yolk extender Triladyl for improved offspring production.

Caffeine attenuates spermatogenic disorders in mice with induced chronic scrotal hyperthermia

  • Objective: Chronic scrotal hyperthermia (SHT) can lead to serious disorders of the male reproductive system, with oxidative stress playing a key role in the onset of these dysfunctions. Thus, we evaluated the impact of caffeine, a potent antioxidant, on cellular and tissue disorders in mice with chronic SHT. Methods: In this experimental study, 56 adult male NMRI mice were allocated into seven equal groups. Apart from the non-treated control group, all were exposed to heat stress. Two groups, termed "preventive" and "curative," were orally administered caffeine. The preventive mice began receiving caffeine immediately prior to heat exposure, while for the curative group, a caffeine regimen was initiated 15 consecutive days following cessation of heat exposure. Each treated group was subdivided based on pairing with a positive control (Pre/ curative [Cur]+PC) or a vehicle (Pre/Cur+vehicle). Upon conclusion of the study, we assessed sperm characteristics, testosterone levels, stereological parameters, apoptosis, antioxidant and oxidant levels, and molecular markers. Results: Sperm parameters, testosterone levels, stereological parameters, biochemical factors (excluding malondialdehyde [MDA]), and c-kit gene expression were significantly elevated in the preventive and curative groups, especially the former, relative to the other groups. Conversely, expression levels of the heat shock protein 72 (HSP72) and nuclear factor kappa beta (NF-κβ) genes, MDA levels, and apoptotic cell density were markedly lower in both caffeine-treated groups relative to the other groups, with more pronounced differences observed in the preventive group. Conclusion: Overall, caffeine attenuated cellular and molecular abnormalities induced by heat stress in the testis, particularly in the mice treated under the preventive condition.
