The objectives of this research are to investigate coagulation efficiencies of two coagulants l.e., alum and polyaluminum chloride and to understand effects of the coagulants on membrane fouling in microfiltration. The turbidity of supernatant from alum coagulation was increased with increasing doses whereas the turbidity from PACl coagulation was maintained at the low values. The observed injection volume of PACl for the same removal was approximately 30 percent less than alum, which produced a low sludge volume. The settling velocity of PACl flocs was greater than alum flocs. The results corresponded well with floc size measurements. Flux decline from alum coagulation was significant due in part to small sizes of flocs. At the low dose, alum floc had less specific cake resistance than PACl floc. However, as the dosage was increased, the increases in specific cake resistances of alum was substantial. Alum coagulation pretreatment needs careful operation to reduce membrane fouling by flocs. In general, PACl coagulants were more effective than alum coagulants for pretreatment of membrane processes because PACl showed the better performance in coagulation and membrane fouling.
Many reservoirs in Korea and their downstream environments are under increased pressure for water utilization and ecosystem management from longer discharge of turbid flood runoff compared to a natural river system. Turbidity($C_T$) is an indirect measurement of water 'cloudiness' and has been widely used as an important indicator of water quality and environmental "health". However, $C_T$ modeling studies have been rare due to lack of experimental data that are necessary for model validation. The objective of this study is to validate a coupled three-dimensional(3D) hydrodynamic and particle dynamics model (ELCOM-CAEDYM) for the simulation of turbid density flows in stratified Daecheong Reservoir using extensive field data. Three different groups of suspended solids (SS) classified by the particle size were used as model state variables, and their site-specific SS-$C_T$ relationships were used for the conversion between field measurements ($C_T$) and state variables (SS). The simulation results were validated by comparing vertical profiles of temperature and turbidity measured at monitoring stations of Haenam(R3) and Dam(R4) in 2004. The model showed good performance in reproducing the reservoir thermal structure and propagation of stream density flow, and the magnitude and distribution of turbidity in the reservoir were consistent with the field data. The 3D model and turbidity modeling framework suggested in this study can be used as a supportive tool for the best management of turbidity flow in other reservoirs that have similar turbidity problems.

In this paper, oyster shell, diatomaceous earth, and active carbon were used as filter media for treating wastewater produced in paper mills. After filtering, the changes of COD and turbidity were investigated. As the results of estimating the efficiency of wasterwater treatment, porous oyster shell having higher specific surface area in powder was more effective than the others in removal of contaminants in waterwater, especially turbidity.
Cryptosporidium can cause gastrointestinal diseases worldwide, consequently posing public health problems and economic burden. Effective techniques for detecting contaminated oocysts in water are important to prevent and control the contamination. Immunomagnetic separation (IMS) method has been widely employed recently due to its efficiency, but, it is costly. Sucrose floatation technique is generally used for separating organisms by using their different specific gravity. It is effective and cheap but time consuming as well as requiring highly skilled personnel. Water turbidity and parasite load in water sample are additional factors affecting to the recovery rate of those 2 methods. We compared the efficiency of IMS and sucrose floatation methods to recover the spiked Cryptosporidium oocysts in various turbidity water samples. Cryptosporidium oocysts concentration at 1, $10^1$, $10^2$, and $10^3$ per $10{\mu}l$ were spiked into 3 sets of 10 ml-water turbidity (5, 50, and 500 NTU). The recovery rate of the 2 methods was not different. Oocyst load at the concentration < $10^2$ per 10 ml yielded unreliable results. Water turbidity at 500 NTU decreased the recovery rate of both techniques. The combination of sucrose floatation and immunofluorescense assay techniques (SF-FA) showed higher recovery rate than IMS and immunofluorescense assay (IMS-FA). We used this SF-FA to detect Cryptosporidium and Giardia from the river water samples and found 9 and 19 out of 30 (30% and 63.3%) positive, respectively. Our results favored sucrose floatation technique enhanced with immunofluorescense assay for detecting contaminated protozoa in water samples in general laboratories and in the real practical setting.

In this study, we investigated the antimicrobial effects of anti-Salmonella gallinarum-specific IgY separated from egg yolk obtained from layers immunized by S. gallinarum. The comparison tests of vaccination, content of IgY and innoculation number were examined by microscopic observation, turbidity, and pH. The results show that the ratio of anti-S. gallinarum IgY in the total IgY was 23%. Also, the anti-S. gallinarum IgY had selectivity only to S, gallinarum. The 0.3% addition of anti-S. gallinarum-specific IgY resulted in agglutinating clusters of S, gallinarum cells, but the agglomeration didn#t occur in IgY from layers not immunized nor in the control group. Microscopic observation indicated agglomerative cells when IgY was added at 0.2% or higher, and the pH and turbidity examinations revealed that a suppression effect was remarkable in IgY at more than 0.1%. These results suggest the possibility that IgY extracted from eggs and obtained from layers immunized by S. gallinarum can be used to prevent fowl typhoid.
Two step rapid filter system as a pre-treatment for the injected water into aquifer storage and recovery (ASR) in Korea was developed to reduce physical blockage and secure the volume of the injected water. First, single rapid sand filters with three different media sizes (0.4~0.7, 0.7~1.0 and 1.0~1.4 mm) were tested. Only two sizes (0.4~0.7 and 0.7~1.0 mm) satisfied target turbidity, below 1.0 NTU. However, they showed the fast head loss. To prevent the fast head loss and secure the volume of the injected water, a rapid anthracite filter with roughing media size (2.0~3.4 mm) were installed before a single rapid sand filter. As results, both the target turbidity and reduction of head loss were achieved. It was determined that the media size for a rapid sand filter in two step rapid filter system (i.e. a rapid anthracite filter before a rapid sand filter) was 0.7~1.0 mm. In addition, the effects of coagulant doses on the removal of natural organic matter (NOM), which might cause a biological clogging, were preliminarily evaluated, and the values of $UV_{254}$, dissolved organic carbon (DOC) and SUVA were interpreted.
Since polyelectrolytes have been used as conditioners, conventionally only a single polyelectrolyte has been added for sludge conditioning. However, the amount of polyelectrolyte needed for optimal conditioning of sludge is very critical. Overdosing reduces the sludge dewaterbility. In this experimental study, sludge conditioning with single or dual polyelectrolyte was conducted to avoid problems associated with overdosing. Single polyelectrolyte conditioning was conducted by one of cationic, nonionic, and anionic polyelectrolytes. The dual polyelectrolyte conditioning was performed by adding one polyelectrolyte and another one in sequence. The dewaterbility of sludges were measured by SRF(specific resistance to filtration), TTF(time to filter), CST(capillary suction time) respectively. Additionally, parameters such as turbidity, zeta potential, viscosity of conditioned sludges or supematant were measured to evaluate the changed characteristics of sludge by addition of polyelectrolytes. From the experiment results, it was concluded that single polyelectrolyte conditioning had a high probability of overdosing, whereas dual polyelectrolyte conditioning resulted in a better dewaterbility and less chance of overdosing. But, it was also found that dosing sequence in dual conditioning was very important according to the characteristics of sludge. Parameters such as viscosity, turbidity, zeta potential were found to be useful as a means of evaluating sludge dewaterbility.
Several contaminated bacteria such as Lactobacillus brevis and Pediococcus damnosus in beer production cause beer spoilage by producing off flavours and turbidity. Detection of these organisms is complicated by the strict anaerobic conditions and lengthy incubation times required for their cultivation, consequently there is a need for more rapid detection methods. Recently, two contaminated strains were isolated from vessel of beer production and identified as Lactobacillus species by API kit identificaton as well as 16S-23S ITS sequencing analyses. Two isolated strains were named as Lactobacillus sp. HLA1 and Lactobacillus HLB2, respectively. A polymerase chain reaction (PCR) method was developed for the rapid and specific detection of Lactobacillus sp.. Two sets of primer pairs (HLA1-F/HLA1-R and HLB2-F/HLB2-R) were designed for the amplification of a 1576 base pair (bp) fragment of the HLA1 16S-23S rRNA gene and 1888 bp fragement of the HLB2 16S-23S rRNA. Amplified PCR products were highly specific to detect corresponding bacteria when other contaminated strains were used as PCR templates. However, detection of both strains were limited when $100{\mu}{\ell}$ of cultured samples were mixed with $100m{\ell}$ of beer sample in arbitrary manner. The sensitivity of the assay still needs to be improved for direct detection of the small amounts of bacteria present in beer.
This study was carried to investigate the effect of humic acid and kaloin which cause the turbidity and organic substance component for optimization of drinking water treatment process using the membrane separation. Also we were ovserved the optimum operating condition which flux was stabilized, while specific resistance value in membrane was minimized. As the result, the membrane separation was operated at low specific resistance value with the increase of the pressure. And then, cake load decreased by high velocity with the increase of the linear velocity, and the tendency in which specific resistance value and flux increased. Therefore, we confirmed the optimum operating condition as pressure $2.0 kgf/cm^2,$ 0.92 m/sec linear velocity.
To understand the effects of the powdered manganese nodule and sea bottom sediment pumped up with nodules on the mining process, the shattering ratio of manganese nodule and their physical properties are analyzed. The self shattering ratio and crushing shattering ratio are about 27% and about 3%, respectively. Then total shattering ratio is about 30%. The initial turbidity of the powdered manganese nodule and the bottom sediment show high, i.e., about 3,100 and 1,850 respectively. But their turbidities decrease rapidly with time. After 1 hour, turbidity of the powdered manganese nodule drops to about 1,570 and that of the bottom sediment to 1,310. The turbidity of Na-bentonite changes from 820 to 730 after 1 h and to 700 after 2 h. The viscosity of powdered manganese nodule is $1.4{\sim}1.5cP$, and the viscosity of bottom sediment is less than 1 cP. The viscosity fo Na-bentonite is initially 37.2 and increase with time to 86.4 cP after 30 min. The high initial turbidity of powdered manganese nodule is due to dark color of the powder. The high specific gravity makes rapid precipitation and then decreases the turbidity rapidly. The bottom sediment shows high initial turbidity because of easy suspension with very fine particle size. But it cannot be hydrated and formed gel in suspension, then it is easily precipitated. However Na-bentonite is hydrated to the expended state and makes gel state, then it shows high turbidity and high viscosity. These physical properties of the powdered manganese nodule suggest that the powder of manganese nodule should not make scaling inside of lifting pipe or pump. And the bottom sediment lifted up with manganese nodule should not play the role of drilling mud shch as Na-bentonite.
