슬러지의 탈수성(脫水性)에 대한 농도(濃度)의 영향(影響)과 케이크 폐색현상(閉塞現象)의 해석(解析) (Effect of sludge concentration on sludge dewaterability and cake clogging analysis)

  • Although the specific resistance to filtration is the most frequently employed means for characterizing dewaterability of a sludge, it presently is not possible to design nor to prediet performance of dewatering facilities using traditional linearized parabolic filtration equation, that is, the specific resistance model because of theoretical and practical inadequacies of the concept. Limitations of the specific resistance model reflect the need to examine fundamental sludge properties and filtration behaviors affecting dewaterability. From this study, two major limitations of the specific resistance model were noted. First, specific resistance values are very dependent on the sludge concentration because of the variations of particle size distribution and cake clogging to occur when surface area mean diameter is less than $25{\mu}m$ for activated sludge, $18{\mu}m$ for water treatment plant sludge. Second, nonparabolic filtration behavior can result from cake clogging, caused by the migration of fine particles into the cake pores, accelated by skin effect with highly compressible sludges.

Biological Control of Powdery Mildew by a Hyperparasite, Ampelomyces quisqualis 94103: From isolation to a commercial biofungicide product.

  • Total 291 isolates of Ampelomyces quisqualis were obtained from 32 species of powdery mildew fungi and a selected isolate, Ampelomyces quisqualis 94013 (AQ94013) effectively hyperparasitized 6 species of Sphaerotheca and one species of Erysiphe which cause serious damage on many important crops in Korea. Moreover, AQ94013 showed antagonistic effects against 12 major fungal plant pathogens as well. Results indicated that the present isolate is not a host specific hyperparasite and has a broad spectrum of biocontrol potential. Providentially, AQ94013 revealed resistance to a number of agrochemicals so as to be applied with the chemicals reciprocally.(중략)

  • In spite of the development of highly sophisticated instrument, the precise quantitation of plant hormones still has many difficulties. Due to their high specificity, sensitivity and minimal sample purification steps, immunological assays have been widely applied for plant hormone assay. Enzme-linked immunosorbent assay technique for the determination of plant hormones was developed by Voller in 1978. Immunological assays are accomplished by competition of labeled tracer antigen and unlabeled antigen for a limited number of specific antibodies. The use of enzyme as replacement labels for radioisotopes enabled much of the sensitivity and specificity of radioimmunoassay (RIA) to be retained but without the inherent disadvantage of high capital cost, potential health hazard, and short shelf life of the labeled reactants.

  • Applying digital network technology for advanced nuclear plant requires deterministic communication for tight safety requirements, timely and reliable data delivery for operation-critical and mission-critical characteristics of nuclear plant. Communication protocols, such as IEEE 802/4 Token Bus, IEEE 802/5 Token Ring, FDDI, and ARCnet, which have deterministic communication capability are partially applied to several nuclear power plants. Although digital communication technologies have many advantages, it is necessary to consider the noise immunity from electromagnetic interference (EMI), electrical interference, impulse noise, and heat noise before selecting specific digital network technology for nuclear plant. In this paper, we consider the token frame loss and data frame loss rate due to the link error event, frame size, and link data rate in different protocols, and evaluate the possibility of failure to meet the hard real-time requirement in nuclear plant.

Marker Genes for in Vitro Selection of Transgenic Plants

  • The use of a marker gene in a transformation process aims to give a selective advantage to the transformed cells, allowing them to grow faster and better, and to kill the non-transformed cells. In general, the selective gene is introduced into plant genome along with the genes of interest. In some cases, the marker gene can be the gene of interest that will confer an agronomic characteristic, such as herbicide resistance. In this review we list and discuss the use of the most common selective marker genes on plant transformation and the effects of their respective selective agents. These genes could be divided in categories according their mode of action: genes that confer resistance to antibiotics and herbicides; and genes for positive selection. The contention of the marker gene flow through chloroplast transformation is further discussed. Moreover, strategies to recover marker-free transgenic plants, involving multi-auto-transformation (MAT), co-transformation, site specific recombination and intragenomic relocation of transgenes through transposable elements, are also reviewed.

안동오이 대목을 이용한 수박 덩굴쪼김병 방제 (Control of Fusarium Wilt of Watermelon with the Root-Stock Grafting of Sicyos angulatus L.)

  • Watermelon plants grafted with the root-stock of wild-cucumber (Sicyos angulatus) were not infected by Fusarium oxysporum f.sp. niveum in pot inoculation and infected fields tests. Controlling efficacy of the root-stock grafting with S. angulatus on Fusarium wilt of watermelon was more excellent than that of the root-stock grafting with Lagenaria siceraria. The isolates of Fusarium oxysprum from cucurbitaceae plants had a certain host-specific pathogenicity, but they did not express the absolute one forma specialis-one host-plant phenomenon by the root dipping inoculation. The pathogenic isolates of Fusarium oxysproum from cucurbitaceae crops did not infect the root-stock plant such as S. angulatus, L. siceraria and Cucurbita ficifolia. The fast-wilting of watermelon caused by uncertain agents was observed in watermelon plant grafted with L. siceraria in the continuously cropping fields, but it was not observed in watermelon plants grafted with S. angulatus in the same fields.

Rhizobacterial Exopolysaccharides Elicit Induced Resistance on Cucumber

  • The role of exopolysaccharides (EPSs) from a plant growth-promoting rhizobacterium, Burkholderia gladioli IN26, on elicitation of induced systemic resistance was investigated. A purified EPS induced expression of PR-1a::GUS on tobacco and elicited induced resistance against Colletotrichum orbiculare on cucumber. The maximum level of disease protection was noted when seeds were soaked in 200 ppm of the EPS. Our results indicate that EPS from specific rhizobacteria can elicit induced resistance and suggest that bacterial EPS might be a useful elicitor of resistance under field conditions.

Role of Siderophores in Biocontrol of Fusarium solani and Enhanced Growth Response of Bean by Pseudomonas fluorescens GL20

  • Plant growth-promoting Psudomonas fluorescens GL20 was isolated from a ginseng rhizosphere on chrome azurol Sagar. P. fluorescens GL20 produced a large amount of hydoxamate siderophore in an iron-deficient medium. The siderophore showed significantly high specific activity of 20.2 unit. Using an in vitro antifungal test, P. fluorescens GL20 considerably suppressed growth of phytopathogenic fungus Fusarium solani, inhibiting spore germination and germ tube elongation. In pot trials of kidney beans with P. fluorescens GL20, disease incidence was remarkably reduced up to $68{\%}$ compared with that of F. solani alone, and plant growth was also increased nearly 1.6 fold as compared to that of the untreated control, promoting elongation and development of the roots. These results indicate that the plant growth-promoting activity of P. fluorescens GL20 can play an important role in biological control of soil-borne plant disease in a rhizosphere, enhancing the growth of plants.

Genomics and Biotechnology of Plant Adaptation to the Environment

  • The molecular basis of plant adaptation to the environment is one of the most complex areas of plant science and an understanding of this subject is critical to our ability to feed an increasing word population. Research on plant adaptation to the environment often involves two complementary approaches; top-down studies of well-adapted plants with the goal of identifying and describing the biochemical basis of adaptation and bottom-up targeted analysis of specific biochemical mechanisms with the goal of understanding how these mechanisms contribute to overall plant performance. This brief review will provide examples of both of these approaches by describing the study of sorghum's adaptation to dry environments and the role of a blue light responsive chloroplast promoter that helps protect plants against damage by high irradiance.

  • Long Term Asset Management(LTAM) means a plan developed by using LCM(Life Cycle Management) process for optimum life cycle management of significant plant assets at each plant across the fleet. As a part of development of LTAM Strategies on nuclear turbines, a method so as to determine the future failure rates for low pressure turbine facilities at a nuclear plant was studied and developed by using both plant specific and industry-wide performance data. INPO's EPIX data were analyzed and some failure rate evaluation values considering preventive maintenance practices were calculated by using EPRI's PM Basis software. As the result, failure rate functions applicable to a priori and a posteriori replacement of low pressure turbines at a nuclear plant were developed and utilized in an assessment of economics of LCM alternatives on the nuclear turbine facilities in the respects of 40-year and 60-year operation bases.
