Identification of the Chicken Meat Allergens (닭고기 중 알레르기 유발성분의 동정)

  • The chicken meat has been reported as one of the food causing allergic reactions predominantly to Korean. At present, several in vitro tests for immunoglobulinG (IgG)-mediated as well as IgE-mediated food allergy are available. 13 clinically chicken meat-allergic patients were investigated together with 4control subjects for identification of chicken meat-specific reactivity by ELISA. Also, protein profile and IgE, IgGtotal and IgG4-reacting allergens were detected by means of sodium dodecyl sulfate-polyacrylamide gel electro-phoresis (SDS-PAGE)and immunoblotting. Chicken meat extracts were prepared as raw, heated, heat and simulated gastric fluid (SGF) treated samples to characterize the stability of allergen to physicochemical treatment. SDS-PAGE revealed 9~200 kDa bands. And in immunoblotting 7 sera were identified most major bands between 10 and 78 kDa. In case of IgE, six proteins (17, 26, 35, 40, 78 kDa) were predominant in heat-treated extract, and the one (35 kDa) was present in SGF-treated preparations. In case of IgG$_{total}$ and IgG4, most of them showed a patters simmilar to IgE. There were significant differences (P<0.05) in IgE, IgG$_{total}$ , IgG4 Abs to chicken meat between the allergic and control subjects in ELISA. In addition, the concentration of IgG4Abs in the challenge-positive subjects was significantly higher than that of control subjects. It is considered that the specific IgE response to chicken meat was rarely prevalent to Koreans. However, the specific IgG4 response play an important role in the development of allergic symptoms.

Effect of Diet Supplements on the Production of Anti-Dental Caries Hen's Eggs by Immunization of Streptococcus mutans (Streptococcus mutans 균의 면역과 사료첨가제가 충치 예방용 계란의 생산에 미치는 영향)

  • To increase IgY in egg yolks, hens were fed a feed supplemented with kelp meal $4\%$ cinnamon $0.3\%$ and mint $2\%$ respectively, and immunized 5 times with Streptococcus mutans(S. mutans) at 2 week intervals. Groups fed experimental feeds without immunization showed higher laying rate than the control group, without supplementary feed and immunization. After the immunization, the laying rates had been decreasing due to the stress of immunization. The laying rate was recovered after the termination of immunization. Egg weight was not affected by the immunization but diets. Feed intake was dependent on the laying rate. Total IgY concentration in eggs laid from hens fed feeds containing supplementary feeds was higher than that of control. Especially, total IgY was increased up to $7.9\%$ in eggs laid from hens fed feeds supplemented with $4\%$ of kelp meal. Anti-S. mutans IgY was detected at 4 weeks after first immunization. Activity of anti-S. mutans IgY was sustained at 5 week after the final immunization. As the average concentration of specific IgY during the experimental period showed that eggs from hens fed the feed containing $4\%$ of kelp meal increased the specific IgY by $8.5\%$ kelp meal supplement improved specific IgY production by immunization.

Induction of Rice Allergen-Specific IgE Synthesis by KU8l2 Cells (KU812세포에 의한 쌀 알레르겐 특이적 IgE항체 합성의 유도)

  • In vitro IgE class switching could be induced through co-culture of CD40L-expressing KU812 cells and CD40-expressing B cells in the presence of IL-4 or IL-13. It has been generated several B cell lines, which produce rice allergen (RA)-specific IgM antibody by in witγo immunization (IVI) using peripheral blood lymphocyte (PBL). In this study, induction of RA-specific IgE antibody by KU812 cells was attempted. Before co-culture, we determined the CD40 expression in RA-specific B cell lines, RA9G11 and the CD40 ligand (CD40L) expression in activated KU812 cells by treatments with phorbol myristate acetate (PMA) and ionomycin for 6 hrs. Flow cytometric analysis shown that RA9G11 and activated KU812 cells expressed high level of CD40 and CD40L, respectively. RA9G11 cells were cultured with activated KU812 cells for 12 days in the presence of IL-4 for IgE class switching. Mature $C{\varepsilon}$ mRNA level and RA-specific IgE spot forming cells (SFC) were observed in all culture condition, and especially, high level of RA-specific IgE synthesis was determined the same ratio of RA9G11 and activated KU812 cells in the presence of 50U IL-4. Therefore, induction of RA-specific IgE synthesis by activated KU812 cells can be contributed in the application for allergic therapy and prevention.

Measles Specific IgG in Adolescents (청소년기의 홍역 특이 IgG)

  • Purpose : To determine the age-specific seropositivity and the level of measles specific IgG in vaccinated adolescents, the measles specific IgG titers were measured from the sera of junior & senior high school students in Seoul, Korea. Methods : From May to July in 1996, 216 sera were collected from junior & senior high school students(male : 110, female : 106) aged 12 to 17 years who were vaccinated and had no history of natural measles. Measles specific IgG titers were measured using ELISA method($SIA^{TM}$ Measles IgG Kit, Sigma Co. St. Louis, Mo). Results : The seropositivity of measles specific IgG in vaccinated adolescents aged 12 to 17 years was 91.2%. There were no significant correlations between age and seropositivity as well as between age and measles specific IgG level. Conclusions : We consider that our data suggest the necessity of revaccination of measles in adolescents. Further considerations should be required to establish an appropriate revaccination method and to evaluate the efficacy of revaccination of previously vaccinated adolescents.

  • As the allergic diseases in patients are increasing, it is important to find out the allergens. A multiple antigen simultaneous test(MAST) is a simple method for in vitro measurement of allergen-specific IgE antibodies. This study was performed to evaluate the relationship between allergen-specific IgE antibodies, serum total IgE and peripheral eosinophil count in the allergic patients. According to the results of the study, the total IgE positive rate(above class 2) from the inhalent is 96.97%, and that from food panel is 98.06%. The research showed that the positive rate of the allergen-specific IgE was House dust 51.52%, D. farinae 45.46%, Cat 31.99% in inhalent panel, and 55.34%, 42.72%, 34.96% in food panel. Serum total IgE was associated with allergy, however, allergy was not always associated with eosinophilia.

Effect of Charcoal on the Production of Anti-Salmonella gallinarum IgY (Anti-Salmonella gallinarum Immunoglobulin 생산을 위한 계란의 이용)

  • In order to produce the antibody rich eggs against Salmonella gallinarum(S.G.) causing fowl typhoid, the productions of immunoglobulin in eggs were compared and examined with the feed additives, the variety of adjuvants in vaccines to layers, and the existence of additive antigens other than target microorganism. The examination of the average contents of specific IgY in immunized group by supplying hardwood charcoal showed that the group supplied with 0.5% hardwood charcoal had the highest contents, implying that the supply of hardwood charcoal promoted the production of specific IgY. Adjuvant appeared to have little effect on the average contents of total IgY, but specific IgY contents increased in the immunized group with Freund's adjuvant. Addition of BCG in adjuvant treatment increased specific IgY however, this feature was not seen in aluminum hydroxide treated group. Immunization at 15 week layers resulted in higher laying rate than immunization at 21 week and addition of hardwood charcoal in feed recovered laying rate. It was therefore, concluded that the feed supplement, such as hardwood charcoal followed by a proper immunization program concerning adjuvant, vaccination period and supplementary microorganism hastened the production of IgY.

House dust mite-specific immunoglobulin E and longitudinal exhaled nitric oxide measurements in children with atopic asthma

  • Purpose: House dust mite (HDM) has been suggested to be the most important aeroallergen responsible for atopic asthma in Korea. We aimed to investigate that specific IgE antibodies to HDM and other common indoor aeroallergens contribute differently to total serum IgE and show different relationships with longitudinal fractional exhaled nitric oxide (FeNO) measurements in Korean atopic asthmatic patients. Methods: A total of 193 children aged 8 to 16 years with intermittent or mild persistent atopic asthma were recruited. Sera were assayed for total IgE and specific IgE antibodies to HDM and other common indoor allergens. FeNO was serially measured 10 times or more over 2 years when subjects were not receiving controller medications. Results: In 152 children who completed the study, IgE antibodies to specific HDM were more prevalent than those to other common indoor aeroallergens. In addition, IgE antibody titers to HDM were the strongest contributor to total IgE increases. Furthermore, only HDM-specific IgE antibody titer significantly correlated with maximum FeNO (r=0.21, P=0.029) and the rate of FeNO higher than 21 parts per billion (ppb) (r=0.30, P=0.002). Eight patients (5%) were found to have maximum FeNO of 21 ppb or less, suggesting the presence of a low FeNO phenotype among atopic asthmatic patients. Conclusion: The quantity of HDM-specific IgE antibody provides a possible explanation for increases of total IgE and significantly correlates with the amount and frequency of FeNO increases in Korean atopic asthmatic patients.

Detection of Serum IgE Specific to Mite Allergens by Immuno-PCR

  • Background: Although a skin test is the primary option for detecting allergen-specific IgE in clinics, the serum IgE immunoassay is also important because it allows for the diagnosis of allergy without any accompanying adverse effect on the patient. However, the low detection limit of IgE levels by immunoassay may restrict the use of the method in some occasions, and improving its sensitivity would thus have a significant implication in allergy-immunology clinics. Methods: In this study, we attempted to detect specific serum IgE by using immuno-polymerase chain reaction (IPCR) which combines the antigen-antibody specificity of enzyme-linked immunosorbent assays (ELISAs) with the amplification power of PCR. Results: Our results demonstrated that Blo t5-specific serum IgE can be detected by IPCR with a 100-fold higher sensitivity than ELISA, and cross-reactivity of serum IgE to other mite allergens is able to be analyzed by using only $0.3{\mu}l$ of serum sample. Use of real-time IPCR seemed to permit more convenient determination of specific serum IgE as well. Conclusion: We believe that IPCR can serve as a valuable tool in determining specific serum IgE, especially when the amount of serum sample is limited.

Effects of Socheongryong-Tang on Immunoglobulin Production in Asthmatic Mice (소청용탕(小靑龍湯)이 천식(喘息)이 유발된 생쥐의 면역 글로불린 분비에 미치는 영향)

  • Objectives : Socheongryong-Tang(小靑龍湯, SCRT), a herbal remedy, has been widely used to treat respiratory disease such as cough and asthma in Oriental countries. Recent years SCRT was known as anti-allergic agent. However, its therapeutic mechanisms including immunoglobuline such as IgE, IgG1, IgG2a productions are unclear. Methods : We investigated the effects of SCRT on levels of antigen specific total antibody, IgE, IgG1, IgG2a using ELISA method in serum from allergen-induced asthma mice. Results : SCRT decreased level of antigen specific IgE significantly. And SCRT treated mice showed downward tendency of IgG1, a Th1 relative antibody, level. But, SCRT did not affect levels of antigen specific total antibody and IgG2a, a Th1 relative antibody. Conclusions : we demonstrated the strong possibility of SCRT as a complementary or alternative drug to western drug also demonstrated that regulation of Th1/Th2 imbalance may be one of mechanism contributed to treatment for respiratory disease by SCRT.

Egg Antibody Farming and IgY Technology for Food and Biomedical Applications

  • It has been recognized that the hen, like its mammalian counterparts, provides young chicks with antibodies as protection against hostile invaders. This system facilitates the transfer of specific antibodies from serum to egg yolk, and provides a supply of antibodies called immunoglobulin Y(IgY) to the developing embryo and the hatched chick. The protection against pathogens that the relatively immune-incompetent newly hatched chick has, is through transmission of antibodies from the mother via the egg. Egg yolk, therefore, can be loaded with a large amount of IgY against pathogens which can immobilize the existing or invading pathogens during the embryo development or in day-old chicks. Thus, the immunization of laying hens to various pathogens results in production of different antigen-specific IgY in eggs. Egg yolk contains 8∼20 mg of jmmunoglobulins (IgY) per ml or 136∼340 mg per yolk suggesting that more than 30 g of IgY can be obtained from one immunized hen in a year. By immunizing laying hens with antigens and collecting IgY from egg yolk, low cost antibodies at less than $10 per g compared to more than $20,000 per g of mammalian IgG can be obtained. This IgY technology opens new potential market applications in medicine, public health, veterinary medicine and food safety. A broader use of IgY technology could be applied as biological or diagnostic tool, nutraceutical or functional food development, oral-supplementation for prophylaxis, and as pathogen-specific antimicrobial agents for infectious disease control. This paper has emphasized that when IgY-loaded chicken eggs are produced and consumed, the specific antibody binds, immobilizes and consequently reduces or inhibits the growth or colony forming abilities of microbial pathogens. This concept could serve as an alternative agent to replace the use of antibiotics, since today, more and more antibiotics are less effective in the treatment of infections, due to the emergence of drug-resistant bacteria.
