The therapeutic effect of Drynariae Rhizoma in a mouse model of allergic asthma (천식 모델 마우스에서 골쇄보의 항천식 효과)

  • Objective : Allergic asthma is a chronic airway disease that affects millions of people in the developed world. The disease is characterized by concurring airway inflammation, Th2 cytokine production, increased mucus secretion, airway hyperresponsiveness (AHR) to inhaled antigen, and pulmonary fibrosis. To investigate the therapeutic and anti-asthmatic effects of Drynariae Rhizoma (DR), we examined the influence of DR on the development of pulmonary eosinophilic inflammation and airway hyperresponsiveness in a mouse model of allergic asthma. Methods : In this study, BALB/c mice were systemically sensitized to ovalbumin (OVA) followed intratracheally, intraperitoneally, and by aerosol allergen challenges. We investigated the effect of DR on airway hyperresponsiveness, pulmonary eosinophilic infiltration, various immune cell phenotypes, Th2 cytokine production and OVA specific IgE production in a mouse model of asthma. Results : In asthmatic mice, we found that DR.treated groups had suppressed eosinophil infiltration, allergic airway inflammation and AHR by suppressing the production of IL-5, IL-13 and OVA specific IgE. Conclusions : Our data suggest that the therapeutic mechanism by which DR effectively treats asthma is based on reductions of Th2 cytokines (IL-5), eotaxin, OVA-specific IgE production and eosinophil infiltration.

Noninvasive Detection of Specific Diagnostic Biomarkers for Atopic Dermatitis

  • The diagnosis of atopic dermatitis (AD) includes a test that checks allergen-mediated skin reactions and a method of measuring the total IgE and allergen-specific IgE in blood. However, these test methods are performed directly on the patient, which cause some pain or discomfort. In addition, the skin response test or IgE may result in false negative in about 20% of patients. In the present study, to identify specific biomarkers, HaCaT cells were used as a human keratinocyte that make up the skin, were treated IL-4 and IL-13 for 24 hours to induce a situation similar to keratinocytes in AD patients. In the HaCaT cells, pro-inflammatory cytokine such as IL-5, IL-6, and MCP-1 were increased by IL-4 and IL-13 and skin barrier proteins was reduced by IL-4 and L-13. This results showed that a situation similar to the stratum corneum of an actual patient is induced in HaCaT cells. And then the secretions of Kallikrein (KLK) 5 and KLK7 protease were checked by enzyme-linked immunosorbent assay (ELISA). It was specifically increased by IL-4 and IL-13. This showed that AD-related protease can be detected at the protein level using keratinocytes that can be taken in a non-invasive manner and suggested the possibility of applying it to AD diagnosis.

A seroepidemiological survey for toxocariasis in apparently healthy residents in Gangwon-do, Korea

  • We investigated the sero-prevalence of toxocariasis among healthy Korean adults in 1999. A total of 314 sera from normal inhabitants in Whachon-gun, Gangwon do, Korea was examined for specific antibody levels against excretory-secretory products of second stage larvae of Toxocara (TES). The presence of cross-reactions with other helminthiases such as cysticercosis, paragonimiasis, sparganosis or clonorchiasis was also checked by specific IgG ELISA. Sera showing positive reaction against TES were also tested by IgG immunoblot and by IgE ELISA. Out of 314 subjects, 16 was found to be positive by TES IgG ELISA and immunoblot, among whom 12 were also positive by TES IgE ELISA. Among the 16 seropositive samples, two sera showed positive reaction against Paragonimus and sparganum antigen, respectively. These results inferred that cross-reactions were negligible between toxocariasis and other helminthiases. Toxocariasis seroprevalence among Korean rural adults was detected to be approximately 5%.

Regulation of IgE-Mediated Food Allergy by IL-9 Producing Mucosal Mast Cells and Type 2 Innate Lymphoid Cells

  • Due to the increasing prevalence and number of life-threatening cases, food allergy has emerged as a major health concern. The classic immune response seen during food allergy is allergen-specific IgE sensitization and hypersensitivity reactions to foods occur in the effector phase with often severe and deleterious outcomes. Recent research has advanced understanding of the immunological mechanisms occurring during the effector phase of allergic reactions to ingested food. Therefore, this review will not only cover the mucosal immune system of the gastrointestinal tract and the immunological mechanisms underlying IgE-mediated food allergy, but will also introduce cells recently identified to have a role in the hypersensitivity reaction to food allergens. These include IL-9 producing mucosal mast cells (MMC9s) and type 2 innate lymphoid cells (ILC2s). The involvement of these cell types in potentiating the type 2 immune response and developing the anaphylactic response to food allergens will be discussed. In addition, it has become apparent that there is a collaboration between these cells that contributes to an individual's susceptibility to IgE-mediated food allergy.

Immunological Assessment of Respiratory Allergy Status for the Swine Farm Workers in Gyeonggi Province of Korea (경기 일부지역 양돈 축산인들의 호흡기 알레르기 관련 면역학적 지표 분석)

  • Objectives: Animal husbandry workers could be exposed to various work hazards including toxic gases, chemicals such as pesticides or organic dust. Immunological evaluation focusing on respiratory allergic hypersensitivity occurrence was under-taken for swine farm workers as a part of the study on immunologic status of dairy barn, swine confinement, and poultry farm workers. Materials and Methods: Peripheral bloods were collected from 25 workers at the year of 2001 and 12 workers at the year of 2012 from swine farms located at Gyeonggi province, Korea. Seven adults not involved with animal husbandry were recruited at the year of 2001 from the same residential area as the swine farm workers'. Level of plasma IgE and 20 respiratory allergen-specific IgE were evaluated using commercially available ELISA kit. Results: Plasma IgE level was approximately five-fold higher in the swine farm workers regardless of the sampling year than the control subjects. Plant allergens from outdoor environments such as golden rod, pigweed, Russian thistle, or ragweed were the major allergens with positive reaction(allergen specific IgE${\geq}$0.7 IU/mL) for the swine farm workers at 2001 year. Meanwhile, house dust mite(Dermatophagoides farinae, D. pteronyssinus) and cockroach, typical indoor allergens in Korea, were the major respiratory allergens for the swine farm workers at 2012 year. Conclusions: Overall, even though our results are primitive, the results suggest that immunological function of swine farm workers could be modulated toward type-2 reactivity.

The Effects of Sinapis Semen, Raphani Semen, and mixture decoction on the Asthmatic Mouse Model (백개자, 나복자 및 두 배합 약물의 천식 동물 모델에 대한 효과)

  • Objectives : To clarify the possible effects of Sinapis Semen and Raphani Semen on the development of pulmonary eosinophilic inflammation in a asthmatic mouse model. Methods : BALBav/c mice were sensitized to OVA followed intratracheally and by aerosol allergene challenges. We investigated the effect of Sinapis Semen and Raphani Semen on airway hyperresponsiveness, eosinophiic infitratio, immune cell phenotype, The2 cytokine product, and OVA-spedific IgE production. Results : Total lung cells, eosinophils, and lung leukocytes, OVA specific IgE levels, and Th 2cytokine levels such as IL-5, IL-13, IL-17, TNF-alpha, and eotaxin in BALF were reduced compared with those of OVA sensitized asthma mice (control). The absolute numbers of $CD3^+$, $CD3^+/CD69^+$, $CD3^-/CCR3^+$, $CD4^+$, $CD8^+$, $Gr-1^+/CD11b^+$, $B220^+/CD22^+$, $B220^+/IgE^+$ cells in lung tissiues significantly reduced compared to those of control. Specially total lung cells in BALF and the absolute number of $CD3^+/CD69^+$ and, $B220^+/IgE^+$ cells in lung tissiue effectively reduced in Sinapis Semen plus Raphani Semen compared to those of Sinapis Semen and Raphani Semen. Conclusions : These results indicate that Sinapis Semen plus Raphani Semen has deep inhibitory effects on airway inflammation and hyperresponsiveness in asmatic mouse model and also has effect of suppression of IL-5, IL-13, IL-17, OVA specific IgE production in BALF. The results verified that Sinapis Semen, Raphani Semen, and Sinapis Semen plus Raphani Semen could act as a immunomodulator which possess anti-inflammatory and anti-asthmatic property by modulating the relationship of Th1/Th2 cytokine imbalance.

  • Porcine epidemic diarrhea (PED), transmissible gastroenteritis (TGE) are an acute viral enteritis. colibacillosis by E coli is a microbial enteritic disease in suckling piglets[1]. These infectious intestinal diarrheal diseases cause severe diarrhea to suckling piglets, so that lead to enormous economical loss in swine-product industries. Ig-Top (AD Biotech, Korea) is a immunomodulator with IgY the specific yolk-antibody for PED, TGE and E. coli and oligosaccharide. The purpose of this study was to investigate protective effects against PED virus, TGE virus E.coli and in suckling piglets by oral administration of the Ig-Top. (omitted)

Clinical and laboratory findings of childhood buckwheat allergy in a single tertiary hospital

  • Purpose: Buckwheat allergy is one of the most severe types of food allergy in some countries, especially among children. However, few studies have investigated this condition. The aim of this study was to report the clinical and laboratory findings in Korean children with buckwheat allergy. Methods: Thirty-seven subjects, aged 1 to 14 years, were enrolled by retrospective medical record review from January 2000 through May 2015 at the Department of Pediatrics in Ajou University Hospital. The demographic profile, previous exposure to buckwheat pillows, clinical symptoms, and laboratory findings of each subject were recorded. Results: Twenty-six of the 37 children had immediate-type allergic symptoms to buckwheat, while 11 subjects were tolerant to buckwheat. Seventeen out of 26 buckwheat allergic children (65.4%) had anaphylaxis. The median buckwheat specific IgE level in the buckwheat allergic group ($7.71kU_A/L$) was significantly higher (P<0.001) than in the buckwheat tolerant group ($0.08kU_A/L$) with an optimal cutoff value of $1.27kU_A/L$ (sensitivity 84.6%, specificity 100%). When adjusted for age, the difference between the 2 groups showed no statistical significance (P=0.063). In subjects who had anaphylaxis, buckwheat-specific IgE levels ranged from 0.37 to $100kU_A/L$. Conclusion: Almost two-thirds of buckwheat-allergic children had anaphylaxis, and a wide-range of buckwheat specific IgE levels were observed in these children. Anaphylaxis occurred in a subject with a remarkably low IgE level ($0.37kU_A/L$).

The Effects of Maekmoondong-tang and Jeongcheonhwadamgangki-tang on Immune Cell and Serum OA-specific IgE in BALF in Rat Asthma Model (맥문동탕과 정천화담항기탕이 알레르기 천식 모델 흰쥐의 BALF내 면역세포 및 혈청 IgE에 미치는 영향)

  • Objectives: Maekmoondong-tang and Jeongcheonhwadamgangki-tang are herbal decoctions which have been used as traditional therapeutic agents for asthma. This study was performed to investigate the effects of Maekmoondong-tang and Jeongcheonhwadamgangki-tang on immune Cell & serum OA-specific 19B in broncho-alveolar lavage fluid (BALF) in rat asthma model. Methods: Rats were sensitized with ovalbumin (OA); at day I the sensitized group and Maekmoondong-tang and Jeongcheonhwadamgangki-tang groups were systemically immunized by subcutaneous injection of 1mg OA and 300mg of $Al(OH)_3$ in a total volume of 2ml. At the same time, 1ml of 0.9% saline containing $6{\times}10^9$ B. pertussis bacilli was injected by i.p. for 14 days. After systemic immunization, the rats received local immunization by inhaling 0.9% saline aerosol containing 2% (wt/vol) OA. A day after local immunization, BAL fluid was collected from the rats. A day after local immunization, rats were orally administered with each of Maekmoondong-tang and Jeongcheonhwadamgangki-tang extract for 14 days. Lymphocyte, CD4+T-cell CD8+ T-cell counts, CD4+/CD8+ ratio in BALF, change of serum OA-specific IgE level, and CD4+T-cell CD8+T-cell percentage in the peripheral blood were measured and evaluated. Results: Maekmoondong-tang and Jeongcheonhwadamgangki-tang showed a suppressive effect on the rat asthma model. Maekmoondong-tang decreased total cell, lymphocyte, CD4+T-cell, CD8+T-cell in BALF, and serum OA specific 19B level as compared with the control group, whereas Maekmoondong-tang decreased CD4+/CD8+ ratio in BALF with statistical nonsignificance as compared with the control group. 1 decreased total cell, CD4+T-cell, CD4+/CD8+ ratio in BALF, and serum OA specific IgE level, whereas Jeongcheonhwadamgangki-tang decreased ymphocyte, and CD8+T-cell in BALF with statistical nonsignificance as compared with the control group. CD4+T-cell and CD8+ T-cell percentage in peripheral blood were not changed significantly in all the experimental groups. Conclusions: This study shows that Maekmoondong-tang and Jeongcheonhwadamgangki-tang have a suppressive effect on asthma. Maekmooruiong-tang and Jeongcheonhwadamgangki-tang would be useful asthma treatment agents.

Gagam-Gongjin-dan Extract Attenuates Immune Responses to Ovalbumin in Balb/c Mice (Balb/c 마우스에서 Ovalbumin 면역반응에 대한 가감공진단(加減拱辰丹) 추출물의 억제효과)

  • Objectives : Gagam-Gongjin-dan (GGD) composited with Cervi parvum Cornu, Corni Fructus, Angelica gigantis Radix, Lycii Fructus, Dioscoreae Rhizoma, Citri Pericarpium, Gastrodiae Rihzoma, Agastachis Herba, Cassiae cortkex, Scutellariae Radix, Schisandrae Fructus has been traditionally used for chronic diseases or weakness after illness in oriental countries. However, little is known about the effects of methanol extract of GGD on immune responses to ovalbumin (OVA) plus alum. Therefore, the purpose of this study was to investigate the effects of GGD on immune responses to ovalbumin plus alum in Balb/c mice. Methods : In this study, the extract of GGD was prepared by extracting with methanol for 7 days. The extract was freeze-dried following filtration through vacuum distillation system. Mice were orally administrated with or without GGD extract of different doses (50-200 mg/kg/day) for 30 days. We examined the effects of GGD extract on the serum levels of total IgE, OVA-specific IgE, IgG1, IgG2a, and CTACK/CCL27 production and CCR10 expression in lymph node cells and body weight change and foot pad swelling responses in ovalbumin treated Balb/c. Results : The oral administration of GGD dose-dependently reduced the serum levels of total IgE, OVA-specific immunoglobulin (IgE, IgG1 and IgG2b) and CTACK/CCL27 production in ovalbumin treated BALB/c mice. The levels of CCR10 expression from lymph node cells of OVA treated mice were markedly suppressed by treatment with GGD in a concentration dependent manner. Furthermore, foot pad swelling responses were also markedly suppressed by GGD. However, body weight were significantly increased dose dependently by GGD treatment. Conclusions : These results suggest that GGD treatment suppresses immune responses to ovalbumin, and these properties may contribute to allergic disease care.
