Genetic diversity and population structure of endangered Neofinetia falcata (Orchidaceae) in South Korea based on microsatellite analysis

  • Population genetic assessment is essential for the conservation and management of endangered and rare plants. Neofinetia falcata is endangered epiphyte orchid and protected by law in Korea. In Korea, this species is only found on islands in the South Sea of Korea (including Jeju-do) and the southern coast of the Korean Peninsula. We developed nine microsatellite makers to assess the genetic diversity and population genetic structure of three populations of N. falcata. The genetic diversity at the species level was low, which can be attributed to inbreeding or fragmentation into small, isolated populations. A recent bottleneck was detected in one population, likely due to overcollection. N. falcata exhibited moderated levels of differentiation among populations, with the three populations were divided into two clusters based on genetic structure. The genetic diversity and structure of N. falcata are affected by restricted gene flow by pollen or seeds due to isolation and geographic distance. Strategies for in situ and ex situ conservation of this species are been proposed based on the results of our study.

Phylogeography of the economic seaweeds Chondrus (Gigartinales, Rhodophyta) in the northwest Pacific based on rbcL and COI-5P genes

  • The red algal genus Chondrus have long been used as raw materials for carrageenan and dietary fiber in health foods. Despite the importance of genetic information in safeguarding natural seaweed resources, knowledge of the population genetics of Chondrus in the northwest Pacific is limited. In this study, genetic diversity and phylogeographic structure of 45 populations (777 specimens) of Chondrus from Korea, China, and Japan were evaluated based on mitochondrial COI-5P gene sequences, and phylogenetic relationships were confirmed based on plastid rbcL gene sequences. Molecular analyses assigned the specimens in this study to three Chondrus species: C. nipponicus, C. ocellatus, and C. giganteus; phenotype-based species classification was impossible owing to their high morphological plasticity. We found moderate intraspecific genetic diversity and a shallow phylogeographic structure in both for C. nipponicus and C. ocellatus, and low intraspecific genetic diversity in C. giganteus. Each of the three species exhibited high-level intraspecific gene flow among regions based on the most common haplotypes (CN1 for C. nipponicus, CO1 for C. ocellatus, and CG1 for C. giganteus). Our comprehensive genetic information provides insights into the phylogeographic patterns and intraspecific diversity of the economically important Chondrus species. It also highlights the need to conserve existing natural Chondrus resources through continuous monitoring of genetic diversity and phylogeographic pattern.

Do Physiognomically Designated Protected Areas Match Well with Ecological Data based upon Diversity Indices and Ordination? Implications for Urban Forest Conservation

  • We surveyed the vegetation of an ecological landscape preservation area (legally protected conservation areas or national parks) and the surrounding areas of Mt. Cheonggye, Republic of Korea, to explore the conservation implications for preservation areas and surrounding transition areas. We calculated diversity indices to identify the properties of the preservation and surrounding areas that are relevant to conservation efforts. We then compared the plant community composition between the areas using field and quadrat surveys in the preservation and surrounding areas. The cover of the dominant species in all tree and herb layers was markedly higher in the preservation area than in the peripheral zones. The species richness indices were significantly higher in the preservation area than in the peripheral zones. Ordination using detrended canonical correspondence analyses showed that the cover of the dominant tree species and rocks could explain the distribution of plant species in the Cartesian space of the ordination. Our results demonstrate that physiognomically designated protected areas match well with ecological data based on diversity indices and ordination analyses and that disturbances in the areas surrounding the ecological landscape of preservation areas can have considerable impacts on plant diversity indices. Hence, the preservation and management of surrounding areas are essential conservation elements for protecting the entire ecological landscape of preservation areas.

점봉산 생물권 보전지역내 온대낙엽수림에서 미소환경요인과 식생요인의 공간분포와 상관 분석 (Relationships Between the Spatial Distribution of Vegetation and Microenviromnent in a Temperate Hardwood Forest in Mt. Jrnbong Biosphere Reserve Area, Korea)

  • 생태계 내에서 미소환경요인과 식생 요인의 상호관련성을 밝히는 일은 생태계의 구성원리와 생태계의 관리에 많은 도움을 준다. 생물권보전지역으로 지정된 점봉산 일대의 온대낙엽수림에서 식생요인과 환경요인의 공간적 이질성을 정량화하고, 임상식생의 공간분포에 미치는 미소환경요인의 영향을 파악하였다. 대부분의 환경요인과 식생요인은 영구방형구내 미소지소간에 변이를 나타내었다. 환경요인 중에서 공간의존도가 높은 요인들은 지형적요인(고도, 경사도), 낙엽층 두께, 토양수분 함량 및 상대광도이었고, 공간의존거리가 상대적으로 짧은 요인은 낙엽층 두께, 상대광도 및 마그네슘 함량이었다. 식생요인 중에서 아교목층의 피도, 가을 초본층의 피도 및 철쭉꽃의 피도를 제외하고는 모든 변수들의 공간의존도는 높은 편이었다. 교목층의 피도, 관목층의 피도 및 가을 초본층의 종다양성 지수의 공간의존거리는 15 m 이내이었고,초봄 초본층의 종풍부도와 종다양성지수 및 가을 초본층의 종풍부도의 공간의존거리는 28∼48 m이었다. 초봄과 가을 임상식물의 종풍부도와 종다양성지수의 공간분포에 영향을 주는 요인으로는 낙엽층 두께, 경사도, 아교목층의 피도, 관목층의 피도, 조릿대의 피도 등이었는데,그 중에서 가장 크게 영향을 주는 요인은 낙엽층 두께로서 낙엽층 두께가 두꺼운 곳일수록 종풍부도와 종다양성지수가 낮은 경향이었다. 본 조사지역에서 낙엽층의 공간 분포에 영향을 주는 요인들로는 바람의 방향, 미소지형 및 관목의 밀집정도로 추정되었다.

Relationship between Tree Species Diversity and Carbon Stock Density in Moist Deciduous Forest of Western Himalayas, India

  • With the growing global concern about climate change, relationship between carbon stock density and tree species has become important for international climate change mitigation programmes. In this study, 150 Quadrats were laid down to assess the diversity, biomass and carbon stocks in each of the forest ranges (Barkot Range, Lachchiwala Range and Thano Range) of Dehra Dun Forest Division in Doon Valley, Western Himalaya, India. Community level carbon stock density was analyzed using Two Way Indicator Species Analysis. Species Richness and Shannon Weiner index was correlated with the carbon stocks of Doon Valley. Positive and weak relationship was found between the carbon stock density and Shannon Weiner Index, and between carbon stock density and Species Richness.

Taxonomy of four scuticociliates(Protozoa: Ciliophora) from coastal waters of South Korea

  • The diversity of scuticociliates (subclass Scuticociliatia Small, 1967) had previously been superficially investigated in Korea and only 10 species were reported according to the National Species List of Korea published in 2019. Here, we identify four scuticociliates, collected from three coastal water samples, by observing protargol-impregnated specimens. As a result, the present species belong to the order Pleuronematida Fauré-Fremiet in Corliss, 1956 and their lower taxonomic classifications are as follows: family Eurystomatellidae Miao et al., 2010 - Eurystomatella sinica Miao et al., 2010; family Pleuronematidae Kent, 1881 - Pleuronema grolierei Wang et al., 2008, P. setigerum Calkins, 1902, and Schizocalyptra aeschtae Long et al., 2007. The family Eurystomatellidae and the genus Schizocalyptra Dragesco, 1968 are reported for the first time in Korea. Considering that the scuticociliates are a species-rich group and very common in most habitats(including freshwater and terrestrial habitats), our findings indicate that we are far from understanding the complete diversity of Korean scuticociliates.

Long-Term Investigation of Marine-Derived Aspergillus Diversity in the Republic of Korea

  • Aspergillus species play a crucial role in terrestrial environments as degraders and are well known for producing various secondary metabolites. Recently, Aspergillus species have been discovered in marine environments, exhibiting adaptability to high salinity and producing diverse secondary metabolites with valuable properties. However, limited research has focused on their marine diversity, leading to inaccurate species identification. The current study addresses this gap by investigating diverse marine habitats in the Republic of Korea, including sediment, seawater, seaweed, and marine animals. From three coasts of the Korean Peninsula, 472 Aspergillus strains were isolated from the various marine habitats. A total of 41 species were accurately identified using multigenetic markers: internal transcribed spacer, calmodulin, and b-tubulin. The findings underscore the importance of accurate identification and provide a basis for elucidating the functional role of marine-derived Aspergillus species in marine ecosystems.

금강수계에서 하상재료에 따른 어류의 종다양성 및 서식지 평가 (Assessment of Inhabitation and Species Diversity of Fish to Substrate Size in the Geum River Basin)

  • In order to establish fundamental data for stream restoration and environmental flow, we investigated inhabitation and species diversity of fish to substrate size in the typical streams of Geum River Basin. Field monitoring including fish sampling was conducted from October 2007 to October 2009. Substrate size was determined according to six different : silt (Si), sand (Sa), fine gravel (Fg), coarse gravel (Cg), cobbles (Co) and boulders (Bo). A total number of fish caught in the 18 sites was 7,649 representing 10 families 50 species, and Si, Sa, Fg, Cg, Co and Bo stations occupied 30, 29, 38, 30, 27 and 17 species, respectively. The most frequently found species in number was pale chum (Zacco platypus, 29.7%, n=2,275) followed by Z. koreanus (22.5%, n=1,720) in total stations. Biological diversity with increase of substrate size from the dominance of part species showed higher values as dominance index, lower and diversity, richness and evenness index. Index of biological integrity (IBI) and qualitative habitat evaluation index (QHEI) scores decreased with increase of substrate size. Therefore, it is necessary to make an effort on stream rehabilitation with evaluation of physical habitat condition by indicator species in order to maintain biodiversity and perform ecological restoration.

화전 후 묵밭의 식생 천이에 따른 종다양성 및 식생 구조의 발달 (Changes of Species Diversity and Development of Vegetation Structure during Abandoned Field Succession after Shifting Cultivation in Korea)

  • 강원도 평창군 일대에서 화전 후 묵밭의 식생 천이에 따른 식생 구조의 발달 과정과 종다양성의 변화를 조사하였다. 방치 후 경과 년 수에 따라 최상층부의 키는 초기에 빠르게 증가하고 후기에 안정화하는 경향을 나타내었다. 10, 20, 50 및 80 년차 묵밭에서 최상층부 식생의 키는 각각 4, 8, 18 및 18 m이었다. 교목층, 아교목층, 관목층 및 초본층의 4층 구조를 갖춘 완전한 형태의 숲으로 발달하기까지는 방치 후 약 35년이 경과해야 하는 것으로 추정되었다. 방치 후 경과 년 수에 따라 식생치 Ivc는 로그 함수적으로 그리고 식생치 Hcl은 직선적으로 증가하는 경향이 있었다. 묵밭의 식생천이에서 종다양성은 천이 중기인 $10{\sim}20$년차에서 최고로 증가한 다음 이후 점차 감소하는 경향을 나타내었다. 본 조사지에서 종다양성은 균등도보다 종풍부도와 매우 밀접한 상관을 나타내었다. 대부분의 목본은 개망초-쑥 단계인 $2{\sim}6$년차에 정착이 이루어져 시간 경과에 따라 이들의 우점도가 변화함으로써 Egler (1954)의 초기 종조성 모델을 따르는 것으로 확인되었다.

Differences in Temporal Variation of Ground Beetle Assemblages (Coleoptera: Carabidae) between Two Well-Preserved Areas in Mt. Sobaeksan National Park

  • Understanding how future climate conditions will be impact on the biodiversity and species composition is important, because biodiversity becomes more important in environment assessment. To understand the biological changes including diversity and species composition over time (temporal variation within a year), the species diversity and composition of ground beetles were investigated in two well-preserved areas in the Sobaeksan National Park using pitfall traps. In addition, relationships between ground beetles and environmental variables were studied by considering temporal variation. We collected 2,146 ground beetle specimens representing 45 species, and individual-based rarefaction curves indicated that similar species richness was found between Geumseon Valley (GV) and Namcheon Valley (NV). The Bray-Curtis matrix comparisons between study sites were characterized by similar ground beetles sample heterogeneity, while temporal variations in abundance, species richness, and ${\beta}-diversity$ of ground beetles showed rather difference over time according to location of study sites. In GV site, minimum temperature was selected as the best predictor for abundance, species richness, and ${\beta}-diversity$ of ground beetles, while those relationships in NV site were more complicated. In conclusion, our study suggests that understanding the different response of ground beetles to climatic variables related to local habitat conditions is important to predict the effect of climate change on biological communities.
