한국 재래식 메주 발효과정에 있어서 지질 및 지방산 조성의 변화 (Changes in lipid and Fatty Acid Composition in Korean Native Meju during Fermentation)

  • 재래식메주의 발효과정에 있어서 지질 및 지방산 조성의 변화를 체계적으로 규명하기 위해 silicic acid column chromatography를 이용하여 중성지질, 당지질, 인지질은 분리하였으며 gas liquid chromatography를 이용하여 지방산 함량은 조사한 결과는 다음과 같다. 1. 침지한 콩과 삶은콩으로부터 얻은 조지질의 분획은 중성지질이 각각 92.47%, 92.35%이었으며, 인지질 및 당지질이 4.64%, 4.88%로 각각 나타났나. 발효과정중 중성지질은 80.59%까지 감소했으며, 당지질과 인지질의 함량은 증가하였다. 2. 침지한 콩과 삶은콩의 비극성 지질에서 triglyceride함량은 89.66%, 87.83%로 주류를 이루었고, sterol ester, free fatty acid, diglyceride 및 sterol이 발효과정중 그 함량이 증가한 반면, triglyceride는 크게 감소했다. 3. 침지한 콩과 삶은콩에서, 조지질, 증성지질, 당지질, 인지질의 지방산 조성은 linoleic acid $54{\sim}58%$, oleic acid $22{\sim}20%$, palmitic acid $10{\sim}12%$, stearic acid $3{\sim}5%$였으며 myristic acid는 trace로 나타났다. 그러나 발효과정 중 당지질에서 myristic acid가 발효 2주때 부터 나타나며 약 2%정도를 유지했다. 4. 메주 발효기간 동안 지방산 함량의 변화는 palmitic acid가 2주째 부터 점차 감소했다. oleic acid와 linoleic acid는 약간씩 감소했으며, linolenic acid는 인지질에서 크게 감소한 반면, 조지질, 중성지질, 당지질에서는 약간씩 증가했다. 5. 발효기간 동안 당지질 분획에서 myristic acid가 증가하였으며, 지방분해효소의 활성은 3주째 최대치를 보였다. palmitic acid, stearic acid는 3주째 까지 증가한 후 차차 감소하였고 linoleic acid는 1주째 부터 6주까지 점차 증가하였다.

국내 콩 유전자원의 지방함량 및 지방산 조성변이 (Variation of Oil Contents and Fatty acid Compositions in Korean Soybean Germplasms)

  • 고품질 콩 품종육성 및 유전자원 연구의 기초자료로 활용하기 위해 국내 콩 주요품종 46계통 및 보존 유전자원 517 계통의 지방함량과 지방산 조성의 변이양상을 분석한 결과를 요약하면 다음과 같다. 1. 한국산 콩 주요 품종의 지방함량 범위는 $15.8%{\sim}20.3%$ 였으며, 평균 18.2%를 나타내었고, 지방함량이 가장 낮은 품종은 1986년에 육성된 보광콩이, 가장 높은 품종은 2000년도에 육성된 새별콩으로 조사되었다. 2. 한국산 콩 주요 품종 46계통의 지방산 조성분석 결과 포화지방산인 C16:0 및 C18:0의 경우 각각 $8.96{\sim}13.23%$$2.55{\sim}4.20%$ 범위로 존재하였고, 불포화지방산인 C18:1, C18:2 및 C18:3의 범위는 각각 $16.33{\sim}36.41%,\;42.32{\sim}58.84%$$6.98{\sim}10.72%$를 나타내었다. 3. 국내 보존 유전자원 517계통의 지방산 조성을 분석한 결과 C16:0은 8.50%(IT 154681)${\sim}14.78%$(IT 154558), C18:0은 1.86%(IT 022331)${\sim}4.78%$(KLG 10738), C18:1은 13.29%(IT 178710)${\sim}43.30%$(IT 154657), C18:2는 38.21%(IT 156004)${\sim}61.51%$(IT 178710) 및 C18:3이 5.03(IT 188085)%${\sim}11.48%$(IT 153412)의 범위를 나타내었다. 4. 콩 함유 지방산 중 포화지방산에 속하는 C16:0 및 C18:0은 불포화지방산인 C18:1과 고도의 부의 상관을 나타내었으며, 불포화지방산인 C18:1은 다른 종류의 불포화지방산인 C18:2 및 C18:3과 고도의 부의 상관을 나타내었고, C18:2와 C18:3 사이에는 고도의 정의상관이 인정되는 것을 확인할 수 있었다.

  • 전통식품인 콩나물의 고부가가치화를 위한 우량 나물콩 품종 육성의 기초 자료를 제공하고자 특성이 다른 5종의 장려품종 나물콩으로 재배한 콩나물의 지방산 및 식이섬유 함량을 분석한 결과는 다음과 같다. 1. 생콩나물과 데친 콩나물의 지방산 조성은 큰 차이가 없어 가열 처리가 지방산 조성에는 영향을 끼치지 못하였다. 2. 전체 콩나물과 콩나물의 자엽 부위 지방산 조성은 원료콩과 비슷하였으나 배축 부위는 원료콩에 비해 포화지방산 함량이 높았으며 변이도 자엽 부위보다 컸다. 3. 가열 처리된 콩나물이 생콩나물보다 식이섬유 함량이 높았다. 4. 지방산 조성과 식이섬유 함량은 품종별로 차이가 있었다.

Evaluation of the quality stability on the soybean flour depending on storage conditions

  • Legumes are good sources of various nutrients. Among legumes, soybean and its flour are accessible foods to consumers. However, in case of soybean flour, there is a disadvantage of easily going rancid. Thus, this study aimed to investigate how the quality of soybean flour changes during storage according to the packaging materials and storage temperatures. The raw and roasted soybean flours were packed in two types of packaging, polyethylene (PE) and polypropylene (PP) film bags respectively, and stored at three different storage temperatures (4, 20, and $45^{\circ}C$) for 1 year. The acid value, conjugated diene value, peroxide value, p-anisidine value, thiobarbituric acid (TBA) value, lipoxygenase activity, and fatty acid content of raw and roasted soybean flours were measured at the point of starting storing, and after 1, 2, 4, 8, and 12 weeks of storage. The acid value of soybean flour was increased for 4 weeks and thereafter significantly decreased (p < 0.0001). The conjugated diene value was significantly increased after 4 weeks storage at $45^{\circ}C$ with PE and PP film bags (p < 0.0001). The peroxide value had no changes during 4 weeks storage at $45^{\circ}C$ with PE and PP film bags, and then those was dramatically increased after 8 weeks (p < 0.0001). The p-anisidine values in all of storage conditions were decreased after 4 weeks. Lipoxygenase activity was decreased at 12 weeks storage at $20^{\circ}C$ and $45^{\circ}C$ with PP film bags. Acid value had positive correlations with p-anisidine value (r = 0.30 and p < 0.0001) and lipoxygenase activity (r = 0.36 and p < 0.0001), and had negative correlations with conjugated diene value (r = -0.45 and p < 0.0001) and peroxide value (r = -0.25 and p < 0.001). Conjugated diene value had a high positive correlation with peroxide value (r = 0.76 and p < 0.0001), but that had a negative correlation with lipoxygenase activity (r = -0.51 and p < 0.0001). Peroxide value had negative correlations with p-anisidine value (r = -0.20 and p < 0.01) and TBA value (r = -0.15 and p < 0.05). The degree of reduction in fatty acid content of raw soybean flour was higher than the roasted soybean flour during the storage. Total fatty acid content had positive correlations with acid value (r = 0.45 and p < 0.0001) and p-anisidine value (r = 0.58 and p < 0.0001), but had a weak negative correlation with conjugated diene value (r = -0.19 and p < 0.01). This study showed how the rancidity of the raw and roasted soybean flours progressed during storage. Thus, our findings can be used as base data to do a further study of finding and developing more stable storage conditions of the soybean flour.

대두(大豆) 및 대두분(大豆粉) 저장중 지방질의 변화 (Changes of Lipids in Whole Soybean and Soy Flour during Storage)

  • 대두(大豆) 및 대두분(大豆粉) 저장중 총지방질(總脂肪質)의 산화지수(酸化指數) 및 지방산조성(脂肪酸組成)을 경시적으로 검토하였다. 저장기간이 경과함으로써 산(酸)값과 과산화물(過酸化物)값이 증가 되었는데, 이와같은 경향은 대두(大豆)보다 대두분(大豆粉)에서, 그리고 저장습도가 높을수록 현저하게 나타났다. 또한 포장재료는 OPP/Al/PE film을 사용함으로써 대두(大豆) 및 대두분(大豆粉)의 산(酸)값과 과산화물(過酸化物)값의 증가를 억제시킬 수 있었을뿐 아니라, 지방산의 변화도 감소되었다. 지방산조성은 저장기간에 따라 18 : 2산(酸)과 18 : 3산(酸)은 감소되었으나 포화지방산(飽化脂肪酸)은 상대적으로 증가되었다.

Fatty Acid Profiles and Stearoyl-CoA Desaturase Gene Expression in Longissimus dorsi Muscle of Growing Lambs Influenced by Addition of Tea Saponins and Soybean Oil

  • This study was conducted to determine the effects of dietary addition of tea saponins (TS) and soybean oil (SO) on fatty acid profile and gene expression of stearoyl-CoA desaturase (SCD) in longissimus dorsi (LD) muscle of growing lambs. Thirty-two Huzhou lambs were assigned to four dietary treatments in a $2{\times}2$ factorial arrangement with main effects of TS (0 or 3 g/d) and SO (0 or 30 g/kg of diet DM). The diet without additives was considered as NTNS (no TS or SO). After a feeding trial for 60 d, four lambs of each treatment were slaughtered to collect the samples of LD muscle. Percentage of trans-11 vaccenic acid was enhanced (p<0.05) in muscle of lambs fed TS and SO. The proportion of total conjugated linoleic acid (CLA) was increased (p<0.05) by SO, but decreased (p<0.05) by TS in LD muscle. The percentage of total saturated fatty acids in muscle was decreased (p<0.05) by addition of TS and SO, while addition of SO increased (p<0.05) the percentage of total polyunsaturated fatty acids. The ratio of cis-9, trans-11 CLA to tran-11 vaccenic acid was decreased (p<0.05) by TS, but increased (p<0.05) by SO. The same effects were observed in SCD mRNA expression. From these results it is indicated that including TS and SO in the diet of growing lambs affect the fatty acid profiles of LD muscle and that the proportion of cis-9, trans-11 CLA in the muscle influenced by TS and SO may be related to the SCD gene expression.

국내산 및 제초제 내성 콩(HS2906)의 일반성분, 무기질 및 지방산 조성 (Proximate Analysis, Mineral and Fatty Acid Composition of Domestic and Glyphosate- Tolerant HS2906 Soybean)

  • Proximate analysis, mineral and fatty acid composition of three conventional domestic soybean cultivars and two imported ones including glyphosate-tolerant HS2906 were evaluated by AOAC method, ICP-AES and gas chromatography. There were several differences in the proximate analysis among three conventional domestic soybean cultivars ; higher crude fat in the cultivar Hwanggumkong, higher crude protein in Pungsankong, and higher carbohydrate and crude ash in Duyukong. The ranges of contents of proximate components of domestic cultivars were similar to the data previously reported. There were no significant differences in proximate analysis between conventional soybean WS82 and glyphosate-tolerant HS2906 ; 23.55-23.90% of crude fat, 34.22-35.55% of crude protein, 6.25-6.45% of crude ash, and 25.35-26.47% of carbohydrate. The mineral and fatty acid compositions of HS2906 were similar to those of conventional soybeans previously reported.

방사선 조사선량에 따른 유지의 이화학적 성질변화 (I) (Changes of the phsico-Chemical Characteristics of oils treated by the ${\gamma}$-ray irradiation (I) -The Extracted soybean oil-)

  • To investigate the oxidative stabilities of the ${\gamma}$-ray irradiated soybean during storage and heating and some physico-chemical characteristics of soybean and the extracted soybean oil (SBO) with/without the ${\gamma}$-ray irradiation were determined. The ${\gamma}$-ray level use in irradiation for soybean were 2.5, 5.0 and 10.0 KGY respectively and Acid Value, Peroxide Value, Conjugated Diene Value, Composed Fatty Acids amounts, and Trans Fatty Acid occurrence were determined for all samples, which were incubated at 45$\pm$1$^{\circ}C$ for 25 days heated at 180$\pm$1$^{\circ}C$ for 30 hours. And these values of the ${\gamma}$-ray treated samples were compared to those of nontreated samples. The results were obtained as follows : 1. According to the increased level of the ${\gamma}$-ray irradiation, there was little difference in Dielectric Constant, Viscosity, and the Induction Period by Rancimat. But, in case of 5.0 KGY, oxidative stability was increased more twice than that of non-irradiation. In the quantity of fatty Acids composition of the extracted soybean oil irradiated with 10.0 KGY, palmitic, oleic and linoleic acids were less increased thanb those of non-irradiation, while stearic, linolenic acids were decreased. In the case of 2.5 KGY irradiation, stearic and oleic acids were increased. 2. The Acid Value of SBO according to the ${\gamma}$-ray irradiation level was almost not change, but was 0.1 lower than that of non-irradiation during incubation (45$\pm$1$^{\circ}C$). The Peroxide Value of SBO with the ${\gamma}$-ray irradiation, was very lower than that of non-irradiation, but its effect on oxidative stability was better of SBO treated with 5.0 KGY and 10.0 KGY. In the Fatty Acids composition of SBO, palmitic, stearic, oleic acids were increased, while linoleic, linolenic acids were decreased during incubation(45$\pm$1$^{\circ}C$). This tendency was more obvious due to the ${\gamma}$-ray level. While heating(180$\pm$1$^{\circ}C$), the Acid Value of SBO treated with the ${\gamma}$-ray irradiation was decreased, the Acid Value of SBO irradiated with 2.5 KGY was the lowest. Also the peroxide Values of SBO treated with 5.0 KGY, 10.0 KGY were very lower than that of non-irradiation. Conjugated Diene Value of SBO was almost unchanged according to the ${\gamma}$-level and heating time. 3. When the methyl linoleate was irradiated with the ${\gamma}$-ray, the Trans Fatty Acid was little produced. In case of SBO with non-irradiation, the trans C18:1 was occured about 6.5~7.9%, but trans C18:2 and C18:3 were not shown, while SBO irradiated with the ${\gamma}$-ray 2.5, 5.0, 10.0 KGY, trans C18:3 and C18:2 amount in SBO were increased according to heating time, but trans C18:3 was little occured. As these results, the effects of the ${\gamma}$-ray irradiation to oil containing food were to cut down the energy for food storage and to increase oxdative stability during storge. And also it was shown to be the best that 10.0 KGY of the ${\gamma}$-ray irradiation would be applied to soybean.

Effect of including n-3/n-6 fatty acid feed sources in diet on fertility and hatchability of broiler breeders and post-hatch performance and carcass parameters of progeny

  • Objective: The present trial was conducted to determine the influence of different dietary fatty acid (omega-3 and omega-6) sources on reproductive performance of female broiler breeders and growth performance and carcass traits of their progeny. Methods: Two hundred and twenty, 25 weeks old Ross-308 male (20) and female (200) broiler breeders were used in the experiment for the period of 6 weeks. All birds were randomly divided into four dietary treatments (containing 2% soybean oil, 2% sunflower oil, 2% flaxseed oil, and 2% fish oil) each with five replicates of one male and ten females. Throughout this experiment hatching performance of broiler breeders, progeny growth performance and carcass parameters were recorded. Results: The results showed that the inclusion of different fatty acid sources in female broiler breeders diet had no significant effects (p>0.05) on number of fertile eggs, post-hatch mortality, and fertility rate. The soybean oil supplemented group had significantly (p<0.05) higher late embryonic mortality compared to other three treatments. Conclusion: It was concluded that inclusion of 2% of different sources of omega-3 and omega-6 fatty acids (especially 2% flax seed oil) in broiler breeders' diet can reduce late embryonic mortality. The other reproductive characteristics of parents and growth and carcass characteristics of progeny remained unaltered by dietary sources of omega-3 and omega-6 fatty acids.

Genetics and Breeding for Modified Fatty Acid Profile in Soybean Seed Oil

  • Soybean [Glycine max(L.) Merr.] oil is versatile and used in many products. Modifying the fatty acid profile would make soy oil more functional in food and other products. The ideal oil with the most end uses would have saturates(palmitic + stearic acids) reduced from 15 to < 7%, oleic acid increased from 23 to > 55%, and linolenic acid reduced from 8 to < 3%. Reduced palmitic acid(16:0) is conditioned by three or more recessive alleles at the Fap locus. QTLs for reduced palmitic acid have mapped to linkage groups(LGs) A1, A2, B2, H, J, and L. Genes at the Fad locus control oleic acid content(18:1). Six QTLs($R^2$=4-25%) for increased 18:1 in N00-3350(50 to 60% 18:1) explained four to 25% of the phenotypic variation. M23, a Japanese mutant line with 40 to 50% 18:1 is controlled by a single recessive gene, ol. A candidate gene for FAD2-1A can be used in marker-assisted breeding for high 18:1 from M23. Low linolenic acid(18:3) is desirable in soy oil to reduce hydrogenation and trans-fat accumulation. Three independent recessive genes affecting omega-3 fatty acid desaturase enzyme activity are responsible for the lower 18:3 content in soybeans. Linolenic acid can be reduced from 8 to about 4, 2, and 1% from copies of one, two, or three genes, respectively. Using a candidate gene approach perfect markers for three microsomal omega-3 desaturase genes have been characterized and can readily be used in for marker assisted selection in breeding for low 18:3.
