Effects of Source-Sink Alteration on Dry Matter Accumulation and Protein Content in Soybean (콩에서 Source-Sink변경이 건물축적과 단백질함량에 미치는 영향)

  • Effects of leaf and pod removal on changes in dry weight and protein content of soybean [Glycine max (L.) Merr.] cultivar 'Hwangkeumkong' were measured at the research farm of Korea University in 1992. The upper 40% and lower 60% of leaves and pods were subjected to treatments at the growth stage of beginning pod(R3). Leaf dry weight of lower part was increased by upper leaf-lower pod removal, but seed dry weight of upper part was decreased. The upper leaf-lower pod removal also increased stem dry weight and decreased upper pod dry weight. Protein content was higher in lower leaves than upper leaves. Seed protein contents were decreased both upper leaf-lower pod removal and lower leaf-upper pod removal treatments. The upper leaf-lower pod removal showed the highest leaf and the lowest seed protein quantities among the five treatments. These results indicated that protein sources were moved from lower to upper parts but weak in remobilization from leaves for the long distance translocation during the reproductive growth period of soybean plants.an plants.

  • This experiment was carried out to compare chemical composition, TDN yield of corn-soybean intercropping and corn monocropping forage plants at different harvesting time and obtained the following results. 1. In both cropping systems, the content of chemical composition of forage were changed same pattern in growing stage. The content of crude protein in corn-soybean intercropping forage at yellow stage increased more than that of corn nonocropping forage, while the crude fat in corn monocropping forage plants increased than that of corn-soybean intercropping forage plants at mature stage. 2. The crude fiber, crude ash, ADF content of forage plants in both cropping system decreased same pattern in growing period, however, NFE content of forage increased with maturity. 3. TDN yield of corn-soybean intercropping and corn monocropping forage plants at yellow stage obtained similar results and TDN yield per 10a in intercropping and monocropping were 1006.lkg and 978.6kg, respectively. 4. Consequently, corn-soybean interaopping system could be increased crude protein yield without decreasing of dry matter yield in comparison with corn monocropping system for corn silage.

  • Efficient in vitro regeneration system is essential for the successful crop breeding of soybean (Glycine max (L.) Merr.) using the new biotechnology. The genotype of donor plants strongly influences the establishment of tissue culture system. Therefore, the screening of genotypes with excellent tissue culture ability is very important for soybean genetic improvement. In this study, we report the tissue culture efficiency of 21 soybean cultivars belong to Korean soybean core-collection and two foreign cultivars (Jack and Maverick). The Kwangan, Anpyeong and Seonam are share close genetic relationship in 21 cultivars and these three cultivars were observed the high frequency of germination and regeneration. Furthermore, the high tissue culture abilities were also observed in the Williams 82 used in reference genome sequencing and the two foreign cultivars. The transformation of pBAtc:tRNA with bar gene was performed by Agrobacterium tumefaciens in the cultivars with high tissue culture ability. Transformation of the bar gene was identified by PCR analysis in Kwangan, Pungwon, Seonam, and Maverick. Our results provide useful information for the breeding of various soybean cultivars by plant biotechnology such as, genome editing.

Role of Mesophyll Morphology in Determination of Leaf Photosynthesis in Field Grown Soybeans (포장생육대두의 엽광합성과정에서 엽육세포 형태의 역할)

  • Photosynthetic variation in field grown soybean [Glycine max (L.) Merr. cv Hodgson78] was studied in relation to leaf anatomical variation. Variations in mesophyll morphology were accentuated by manipulating source and sink size. At R3 stage, two treatments were started: one was thinning and continu-ous debranching(6. 5 plants rather than 26 plants per m of row and remaining plants were debranched weekly), and the other was continuous partial depodding (allowing only one pod to develop at each mainstem node). Gas exchange characteristics, mesophyll cell volume and surface area per unit leaf surface, and microclimatic parameters were measured on the intact terminal leaflet at the 10th node. Observations were made 5 times with 3 to 4 day intervals starting R4 stage. Two models were used to compute leaf photosynthetic rates: one considered no effect of mesophyll morphology on photosynthesis, and the other considered potential effects of variations in mesophyll cell volume and surface area on diffusion and biochemical processes. Seventy nine percent of total photosynthetic variations observed in the experiment was explained by the latter, while 69% of the same variations was explained by the former model. By incorporating the mesophyll morphology concept, the predictability was improved by 14.6% in the field condition. Additional Index Words: photosynthesis model, leaf anatomy, Glycine max (L.) The Development of a New Soybean Strain Without Kunitz Trypsin Inhibitor, Lectin, and 7S α' Subunit Protein (쿠니츠트립인히비터, 렉틴 및 7S α' 서버유닛 3가지 단백질이 없는 콩 계통의 개발)

  • Soybean [Glycine max (L.) Merr.] seeds contain an average of 40% protein on a dry weight basis, but they also contain antinutritional elements such as lectin, Kunitz trypsin inhibitor (KTI), and 7S α'- subunit protein. The objective of this research was to develop a new soybean genotype with triple recessive alleles for these elements. Three parents (Gaechuck#2, PI506876, and Le-16) were used to develop the genetic population, and the presence of lectin and KTI protein was detected using Western blot while 7S α' subunit protein was detected using SDS-PAGE. One F3 plant strain with proper agronomical traits such as type, height, seed quality, and 100-seed weight was selected. The genotype of the developed strain is titilelecgy1cgy1, that is KTI, lectin, and 7S α' subunit protein free. The new strain has a purple flower, determinate growth habit, and light yellow pods at maturity. The seed has a buffer hilum and is yellow in color. The new strain's height was 58 cm compared to the Daewonkong cultivar at 46 cm, and its 100-seed weight was 27.1 g, smaller than the Daewonkong at 29.0 g. This is the first new soybean strain with the titilelecgy1cgy1 genotype, and it can be used to improve yellow soybean cultivars of high quality and function.

Genetic Diversity and Population Structure of Korean Soybean Landrace [Glycine max(L.) Merr.]

  • Two hundred and sixty Korean soybean landrace accessions were analyzed for polymorphism at 92 simple sequence repeat(SSR) loci. The 995 identified alleles served as raw data for estimating genetic diversity and population structure. The number of alleles at a locus ranged from three to 27 with a mean of 10.4 alleles per locus. $F_{ST}$ values estimated by analysis of molecular variance(AMOVA) using SSR data set were 0.018, 0.027, and 0.016 for usage, collection site and maturity groups, respectively, indicating little genetic differentiation. The model-based clustering analysis placed the accessions into three clusters(K=3) with 0.0503 of $F_{ST}$, indicating moderate genetic differentiation. Duncan's Multiple Range Test at K = 3 on the basis of 18 quantitative traits revealed that one cluster was mainly differentiated from the other two clusters by seed related traits and the other two clusters were differentiated from each other by biochemical traits. Genetic structure of Korean soybean landraces was differentiated by model-based clustering and supported by their phenotypic traits in part. This preliminary study could be the first step towards more efficient germplasm management and utilization of soybean landraces and helpful in association studies between genotypic and phenotypic traits in Korean soybean landraces.

Identification of SNPs tightly linked to the QTL for pod shattering in soybean[Glycine max (L.) Merr.]

  • The pod shattering or dehiscence is essential for the propagation of pod-bearing plant species in the wild, but it causes significant yield losses during harvest of domesticated crop plants. Identifying novel molecular makers, which are linked to seed-shattering genes, is needed to employ the molecular marker-assisted selection for efficiently developing shattering-resistant soybean varieties. In this study, a genetic linkage map was constructed using 115 recombinant inbred lines (RILs) developed from crosses between the pod shattering susceptible variety, Keunol, and resistant variety, Sinpaldal. A 180 K Axiom(R) SoyaSNPs data and pod shattering data from two environments in 2001 and 2015 were used to identify quantitative trait loci (QTL) for pod shattering. A major QTL was identified between two flanking single nucleotide polymorphism (SNP) markers, AX-90320801 and AX-90306327 on chromosome 16 with 1.3 cM interval, 857 kb of physical range. In sequence, genotype distribution analysis was conducted using extreme phenotype RILs. This could narrow down the QTL down to 153 kb on the physical map and was designated as qPDH1-KS with 6 annotated gene models. All exons within qPDH1-KS were sequenced and the 6 polymorphic SNPs affecting the amino acid sequence were identified. To develop universally available molecular markers, 38 Korean soybean cultivars were investigated by the association study using the 6 identified SNPs. Only two SNPswere strongly associated with the pod shattering. These two identified SNPs will help to identify the pod shattering responsible gene and to develop pod shattering-resistant soybean plants using marker-assisted selection.

Response of Leaf Water Potential and Growth Characteristics to Irrigation Treatment in Soybean

  • Soybeans [Glycine max (L.) Merr.] are frequently exposed to unfavorable environments during growing seasons and water is the most important factor limiting for the production system. The purpose of this study was to determine the leaf water potential changes by irrigation, and to evaluate the relationships of leaf water potential, growth and yield in soybeans. Three soybean cultivars, Hwangkeumkong, Shinpaldalkong 2, and Pungsannamulkong, were planted in growth chamber and field with irrigated treatments. Leaf water potential of three soybean cultivars was positively correlated with leaf water content during vegetative and reproductive growth stages in growth chamber and field experiments. Leaf water potentials measured for three soybean cultivars under growth chamber were higher than those of under field conditions. Higher leaf water potential with irrigated plots under field was observed compared to conventional plots during reproductive growth stages. Leaf water potentials of three soybean cultivars were continually decreased during reproductive growth stages under field and there was no significant difference among them. Number of leaves, leaf water content, pod dry weight, number of seeds and seed dry weight with irrigated plots were higher than those of conventional plots. The results of this study suggested that leaf water potential could be used as an important growth indicator during the growing season of soybean plants.

Phytochemical Constituents from the Leaves of Soybean [Glycine max (L.) Merr.]

  • Phytochemicals study from the leaves of soybean [Glycine max (L.) Merr.], one of Korean edible plant materials were investigated through various chromatographic procedures. The methanolic leaves extracts of soybean yielded 16 phytochemicals, including 5 isoflavones 1-5, 3 flavones 6-8, 1 flavonol 9, 2 pterocarpans 10 and 11, 2 phenolic compounds 12 and 13, 2 phytosterols 14 and 15, and 1 sugar alcohol 16. The structures were fully characterized by analysis of physical and spectral data and were defined clearly as 4,5,7-trihydroxyisoflavone (1), 4,5,7-trihydroxyisoflavone-7-O-$\beta$-D-glucopyranoside (2), 4,7-dihydroxy-6-methoxyisoflavone (3), 4,7-dihydroxyisoflavone (4), 4,7-dihydroxyisoflavone-7-O-$\beta$-D-glucopyranoside (5), 5,7,4'-trihydroxyflavone (6), 3',4',5,7-tetrahydroxyflavone (7), 3',4',5-trihydroxyflavone-7-O-$\beta$-D-glucopyranoside (8), 3,4',5,7-tetrahydroxyflavonol (9), coumestrol (10), glyceofuran (11), 4-hydroxybenzoic acid (12), methyl-4-hydroxybenzoate (13), soyasapogenol B (14), stigmasterol (15), and D-mannitol (16), respectively. Among them, phytochemicals 7-16 were reported for the first time on the isolation and confirmation from the leaves of this species. These results suggest that the leaves extracts of soybean may possess possible health related benefits to human due to the isolated phytochemicals 1-16 which have been well known potential effects on various chronic diseases.

A Stack of Recessive Alleles of Kunitz Trypsin Inhibitor, Lectin, and Stachyose in Soybean (콩에서 쿠니츠트립인히비터, 렉틴 및 스타키오스에 대한 열성 유전자의 집적)

  • Soybean [Glycine max (L.) Merr.] is one of the major food sources of protein, oil, carbohydrates, isoflavones, and other nutrients for both humans and animals. However, soybean seeds contain antinutritional factors, such as lectin protein, Kunitz Trypsin Inhibitor (KTI) protein, and stachyose. The objective of this research was to stack recessive alleles for development a triple recessive genotype, titilelers2rs2, with low KTI protein, lectin protein, and stachyose contents. Three parents (Gaechuck#2, PI200508, and 14G20) were used to develop the breeding population. The presence or absence of the lectin and KTI proteins was detected by western blotting. The stachyose content in mature seeds was determined by HPLC. Agronomic traits, such as plant type, plant height, maturity date, lodging, seed quality, and 100-seed weight, were evaluated for the four $F_3$ plant strains. One $F_4$ plant strain with the desired agronomical traits was selected. One new strain with the triple recessive titilelers2rs2 genotype was developed. The plant height of the new strain was 51 cm and the 100-seed weight was 31.0 g. The new strain had a yellow seed coat and yellow hilum. The stachyose content of the new strain was 3.8 g/kg. One strain developed in this research will be used to produce improved yellow soybean cultivars that are free of lectin and KTI proteins and low in stachyose content.
