Studies on the Processing of Sardine Meal Koji Using Aspergillus spp. (Aspergillus spp.를 이용한 Sardine Meal Koji제조에 관한 연구)

  • To substitute the soy-sauce 'koji' used in fish sauce processing with molded sardine meal(MSM) 'koji', the culture conditions of MSM be inoculated with Aspergillus spp. were investigated. To prepare the MSM, Asp. awamori(KFCC. 11439), Asp. guercinus (KFCC. l1595), Asp. niger(KFCC. 11239), Asp. oryzae(KFCC. 32343), and Asp. sojae(KFCC. 11559) were inoculated upon the chopped sardine with $20\%$ (w/w) corn starch after steriling it at $121^{\circ}C$ for 15min. Sporulation time cultured with Asp. spp. at $30^{\circ}C$ was 48hrs and color of mycelium on the surface of MSM inoculated with Asp. awamari and Asp. oryzae were black, that of MSM inoculated with the other strains were yellowish brown. The activity of protease and lipase from the MSM were increased till the 72hrs of culture at $30^{\circ}C$, while the content of trimethylamine was decreased after 96hrs of culture period at same condition with exception of Asp. niger. Asp. oryzae and Asp. sojae showed superior in pro-tease and lipase activity in comparison with the other strains, and maximum activity of protease and lipase of MSM was observed after 72hrs of culture period. The optimum pH and temperature for the activity of MSM inoculated with Asp. oryzae and Asp. sojae were pH 9.0, $30\~35^{\circ}C$ and pH $6\~7$, $35^{\circ}C$, respectively.

Comparative Efficacy of Different Soy Protein Sources on Growth Performance, Nutrient Digestibility and Intestinal Morphology in Weaned Pigs

  • To elucidate the efficacy of different soy protein sources on piglet's performance, a total of 280 weaned piglets ($Duroc{\times}Yorkshire{\times}Landrace$, $23{\pm}3$ d of age, $5.86{\pm}0.45$ kg initial BW) were allotted to 5 treatment diets comprising soybean meal (SBM), soy protein concentrate (SPC), Hamlet protein (HP300), fungal (Aspergillus oryzae) fermented soy protein (FSP-A), and fungal plus bacterial (A. oryzae+Bacillus subtilis) fermented soy protein (FSP-B), respectively. Experimental diets for feeding trial were formulated to contain each soy protein sources at 8% level to corn-whey powder basal diet. There were 14 pigs per pen and 4 pens per treatment. Experimental diets were fed from 0 to 14 d after weaning and then a common commercial diet was fed from 15 to 35 d. Also for ileal digestibility studies, 18 pigs were assigned to 6 dietary treatments as N-free, SBM, SPC, HP300, FSP-A and FSP-B with T-canulation at distal ileum for 6 days. At $14^{th}$ d of experimental feeding, the ADG was significantly higher (p<0.05) in SPC fed diet as compared with others. Similar trend was noticed during the 15-35 d and overall study (0-35 d). All the processed soy protein sources tested in this experiment improved (p<0.05) growth than SBM during overall study. The nutrient digestibility of GE, DM, CP and Ca showed lower (p<0.05) values in SBM and FSP-A fed groups than SPC and FSP-B treatments. The apparent ileal digestibility of TEAA, non-TEAA and TAA showed lower (p<0.05) in SBM treatments compared with other soy protein sources. The true ileal digestibility of TEAA, non-TEAA and TAA were lower (p<0.05) in SBM fed group than SPC and HP300 treatments, and lower than FSP treatments though they didn't achieve significant difference (p>0.05). Villous height and crypt depth was not affected by dietary treatments. In conclusion, the growth and digestibility of nutrients in weaned pigs fed SPC was superior to others. Also FSP-A and FSP-B showed improved performance than those fed SBM.

Phytase Production by Rhizopus microsporus var. microsporus Biofilm: Characterization of Enzymatic Activity After Spray Drying in Presence of Carbohydrates and Nonconventional Adjuvants

  • Microbial phytases are enzymes with biotechnological interest for the feed industry. In this article, the effect of spray-drying conditions on the stability and activity of extracellular phytase produced by R. microsporus var. microsporus biofilm is described. The phytase was spray-dried in the presence of starch, corn meal (> $150{\mu}m$), soy bean meal (SB), corn meal (< $150{\mu}m$) (CM), and maltodextrin as drying adjuvants. The residual enzyme activity after drying ranged from 10.7% to 60.4%, with SB and CM standing out as stabilizing agents. Water concentration and residual enzyme activity were determined in obtained powders as a function of the drying condition. When exposed to different pH values, the SB and CM products were stable, with residual activity above 50% in the pH range from 4.5 to 8.5 for 60 min. The use of CM as drying adjuvant promoted the best retention of enzymatic activity compared with SB. Spray drying of the R. microsporus var. microsporus phytase using different drying adjuvants showed interesting results, being quite feasible with regards their biotechnological applications, especially for poultry diets.

Bacillus subtilis Fermentation for Enhancement of Feed Nutritive Value of Soybean Meal

  • In order to increase the nutritional quality of soybean meal (SBM) as an animal feed, Bacillus subtilis TP6, a previously isolated strain from an Indonesian traditional fermented soybean food, Tempeh, was used as a starter organism for solid-state fermentation. In the pre-treated SBM with water content of 60% (v/w), B. subtilis TP6 was grown to a maximum viable cell number of $3.5{\times}10^9CFU/g$. Compared to control, crude protein in Bacillus fermented SBM was increased by 16%, while raffinose, stachyose, and trypsin inhibitors were reduced by 31, 37, and 90%, respectively. The Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis showed that proteins in the fermented SBM were remarkably hydrolyzed into smaller molecular masses, resulting in a decrease in large sized proteins. Our data suggested that B. subtilis fermentation could increase the nutritive value of SBM through reduction of anti-nutritive factors and improvement of protein quality by hydrolysis of soy protein. In addition, B. subtilis TP6 produced a functional ingredient, poly-${\gamma}$-glutamic acid which has various health benefits.

Study on the Use of Sardine Meal Koji and Autolysates from Sardine Meat in Rapid Processing of Sardine Sauce (자가 소화액 및 정어리 기질 코오지를 이용한 속성 정어리 액젓 제조에 관한 연구)

  • Rapid production of sardine sauce using sardine meal koji and autolysate from sardine meat was investigated. The sardine meal koji was prepared by mixing sterillized sardine meal with $10\%$ flour and $10\%$ soy sauce koji, and cultivating the mixture for 48hrs at $30^{\circ}C$ and $80\%$ R. H. For sardine autolysate preparation, chopped sardine was mixed with water (10:8= sardine:water, w/w) and autolyzed for 6hrs at $550^{\circ}C$. The optimum temperature and salinity were $40^{\circ}C$ and $15\%$ for rapid fermentation of sardine sauce. Sardine sauce were prepared experimentally under 10 kinds of conditions and fermented for 20 days. The excellent effects of sardine meal koji and autolysates from sardine meat on rapid processing of sardine sauce were showed in enhancing its flavor and advancing of nitrogenous compounds.

Replacing Fish Meal with a Mixture of Plant and Animal Protein Sources in the Diets of Juvenile Olive Flounder Paralichthys olivaceus (동·식물성 단백질원료 혼합을 이용한 치어기 넙치(Paralichthys olivaceus) 사료 내 어분대체)

  • This study was conducted to investigate the replacement of fish meal (FM) with a plant and animal protein mixture (wheat gluten, soy protein concentrate, tankage meal and poultry by-product meal) in the diets of juvenile olive flounder Paralichthys olivaceus. The basal diet was formulated to contain 65% FM (Con). Four other experimental diets were formulated with alternative proteins replacing 20%, 30%, 40% and 50% of FM (FM20, FM30, FM40 and FM50, respectively). Taurine and betaine were added to the FM replacement diets. Triplicate groups of fish (mean±SD, 5.41±0.01) were fed the diets to apparent satiation for 15 weeks. After the feeding trial, no significant differences were found between any dietary groups in growth performance, feed utilization, survival, hematological parameters or whole-body composition. This result indicates that a proper mixture of the four protein sources with taurine and betaine supplements can be used as FM replacement to reduce FM levels from 65% to 32.5% in juvenile olive flounder diets.

  • Isoflavones are naturally occurring plant chemicals belonging to the 'phytoestrogen' class. The isoflavones are strikingly similar in chemical structure to natural estrogens. The phenolic ring is a key structural element of most compounds that bind to estrogen receptors. Dietary components that recently have received attention for their action as phytoestrogens are soy isoflavones. Soy products are the most significant dietary sources of isoflavones. Recently It is concerned clinical nutrition of isoflavone that is driven by reason of alternative sources of exogenous estrogen are constantly being needed. Estrogen therapy after the menopause offers protection from cardiovascular disease, reduces the extent of osteoporosis and relieves menopausal symptoms. Exogenous estrogen treatment is a fear of possible increased risk of developing breast cancer and because of side effects. Daily intake of soybean or soy food can affirmative effect to disease occurrence, that is based on mechanical investigation, experimental results of animals and human. Research into isoflavone is going on various field to relieve hormone - dependent disease such as cancer, menopausal symptom, cardiovascular disease and osteoporosis. Isoflavone is plenty in soybean meal, soy by-product, but only limited information is available on isoflavone efficacy into animal husbandry. Thus we conducted three experiments to investigate the effects of dietary isoflavone on productivities, antioxidative responses and bone metabolism in poultry. Dietary supplementation of isoflavone resulted in preventing the lipid oxidation of plasma and egg yolk. Dietary isoflavone improved bone development in egg-type growing chicks and broilers in terms of tibial strength. It was suggested that the proper use of feed additives such as isoflavone might provide means of improving antioxidative effect, skeletal strength, egg and eggshell quality.

  • Soybean meal was widely used as a protein source in pig feedstuff because it has a good amino acid balance compared with other vegetable sources. However, soybeans contain trypsin inhibitors and other antinutritional factors which can lead to lower the digestibility of amino acid, and consequently reduce the growth performance. Heat treatment of soybeans is helpful shown to decrease the antinutritional factors and elicit an improved growth performance. Additionally, microbial processe using(HP 100, HP 200 and HP 300), and non-protein constituent removal are suggested to improve the growth performance and nutrient digestibility. Inadequate heat treatment of soybeans gives no damage to adult pig, but it has been shown to decrease nutrient digestibility in young pig. So, soy protein concentrate (SPC) and Isolated soy protein(ISP) were more widely used for nursery pigs than growing and finishing pigs, since SPC and ISP have similar characteristics as milk product.

어린 병아리에서 isoflavones 급여가 골격의 회분 함량 및 물리적 성질에 미치는 영향

  • Considerable beneficial effects on osteoporosis from soy intake in postmenopausal women are being reported (Murkies et al.1998, Arjmandi et al. 1996). Isoflavones(IF), one of phytoestrogenic substances in the soybean, have been suggested for the effect. Our preliminary study showed that even the young chicks fed IF-rich diet tended to have higher bone ash content than those fed IF-poor diet. IF have been reported to decrease bone ash content than those fed IF-poor diet, IF have been reported to decrease bone loss by reducing bone resorption. Soybean meal has been one of the most important protein sources in poultry diets. We assume that the IF intake through dietary soybean meal could give meaningful influences on the birds. This study was carried out to determine effects of dietary IF from soybean sources on bone development in young chicks. Soy protein concentrate (SPC, IF-poor)and an IF concentrate(Phyto-Nutramin) were formulated together with purified-type ingredients to provide three different levels(25, 240 and 480 mg/kg) of total IF. Control diet(240 mg/kg) was prepared with isolated soyprotein(ISP, IF-rich). The diets were fed to 96, 3-day old, layer-type, male chicks, divided into 4 treatments with 3 replications for 3 weeks. Chicks fed the ISP diet had better gain/feed ratios than fed the SPC diets. Birds fed the diets with higher levels of IF tended to show higher values in serum total and ionized Ca% and tibial bone density, length % ash, stiffness and strain. This trend however, appeared less significant at the end of third week. No noticeable differences in sizes of comb and testicle and serum alkaline phosphatase activities were observed among the dietary groups. These results suggest that dietary isoflavones from soybean sources could be associated with chemical composition and physical properties of bone in sizes of comb and testicle and serum alkaline phosphatase activities were observed among the dietary groups. These results suggest that dietary isoflavones from soybean sources could be associated with chemical composition and physical properties of bone in young chicks.

Effect of replacing corn with soy hulls on nutrient digestibility of growing pigs

  • The objective of this study was to determine the effect of replacing corn with soy hulls on nutrient digestibility of growing pigs. Three experimental diets were tested using a 3 × 3 Latin square design using three barrows per group (Landrace × Yorkshire × Duroc, average initial body weight of 36.9 ± 4.0 kg) in individual metabolic cages. The dietary treatments were the control (CON) basal diet (corn-soybean meal based diet), whereas in the test diets, soy hulls were included at 5% (H1), 10% (H2), gradually replacing corn. The daily feed allowance was adjusted to 2.7 times the maintenance requirement for digestible energy (DE) (2.7 × 110 kcal of DE/kg BW0.75). Intake of crude fiber (CF) was different among treatments (p < 0.05). Pigs fed with H2 diet excreted higher concentrations of dry matter (DM), and CF than pigs fed with H1 diet and basal diet (p < 0.05). Pigs fed with H1 and H2 diets excreted higher concentrations of crude protein (CP) than pigs fed with CON diet (p < 0.05). The apparent total tract digestibility (ATTD) of CF from H1 and H2 diets were higher than CON diet (p < 0.05). Pigs fed with H2 diets were significantly lower (p < 0.05) the ATTD of CP than the pigs fed with CON and H1 diets. The current data suggest that 10% inclusion soy hulls can slightly decrease CP digestibility. However, soy hulls considerably increased CF digestibility.It is concluded that 5% soy hulls may be a usable alternative to corn in growing pig diets.
