Brown Color Characteristics and Antioxidizing Activity of Doenjang Extracts (된장의 지용성ㆍ수용성추출물에 대한 갈색 특성 및 항산화 효과)

  • Brown color characteristics and antioxidizing activities were investigated for Doenjang under different processing conditions. Doenjang A was prepared directly from Meju and saline solution whereas Doenjang B was Prepared after separating soy sauce by soaking for 45 days. Both Doenjangs were aged for up to 180 days. Antioxidizing activity was studied in relation to the brown color characteristics using fat-soluble extract and water-soluble extract of Doenjang. The intensity of brown color was higher in the water-soluble Doenjang extract than the fat-soluble Doenjang extract. In the UV-VIS scanning spectra, water-soluble Doenjang extracts showed significant changes as the aging proceeded, but fat-soluble Doenjang extract did not. Antioxidizing activity of fat-soluble Doenjang extract increased as the aging period extended; however, no significant difference was detected in the water-soluble extract. Overall, Doenjang A showed higher contents of amino acids, reducing sugar, brown color, and antioxidizing activity, and the antioxidizing ability was higher in water-soluble Doenjang extract rather than in the fat-soluble Doenjang extract.

Effect of Pine Needle and Green Tea Extracts on the Survival of Pathogenic Bacteria (솔잎과 녹차 추출물이 식중독세균의 생존에 미치는 영향)

  • The sensitivity of various pathogenic bacteria(Listeria monocytogenes, Staphylococcus aureus, Aeromonas hydrophila, Escherichia coli O157:H7 and Salmonella typhimurium) to the pine needle and green tea extracts was tested. Water extract of pine needle(PNW), 70% ethanol extract of pine needle(PNE), water extract of green tea(GTW) and 70% ethanol extract of green tea(GTE) were prepared for the test of antibacterial activty. Tryptic soy broth(TSB) containing 0∼2%(w/v) of pine needle and green tea extracts were inoculated with 10$\^$5/∼10$\^$6/ cells/ml of each bacterium and incubated at 35$\^{C}$ for 24 hours. The standard plate count method was used to measure the inhibitory effect of the extracts. Minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) were derived from the survival curves of pathogenic bacteria. Antibacterial activities of the pine needle and green tea extracts were compared with that of sodium benzoate, a preservative, by clear zone test. L. monocytogenes, S. aureus and A. hydrophila were completely inhibited at 0.4∼1.6% level while E. coli and S. typhimurium were very resistant to the pine needle extracts. Green tea extracts completely inhibited all strains tested at 0.2∼1.0% level and bactercidal to all strains except L. monocytogenes at 0.5∼2.0% level. Antibacterial activities of pine needle and green tea extracts were stronger than that of sodium benzoate. The order of antibacterial activities of pine needle and green tea extracts to the pathogenic bacteria was GTE > GTW > PNE > PNW. This result suggests that green tea extracts can be used as an effective natural antibacterial agent in food.

Sensory Characteristics and Cross-cultural Acceptability of Sweet Crispy Chicken (Dakgangjeong) Prepared Using Sauces with Different Ethnic Korean Style Flavors among Korean and Chinese Consumers (다양한 한국식 소스를 이용한 닭강정의 관능적 특성과 중국 및 한국 소비자의 교차문화적 소비자 기호도)

  • This study investigated sensory characteristics and cross-cultural consumer acceptability of sweet crispy chicken (Dakgangjeong) prepared with six types of Korean-style sauces among Korean and Chinese consumers. The main ingredient(s) of each sauce was soy sauce (SOY), Japanese apricot extract and soy sauce (JASOY), gochujang (SPICY), minced garlic (GARL), and ketchup (KET-I and KET-II); KET-I and KET-II were modified to possess ethnic Korean flavors. In general, Korean and Chinese consumers preferred all types of Dakgangjeong, except for GARL and SPICY, respectively. Least preferred products of each country had the lowest familiarity rating among consumers of the respective countries. Similar to previous studies, these results showed that familiarity is an important factor affecting consumer preference in a cross-cultural context. Particularly, it was found that higher familiarity of the product was not found to influence consumer to like a product, but rather low familiarity seemed to affect consumers to reject a product.

  • Leuconostoc mesenteroides CJNU 0705 was isolated from a breast milk sample and identified by 16S rRNA gene sequencing and confirmed by its ability to produce dextran from tryptic soy agar plates supplemented with 2% sucrose. This strain can absorb various heavy metals including lead (Pb) and cadmium (Cd) which are both found in fine dust and have been shown to be harmful to human skin. In addition, Leu. mesenteroides CJNU 0705 has demonstrated antimicrobial activity against Propionibacterium acnes, the primary causative agent of acne. Given these traits it was natural to evaluate the use of this strain in the fermentation of several natural extracts from green tea, carrot, annual wormwood, parsley, broccoli, and corn silk, which are known to improve skin health, to see if it could increase their dextran content when supplemented with no sucrose, 2% sucrose, or 2% sucrose and 3% yeast extract. The extracts supplemented with both yeast and sucrose were found to produce the most dextran, which was confirmed by the scanning electron microscope (SEM) images. These results suggest that Leu. mesenteroides CJNU 0705 and its fermented natural extracts could be used as functional materials for improving human skin health.

Medium optimization for growth of Bacillus amyloliquefaciens ISP-5 strain and evaluation of plant growth promotion using lettuce (Bacillus amyloliquefaciens ISP-5 균주의 배지 최적화 및 상추를 이용한 식물 생장 촉진 평가)

  • Bacillus sp. is a useful strain for agriculture because it promotes plant growth and controls plant pathogens through a variety of mechanisms. In this study, we obtained a microbial preparation with a high number of viable cells by culturing newly isolated soil bacteria on an optimized medium. Subsequently, we applied this preparation to lettuce to enhance its growth and yield. First, B. amyloliquefaciens ISP-5 was isolated from soil. Next, optimization of culture medium was carried out using 5 L scale fermenters. When culturing B. amyloliquefaciens ISP-5 on this optimized medium, the number of viable cells was approximately 1000 times higher than that obtained from culturing on the commercial medium. Afterwards, the plant growth promotion properties of the ISP-5 strain were evaluated using lettuce as a test plant. Foliar spray treatment of lettuce was carried out by inoculating half the standard concentration suspension (0.5 × 107 cfu/ml). As a result, leaf width increased by 8.6% and leaf length increased by 12.9% compared to the control group. Live weight also increased by 24.2% and dry weight by 23.9%. Considering the results from field test, B. amyloliquefaciens ISP-5 showed potential as a plant growth-promoting bacteria.

Stability of Anti-Yeast Activities and Inhibitory Effects of Defatted Green Tea Seed Extracts on Yeast Film Formation (탈지 녹차씨 추출물의 항효모 활성 안정성 및 산막 형성 억제능 평가)

  • Water and 75% ethanol extracts were prepared from defatted green tea seeds and evaluated for their anti-yeast activities. The antimicrobial activities of defatted green tea seed extracts (DGTSEs) were tested against food-spoilage bacteria, yeasts, and molds. DGTSEs exhibited antimicrobial activities with minimum inhibitory concentrations of $39{\sim}1,250{\mu}g/mL$ against three bacteria, two molds, and all tested yeast strains. Ethanol extract showed higher antimicrobial activity than water extract. The stability of anti-yeast activities of DGTSEs was examined under different conditions of temperature, pH, and NaCl concentrations. The anti-yeast activities of DGTSEs were stable at pH 3~9, 0~20% NaCl, and $100^{\circ}C$ for 30 min. However, anti-yeast activities of DGTSEs decreased upon heating at $70^{\circ}C$ for 24 h or $121^{\circ}C$ for 15 min. DGTSEs were applied to food models to determine their inhibitory effects on yeast film formation. Water and 75% ethanol extracts were effective in preventing yeast film formation at concentrations more than 156 and $39{\mu}g/mL$ in soy sauce, 156 and $78{\mu}g/mL$ in pickle sauce, and 78 and $39{\mu}g/mL$ in kimchi, respectively.

Isolation and Identification of Growth Inhibition Substance on L. monocytogenes from Dystaenia takesimana Kitagawa (섬바디로부터 L. monocytogenes에 대한 생장억제 물질의 분리 및 구조동정)

  • The ethanol extracts and its n-hexane fraction of Dystaenia takesimana Kitagawa exhibited growth inhibition on Listeria monocytogenes ATCC 19111, ATCC 19112, ATCC 19113, ATCC 19114 and ATCC 15313. The minimum inhibitory concentration of the ethanol extract and its n-hexane fraction were 50 ppm and below 30 ppm on Listeria monocytogenes respectively. By silica gel column chromatography, the active fraction A8 was obtained from the ethanol extract of Dystaenia takesimana Kitagawa. After three times of column chromatography, the SBD-1 and SBD-2 were separated from the A8 fraction of the ethanol extract of Dystaenia takesimana Kitagawa. Antimicrobial activity of the SBD-l and SBD-2 was lower than that of the A8. And the A8 exhibited growth inhibition on five strains of Listeria monocytogenes at the level of $10{\sim}30$ ppm and the bactericidal effect was confirmed at same the level. The purified antimicrobial active compound was identified as (9z)-heptadeca-l,9-dien-4,6-diyn-3,8-diol, falcarindiol, by EI/MS, $^{1}H-NMR$ and $^{13}C-NMR$.

Development of Jelly Fortified with Lactic Acid Fermented Prickly Pear Extract (백년초 젖산 발효액을 이용한 젤리의 제조)

  • Rheological properties and sensory characteristics of prickly pear jelly made from κ-carrageenan and glucomannan were examined. Prickly pear extract (PPE) fermented by Lactobacillus casei LS (isolated from soy milk curd residue) exhibited pH 3.12, 1.15% acidity (lactic acid) and 2.7 × 10/sup 9/ CFU/mL lactic acid bacteria. In the effect of concentration of fermented PPE, the jelly containing 4% fermented PPE had higher springiness and slightly lower hardness than that without fermented PPE, and was superior to color and taste. The hardness and brittleness of jellies were enhanced by the addition of 0.25% citric acid. The addition of 0.25% ascorbic acid resulted in the production of soft jelly like pudding. In the effect of concentration of κ-carrageenan, the hardness and springiness of jellies were increased by adding 0.75% κ-carrageenan, but was decreased by adding at the 0.9% level. In the overall preference of jellies including sweet and sour taste, higher score was obtained by the fortification of 0.6% and 0.75% κ-carrageenan, 0.25% citric acid and 30% fructose syrup.

Optimum Formulation of Kochujang Seasoning Sauce with the Addition of Fruit and Vegetable Extract for Pork Bulgogi (돼지불고기용 과채열수추출액 함유 고추장양념소스 최적 배합비 개발)

  • This study was carried out to investigate the antioxidative effect of Kochujang seasoning sauce with the addition of fruit and vegetable extract (FVE) for pork Bulgogi, as well as to establish the optimum mixture ratio of ingredients using mixture experimental design (MED). During 5 months of storage at $-25^{\circ}C$, the thiobarbituric acid (TBA) values of the seasoned pork containing soy sauce (control), Kochujang added group (KG) and Kochujang and FVE added group (KFVEG) were remarkably lower than that of unseasoned pork. Among the seasoned porks, the antioxidative effect of KG was much higher than the control. Moreover, TBA values of KFVEG were significantly lower than those of KG. Therefore, the lipid oxidation stability of seasoned pork Bulgogi was improved by the addition of Kochujang, red pepper powder and FVE into the seasoning sauce. When Kochujang was used in seasoning sauce, the preference scores of pork Bulgogi increased with the increasing amount of red pepper powder and FVE. The optimum mixture ratios of seasoning sauce for pork Bulgogi established by the MED were Kochujang 0.04, red pepper powder 0.20, FVE 0.39 and water 0.37.

Quality Characteristics of Doenjang Prepared with Fermented Hwangchil (Dendropanax morbifera) Extract (황칠 발효액을 첨가한 된장의 품질 특성)

  • This study was performed to investigate the quality characteristics of various salted (8, 10, and 12%) Doenjang prepared with fermented Hwangchil (Dendropanax morbifera) extract as well as the effect of Bacillus methylotrophicus S8 strain, isolated from soy sauce, as a starter culture. After fermentation, the total cell number of low-salted Doenjang (8% and 10%) prepared with fermented Hwangchil was lower than that of Doenjang without fermented Hwangchil. Low-salted Doenjang (8% and 10%) showed higher total acidity, amino acid, and organic acid levels as well lower pH levels than high-salted Doenjang (12%), indicating active fermentation. The highest levels of lactic acid (48.1 mg/L) and glutamic acid (549.0 mg/L) were observed in low-salted Doenjang (8%) fermented with B. methylotrophicus S8 strain. The low-salted Doenjang (8%) fermented with B. methylotrophicus S8 also showed the highest score in overall preference of sensory evaluation. These results indicate that Hwangchil extract fermented with B. methylotrophicus S8 can be used as a starter for making low-salted Doenjang, resulting in improved palatability, and inhibition of abnormal fermentation.
