Freezing Injury of Evergreen Broad-Leaved Trees in Warm-Temperature in the Southern Region in Korea -A Case of Machilus Thunbergii Siebold & Zucc. on the Jungma-dong, Gwangyang-city, Jeollanam Province- (한국 남부지방 난온대성 상록활엽수의 동해피해 -전라남도 광양시 중마동의 후박나무를 중심으로-)

  • This study was carried out to clarify the climatic factors of the freezing injury for the judgement on the adaptation areas of evergreen broad-leaved trees. We surveyed and analyzed the climatic factors of the freezing injury to Machilus thunbergii Siebold & Zucc. on the streets with the analyzation of planting grounds, soil conditions and the surrounding buildings. This study showed that only the minimum air temperature factor out of the other climate elements, which were the annual precipitation; the average annual temperature; the average monthly temperature of january; the average monthly minimum temperature of January; the average temperature of the coldest month; the warmth index and the coldness index, was matched up with the previous theories and reports on the freezing damages on the evergreen broad leaved trees and Machilus thunbergii Siebold & Zucc. The freezing injury of Machilus thunbergii Siebold & Zucc was occurred when the mean minimum temperature of the coldest month(TMC) in winter season fell down below $-4.1^{\circ}C$ and the temperature fell down below $-9.2^{\circ}C$. The freezing damage on Machilus thunbergii Siebold & Zucc surrounded by high buildings were less than those surrounded by low buildings or at non buildings.

Effect of Cattle-Manure Application on Soil Chemical Properties and Crop Yields in Rice-Forage Cropping System

  • The steady increase in livestock industry has greatly required the stable production of food and forage crops. As an alternative, rice-forage cropping system has been attempted in several southern areas. The present study was performed to understand whether an application of cattle-manure compost affects soil chemical properties and crop productivity in rice-forage cropping system, rice ${\rightarrow}$ summer oat ${\rightarrow}$ rye, in Jangheong county, south Jeolla province from 2013 to 2014. Treatments was composed of control (no compost), CM1 (compost application before rice transplanting), and CM2 (two-times compost application, before rice transplanting and after rice harvest), and inorganic fertilizers (N, P, and K) were equally dressed in all plots. Yields of rice were not significantly different between treatments, however, oat production was 1.25-fold higher in CM1 and CM2. Nutrient uptake amounts of rye were higher in CM2 than CM1 and control. Total nitrogen in soil was maintained stable level during crop cultivation. And soil organic matter contents in all treatments were increased by crop residue. Available P_2O_5$ and exchangeable K were increased by cattle manure application. Therefore, it suggested that the amount of nutrient by forage crop residue should be considered in rice-forage multiple cultivation.

Pasteurella anatipestifer infection in ducklings in Korea (오리의 Pasteurella anatipestifer 감염증 발생)

  • This study was conducted to investigate the cause of a new duck disease occured in southern part of Korea. A meat type duck farm located in Kangjin, Chonnam Province had experienced outbreaks of septicemic disease at around 20 days of age in nearly every batch of ducklings from early spring to early summer in 1989. Main symptoms of the birds were eye and nasal discharge, depression, inappetence, diarrhea and nervous signs such as tremor and ataxia. Some birds died suddenly without any signs. Mortality reached from 20% to 80% in severe cases. The autopsy findings of the affected ducklings revealed consistantly severe airsacculitis, fibropericarditis, perihepatitis and occasionaly enteritis and distended ureter with urate deposit. A rod shaped gram-negative bacterium was isolated purely from brain and liver of the diseased ducks by culturing the specimens on blood agar for 48 hours in candle jar. The isolate neither produced hemolysis nor grew on MacConKey Agar. It formed colony relatively slowly being recognizable at least 36 hours after culturing, reaching colony size of about 1mm in diameter at 48 hours culture. The colony looked iridescent under oblique light and had muddy odor. The isolate did not ferment carbohydrates tested but produced gelatinase, hippuricase and oxidase which were considered as characteristics of P anatipestifer. The isolate induced similar signs and lesions when infected experimentally into ducks of 3 to 38 days age via intraperitoneum or intratrachea. However it did not produce any clinical signs wen inoculated via intranasal route. It produced only mild signs in chicken just injected with a very large dose. The bacteria did not produce any signas or lesions in mice. It was concluded through biochemical and physiological tests and animal inoculation tests that the new disease was caused by P anatipestifer.

An Affinity analysis for Rural Amenity Resources according to the Life-Styles of Urbanites (도시민의 라이프스타일에 따른 농촌어메니티자원 선호도 분석)

  • The demand of rural tourism industry has increased among the urbanites in South Korea, in due to the increase of leisure activity and the emergence of ageing society. Rural amenity resources are gaining various interests, in the value creation and promotion of tourism. In this study, the propensities of city dwellers were separated by life-style classification, and each affinity to the rural amenity resources was examined in accordance with the separation. A questionnaire survey of urbanites in the southern area of Gyeonggi-do, the most populous province in South Korea, was conducted to analyze the preference of city dwellers about rural amenity resource and life-style of themselves. For statistical verification, $IBM^{(R)}$ $SPSS^{(R)}$ Statistics 20 software was used for frequency, reliability, factor and multiple regression analysis of this research. The results of the statistical analyses found a noticeable characteristic in life-style classification. The affinities of urbanites can be classified into four congregations of life-style factors in this statistical model. Each congregation of the factors was named as 'Self-development-oriented', 'Leisure-oriented', 'Achievement-oriented', and 'Culture-oriented' life-style, to represent the characteristics for convenience' sake. Among these styles, only 'Self-development-oriented' and 'Achievement-oriented' showed the positive correlation with rural amenity resources in the multiple regression analysis. In addition, the rural amenity resources were also analyzed in accordance with the life-styles classification of urbanites. City dwellers showed the highest interest to the 'natural resource management facility resource' in natural resources, the 'traditional heritage resource' in cultural resources, and the 'community resource' in social resources. Meanwhile, they showed less interest to 'agricultural and scenery resources' in natural resources, 'specialty production resource' in cultural resources, and 'cooperative farming' in social resources. These characteristics can be constructed as meaning that the urbanites who concern self-development and achievement of their lives have high interest in rural amenity resources, and the main interest of them is not 'return-to-the-farm'(歸農) but 'return-to-the-home'(歸村).

A new endemic focus of Heterophyes nocens and other heterophyid infections in a coastal area of Gangjin-gun, Jeollanam-do

  • To know the prevalence of heterophyid trematodes among inhabitants of a southern coastal village, i.e., Sacho-ri, Gangjin-gun, Jeollanam-do(Province), 82 stool samples were examined on helminth eggs and protozoan cysts using Kato-Katz and formalin-ether sedimentation techniques. Total 33 people(40.2%) were positive for trematodes(Heterophyes nocens; 15 people, Pygidiopsis summa; 3, Metagonimus sp.; 4, Clonorchis sinensis; 7, Gymnophalloides seoi; 6) and/or protozoa(Entamoeba coli; 3). Among intestinal trematode egg positive cases, 17 were treated with praziquantel and their whole diarrheic stools were collected after purgation. Adult flukes of H. nocens(number of specimens=1,294), P. summa(386), Stellantchasmus falcatus(5), Stictodora lari(4), and Heterophyopsis continua(1) were collected using a stereomicroscope. To know the source of human H. nocens infections in this village, metacercarial infections in mullets(10) were examined and most H. nocens metacercariae(101/105, 96.2%) were found in the trunk portion. From above results, the surveyed coastal village has been newly known as an endemic focus of human H. nocens infection and consuming raw mullets was the presumable source of human heterophyid infections.

The Deformation Properties and their Formative Processes in Ogcheon Terrain around Ogcheon Town, North Chungcheong Province, Korea (옥천대(沃天帶)의 변형특성(變形特性)과 그 형성(形成) 과정(過程) -충북(忠北) 남서단(南西端)을 예(例)로 하여-)

  • The studied area is situated in tho southern part of the Ogcheon fold belt, where the "Ogcheon Group" is widespread with Jurassic and Cretaceous intrusions. The regional stratigraphy may be divided into three formations, the lower pebble bearing phyllitic, the middle dark grey phyllitic, and the upper black phyllitic formations. For the purposes of the present study, the area has been partitioned to three structural subareas based on major fold axes and fault line. The main subjects of the research have been discussed from two different points, multiple deformation and minor-micro fold styles. The former is analyzed by pebble elongation, folding and lineation in a pebbly formation as well as schistosity, crenulation cleavage and crenulated lineation in the phyllitic formation. The later describes the characteristic features of fold style in each formation and structural subarea. Although minor fold axes within broad pelitic rocks usually tend to trend northeast and to plunge northward, most of these were probably formed by two stages, first a similar fold phase and second a kink fold phase. Measured structural elements indicate that crenulation cleavage in phyllite formed parallel to fold axes of folded pebble followed a NE phase of first deformation and a fold axes of pebbles diagonal to bedding of phyllite are represented by a NW phase of a second deformation. Microscopically, quartz and mica grains form a micro fold enabling one to establish tectonic levels which occur in different deformation modes in each stratigraphic sequence. Microtextures such as crenulation cleavage, kink band, aggregate band of mica and pressure shadows of porphyroblast of quartz related to qarnet and staurolite may suggest the time relation of crystallization and tectonism. The result of this study may conform that three deformation phase, NE first phase-NE second phase-NW phase, occurred in the area.

A Study of the Microstructure and Provenance Area of Bronze Spoons Excavated from the Ok-dong Site in Andong (안동 옥동유적 출토 청동시(靑銅匙)의 미세조직 및 산지연구)

  • A series of scientific analyses, including ICP-AES, microstructure, and TIMS, were carried out on 5 bronze spoons among 14 bronze artifacts excavated from the Ok-dong site in Andong, North Gyeongsang Province. The results of analysis showed that the bronze spoons were formed from an alloy of no more than 75% Cu and 21~23% Sn, placing it in the category of brassware, with trace elements such as Pb, Fe, and Zn. In particular, plumbum Pb contents of which traces of 0.04% were found, appears to have been added as an impurity during smelting. The microstructure of the bronze spoons suggests that the casting process involved using a spoon-shaped hollow mold, followed by an ropid-cooling process after the shape was perfected at approximately $586^{\circ}C$. After comparison of data measured from thermal ionization mass spectrometry (TIMS) of galena from Korea, China, and Japan, it appears that the raw materials for the bronze spoons included galena (lead sulfide) produced from the southern part of China.

Among these species, three species and a genus Zodiomyces (L. proliferans, Z. subseriatus and Z. vorticellarius) are newly reported to the flora of southern Korean region.

Leaf Blight of Kiwifruit Caused by Pestalotiopsis menezesiana (Bress. & Torr.) Bissett and Its Ecopathology (Pestalotiopsis menezesiana에 의한 참다래 잎마름병과 발생생태)

  • Etiological and ecopathological studi88888es on the leaf blight of kiwifruit (Chinese gooseberry), a disease newly found in 1993 in Korea, were carried out in this experiment. The leaf blight symptom was prevalent in the southern area of Chonnam Province where kiwifruits were widely cultivated. The disease occurred from July to fall, causing early defoliation of kiwifruit vines. Small brownish circular or irregular lesions appeared initially on the leaves, starting from the marginal leaf areas, and enlarged. Afterwards small grayish black granules (acervuli) were formed on the lesions. The color of old lesions changed to grayish brown or grayish white. The causal fungus was identified as Pestalotiopsis menezesiana (Bress. & Torr.) Bissett according to its morphological characters and pathogenicity (pathogenic to Chinese gooseberry and grapevine, but not to other hosts of Pestalotiopsis spp.such as tea, apple, persimmon, and chestnut). As P. longiseta and P. neglecta were reported recently in Japan as pathogens causing similar blight symptoms in kiwifruit, P. menezesiana is a new leaf blight pathogen in kiwifruit. Development of the disease was stimulated by wounding, prevailing wind, and drought. The causal fungus was proven to be overwintered in the diseased leaves on the ground surface.

Optimization of Polyethylene Glycol-Mediated Transformation of the Pepper Anthracnose Pathogen Colletotrichum scovillei to Develop an Applied Genomics Approach

  • Colletotrichum acutatum is a species complex responsible for anthracnose disease in a wide range of host plants. Strain C. acutatum KC05, which was previously isolated from an infected pepper in Gangwon Province of South Korea, was reidentified as C. scovillei using combined sequence analyses of multiple genes. As a prerequisite for understanding the pathogenic development of the pepper anthracnose pathogen, we optimized the transformation system of C. scovillei KC05. Protoplast generation from young hyphae of KC05 was optimal in an enzymatic digestion using a combined treatment of 2% lysing enzyme and 0.8% driselase in 1 M NH4Cl for 3 h incubation. Prolonged incubation for more than 3 h decreased protoplast yields. Protoplast growth of KC05 was completely inhibited for 4 days on regeneration media containing 200 ㎍/ml hygromycin B, indicating the viability of this antibiotic as a selection marker. To evaluate transformation efficiency, we tested polyethylene glycol-mediated protoplast transformation of KC05 using 19 different loci found throughout 10 (of 27) scaffolds, covering approximately 84.1% of the entire genome. PCR screening showed that the average transformation efficiency was about 17.1% per 100 colonies. Southern blot analyses revealed that at least one transformant per locus had single copy integration of PCR-screened positive transformants. Our results provide valuable information for a functional genomics approach to the pepper anthracnose pathogen C. scovillei.
