계면활성제 미생물반응기의(혼합 VOCs) 생분해 II: 미생물의 군집해석 (Biodegradation of VOC Mixtures using a Bioactive Foam Reactor II: Analysis of Microbial Community)

  • Toluene을 유일 탄소원으로 배양한 혼합미생물 군으로부터 순수미생물을 분리배양한 후, 염기서열을 분석하여 Pseudomonas sp. TDB4 균주를 얻었다. 이 균주의 VOC 분해능을 평가하기 위해 toluene, benzene, p-xylene, styrene을 대상기질로 하여 회분식 생분해실험을 진행하였으며, 단일 기질인 경우 toluene, benzene, styrene, p-xylene의 순으로 분해능이 좋은 것으로 나타났다. 또한 VOCs 혼합물을 주입하였을 경우에는 모든 조합조건에서 styrene, toluene, benzene, pxylene의 순서로 쉽게 분해하였다. 이러한 결과로부터 VOC가 공존할 경우 toluene과 p-xylene은 다른 VOC의 분해를 촉진시키지만 styrene은 저해물질로 작용함을 알 수 있었다. 한편 연속식 계면활성제 미생물 반응기(Bioactive Foam Reactor, BFR)에 혼합미생물 배양액과 TDB4 배양액을 접종하고 장기 운전하면서 운전특성과 미생물 동적변화를 확인하였다. VOCs 혼합물질 유입농도가 250 ppm 이상에서는 처리효율이 급격히 저하되었는데, FISH 분석 결과에 따르면 동일한 시점에서 TDB4가 차지하는 비율이 전체 미생물의 20% 미만으로 감소하여 BFR의 처리효율과 미생물상은 매우 밀접한 관련이 있음을 알 수 있었다. 본 연구 결과 FISH 분석법을 계면활성제 미생물 반응기의 미생물 군집변화 해석에 유용하게 사용할 수 있음을 확인하였으며, 미생물 반응조를 장기간 운전할 때에는 안정적인 처리효율을 유지하기 위하여 주기적인 식종을 해주는 것이 적절하다고 판단된다.

  • The aim of the present investigation is to determine the nutritional composition of various insects and their potential uses as alternative protein sources in animal diets. The feeding industry requires production systems that use accessible resources, such as feed resources, and concentrates on the potential impacts on production yield and nutritional quality. Invertebrate insects, such as black soldier flies, grasshoppers, mealworms, housefly larvae, and crickets, have been used as human food and as feed for nonruminants and aqua culture while for ruminants their use has been limited. Insects can be mass-produced, participating in a circular economy that minimizes or eliminates food- and feed-waste through bioconversion. Although the model for formula-scale production of insects as feed for domestic animals has been explored for a number of years, significant production and transformation to being a conventional protein resource remains to be deeply investigated. This review will focus on the nutritional composition of various insects and their potential use as alternative protein sources, as well as their potential use to promote and support sustainable animal production. Furthermore, nutritional compositions, such as high protein, lauric acid omega 6, and omega 3, and bioactive compounds, such as chitin, are of great potential use for animal feeding.

Taste-Active and Nutritional Components of Thai Native Chicken Meat: A Perspective of Consumer Satisfaction

  • The taste-active and nutritional components of Thai native, broilers, black-boned, and spent hen chickens were analyzed. The amounts of tasty amino acids especially glutamic acid were the highest in Thai native chicken. The black-boned chicken had the highest arginine content, related to the least amount of consumer satisfaction. Concerning nutritional quality, choline, and taurine were deemed important for brain function. The black-boned chicken showed the highest choline and taurine contents, unlike that of the spent hens. In contrast, broilers presented the highest betaine content, which might be attributed to their lipid metabolism. L-carnitine content was abundant in black-boned and Thai native chickens. Moreover, the amounts of essential amino acids were high in Thai native chicken. In conclusion, black-boned chicken proved to be an excellent nutritional source for health-conscience consumers, whereas the Thai native chickens were flavourful and delicious.

Inhibitory Components from Glycosmis stenocarpa on Pepper Mild Mottle Virus

  • The goal of this study was to identify a source of natural plant compounds with inhibitory activity against pepper mild mottle virus (PMMoV). We showed, using a half-leaf assay, that murrayafoline-A (1) and isomahanine (2) isolated from the aerial parts of Glycosmis stenocarpa have inhibitory activity against PMMoV through curative, inactivation, and protection effects. Using a leaf-disk assay, we confirmed that 2 inhibited virus replication in Nicotiana benthamiana. Using electron microscopy, we found that a mixture of the virus with 2 resulted in damage to the rod-shaped virus.

Nabag (Zizyphus spina-christi) Extract Prevents Aberrant Crypt Foci Development in Colons of Azoxymethane-Treated Rats by Abrogating Oxidative Stress and inducing Apoptosis

  • Zizyphus spina-christi (ZSC) fruit is a rich source of bioactive compounds but any medicinal properties in chemoprevention of colon cancer have hitherto not been studied. The aim of the present study was to examine in vivo protective effects of ZSC water extract on colon carcinogenesis in azoxymethane (AOM)-treated rats. Our results showed that ZSC significantly reduced AOM-induced colonic aberrant crypt foci development and AOM-induced oxidative stress as indicated by restoration of endogenous glutathione depletion and abrogating the impairment of total antioxidant capacity. Caspase-3 cleavage, which has been considered as an apoptotic index, was almost undetectable in AOM-treated rats and ZSC exhibited pro-apoptotic effects evidenced by increased levels of cleaved caspase-3. In the studied model, our findings provide the first in vivo evidence that ZSC extract could inhibit the early stage of colon carcinogenesis by preventing oxidative stress and inducing apoptosis.

별 불가사리(Asterina pectinifera) 조직별 초산추출물의 생리활성 탐색 (Biological Activities of Acidic Extracts of the Starfish Asterina pectinifera)

  • The present study was performed to examine the contraction and relaxation responses of the smooth muscles, and search for antimicrobial and antioxidant activities in the tissues, of the starfish Asterina pectinifera. Frozen samples were extracted with distilled water containing 1% acetic acid. Extracts from all tissues showed potent antimicrobial activity against Escherichia coli D31. Relatively high levels of antimicrobial activity were also detected in the body extracts. Liver, tube feet, and body extracts caused contraction responses in the dorsal retractor muscles (DRM) of the starfish. In contrast, all tissues examined exhibited contractile activity in the esophagus of squid Todarodes pacificus. In addition, liver and gonad extracts caused contraction responses upon application to the intestine of the puffer fish Takifugu pardalis. Relaxation effects on the DRM of starfish were identified in most of the extracts, while no relaxant activity was detected in body extracts. Extracts from all tissues examined also exhibited antioxidant activities. The results of this study suggest that starfish are a potential source of novel bioactive compounds.

Analysis of the Bioactive Metabolites of the Endangered Mexican Lost Fungi Campanophyllum - A Report from India

  • Meghalaya, (in India), in the region of the mega-biodiversity hotspots, is home to a plethora of wild mushrooms. The present study concerns the exploration of the order Agaricales, which includes rare gilled mushrooms considered endangered under IUCN A4c criteria, due to the declining habitat. Electron microscopy of the gill sections revealed an abundance of clamp connections, hyphal cell walls, cystidia, and basidia. This rare species which belongs to the family Cyphellaceae, exhibits morphological and molecular differences from the Cyphella spp. Phylogenetic analysis revealed that it formed a clade under the genus Campanophyllum of the order Agaricales, confirmed by both Neighbor Joining (NJ) and Bayesian phylogenetic analysis. Being nutritionally potent along with its efficient antioxidant value, the fungal extract shows significant rise of two-fold in the antimicrobial activity along with the commercial antibiotics. The compound, Phenol, 2, 4-bis (1, 1-Dimethylethyl) (2, 4-DTBP) showed in ample range in the fungal extract along with aliphatic hydrocarbons, terpene, alcohol and volatile organic compounds on further characterization in GCMS. The present study indicates the endangered Campanophyllum proboscideum could be a rich source of natural antioxidants and an effective pharmaceutical agent.

Ecklonia cava Extract Containing Dieckol Suppresses RANKL-Induced Osteoclastogenesis via MAP Kinase/NF-κB Pathway Inhibition and Heme Oxygenase-1 Induction

  • Ecklonia cava, an edible marine brown alga (Laminariaceae), is a rich source of bioactive compounds such as fucoidan and phlorotannins. Ecklonia cava extract (ECE) was prepared using 70% ethanol extraction and ECE contained 67% and 10.6% of total phlorotannins and dieckol, respectively. ECE treatment significantly inhibited receptor activator of nuclear $factor-{\kappa}B$ ligand (RANKL)-induced osteoclast differentiation of RAW 264.7 cells and pit formation in bone resorption assay (p <0.05). Moreover, it suppressed RANKL-induced $NF-{\kappa}B$ and mitogen-activated protein kinase signaling in a dose dependent manner. Downregulated osteoclast-specific gene (tartrate-resistant acid phosphatase, cathepsin K, and matrix metalloproteinase-9) expression and osteoclast proliferative transcriptional factors (nuclear factor of activated T cells-1 and c-fos) confirmed ECE-mediated suppression of osteoclastogenesis. ECE treatment ($100{\mu}g/ml$) increased heme oxygenase-1 expression by 2.5-fold and decreased intercellular reactive oxygen species production during osteoclastogenesis. The effective inhibition of RANKL-stimulated osteoclast differentiation and oxidative stress by ECE suggest that ECE has therapeutic potential in alleviating osteoclast-associated disorders.

  • Medicinal herbs are significant source of synthetic and herbal drugs. In the commercial market, medicinal herbs are used as raw drugs, extracts or tinctures. Isolated active constituents are used for applied research. For the last few decades, phytochemistry (study of plants) has been making rapid progress and herbal products are becoming popular. According to Ayurveda, the ancient healing system of India, the classical texts of Ayurveda, Charaka Samhita and Sushruta Samhita were written around 1000 B.C. The Ayurvedic Materia Medica includes 600 medicinal plants along with therapeutics. Herbs like turmeric, fenugreek, ginger, garlic and holy basil are integral part of Ayurvedic formulations. The formulations incorporate single herb or more than two herbs (poly-herbal formulations). Medicinal herb contains multitude of chemical compounds like alkaloids, glycosides, saponins, resins, oleoresins, sesquiterpene, lactones and oils (essential and fixed). Today there is growing interest in chemical composition of plant based medicines. Several bioactive constituents have been isolated and studied for pharmacological activity. R. cordifolia is an important medicinal plant commonly used in the traditional and Ayurvedic system of medicine for treatment of different ailments. This review illustrates its major constituents, pharmacological actions substantiating the claims made about this plant in the traditional system of medicine and its clinical applications.

오미자씨에서 추출된 유지의 성분 및 항산화 효과 (The Antioxidative Effect and Ingredients of Oil Extracted from Schizandra chinensis Seed)

  • The objectives of this study were to investigate the antioxidant activity of Schizandra chinensis seed oil and its active ingredients. Schizandra chinensis seed oil content extracted with hexane was 36.06%. Schizandra chinensis seed oil extracted with hexane was purified during 20 min at $85^{\circ}C$ with phosphoric acid 0.15% for degumming and 20 min at $80^{\circ}C$ with 3 M NaOH 1% for deaciding. The purified oil consisted of unsaturated fatty acid (88.7%), fatty acid (9.97%), and so on. The major unsaturated fatty acids of purified oil were linoleic acid (71.1%) followed by oleic acid (15.7%), while the main saturated fatty acid was palmitic acid (6.56%). The purified oil was found that contents of phenolic compounds, vitamin A, and E were 1.45 g/100 g, 1494.86 RE/100 g, and 0.58 mg ${\alpha}$-TE/100 g, respectively. Schizandra chinensis seed oil exhibited strong antioxidant activity (91.7%) as compared to grape seed oil and canola seed oil with 87.4% and 85.1% in the DPPH assays. Present results suggest that Schizandra chinensis seed oil could be potentially used as bioactive source for health and preventing numerous diseases.
