Plant Regeneration from Floral Stem Cultures of Nymphoides indica (L.) O. Kuntze. via Somatic Embryogenesis (어리연꽃 (Nymphoides indica (L.) O. Kuntze) 화경 배양으로부터 체세포배발생을 통한 식물체 재생)

  • Plant regeneration system from floral stem of Mymphoides indica via somatic embryogenesis was established. After four weeks of culture onto 1/2MS medium containing 2,4-D, pale-yellow globular structures and calluses were formed on the cut surface of floral stem explants. Upon transfer to 1/2MS basal medium, pale-yellow globular structures were developed into somatic embryos and normal plantlets. These results indicated that pale-yellow globular structures and calluses from floral stem were globular embryos and embryogenic calluses, respectively. The frequency of embryogenic callus formation from floral stem was reached to nearly 100% when floral stem was cultured onto 1/2Ms medium supplemented with low concentration of 2,4-D (0.1 to 0.3 mg/L). However, the higher concentration of 2,4-D resulted in decrease of the frequency of embryogenic callus formation. In this study, low concentration of 2,4-D had a stimulative role in embryogenic callus formation, whereas BA showed inhibitory role in callus formation. In comparison to floral stem, leaf explants showed low frequency of embryogenic callus formation. The highest frequency of embryogenic callus formation from leaf explants was 9.5% when leaf explants were cultured onto 1/2MS medium supplemented with 0.3 mg/L of 2,4-D. The plant regeneration system of Nymphoides indica established in this study, might be applied to mass proliferation, conservation of genetic resources and genetic transformation for molecular breeding.

Production of Triploid Somatic Hybrids Between Mandarin and Grapefruit through Electrofusion

  • Somatic hybrids were produced by electrofusion between embryogenic callus protoplasts of 'Syougun' mandarin and leaf protoplasts of grapefruit. Hybridity of the two plants was confirmed by leaf morphological characteristics and random amplified polymorphic DNA (RAPD) analysis. The cpDNA analysis using PCR-RFLP could not distinguish those of both parents. These plants showed normal growth and had chromosome number of 27. These unexpected triploid somatic hybrids might be derived from fused cells between diaploid protoplast of embryogenic calli and diploid protoplast of leaf, because polysomaty, a mixture of haploid cells and diploid cells was observed in the lactose medium-pretreated embryogenic calli of 'Syougun' by flow cytomehy analysis.

Yellowish Friable Embryogenic Callus (YFEC) Production and Plant Regeneration from Immature Embryo Cultures of Domestic Maize Cultivars and Genotypes (Zea may L.) (국내 옥수수 품종 및 계통의 미숙배 배양으로부터 Yellowish Friable Embryogenic 캘러스 (YFEC) 생산과 식물체 재생)

  • Immature embryos of 3 cultivars (Du Me Chal, Mi Baek Chal, Heug Jeom Chal) and 5 genotypes (HW1, KL103, HW3, HW4, KW7) were cultured on medium containing MS salts, Eriksson's vitamins, 1 mg/L 2,4-dichlorophenoxyacetic acid, 25 mM proline, 100 mg/L casamino acid, 3 mM MES, 1.7 mg/L $AgNO_3$ and 20 g/L sucrose (SIM). Frequency of somatic embryo formation on explant of immature embryos showed in HW1 (45.20%), KL103 (5.75%), HW3 (37.20%), HW4 (30.10%), KW70 (55.20%), Mi Baek Chal (18.74%), Heug Jeom Chal (22.41%), Du Me Chal (36.72%) and Hi II type (<10%), respectively. Yellowish friable embryogenic callus (YFEC) such as type II callus of Hi II genotype only produced from the HW3 and Heug Jeom Chal, whereas other cultivars and genotypes were directly formed somatic embryos with late-embryonic stages or expanded yellowish compact somatic embryo with morphological abnormality. The yellowish friable embryogenic callus (YFEC) could be proliferated on the same medium, which were maintained embryogenic capacity for 6 months over. Upon transfer to first regeneration and second regeneration medium, somatic embryos converted to plantlets at a frequency of approximately 100%. However, the expanded somatic embryos with abnormal morphology were slowly proliferated when subcultured on the same medium, and some of them were degenerated or converted to plantlets at a frequency of approximately 25%. Accordingly, The Heug Jeom Chal and HW3 genotype will be further used for development of high frequency transformation system in domestic maize germplasm.

Variations in Sweetpotato Regenerates from Gamma-ray Irradiated Embryogenic Callus

  • Radiation induced and somaclonal variations were investigated in the regenerates from gamma irradiated and controlled embryogenic callus (EC) of sweetpotato cvs., Yulmi and White Star by morphological, RAPD and AFLP analysis. Most (approx. 90%) of the EC produced somatic embryos developed into plantlets after being transferred to the auxin-free medium. The frequency of morphological variants derived from the irradiated callus ranged from 3 to 7.8% compared to 0.1-1.1% of that derived from the non-irradiated. Morphological variants were selected from the regenerates and analyzed by RAPD and AFLP procedures. RAPD polymorphisms of Yulmi and White Star regenerates from irradiated calli were 8.8% and 6.1%, respectively. However, the polymerphisms among regenerates from the non-irradiation treatment in these two cultivars were non-detectable and 3%, respectively. AFLP polymorphisms of Yulmi and White Star regenerates from irradiated calli were 29.9% and 28.6%, respectively. while the frequencies for those form non-irradiated calli were 8.5% and 5.6%, respectively. Initiation of embryogenic callus from mature zygotic embryos in Japanese larch (Larix kaempferi)

  • Embryogenic callus (EC) was created from mature embryos of Larix kaempferi. With the mature embryos, keeping the culture in dark conditions throughout the experiment (38.2%) seemed to give better results than exposing them to 16 h light ($25{\mu}Em^{-2}s^{-1}$) for the first week (21.9%). EC was obtained most frequently from Quoirin and Lepoivre (LP) mediums with 1.0 mg/L 4-amino-3,5,6-trichloropicolinic acid (Picloram), plus 1.0 mg/L benzyladenine (BA) (62.8%) or Litvay's medium (LM) containing 1.0 mg/L p-chlorophenoxyacetic acid (pCPA) plus 1.0 mg/L BA (62.8%) treatment. In both cases, best results were obtained when zygotic embryos were cultured in darkness. As for the effective sucrose concentration on initiation of EC, 29.2 mM sucrose (38.6%) gave the best results.

  • Embryogenic callus induces from shoot tip and leaf segment of Zanthoxylum piperitum for producing somatic embryogenesis and plant regeneration were cultured in vitro on Murashige and Tucker's(MT) medium treated with casein hydrolysate $NH_4NO_3$, $KNO_3$ and plant growth regulator. The most effective somatic embryogensis was observed in the medium added by two fold $NH_4NO_3$(3300mg/l)+2. 4-D 0.1mg/l and $KNO_3$(3800mg/l)+2.4-D 0.1mg/l. Also, MT medium supplemented with casein hydrolysate 700mg/l added by 2, 4-D 0.1mg/l were effective in obtainingn somatic embryos from embryogenic callus The effect ofm MT medium supplemented with casein hydrolysate without 2, 4-D was lower than that with (3300mg/l) 2, 4-D for the formation of somatic embryos.

Optimization of Embryogenic Callus Induction and Plant Regeneration in Orchid Coelogyne cristata

  • An efficient protocol was established for high frequency somatic embryogenesis through a callus culture of Coelogyne cristata. The best frequency of callusing was obtained from a PLB segment (3-5 mm) cultured on MS medium supplemented with coconut water (CW) and a combination of both 3 $mg{\cdot}L^{-1}$ of 2,4-D and BA. When the calli were sub-cultured on the MS medium without any PGRs, the average number of somatic embryos were higher than those with PGRs treatment. NAA is the most critical factor among PGRs, which dramatically hindered for the formation of a somatic embryo. The efficacy of the addition of coconut powder (CP) for somatic embryogenesis was almost the same in all treatments. However, the number of somatic embryos formed distinctly depended on age of the callus. The somatic embryos converted into healthy plants with well-developed shoots on the same medium. Plantlets showed the best responses of root and shoot growth when transferred to $\frac{1}{2}$ MS medium containing 1.5 $g{\cdot}L^{-1}$ of activated charcoal. All plants with above 3.0-cm-high were successfully acclimatized in the greenhouse.

Totipotential, Morphological, Biochemical Comparisons between Nonembryogenic Callus and Embryogenic Callus in Water Dropwort(Oenanthe stolonifera DC) (미나리에서 비배발생캘러스와 배발생캘러스간의 분화능력 및 해부학적, 생화학적 특성비교)

  • The embryogenic callus (EC), from which somatic embryos could be induced, was compared with nonembryogenic callus(NE) to study the origin and features of totipotent cell in water dropwort (Oenanthe stolonifera DC). To induce and maintain of EC and the NE, meristematic stem and immature floret were inoculated in MS media supplemented with 1 mg/L 2,4-D, and with 2.5 mg/L NAA and 5mg/L BA, respectively, The EC was not induced from the NE even after subculturing in MS medium supplemented with 1 mg/L 2,4-D. Plantlets were not regenerated from the NE in hormone-free medium. In histochemical comparison of the EC with the NE by light microscopy, the EC had smaller cells in size, dense cytoplasm, and more starch granules of cells compared to the NE cells. The cell from the EC, as observed by transmission electron microscopy, had smaller vaculoes, well developed ribosomes, mitochondria, and endoplasmic reticulum, whereas the cells from the NE had larger vacuoles and underdeveloped organelles. In protein pattern from NE, EC and Somatic embryo (SE), as analyzed by SDS polyacrylamide gel electrophoresis, different proteins specific for tissue were observed: 17 and 28 KD for NE, 50, 52, 57, 66, 68 KD for EC and 20 KD for SE. DNA polymorphism was also observed between EC and NE as analyzed by RAPD (randomly amplified polymorphic DNA) method. The origin of totipotent stem cell and the relationship between irreversible genomic change arose in differentiation and the loss of totipotency in plant were discussed.

  • Cucumber (Cucumis sativus L.) plants were regenerated through organogenesis and somatic embryogenesis in cotyledon and hypocotyl cultures. The shoots were efficiently formed on the basal region of cotyledons cultured on MS medium containing 1.0㎎/L zeatin and 0.1㎎/L IAA in all cultivars used. Embryogenic calli were formed on hypocotyl segments cultured on MS medium containing 1.0㎎/L 2,4-D in cv. group 'Nakhab' and maintained by consecutive subculture on the same medium every 2-3 weeks without loss of embryogenic ability. Upon transfer to MS basal medium, high frequency somatic embryogenesis was achieved easily from embryogenic callus. Regenerated plantlets through organogenesis and somatic embryogenesis were transplanted to pots and gradually acclimatized to greenhouse condition where they subsequently produced fruits.

Agrobacterium-mediated Transformation of Eleutherococcus sessiliflorus using Embryogenic Calli and the Regeneration of Plants (오갈피(Eleutherococcus sessiliflorus)의 배형성 세포를 이용한 고빈도 형질전환 및 재분화)

  • We have developed a reliable and high-frequency genetic transformation and regeneration system via somatic embryogensis of Eleutherococcus sessiliflorus. Embryogenic callus obtained from seed were co- cultivated with Agrobacterium tumefaciens strain EHA101/pIG121Hm harboring genes for intron-$\beta$-glucoronidase(GUS), kanamycin and hygromycin resistance. Following co-cultivation, two types of samples(fine embrogenic calli and early globular embryo clusters) were cultivated on Murashige and Skoog(MS) medium containing 1 mg/L2.4-D for 3day in dark. Transient expression of GUS gene was found to be higher in the early globular embryo clusters than in the embryogenic calli. Also, co-cultivated period affected expression of GUS gene; the best result was obtained when globular embryo clusters were co-cultivated with Agrobacterium for 3 days. Subsequently, this callus transferred to selective MS medium containing 1mg/L2.4-D, 50mg/L kanamycin or/and 30mg/L hygromycin and 300mg/L cefortaxime. These embryogenic calls were subcultured to the same selection medium at every 2 weeks intervals. Approximately 24.5% of the early globular embryos co-cultivated with Agrobacterium for 3days produced kanamycin or/and hygromycin-resistant calli. Transgenic somatic embryos were converted into plantlets in half strength MS medium supplemented with 3mg/L GA$_3$ kanamycin and were confirmed by GUS histochemical assay and polymerase chain reaction analysis. Genomic Southem blot hybridization confirmed the incorporation of NPT II gene into the host genome.
