Optimization of Headspace Analysis of Volatile Compounds from Oxidized Fish Oil

  • Headspace volatile compounds of oxidized fish oil were analyzed by the combination of hexane solvent or solid phase microextraction, gas chromatography and mass selective detector. The optimum condition of headspace analysis by hexane trapping was 23 min absorption time, 96$^{\circ}C$ sample temperature and 20 mL/min air flow rate. The numbers of volatile compounds identified by solvent trapping and SPME were 35 and 14, respectively. Groups having the largest amount and many kinds were hydrocarbons and aldehydes, respectively. The numbers of aldehydes were 15 and 6 for solvent trap and SPME, respectively. These basic data could be used as indicators for the quality changes of fish oil.

  • 피등어꼴뚜기를 가열가공할 때 발생하는 취기를 제거할 목적으로 취기성분을 gas chromatography로 공석한 것을 단보로서 요약하면 다음과 같다. 1. 피등어꼴뚜기 취기성분 추출에는 methanol이 가장 적합하였다. 2. 빙수trap에서 25개 peak, dry ice-acetone trap에서 32개 peak가 검출되었고, 그 중 acetic acid, butyric acid, iso-valeric acid, valeric acid, caproic acid 등 5성분을 동정하였다. 3. 액체질소 trap의 head space vapor에서 11개 peak가 검출되었고, 그 중 mothylamine, trimethylamine, dimethylamine, ethylamine, isopropylamine등 5성분을 동정하였다.

추출방법에 의한 아니스의 휘발성 성분 조성 비교 (A Comparison of Different Extraction Methods for the Volatile Components of Anise(Pimpinella anisum L.))

  • 식물체의 향기성분을 분리하는데 사용되는 몇가지 방법을 비교하기 위해서 headspace(purge & trap)법, simultaneous distillation extraction(SDE)법, steam distillation법, 용매추출법 등으로 분리된 휘발성 성분의 조성차이를 분석하여 각각의 사용된 방법들의 특징을 비교하였다. 휘발성 성분 분석시료로는 Anise(Pimpinella anisum L.)씨앗을 사용하였으며 분리된 휘발성분중 GC/MSD를 이용하여 43종의 성분을 확인하였다. Headspace (purge & trap)법에서는 휘발성이 강한 2-butanol, pentanal, 3-hexen-1-ol 등과 같은 alcohol류 및 aldehyde류 성분들을 주요성분으로 확인할 수 있었으며, 용매추출에서는 비점이 높은 성분들인 myristic acid, oleic acid 등의 acid류 성분들과 ethyl hexadecanoate, ethyl octadecanoate 등과 같은 ester 화합물들이 주요성분으로 확인되었다. SDE법과 steam distillation법에서 추출된 성분의 조성은 유사하였으며 terpene 화합물들이 주요성분으로 확인되었다. Anise의 주성분인 anethole은 SDE법, headspace(purge & trap)법으로 추출하였을때 가장 많이 추출되었다.

흡착제를 이용한 수중의 미량 이취물질 분석법 개발에 관한 연구 (A Study on the Development of Analytical Method for Micro-Odorous Compounds in Water)

  • Even if odorous compounds remained very low concentration in water, it caused strong odor. Because Geosmin and most of odorous compound had very vaporization, those were difficult to analyze with GC/MSD and Purge & Trap. So, we needed pre-treatment method for decreasing amounts of extracting solvents, improving recovery efficiencies and increasing analytical efficiencies. This study developed efficient technology for analyzing odorous compounds, using various adsorbents and extracting solvents. The optimum adsorbent was XAD resins. Especially, XAD-2, XAD-7 and XAD-2010 were superior, but XAD-2 of these and MTBE was the optimum extraction solvent. Other extraction solvent's efficiency was in order of MTBE>Dichloromethane>n-Hexane>Diethylether. The optimum NaCl dosage for increasing efficiency was 5g in liquid-liquid extraction method. The shaking time(0∼24hr) had no concern with adsorption efficiency. The optimum adsorbent was XAD-2 resin and extraction solvent was MTBE. Dosing NaCl, adsorption efficiency was increased in liquid-liquid extraction method, but NaCl has no effect on liquid-solid extraction method. In this experimental results, this method will apply to not only Geosmin but other well-known odorous compounds (2-MIB, IBMP, IPMP, TCA) and algae toxins (Mycrocystin, Anatoxin etc)

Identification of Coffee Fragrances Using Needle Trap Device-Gas Chromatograph/Mass Spectrometry (NTD-GC/MS)

  • A fast and simple sampling and sample preparation device, (NTD) has been developed and applied to sample and analyze volatile components from ground coffee beans. Coffee fragrances and other volatile organic compounds (VOCs) were sampled by the NTD and then analyzed by gas chromatograph-mass spectrometry (GC/MS). Divinylbenzene (DVB) particles (80/100 mesh size) were the sorbent bed of the NTD. More than 150 volatile components were first identified based on the database of the mass library and then finally 30 fragrances including caffeine were further confirmed by comparing experimental retention indices (i.e. Kovat index) with literature retention indices. Total sampling time was 10 minutes and no extra solvent extraction and/or reconstitution step need. Straight n-alkanes (C6-C20) were used as retention index probes for the calculation of experimental retention indices. In addition, this report suggests that an empty needle can be an alternative platform for analyzing polymers by pyrolysis-GC/MS.

Removal of OH Spectral Interferences from Aqueous Solvents in Inductively Coupled Plasma-Atomic Emission Spectrometry (ICP-AES) with Ar Cryogenic Desolvation

  • The spectral interferences of OH from aqueous solvents in ICP-AES have been studied and eliminated using a cryogenic argon trap. The prominent lines of Bi I 306.772 nm, Al I 309.271 nm, and V II 310.230 nm, which are very seriously overlapped with the OH band, were examined. With an extended torch and high tangential flow of 20 L/min, water vapor from air entrainment was prevented. The combination of a condenser and argon cryogenic trap was able to eliminated most of water vapor carried by the argon sample gas. Removal of OH spectral interference could extend the linearity of the calibration curve 5-10 times on the lower concentration for ICP-AES. Interference Equivalent Concentration (IEC) has been reduced to 5.6, 5.9, and 12.4 times for Bi, Al and V, respectively.

의왕시 백운산 주변 유류 오염도 조사 및 현장 복원 기초실험 (Survey of the oil contaminated level and preliminary field bioremediation test in the Mountain Baegun at Uiwang city)

  • 본 연구의 목적은 경기도 의왕시 소재 백운산 주변의 유류로 인한 토양오염도를 조사하고 복원에 필요한 기초자료를 마련하는 데 있다. 토양의 유류오염도를 측정하기 위해 BTEX와 TPH를 분석하였으며, 토양의 물리적 특성은 pH, 유기물함량, 함수율, 투수계수, 비중, 공극율을 측정하였다. BTEX는 GC-FID와 Purge & Trap을 사용하여 분석하였으며, TPH는 가속용매추출기(Accelerated Solvent Extractor)를 이용하여 토양시료를 전처리 하였으며 GC-FID로 분석하였다. 분석 결과 TPH의 경우 최고 24,773mg/kg, BTEX는 101.7mg/kg가 검출되어 TPH의 토양오염대책기준인 5,000mg/kg, BTEX의 토양오염우려기준인 80mg/kg을 많은 지점에서 초과하고 있었다. 따라서 백운산 지역은 오염지역의 토양복원이 반드시 필요함을 알 수 있다. 한편 연구대상 지역 중 오염도가 높은 지점에서 본 연구실에서 개발한 미생물제제를 사용하여 5주 동안 오염도의 회복정도를 조사하였다. 실험결과 3주만에 대부분의 오염물질이 분해되었고, 5주 후에 초기농도의 95%의 TPH가 분해됨을 확인할 수 있었다. 따라서 백운산 토양오염 현장에는 본 연구팀이 사용한 미생물제제를 이용하여 복원하는 것이 매우 효과적인 것으로 사료된다.

용매 포집장치가 연기 중 Carbonyl 화합물의 분석에 미치는 영향 (The Effect of Solvent Trap Apparatus on the Analysis of Carbonyl Compounds in Cigarette Smoke)

  • Among the most popular means of mainstream smoke collection for specific analysis are solvent traps. Determination of the carbonyl compounds as their 2,4-dinitrophenylhydrazones(DNPHs) by HPLC offers increased sensitivity and reproducibility in the analysis of mainstream cigarette smoke. In this work, the mainstream smoke of a reference cigarette(lR4F) was collected in four kinds of gas washing bottles and their effects on the analysis of carbonyl compounds were investigated. The results show that the porosity of fritted glass filter in gas washing bottles exerted profound effect on the composition of the carbonyl compounds in mainstream smoke.

시료상층부 원판 형태 단일 다공성 물질을 이용한 바닐라 향수의 휘발성 아로마 성분 추출 분석 (Analysis of volatile aroma compounds from vanilla perfume using headspace disk type monolithic material sorptive extraction)

  • 본 연구에서는 시료상층부 원판 형태 흡탈착 추출법(HS-MMSE)을 개발하여 검증하고 바닐라 향수의 휘발성 아로마 성분들의 기체 크로마토그래피-질량 분석법에 적용하였다. HS-MMSE 방법은 실리카에 octadecyl silane (ODS)과 활성탄을 결합하여 만든 단일 다공성 물질(monolithic material, MonoTrap)을 흡착제로 이용한다. MonoTrap은 시료상층부에서 아로마 성분들을 흡착하고 100 ${\mu}L$ 소량의 용매로 추출한다. HS-MMSE를 이용하여 바닐라 향수로부터 12개의 성분들을 성공적으로 분석하였다. 그 중 benzyl acetate, linalyl acetate, vanillin, ethyl vanillin을 이용하여 추출 효율에 영향을 주는 파라미터들에 대한 실험을 통해 추출 조건을 최적화하였다. 또한, 대상 물질들에 대한 분석 방법의 검증 실험을 진행하였다. 그 결과, 검출한계, 정량한계는 각각 8.35~13.76 ng, 27.82~45.88 ng 범위였고 $r^2$값이 0.9888 이상인 우수한 직선성과 양호한 회수율 및 재현성을 얻었다. 이러한 결과들을 바탕으로 원판 형태 MonoTrap을 이용한 HS-MMSE 방법은 바닐라 향수의 휘발성 아로마 성분들을 분석하는데 효과적인 방법임을 확인하였다.
