호도약침액(胡桃藥鍼液)이 가토(家兎) 뇌(腦)의 Synaptosome에서 Oxidant에 의한 물질이동계(物質移動系)의 장애(障碍)에 미치는 영향(影響) (The Effect of Juglandis Semen Extract Solution on Oxidant-Induced Alteration of Glutamate Uptake in Rabbit Brain Synaptosome)

  • This study was undertaken to determine whether Juglandis semen extract solution (JLS solution) exerts protective effect against oxidant-induced inhibition of glutamate uptake by synaptosomes. Synaptosome was prepared from rabbit brain cortex. Glutamate uptake increased by incubation time during 10 minutes, which was significantly inhibited by 1mM t-buthylhydroperoxide(t-BHP). JLS solution prevented t-BHP-induced inhibition of glutamate uptake in a dose-dependent manner. t-BHP reduced glutamate uptake in dose-dependent fashion, which was significantly prevented by 2% JLS solution. t-BHP(1mM) and $ascorbate/Fe^{2+}(50/1{\mu}M)$ increased lipid peroxidation in synaptosomes by 5-fold, and it was significantly prevented by 2% JLS solution. $HgCl_2(0.1mM)$ inhibited glutamate uptake and increased lipid peroxidation. These changes were prevented by 2% JLS solution. Synaptosomal Na-K-ATPase activity was inhibited by t-BHP(1mM) and $H_2O_2(50mM)$, which was prevented by 2% JLS solution. The results indicate that JLS solution prevents oxidant-induced inhibition of glutamate by synaptosomes, and this may result from inhibition of lipid peroxidation induced by oxidants.

Saccharomyces uvarum에 의한 중금속 생체흡착에 관한 연구 (Biosorption of Heavy Metals by Saccharomyces uvarum)

  • The waste biomass of Sacchromyces uvarum, used in fermentation industries to produce ethanol, were studied for their ability to absorb various heavy metal ions. Heavy metal ions studied in this research were Cd, Co, Cr, Cu, Ni and Pb. The order of the sorption capacity was Pb>Cu>Co=Cr=Cd>Ni. The living Sacchromyces uvarum exhibited higher metal-uptake capacity than the dead Sacchromyces uvarum. After we compare the uptake capacity of the Sacchromyces uvarum for individual metal ions with for a mixture of them, the following was observed: in the mixed heavy metal solution the uptake capacity was decreased than the one heavy metal solution. The selective uptake was observed when all . the heavy metal ions were dissolved in a mixed solution. The adsorption isotherm modelling was decribed with the Langmuir and Freundlich model. The results were in good agreement with the Langmuir model.

Water Uptake and Tensile Properties of Plasma Treated Abaca Fiber Reinforced Epoxy Composite

  • This work presents the tensile properties and water uptake behavior of plasma treated abaca fibers reinforced epoxy composites. The composites were prepared by vacuum assisted resin transfer molding. The effects of treatment on tensile properties and sorption characteristics of abaca fiber composites in distilled water and salt solution at room temperature were investigated. The tensile strength of the composites increased with plasma treatment. With plasma treatment, an improvement of 92.9% was obtained in 2.5 min exposure time in plasma. This is attributed to high fiber-matrix compatibility. Less improvement on tensile properties of hybrid treatment of sodium hydroxide and plasma was obtained. However, both treatments reduced overall water uptake in distilled water and salt solution. Hydrophilicity of the fibers decreased upon plasma and sodium hydroxide treatment, which decreases water uptake.

갈색해조류에 의한 카드뮴의 흡착 및 탈착 특성 (Characteristics of Cadmium Biosorption and Desorption by Brown Marine Algae)

  • The biosorption and desorption of Cd were carried out using brown marine algae, known as the good biosorbent of heavy metals. The content of alginate bound to light metals could be changed by the physical and the chemical pretreatment of Sargassum fluitans biomass. The Cd uptake was independent of the alginate content. In case of protonated biomass, Cd uptake was the lowest because the alginic acid of biomass was dissolved to cadmium solution during the biosorption. The maximum Cd uptake of Sargassum biomass was ranged from 79 mg/g to 139 mg/g. In case of raw biomass, the higher the alginate content of biomass, the higher was the Cd uptake. 100% of Cd and light metals sorbed in the biomass were eluted at 0.1N HCI(pH 1.1). However, the elution efficiency in $CaCl_2$ and $Ca{(NO_3)}_2$solution was varied by the concentration, the solid to liquid ratio and the pH of calcium solution. The distribution coefficient between Cd and protons in the desorption solution at pH ranged from 1.6 to 2.9 was observed on the constant stoichometric coefficient(1.3).

보존용액에 함유된 당, 살균제 및 ABA가 절화국화의 수확후 품질에 미치는 영향 (Effect of Sucrose, Germicides and ABA in the Preservatives on Postharvest Quality of Cut Chrysanthemum 'Kyoungsubang`)

  • An investigation was conducted to elucidate the effect of sucrose, germicides and abscisic acid (ABA) in the preservatives on postharvest quality, such as fresh weight, solution uptake, vase life, diameter and thickness of flower, and chlorolhyll of leaf in cut chrysanthemum 'Kyoungsubang`. Fresh weight of cut chrysanthemum was highest in 250mg/L aluminum sulfate [{TEX}$Al_{2}(SO_{4})_{3}${/TEX}]+3% sucrose and 250 mg/L 8-hydroxyquinoline sulfate (8-HQS)+ 3% sucrose. Addition of sucrose and germicides inhibited microorganisms growth in solution, and apparently promoted solution uptake. The increased fresh weight seems to attribute to the elevated uptake. Germicide 8-HQS was more effective than {TEX}$Al_{2}(SO_{4})_{3}${/TEX} in the aspect of solution uptake and fresh weight. Treatment of 250 mg/L 8-HQS +3% sucrose showed the longest vase life (24.67±2.52 days) and the lowest rate of leaf chlorosis (or senescence). Addition of {TEX}$Al_{2}(SO_{4})_{3}${/TEX} and sucrose accelerated leaf chlorosis and increased diameter of flower, respectively. Sucrose treatment prolonged the vase life of flowers. Except control and 250 mg/L 8-HQS, content of chlorophyll was rapidly decreased after 6 days at upper, middle and lower leaves in order.

양액재배에서 감귤나무의 무기양분 흡수 (Nutrient Absorption by Citrus unshiu Marc. Grown in Out-Door Solution Culture)

  • 노지에서 분무식 양액재배를 이용하여 감귤나무(Citrus unshiu Marc. cv. Miyagawa Wase)의 무기양분과 물 흡수 실험을 수행하였다. 양액의 농도는 세 수준으로 나누어 공급한 뒤 주기적으로 양액을 채취하여 무기양분과 물 흡수량을 측정하였고, 생체중의 변화를 조사하여 수체 증가량을 알아보았다. 물의 흡수는 감귤나무의 수체와 시간이 경과할수록 증가하였으나, High 처리구에서는 Medium과 Low보다 적게 흡수되었다. 영양생장(5~7월)과 과실형성(8~9월)기에 생체중과 양분흡수 증가가 가장 크게 일어났다. 양분의 흡수는 양액 공급 농도에 따라 차이가 컸으며, 높을수록 더 많이 흡수하는 경향이었고, $NH_4-N$, S, P, Mg, Fe보다 $NO_3-N$, K, Ca가 많았다. 또한 감귤잎 중 무기양분 함량도 공급되는 양액의 농도가 높을수록 증가하였다. 감귤나무의 주요 원소의 양액 농도 조성은 N, P, K, Ca, Mg 경우 각각 27.1, 16.5, 66.0, 80.0, $24.0mg\;L^{-1}$이 적당힌 것으로 사료된다.

Effect of sodium on transmembrane calcium movement in the cat ileal longitudinal muscle

  • To get a better insight into the exxistence and the role of a Na-Ca exchange mechanism in smooth muscle, the effect of Na substitution with sucrose on tension development, cellular Ca uptake and $^{45}Ca$ efflux was investigated using isolated cat ileal longitudinal muscle strips. Experimental results were summarized as follows;1) Exposure of the cat ileal longitudinal muscle to Na-free solution induced a contraction, and the magnitude of the contraction increased after incubation of the muscle strips with ouabain ($2{\times10^{-}5}$M) for 1hr. 2) Cellular Ca uptake in Na-free solution increased with an increase in Na content of the Na-loading media, and a linear relationship existed between tissue Na content and cellular Ca uptake for 10 min 3) After tissues were equilibrated in PSS containing $^{45}Ca$ for 2hr, cellular Ca uptake decreased with rising the external Na concentration. 4)Removal of medium Na or inhibition of the Na-K pump decreased the rate of $^{45}Ca$ efflux. These results strongly suggested that Na substitution increases cellular Ca uptake and decreases the rate of $^{45}Ca$ efflux via a Na-Ca exchange mechanism.

Ammonium and Nitrate Uptake and Utilization Efficiency of Rice varieties as Affected by Different N-Concentrations

  • To find out the optimum mixture ratio of ammonium and nitrate on rice plant, 4 rice varieties were examined during 14days after transplanting in hydroponics with the different ratio of ammonium to nitrate(100 : 0, 75: 25,50: 50, 25: 75 and 0: 100). The highest N uptake from solution and the maximum plant dry weight were $60\~70\%$ ammonium and $30\~40\%$ nitrate mixture treatment both in Japonica and Tongil type rice plants. And with the same varieties N-uptake and N use-efficiency were compared between 10.0 mM and 1.0 mM nitrogen using $70\%$ ammonium and $30\%$ nitrate for 24 days after transplanting. Rice plants absorbed more nitrogen$(131\~145\%)$ in 10.0mM than 1.0mM treatment but accumulated N in rice plants were almost the same in both treatment. Among the tested rice cultivars, dry matter production and total accumulative nitrogen in rice plants were much high in Tongil type than japonica type rice cultivars. N-recovery ratios of rice plants from uptake N were $90.8-99.0\%$ in low concentration N solution(1.0 mM), but $69.4-81.7\%$ were observed in high concentration N solution(10.0 mM). It means that suppling low concentration N steadily will be better to prevent loss of N without reducing of growth in rice plants.

옹기 화병이 절화 거베라의 수명 및 미생물 증식에 미치는 영향 (Effects of Earthenware Vase on Vase Life of Cut Gerbera Flowers and Microbial Multiplication)

  • 미세다공성의 옹기 화병의 절화수명 연장효과를 알아보기 위하여 거베라 절화를 식물재료로 하여 관행의 자기 및 유리 화병과 절화수명을 비교하였다. 수돗물과 절화보존제가 들어있는 유리, 자기, 옹기 화병에 거베라 'Honeymoon'과 'Golden Time'을 꽂고 절화수명과 미생물 수를 조사하였다. 그 결과, 두 품종 모두 옹기 화병에서 절화수명이 길었으며, 절화보존용제가 수돗물에 비해 절화수명을 연장시켰다. 특히 절화보존제를 넣은 옹기 화병에서 두 품종 모두 절화수명이 14일 이상으로 가장 길었다. 누적 흡수량은 절화보존용제를 넣은 옹기 화병에서 'Honeymoon'이 19.2mL, 'Golden Time'이 15.8mL로 가장 많았다. 생체중은 대체로 옹기에 꽂은 절화가 다른 재질의 화병에 꽂은 것에 비해 컸으며, 절화보존제 처리가 생체중 감소를 지연시켰다. 특히 옹기에 절화보존제를 처리한 경우 처리 8일 후에도 초기 생체중의 89% 수준을 유지하여 수분 흡수가 원활했음을 알 수 있었다. 미생물수 측정 결과, 'Golden Time'은 옹기 화병 내의 보존용액의 총균수가 다른 재질의 화병에 비해 적게 나타났다. 'Golden Time'의 총균수는 8일째 옹기 화병 내의 보존용액에서 435cfu/mL 정도로 자기나 유리 화병 내의 보존용액보다 20-30% 수준으로 낮게 나타났다. 이상의 결과로 옹기 화병은 보존용액 내 미생물의 증식을 지연시켜 절화 거베라의 물올림을 유지시켜 주기 때문에 도관 폐쇄에 의한 화경굽음 현상이 억제되었고, 절화수명도 길어졌다고 볼 수 있다. 따라서 미세다공성의 옹기는 화병의 좋은 재질이며, 절화보존제는 그 기능을 향상시킬 수 있을 것이다.
