Changes in the Cell Wall Components and Cell Wall-Degrading Enzyme Activities of Jujube Fruits during Maturation (대추 성숙중의 세포벽 성분과 세포벽 분해효소의 활성 변화)

  • This paper was investigated the changes of the cell wall components, enzyme activities during ripening of jujuba fruits for elucidating the softening metabolism of jujuba fruits. Firmness were decreased during ripening. Moisture content did not show any notable cahanges until ripening stage but they decreased a little In overripe jujuba fruits. Polygalacturonase activities were not detected at nature green stage and $\beta$-galactosidase activities were until turning stage. But polygalacturonase activities in ripening and overripening were 51.31 and 100.72 units/100g-fr, wt. respectively. $\beta$-galactosidase activities were 16.05 and 182.55units/100g-fr. wt. in the same stages. The content of water-soluble protein was increased in overripening. Stage the contents of cell wall and alcohol-insoluble material were. decraesed during maturation, but water-soluble material was increased. The pectin and alkali-soluble hemicellulose were increased until ripening stage, but decreased in overripe jujube fruits. The total pectin and insoluble pectin during ripening, but decreased in overripe jujuba fruits.

Comparison of physico-chemical components on citrus varieties (감귤 품종별 이화학적 성분 비교)

  • Physico-chemical components which are closely related to processed products were investigated on 10 varieties of Cheju citrus fruits. Juice ratio of Hungjin was the highest, 49.2%, while Sankyool was the lowest, 4.2%, which showed great differences among varieties. Peel ratio of Dangyooja was the highest, 46.1%, while both Navel orange and Hungjin were relatively low. Soluble solid($^{\circ}$Brix) was relatively high In Sankyool, Meiwa Kumquat, Sambokam and Iyo. Acid content was the highest, 4.86% in sudachi and relatively high in Sankyool, Natsudaidai and Dangyooja. The $^{\circ}$Brix to acid content ratio was 13.9 in Meiwa Kumquat and more than 10 in Navel orange and Hungjin. Total sugar contents of Juice were 2.78∼10.94%, while reduced sugar contents were 1.63∼6.38% which showed higher in Meiwa Kumquat, Iyo and Navel Orange. Hesperidin and naringin, the sources of biiter taste and cloudness were low in Hungjin and Iyo. Soluble solid($^{\circ}$Brix) of citrus juice showed highest statistical relationship(r=0.907) with total sugar, and was highly significant at 1% level.

Biochemical Studies on Antler (Cervus nippon taiouanus) (VI) Comparative Study on the Effect of Lipid Soluble Fractions of Antler Sponge and Velvet Layers and Pantocrin on the Aldolase Activity in the Rat Spinal Nerves (녹용의 약효 성분에 관한 연구(VI) 녹용의 지용성 성분 및 Pantocrin이 흰쥐 척수 신경의 Aldolase 활성에 미치는 영향)

  • In the present study, in attempt was made to observe the effect of lipid components in pantocrin and antler velvet and sponge layers extracts on the aldolase activity in the rat spinal nerves. The antler chloroform-methanol (2:1) extracts are known to be composed of neutral lipids, cholesterol, cerebrosides, spingomyelin, phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl serine and gangliosides. The antler extracts were studied on the aldolase activity in the rat spinal nerves. The aldolase activity was measured by the method of Lehninger. To investigate the components of tile extracts which affect the enzyme activity, the chloroform-methanol (2:1) extracts of antler were fractionated into petroleum ether soluble and insoluble fractions and their effects on the enzyme activity were compared. It was found that the chloroform-methanol (2:1) extracts of the antler sponge and velvet layers as well as those petroleum ether soluble fractions obtained from the chloroform-methanol (2:1) extracts, decrease the aldolase activity by 18-23%. However, pantocrin showed increasing effect on the aldolase activity by 42.6%. The petroleum ether insoluble fraction of the chloroform-methanol (2:1) -extract showed no significant increasing activity (about 13%) on the spinal nerves aldolase. The components of pantocrin ind the petroleum ether insoluble fractions were attempted to analysize by thin layer (silica gel) and gas liquid chromatography.

Optimization of Extraction Conditions for Soluble Ginseng Components Using Microwave Extraction System under Pressure (가압형 마이크로웨이브 추출장치를 이용한 가용성 인삼성분의 추출조건 최적화)

  • Microwave extraction system equipped with closed vessels was applied to confirm its capability of extracting ginseng solubles including saponin, which was monitored to optimize extraction con ditions by response surface methodology. Total yield increased with the decrease in particles size and more extraction steps. Soluble components were completely extracted by operating the system within 6 min per one step, which should be repeated by 3 or 4 times. Optimized conditions for maxi mum extraction of response variables(total yield, crude saponin) were 40~50% of ethanol concentration, about 140oC of extraction temperature, and within 6 min of the extraction time. The extraction effi ciency of total yield was dependent on the decrease of ethanol concentration, while crude saponin content was favored on the higher degree of ethanol concentration. According to the temperature guideline, below 90oC of extraction temperature, the optimal ranges of extraction conditions were predicted as 30~50% of ethanol concentration and 2.5~6.5 min of extraction time. Estimated values of total yield and crude saponin were in good agreement with experimental values.

  • This study was conducted to manufacture the soluble micro-fiber and to synthesize low melting polymer for the interior fabric not to use the polyurethane resin causing some problems. Low melting polyester for weft yarn was introduced by adding 30-40 mol% ratio of isophthalic acid to a main chain of polyethylene terephthalate to decrease the melting temperature up to heat setting temperature. Micro-fiber for warp yarn consisted of both soluble and insoluble components with multi-layered structure. When the soluble micro-fiber was treated by alkaline hydrolysis with 3-5% concentration of NaOH, it showed the turning point at 28% weight loss since soluble polyester was hydrolyzed approximate five times faster than regular polyester.

  • Effect of ginsenoside mixture, ginsenoside $Rb_1$,$Rb_2$,$Rg_1$ and the fat soluble fraction of the roots of Panax ginseng C.A. Meyer on the activity of glucokinase (GK) in vitro has been observed and found that GK activity was increased about 15c1c at the concentration of ginsenoside mixture and/or the fat soluble fraction being $10^{-7}$,$10^{-5}$%. It was also observed that glucose uptake by rat liver was increased in the presence of either ginsenoside mixture or the fat soluble fraction by perfusion technique. Ginsenoside mixture stimulated various enzymes related to glucose metabolism, however, both ginsenoside mixture and the fat soluble fraction did not stimulate GK activity as expected. Primary culture of liver cells showed that the ginsenoside mixture and the fat soluble fraction increased GK activity significantly and they stimulated the GK activity synergistically in the co-presence of insulin.

  • The purpose of this study was to investigate the possibility to reduce the toxicity of oil based root canal sealers containing calcium hydroxide using MTT & agar overlay assays. Thus some formulations of traditional root canal sealers were replaced with oil-soluble solvents and experimental root canal sealers manufactured. In MTT assay, Cell viability of all experimental sealers in addition with oil soluble solvents were observed significantly higher than both control groups, especially according to replace zinc and/or calcium ion components. Also agar overlay assay was appeared moderate to no cell responses into modifying both zinc and/or calcium ion components and oil soluble solvent weight. Authors found the reducing effect of cell toxicity through significant role of oil soluble solvent factor into root canal sealer containing calcium hydroxide.

Discrimination of Turkish Propolis from Different Geographical Origins by NMR Spectroscopy

  • In a study utilizing NMR spectroscopy and chemometrics, propolis samples from seven diverse geographic regions across Turkey were analyzed. To identify the optimal method for studying both the antimicrobial properties and compositional variations of propolis from different regions, we investigated metabolite extraction using three solvents: water only, ethanol only, and sequential water-ethanol extraction for residual components. Notably, water-soluble components exhibited significant variation among the samples, which is particularly interesting considering the potability of propolis in water-based solutions. Furthermore, the Muğla sample displayed a distinct water-soluble profile, likely due to its unique coastal location on the Aegean Sea. This specific climate may influence the propolis' chemical composition, resulting in a different mixture of components. Interestingly, the Muğla sample contained pharmaceutically active compounds like cinnamate, ferulate, and verapamil. This research establishes a valuable foundation for further exploration of propolis' antimicrobial potential.

Temporal Variations in Optical Properties and Direct Radiative Forcing of Different Aerosol Chemical Components in Seoul using Hourly Aerosol Sampling (서울지역 시간별 에어로솔 자료를 이용한 화학성분별 광학특성 및 직접 복사강제력의 시간 변화 분석)

  • Temporal variations of optical properties of urban aerosol in Seoul were estimated by the Optical Properties of Aerosols and Clouds (OPAC) model, based on hourly aerosol sampling data in Seoul during the year of 2010. These optical properties were then used to calculate direct radiative forcing during the study period. The optical properties and direct radiative forcing of aerosol were calculated separately for four chemical components such as water-soluble, insoluble, black carbon (BC), and sea-salt aerosols. Overall, the coefficients of absorption, scattering, and extinction, as well as aerosol optical depth (AOD) for water-soluble component predominated over three other aerosol components, except for the absorption coefficient of BC. In the urban environment (Seoul), the contribution of AOD (0.10~0.12) for the sum of OC and BC to total AODs ranged from 23% (spring) to 31% (winter). The diurnal variation of AOD for each component was high in the morning and low in the late afternoon during the most of seasons, but the high AODs at 14:00 and 15:00 LST in summer and fall, respectively. The direct negative radiative forcing of most chemical components (especially, $NO_3{^-}$ of water-soluble) was highest in January and lowest in September. Conversely, the positive radiative forcing of BC was highest in November and lowest in August due to the distribution pattern of BC concentration.

Carbohydrates Analyses of Korean Yam(Dioscorea) Tubers (한국산 마의 당질 분석)

  • Yam(Dioscorea) tubers were used as one of the dietary starches in Korea, China and Japan because of its characteristic properties. Yam tubers were powdered after freeze drying the sliced tubers to investigate carbohydrates. Chemical properties such as proximate components and carbohydrates were studied using three varieties of Korean yams, namely. D. batatas, D. aimadoimo and D. japonica. The proximate components of yam tubers showed that the major components of Korean yam tubers were carbohydrates and moisture. The main components of free sugars in yams were identified as fructose, glucose, sucrose and maltose. In addition to these four sugars, there were two unidentifiable peaks whose areas are too big to ignore. These results were obviously different from other reported data of free sugars in yams. The main components of total sugars were mannose and glucose.
