Epirubicin Inhibits Soluble CD25 Secretion by Treg Cells Isolated from Diffuse Large B-cell Lymphoma Patients

  • Objective: To investigate the effect of epirubicin on soluble CD25 (sCD25) secretion by CD4+CD25+ regulatory T (Treg) cells isolated from diffuse large B-cell lymphoma (DLBCL) patients. Methods: Treg cells were isolated from the peripheral blood mononuclear cells isolated from the newly diagnosed DBLCL patients. The concentration of sCD25 in the supernatant was determined with a commercial sCD25 (IL-2R) enzyme-linked immunosorbent assay (ELISA) kit. The fluorescence intensity of CD25 was detected by flow cytometry. Results: Cell survival rate was significantly decreased along with the increase of epirubicin concentration after treatment for 24 h. There was also a significant difference in the concentration of sCD25 between the epirubicin group and the control group (P<0.01). A positive correlation between the Treg cells survival rate and the concentration of sCD25 was detected (r=0.993, P<0.01). When equal numbers of CD4+CD25+ Treg cells of the epirubicin group and the control group were cultured for another 24 h without epirubicin the CD25 fluorescence intensity on the surface of Treg cells was obviously higher in the epirubicin group than that in the control group (P<0.01), while the sCD25 concentration in the supernatant in the epirubicin group was significantly lower than that in the control group (P<0.05). Conclusion: Epirubicin may improve the body's immune functions by inhibiting the sCD25 secretion by Treg cells in DLBCL patients.

Characteristics of Heavy Metal Releases from the Abandoned Dogog Mine Tailing in Korea (도곡광산 광미의 중금속 용출 특성)

  • Objective of this research was to assess the release characteristics of metals from the mine tailing to base the prediction of metal load potential from tailing to soils. Water-soluble concentrations of Cd, Cu, Pb and Zn released from mine tailing after 2 hrs were 2.31, 129.38, 17.17, and 287.53 mg/kg, respectively, as compared to 1.6, 128, 108, and 142 mg/kg that were extractable by 0.1 M HCl. Kinetics of metal releases followed the power function model significantly indicating that more of water soluble fractions of those metals released at the initial short time, followed by a slow increase. Concentrations of metals released from tailing by water and 0.1 M HCl were in the orders of Zn > Cu > Pb > Cd. The breakthrough curve from the column experiment showed that concentrations of Cd, Cu, and Zn reached at highest after one pore volume, but that of Pb reached highest after five pore volumes when 0.1 M HCl was used as eluent. The release rate of Cd from mine tailing was the fastest but Pb was the slowest. The cumulative mass of metal released by 0.1 M HCl was in the order of Zn > Cu > Pb > Cd after nine pore volume elution.

  • Background level of heavy metals In soils (316 points by 15 classifications of land use) was investigated by two test methods, 0.1N HCl(1N HCl for As) extraction and aqua regia extraction methods. The average concentrations of aqua regia extractable heavy metals in soil(n=316) was 6.24(As), 0.25(Cd), 37.99(Cr), 24.10(Cu), 0.04(Hg), 25.68(Pb), 22.59(Ni), 106.11(Zn) mg/kg, respectively. Also the average concentrations of 0.1N HCl extractable heavy metals was 0.06(As), 0.08(Cd), 0.27(Cr), 3.78(Cu), 4.02(Pb), 12.5(Zn), 0.58(Ni) mg/kg, respectively. The ratio of soluble contents and total contents were 2.6%(As), 32.7%(Cd), 0.7%(Cr), 15.7%(Cu), 15.7%(Pb), 2.6%(Ni), 11.8%(Zn), and the correlation coefficient of soluble contents and total contents were 0.26(As), 0.27(Cd), 0.22(Cr), 0.57(Cu), 0.42(Pb), 0.23(Ni), 0.72(Zn).

Enhancement of Dissolution Rate of Poorly Water-soluble Ibuprofen using Solid Dispersions and Inclusion Complex (고체분산체 및 포접화합물을 이용한 난용성 약물인 이부프로펜의 용출 속도의 증가)

  • Solid dispersions and inclusion complex were prepared for the enhancement of solubility and dissolution rate of poorly water-soluble ibuprofen(IPF) as a model drug. Polyethylene glycol 4000(PEG4000) and polyvinylpyrrolidone(PVP) were used for the preparation of solid dispersion. $2-Hydroxypropyl-{\beta}-cyclodextrin(2-HP{\beta}CD)$ was also used for the preparation of inclusion complex. The solubility of IPF increased as the concentration of PEG4000, PVP and $2-HP{\beta}CD$ increased. Solubilization capacity of $2-HP{\beta}CD$ was increased about 10 times when compared to PEG 4000 and PVP. The dissolution rate of drug from solid dispersions and inclusion complex in the simulated gastric fluid was enhanced when compared to pure IPF and commercial $BR4^{\circledR}$ tablet as a result of improvement of solubility. In case of solid dispersions, dissolution rate of drug was proportional to polymer concentration in the formulation. The marked enhancement of dissolution rate of drug by inclusion complexation with $2-HP{\beta}CD$ was noted. However, dissolution rate of drug from solid dispersions and inclusion complex in the simulated intestinal fluid was not significant because IPF was readily soluble in that condition. From these findings, water-soluble polymers and cyclodextrin were useful to improve solubility and dissolution rate of poorly water-soluble drugs. However, easiness and reliability of preparation method, scale-up and cost of raw materials must be considered for the practical application of solid dispersion and inclusion complex in pharmaceutical industry.

T Regulatory Cell Responses to Immunization with a Soluble Egg Antigen in $Schistosoma$ $mansoni$-Infected Mice

  • The aim of the study is to characterize the phenotypes of $CD4^+$ $CD25^+$ T regulatory cells within the liver granulomas and association with both Foxp-3 gene expression and splenic cytokines. Naive C57BL/6 mice were intravenously injected with multiple doses of the soluble egg antigen (SEA) 7 days before cercarial infection. The immunized and infected control groups were sacrificed 8 and 16 weeks post-infection (PI). Histopathology, parasitological parameters, splenic phenotypes for T regulatory cells, the FOXP-3 expression in hepatic granuloma using real-time PCR, and the associated splenic cytokines were studied. Histopathological examination of the liver revealed remarkable increase in degenerated ova within hepatic granuloma which decreased in diameter at weeks 8 and 16 PI ($P$<0.01). The percentage of T regulatory cells ($CD4^+$ $CD25^+$) increased significantly ($P$<0.01) in the immunized group compared to the infected control at weeks 8 and 16 PI. The FOXP-3 expression in hepatic granulomas increased from 10 at week 8 to 30 fold at week 16 PI in the infected control group. However, its expression in the immunized group showed an increase from 30 at week 8 to 70 fold at week 16 PI. The splenic cytokine levels of pro-inflammatory cytokines, IFN-${\gamma}$, IL-4, and TNF-${\alpha}$, showed significant decreases ($P$<0.05) compared to the infected control group. In conclusion, the magnitude and phenotype of the egg-induced effects on T helper responses were found to be controlled by a parallel response within the T regulatory population which provides protection in worm parasite-induced immunopathology.

Contrast Effect of Citric Acid and Ethylenediaminetetraacetic Acid on Cadmium Extractability in Arable Soil

  • Chelating agents have been proposed to improve the efficiency of phytoextraction of heavy metal hyperaccumulator. However, little studies to elucidate mechanism of chelating agents to increase cadmium (Cd) extractability have been conducted. The objectives of this study were to evaluate effect of different chelating agents on Cd extractability and to determine mechanism of Cd mobilization affected by these agents. An arable soil was spiked with inorganic Cd ($CdCl_2$) to give a total Cd concentration of $20mgCdkg^{-1}$. Ethylenediaminetetraacetic acid (EDTA) and citric acid (CA) were selected and mixed with the arable soil at the rates of 0 and $5mmolkg^{-1}$. The mixture soils were incubated at $25^{\circ}C$ for 4 weeks in dark condition. Concentration of F1 Cd fractions (water soluble) significantly increased with addition of EDTA but did not changed with addition of CA. Especially; concentration of F5 Cd fractions (residual) significantly increased with addition of CA. Increase in water soluble with EDTA might be attributed to complexation of Cd and EDTA. Dissolved organic carbon concentration significantly increased with EDTA addition, but did not with CA implying that considerable amount of CA was decomposed to inorganic carbon by microorganism. Log activity of carbonate ($CO_3{^{2-}}$) which might be generated from CA increased with addition of CA. Increase in residual Cd fraction might be due to precipitation of Cd as $CdCO_3$. As a result, EDTA was effective in increasing Cd extractability, by contrast CA had significant effect in reducing Cd extractability.

Soluble Expression and Purification of the Catalytic Domain of Human Vascular Endothelial Growth Factor Receptor 2 in Escherichia coli

  • Vascular endothelial growth factor (VEGF) plays a key role in angiogenesis through binding to its specific receptors, which mainly occurs to VEGF receptor 2 (VEGFR-2), a kinase insert domain-containing receptor. Therefore, the disruption of VEGFR-2 signaling provides a promising therapeutic approach for the treatment of cancer by inhibiting abnormal or tumorinduced angiogenesis. To explore this potential, we expressed the catalytic domain of VEGFR-2 (VEGFR-2-CD) as a soluble active kinase in Escherichia coli. The recombinant protein was purified and the VEGFR-2-CD activity was investigated. The obtained VEGFR-2-CD showed autophosphorylation activity and phosphate transfer activity comparable to the commercial enzyme. Furthermore, the IC50 value of known VEGFR-2 inhibitor was determined using the purified VEGFR-2-CD. These results indicated a possibility for functional and economical VEGFR-2-CD expression in E. coli to use for inhibitor screening.

Effects of Sulfobutyl Ether $\beta$-Cyclodextrin on Physicochemical Properties of Dexamethasone Dipropionate

  • Complex formation of practically insoluble dexamethasone dipropionate (DDP) with ${\beta}$-cyclodextrin (${\beta}$-CD), dimethyl-${\beta}$-cyclodextrin (DMCD), trimethyl-${\beta}$-cyclodextrin (TMCD), 2-hydroxypropyl-${\beta}$-cyclodextrin (HPCD) and sulfobutyl ether ${\beta}$-cyclodextrin (SBCD) in water was investigated by solubility method at various temperatures. Water solubility of DDP was found to be 1.78 $\mu\textrm{g}$/$m\ell$ at 37$^{\circ}C$. Propylene glycol (PG)-water cosolvent increased the solubility of DDP, but the solubilization was not sufficient (8.93 $\mu\textrm{g}$/$m\ell$ in 20% PG). The addition of CD markedly increased the solubility of DDP in water, and A$\sub$L/ type phase solubility diagrams were obtained with ${\beta}$-CD, TMCD, HPCD and SBCD, where the apparent stability constants of the soluble complexes at 25$^{\circ}C$ were determined to be 1388, 216, 1054, and 1992 M$\^$-1/, respectively. However, DMCD remarkably increased the solubility of DDP, and showed an A$\sub$P/ type diagram, suggesting that DMCD forms a soluble complex of high order with DDP. The stability constant for the DDP-DMCD complex at 25$^{\circ}C$ was determined to be 19132 M$\^$-1/. The thermodynamic parameters were calculated for the inclusion complex formation in aqueous solution. CD (1${\times}$10$\^$-2/M) remarkably decreased the partition coefficients of DDP between isopropyl myristate/water in the order of TMCD < ${\beta}$-CD < HPCD < SBCD < DMCD, and in squalane/water system in the order of HPCD < TMCD < ${\beta}$-CD < DMCD < DMCD $\leq$ SBCD. This finding represents that, in a o/w type cream, cyclodextrin complexation with DDP may result in high concentration of DDP in aqueous phase. The permeation of DDP through a cellophane membrane was highly suppressed by the addition of CD, and the degree of suppression was different among CDs, indicating that CD may control the skin permeation of DDP. The dissolution rates of solid dispersions with CDs were much faster than those of drugs alone and corresponding physical mixtures. All DDP-CD solid dispersions exceeded the equilibrium solubility. Consequently these results suggest that complex formation of DDP with CDs may provide useful means to markedly enhance the solubility, and CDs are useful in the semi-solid preparations such as creams and gels for topical application.

Evaluation on Natural Background of the Soil Heavy Metals in Korea (우리나라 토양의 중금속 자연배경농도 평가)

  • This study was conducted in order to get the scientific background for soil pollution criteria. The 92 soil samples derived from various geological units were taken and analyzed to survey natural heavy metal background levels using aqua regia digestion method and 0.1N HCl extraction method. From these results, the average natural contents of metals were 0.287 mg/kg for Cd, 15.26 mg/kg for Cu, 18.43 mg/kg for Pb, 25.36 mg/kg for Cr, 54.27 mg/kg for Zn, 17.68 mg/kg for Ni, 6.83 mg/kg for As by the aqua regia method, and 0.040 mg/kg for Cd, 0.48 mg/kg for Cu, 3.06 mg/kg for Pb, 0.09 mg/kg for Cr, 1.54 mg/kg for Zn, 0.27 mg/kg for Ni, 0.089 mg/kg for As by the 0.1N HCl extraction method. Ratios of soluble contents and total contents were Cd 0.14, Cu 0.03, Pb 0.17, Cr 0.004, Zn 0.03, Ni 0.02, As 0.013 and the correlation coefficients of soluble contents and total contents were 0.24(As), 0.88(Cd), 0.43(Cr), 0.65(Cu), 0.70(Pb), 0.61(Ni), 0.24(Zn). The correlation factor decreased in the order of Cd > Pb > Cu > Ni > Cr > Zn $\approx$ As.

White Electroluminescence from Bicarbazyl-containing Conjugated Polymers as Single-Emitting Component

  • Two bicarbazyl-containing fluorene copolymers, PFEBCz (95/5) and PFEBCz (75/25), were synthesized for white light electroluminescence from a single emitting polymer. All synthesized polymers were soluble in common organic solvents such as chloroform and toluene. The weight-average molecular weights (Mw) of the PFEBCz (95/5) and PFEBCz (75/25) copolymers were found to be 11,000 and 5,700 with polydispersity indices 1.4 and 1.8. The EL spectrum of the PFEBCz (75/25) device showed bright white-light emission with CIE coordinates of (0.32, 0.34) at 1000 cd/m2, which is very close to that for pure white (0.33, 0.33). This white emission may have been due to strong excimer formation between the bicarbazyl and fluorene polymer backbone. The device exhibited a maximum brightness of 3400 cd/m2 with a maximum efficiency of 0.2 cd/A.
