In this study, we investigated the effect of solid culture medium on the production of cordycepin in Cordyceps militaris. The regression equation was expressed as follows: Y1 = 755.3-58.6625X1+4.79432E-14X2-46.6625X3-5.66269E-14X1X2-0.025X1X3+1.62475E-14X2X3-160.6625X12+0.0125X22-206.9625X32, where, Y represents the value of cordycepin content (㎍/g), X1 corresponds to the weight of M. alternatus in solid culture medium (g/bottle), X2 to the water content of the solid culture medium (%), and X3 to the culture period (day). The solid culture medium was optimized using the response surface methodology, and the optimal medium composition was as follows: the weight of M. alternatus in solid culture medium and water content were 16.2% and 100.7% (20.14 mL water/20 g solid culture medium), respectively, with a culture period of 39 days. Under these conditions, the cordycepin content of the fruiting bodies reached 150.0 ㎍/g (actual value). The supplementation of M. alternatus in solid culture for improved cordycepin content of C. militaris seems to be a promising alternative to wild and solid cultivation.

This study aimed to analyze the selection criteria and priority settings for solid culture medium used in hydroponic crop production in farm households. Expert brain storming was carried out to extract solid culture medium selection criteria for hydroponic farming. As a result, 3 criteria of economy (cost), productivity, and environment, and 9 factors were extracted. A questionnaire survey of hydroponic farm households was conducted in Gyeonggi, Gangwon, and Chooncheong provinces. AHP analysis of the hydroponic solid culture medium selection criteria identified productivity as the most important criterion, chosen by 58.7% of the respondents, followed by economy (28.4%) and environment (12.9%). The 9 factors were rated by the respondents in the following decreasing order of importance: 1, crop yield (28.3%); 2, pest occurrence (18.5%); 3, maintenance/management costs (12.0%); 4, convenience of maintenance/management (13.4%); 5, initial investment cost (11.6%); 6, material energy consumption (6.5%); 7, waste recyclability (4.0%); 8, waste disposal costs (3.4%); and 9 environmental emissions (1.81%). These results imply that hydroponic farm households consider cultivation-related quality factors more important than economic factors, such as price of culture medium or installation cost.
Rapid, real-time detection of pathogenic microorganisms is an emerging and quickly evolving field of research, especially with regard to microorganisms that pose a major threat to public health. Herein, a new method that uses bioimpedance and solid culture medium for the real-time detection of microorganisms is introduced. We fabricated a new impedimetric biosensor by integrating solid media and two plane electrodes attached on two facing sides of an acryl well. During bioelectrical impedance analysis, the solid medium showed the characteristics of a homogenous conductive material. In a real-time impedance measurement, our solid-medium biosensor could monitor bacterial growth in situ with a detection time of ${\sim}4$ hrs. Our data indicate that the solid-medium biosensor is useful for detecting airborne microorganisms, thereby providing a new analytical tool for impedance microbiology.

The antibiotic meroparamycin was produced in the free culture system of Streptomyces sp. strain MAR01. Five solid substrates (rice, wheat bran, Quaker, bread, and ground corn) were screened for their ability to support meroparamycin production in solid-state fermentation. In batch culture, wheat bran recorded the highest antibacterial activity with the lowest residual substrate values. The highest residual substrate values were recorded for both ground corn and Quaker. On the other hand, no antibacterial activity was detected for rice as a solid substrate. The use of the original strength of starch-nitrate medium in the solid-state fermentation gave a lower antibacterial activity compared with the free culture system. Doubling the strength of this medium resulted in the increase in the activity to be equivalent to the free culture. The initial pH (7.0) of the culture medium and 2 ml of spore suspension (1 ml contains $5{\times}10^{9}spores/ml$) were the optima for antibiotic production. The water was the best eluent for the extraction of the antibiotic from the solid-state culture. Ten min was enough time to extract the antibiotic using a mixer, whereas, 60 min was required when shaking was applied. Semicontinuous production of meroparamycin using a percolation method demonstrated a more or less constant antibacterial activity over 4 runs ($450-480{\mu}g/ml$). The semicontinuous production of the antibiotic was monitored in a fixed-bed bioreactor and the maximum activity was attained after the fourth run ($510{\mu}g/ml$) and the overall process continued for 85 days.

Hairy root culture of ginseng is industrially prospected because the cultivation period of ginseng is relatively long. In this study, the effect of medium concentration and sucrose concentration on hairy root culture of ginseng was evaluated. The optimization of ginseng hairy roots transformed by Agrobacterium rhizogene were performed liquid medium. The MS(Murashinge & Skoog basal medium) concentration was selected with 1/2 strength MS and the optimal sucrose concentration was determined at 2-3%(w/v). At the optimum culture condition, The yield (the ratio of weight of grown hairy root cultures to weight of fresh ginseng hairy roots) and production rate of ginseng root were 19.42 times and 5.73 g/l-day. The major ginsenosides were Rb group, Re and Rg1. The produced total ginsenoside content in the solid medium was 9.87 (mg/g) and increased 1.34 times in the liquid medium (13.23 mg/g). In solid culture, the contents of ginsenosides Rb, Re and Rg1 were 2.14, 3.65 and 1.87 mg/g, respectively. In liquid culture, the contents of ginsenosides Rb, Re and Rg1 were 3.54, 4.12 and 2.63 mg/g, respectively.

The effect of the addition of the fresh medium, volume of under solid medium in double layer culture as well as the medium change after pretreating microspores on the production of embryos in microspore culture of hot pepper (Capsicum annuum L.) has been studied. When cultured after heat pre-treatment, changing pretreatment media with fresh culture media proved to be more effective for embryo production rather than supplementing additional culture media. Heat-pretreating for 3 days turned out more effective for embryo production than pretreating for 1 or 2 days. In the case of anther pretreatment, the addition of fresh medium after culture was not effective for embryo production. In pretreating microspores, however, supplementing additional fresh culture media greatly improved embryo yield and quality. The best time point of media addition was 4 days after culture commenced, and the most effective number of times of media addition was one time addition. Moreover, the effective volume of added medium in double layer culture for embryo production was 1.5 ml. The addition of media more than 1.5 ml reduced both embryo yield and quality. Double layer medium was more effective for embryo development than liquid medium. When the volume of under solid medium increased ranging from 3 ml to 7 ml, more cotyledonary embryos were produced in either 5 ml or 7 ml compared to 3 ml, even though the total number of embryos were highest in 3 ml. These results can be used as an important data for establishing an efficient microspore culture system for producing high frequency of normal embryos in hot pepper.

On optimal conditions, 4.1 g/l of the cell mass was obtained at 28$^{\circ}C$ for 3 days. The mycelium were inoculated on 500 g of steamed rice using vinyl bag ($30.6{\times}44$ cm) and incubated at $30^{\circ}C$, 85% humidity for 21 days. Lactone form monacolin K was rapidly increased for 2 days and reached highest concentration of monacolin K (2,930 mg/kg) for 15 days, and monacolin K was decreased after 15 days.
A system was established for induction of multi-shoots and plant regeneration from mesophyll protoplasts of alfalfa, Medicago sativa L. cv. Vernal. Different hormonal effects were tested at each step of protoplast culture, i.e. cell division in modified Kao's liquid medium (K566-7). calli formation on SH semi solid medium, and multi-shoot regeneration from calli on SHa and SHb solid media. Frequency of multi-shoots and plant regeneration was affected by various combinations of phytohormones in final step. The evaluation of multi-shoots induction systems via protoplast culture was discused.
The establishment of cell suspension culture and plant regeneration of the halophytic Leymus chinensis (Trin.) are described in this study for the first time. Callus induction solid medium containing Murashige and Shoog (MS) basic salt, $2.0\;mg\;l^{-1}$ 2,4-dichlorophenoxyacetic acid (2,4-D), and $5.0\;mg\;l^{-1}$ L-glutamic acid with $30.0\;g\;l^{-1}$ sucrose and $4.0\;g\;l^{-1}$ gelrite for solidification induced the highest rate of cell division in Type 1 callus among calli of various types. Liquid medium with the same hormone distribution was therefore, used for cell suspension culture from Type 1 callus. Over a 30 d suspension culture at 100 rpm, great amounts of biomass were accumulated, with 71.07% average daily increment and 22.32-fold total fresh weight increment. Comparison of before and after suspension culture, the distribution of different size callus pieces and the maintenance of callus type were basically unaltered, but a slight increase in relative water contents was observed. To induce the potential of plant regeneration, the directly transferring on plant regeneration solid medium containing MS basic salt, $0.2\;mg\;l^{-1}$$\alpha$-naphthalene acetic acid (NAA), $2.0\;mg\;l^{-1}$ kinetin (Kn), and $2.0\;g\;l^{-1}$ casamino acid and indirectly transferring were simultaneously performed. Even now growth rates of suspension-derived callus on solid medium were approximately half of those of Type 1 callus, but faster somatic embryogenesis was observed. Rooting of all regenerated shoots was successfully performed on half-strength MS medium. All plants appeared phenotypically normal.

We aimed to investigate the proportions of MTB- and NTM-positive tests and the distribution patterns of species isolated by contracted testing agencies in South Korea. Respiratory specimens submitted to contracted testing agencies in South Korea for AFB culture from January 2014 to December 2021 were included (533,713 specimens in total). Trends based on MTB and NTM detection, patient sex and age, culture medium type, and testing year were analyzed. MTB and NTM positive detection increased in the patients. The average ages of MTB- and NTM-positive patients increased in those aged ≥61 years. For solid culture, the MTB detection rate decreased from 5.9% in 2014 to 3.3% in 2018 and increased to 4.7% in 2021; the NTM detection rate increased from 2.1% in 2014 to 3.4% in 2018 and 3.7% in 2021. For liquid culture, the MTB detection rate decreased from 8.3% in 2014 to 5.5% in 2018 and increased to 6.0% in 2021; the NTM detection rate increased from 3.5% in 2014 to 5.5% in 2018 and decreased to 5.3% in 2021. An isolation ratio reversal between MTB and NTM was observed in 2018. In this study, we provide information on mycobacterial isolation rates and species distributions using AFB culture test results from Korea's referral laboratories. Increased MTB- and NTM-isolation rates were observed in individuals aged ≥60 years, indicating the need for regular testing and focused management for them. Expanding liquid culture applications, which show higher positivity rates than solid culture methods, is necessary.
