Kinetic Studies on Production of Pullulan by Aureobasidium pullulans

  • Xinlei studies on the production of pullulan by Aureobasidium pullulans using batch culture in a 15L bioreactor were carried out. The mathematical models were obtained in this study, which provided a reasonable description for the biomass, the product, and the substrate variation with time. The values frets the mathematical models were satisfactorily coincided with the experimental data for the biomass of A. pullulans, the production of pullulan and the utilization of sucrose as the sole carbon source.

Characterization of Formaldehyde-degrading Bacteria Isolated from River Sediment (하천 저질에서 분리한 Formaldehyde 분해 미생물의 특징)

  • A bacterium growing on formaldehyde as a sole carbon source was isolated by the dilution method from an enrichment culture containing formaldehyde. The isolated strain, YK-32, was identified as Pseudomonas sp. by morphological, biochemical, and genetic analyses. Pseudomonas sp. YK-32 completely degraded 0.05% formaldehyde within 24 hrs. The isolated strain had a high level of formaldehyde dehydrogenase activity, which is thought to be one of the important factors for formaldehyde degradation, when cells were cultivated in the presence of formaldehyde.

  • Twenty-nine bacterial isolates capable of growing on aniline as n sole source of carbon and nitrogen were obtained. Ten of these isolates were identified. Nine isolates were Identified as Pseudomonas spp. and one was Acinetobacter sp.. Five strains among 29 isolates had one or several plasmids. Four of these five strains utilized aniline through meta pathway and one through ortho pathway. Pseudomonas acidovorans 4A1 which utilized aniline through meta pathway harbored a plasmid of ca. 230 kilobases shown to be involved in aniline metabolism.

  • An aniline-utilizing microorganism identified as a species of Pseudomonas was isolated from soil contaminated highly with aniline and urea-herbicide. This strain was able to utilize aniline as the sole source of carbon and energy, and was shown to harbor a single large plasmid mediating the aniline assimilation. Subsequent plasmid-curing of this bacterium resulted in the abolishment of the aniline utilizing phenotype and the loss of catechol-C2,3O-oxygenase. The reestablishment of the plasmid, denoted pB10, in cured Pseudomonas sp. via filter surface mating, resulted in restoration of the aniline assimilation abilities and enzyme activity.

Succinic Acid Production by Anaerobiospirillum succiniciproducens ATCC 29305 Growing on Galactose, Galactose/Glucose, and Galactose/Lactose

  • Succinic acid-producing Anaerobinspirillum succiniciproducens was anaerobically grown on galactose, galactose/glucose, or galactose/lactose in order to study its galactose fermentation. Unlike a previous report, A. succiniciproducens was found to efficiently metabolize galactose as the sole carbon source at a rate of 2.4 g/g-DCW/h and produced succinic acid with as high a yield of 87% as with using glucose. When glucose and galactose were present, A. succiniciproducens metabolized both sugars simultaneously. Furthermore, when lactose and galactose coexisted, lactose did not inhibit the galactose fermentation of A. succiniciproducens. Therefore, co-utilization of galactose and other sugars can improve the productivity and economy of bio-based succinic acid processes.

Isolation and Characterization of Novel Alginate-Degrading Pseudoalteromonas sp. Y-4

  • To isolate an alginate-degrading bacterium, we conducted a single colony isolation using a solid medium containing alginate as the sole carbon source. A marine bacterium Y-4 capable of degrading alginate was isolated from seawater. The strain was identified to be Pseudoalteromonas sp., based on morphological, biochemical, 16S rDNA homology, and phylogenetic analyses. Moreover, Pseudoalteromonas sp. Y-4 exhibited alginate lyase activity in the presence of 4% alginate even though many known alginate-degrading bacteria degrade in the range of 0.5-1% alginate. The optimum culture conditions for the Y-4 strain were 2% alginate, pH 8.0, and 3% NaCl at $30^{\circ}C$. The highest alginate lyase activity was also observed under the same conditions. To our knowledge, this is the first reported isolation of a marine bacterium degrading high concentrations of alginate.

Toluene Degradation by Pseudomonas putida in Closed Vial (닫힌 바이얼 안에서 Pseudomonas putida에 의한 톨루엔 분해)

  • The ability of Pseudomonas putida to degrade toluene was studied in toluene-containing vials. The strain grows anaerobically in toluene as a sole source of carbon. When the initial toluene concentrations injected in the vial are varied, the changes of headspace toluene concentration and cell density are observed. We set a model for this vial and simulated the vial reactor using Matlab. With a variation of model parameters, simulated results were compared with the experiment.

Biodegration of Formaldehyde-Releasing Preservatives

  • A strain of Pseudomonas aeruginosa was isolated from the spoiled product and its characteristics on various formaldehyde-releasing preservatives were investigated. This strain, P. aeruginosa FR, could utilize 1.0% of imidazolidinyl urea and 0.2% of DMDM hydantoin as a sole carbon and nitrogen source in the minimal salts medium. With the growth of the strain in minimal salts medium containing imidazolidinyl urea, formic acid was initially accumulated according to the decrease of formaldehyde concentration. It was suggested that formaldehyde dehydrogenase was involved in this oxidation process and could catalyze formaldehyde, imidazolidinyl urea, DMDM hydantoin and quaternium-15, but not bronopol. MICs of this strain to each preservation were 0.03% in formaldehyde, 1.0% in imidazolidinyl urea, 0.2% in DMDM hydantoin, 0.2% in quaternium-15 and 0.1% of EDTA-2Na. But the MICs were diminished about ten times when 0.01% of EDTA-2Na was added to the preservative systems. In actual challenge test, the eyeliner and the pack which contained paraben and imidazolidinyl urea were not able to be protected from this strain, but when 0.05% EDTA-2Na was added the products were sufficiently preserved.

Studies on the receptor for bacteriophage N4 infection (Bacteriophage N4의 receptor에 대한 연구)

  • The evidences that Lam B protein of E. coli is used as a receptor for infections of bacteriophage N4 as well as bacteriophage lambda were obtained from the following experimental results. First, all of the isolated lambda resistant dlones possessing foreign DNA fragments in the plasmids were also resistant to bacteriophage N4, but not to bacteriophage $\phi$ 80, T4 and T7. Second, when the plasmid DNA was treated with various restriction enzymes and ligated to delete the total or a portion of the foreign DNA fragments, the deleted plasmids lost the resistant activities to lambda and N4, simultaneously. Third, after amplification of Lam B protein about 200 times by inducing the protein using maltose as a sole carbon source, the host E. coli became sensitive to both lambda and N4.

Isolation and Characterization of Pseudomonas sp. DCB3 Degrading 1.2-Dichlorobenzene (1.2-Dichlorobenzene을 분해하는 Pseudomonas sp. DCB3의 분리 및 특성)

  • Four bacterial strains able to degrade dichlorobenzene as the sole source of carbon and energy were isolated from soil by selective enrichment culture, and among them, one isolation was the best in the cell growth and identified as Pseudomonas sp. DCB3 by its morphology and physiological properties. Cell growth dramatically increased in a minimal medium containing 500ppm of dichlorobenzene was not detected any more at 72 hours after cultivation. The optimal temperature and initial pH for the growth of the isolated strain were 30$\circ$C and 7.0, respectively. Cell growth was increased by supplementing $(NH_2)_2CO$ and 50 ppm yeast extract as additional nutrients. Therefore, it was suggested that Pseudomonas sp. DCB3 could be effectively used for the biological treatment of wastewater containing dichlorobenzene.
