방선균 분리주 No. 1166이 생산하는 살충성 물질 구조 동정 (Identification of Insecticidal Compounds from Actinomycetes Isolate No. 1166)

  • In the course of screening for insecticidal metabolites from soil microorganisms, Actinomycetes isolate no.1166 was found to produce active metabolites against Musca domestca and Bombyx mori. Three active components from the metabolites were isolated by solvent extraction and chro- matographic techniques and examined their insecticidal activities on Bombyx mori (3rd larvae) by diet feeding bioassay methods. By UV and NMR data analyses, compound I and III were identified as bafilomycin A$_{2}$ and B$_{1}$, respectively and compound II was also estimated to belong to the bafilomycin family from its physico-chemical data and biological properties.

Methanol 자화방선균 MO-16으로부터 항균성 물질의 정제 및 생산조건 (Purification and Production Conditions of Antimicrobial Compound from Methylotrophic Actinomycetes MO-16)

  • A methylotrophic actinomycetes strain MO-16, which produce the antimicrobial compound, was isolated from soil and supposed as Amycolatopsis sp. based on taxonomic studies. The cell-free extract of methanol-grown strain MO-16 showed dehydrogenase activity for methanol and formaldehyde when various electron acceptors were added for oxidation. On the other hand, methanol did not affect the production of antimicrobial compounds, and organic nitrogen sources such as corn steep liquor and peptone were better than inorganic nitrogen sources. These compounds showed broad antimicrobial spectrum to the tested strains such as bacteria and yeast. The antimicrobial comounds were very stable under heat(121$^{\circ}C$), acid(pH2.0), alkali(pH11.0) treatments. These compounds were isolated by ethylacetate extract, silica gel column chromatography and reverse phase HPLC. Two compounds(peak 1 and 2) were detected as antimicrobial compounds through the HPLC analysis. The peak 2 was purified as a single compound and revealed a 98% purity.

세포외 Adenosine Deaminase Inhibitor를 생산하는 방선균의 분리 및 특성 (Isolation and Characterization of Actinomycetes Producing Extracellular Adenosine Deaminase Inhibitor)

  • A strain of actinomycetes producing extracellular adenosine deaminase inhibitor, strain V-8, was isolated from soil. Strain V-8 was gam positive and its cell wall chemotype was decided as cell wall chemotype I from analysis of diaminopimelic acid isomers and sugar pattern. This strain had a wide range of sugar utilization as carbon sources. The optimal pH and temperature for growth were $6.8{\sim}7.0$ and $28{\sim}30^{\circ}C$, respectively. From the morphological, chemotaxonomical characteristics and analysis of various physiological characteristics, the strain V-8 was identified Streptomyces sp.

밭 토양에서 분리한 방선균의 생리적 특성 (Physiological Characteristics of Actinomycetes Isolated in the Upland Soils)

  • 밭 토양에서 분리한 280개 균주의 방선균을 형태적 및 생리적특성으로 구분한 결과 15개 군으로 분류되었으며, 이들 각 군들중 Streptomyces속은 12개군의 87.2% 그리고 Non-Streptomyces속은 3개군의 12.8%로 확인되었다. 또한 전체 분리균주 중 포자체의 Spiral chain형태의 Streptomyces속이 75%로서 우점하였다. Streptomyces속의 12개군을 전자현미경상에서 포자체의 표면형태를 관찰한 결과 2개군은 Spiny형태를 보였고, 10군은 Smooth형태를 갖는 것으로 동정되었다. 또한 분리 동정한 방선균 15개 군을 E. coli. P. aeruginosa 및 S. aureus의 3개 시험균주에 대하여 항균력을 검정한 결과로는 E. coli에서 S-6균주가 $6.23r/m{\ell}$이고 P. aeruginosa에는 S-9균주가 $8.46r/m{\ell}$이며 S. aureus에는 S-2균주가 $7.24r/m{\ell}$로서 다른 방선균에 비하여 가장 높은 항균력을 나타내었다.

Deproteinized Mulberry Leaf Juice - A New Media for Growth of Microorganisms

  • Mulberry being a foliage crop is grown extensively for feeding of silkworms and are also used for cattle feeding. These loaves are highly nutritious, which contain various mineral elements and bio-molecules such as carbohydrates, proteins, lipids and other essential amino acids, etc. In the present study, deproteinized mulberry leaf juice was used for preparation of the medium for cultivation of various types of microbes. Results revealed that deproteinized mulberry leaf juice medium is best for isolation of fungi, bacteria and actinomycetes and this medium can be substituted with synthetic media, which are haying the costly ingredients for isolation and identification of bacteria, fungi and actinomycetes. Further, this deproteinized mulberry juice medium can also be used in mass multiplication of useful/beneficial microbes to enhance soil microflora to improve soil fertility and to avoid root diseases. Perspective enterprises can take up the mass multiplication/large-scale production of useful microbes such as Trichoderma, Rhizobium, Pseudomonas and Bacillus to use in mulberry and in other agricultural crops using deproteinized mulberry leaf juice.

  • The aim of the present research program was to develop a strain of actinomycetes producing extracellular cathepsin B inhibitor. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying various physical and chemical pretreatments. An economical and effective method was developed for the screening of strains producing low molecular weight cathepsin B inhibitor, and consequently a strain (SMF28) among over 700 isolates was selected. Chemotaxonomic and numerical identification were carried out for the isolate. Fifty taxonomic unit characters were tested and the data were analyzed numerically using TAXON program. The isolate was identified as a strain of Streptomyces chromofuscus.

  • To find ${\alpha}$-Glucosidase Inhibitors produced by Actinomycetes, 20 soil samples were tested and 53 Actimycetes were isolated. One of 53 Actinomycetes (strain PM718) showed very potent inhibitory activity in vitro. The morphological and physiological characteristics of strain PM 718 were investigated. The spore morphology. spore chain morphology and spore surface were observed by scanning electron microscope. The inhibitory activity of strain PM718 in vivo has been studied in mice made hyperglycemia by Streptozotocin treatment. The strain PM718 showed signficant reduction of blood glucose level(more than 30%) in mice loaded with maltose.

  • Antitumor antibiotic material was produced by Streptomyces sp. YBE-316 which was isolated from soil, and the optimal culture conditions for the antitumor antibiotic material production were as follows; 2.0% (w/v) sucrose, 0.8% (w/v) polypeptone, 0.4% (w/v) yeast extract, 0.2% (w/v) K$_{2}$HPO$_{4}$, pH 7.0, at 30$\circ$C, 150 rpm and for 100 hours culture. The antitumor antibiotic material had strong antitumor antibiotic activities against most testing tumor cell lines, gram positive and negative bacteria, yeasts, and, especially, Penicillium chrysogenum in fungi.

Isolation and Characterization of Soil Streptomyces Involved in 2,4-Dichlorophenol Oxidation

  • Over 50 morphologically distinctive soil Streptomyces were isolated from various Jocations in the Yongin area in Korea and visually screened for dye-decoloring activities on an agar plate. Two Streptomyces species (AD001 and ND002) showed strong dye-decoloring activities on the plate containing congo-red and new-fuchin dyes, respectively. Also, the liquid culture supernatants of these species showed 2,4-dicholophenol (DCP) oxidation activities only in the presence of hydrogen peroxide, a characteristic of Actinomycetes lignin-peroxidase (ALiP)-P3 isoform found in dye-degrading S. viridosporus T7A and S. badius 252. Based on their dye-decoloring capabilities and the 2,4-DCP oxidation kinetic data, it is suggested that these Streptomyces secrete not-yet-characterized extracelluar enzyme(s), whose activities are very similar to the ALiP-P3 enzyme.

  • 본 연구에서는 독성이 적고, 활성이 높으면서 안정한 새로운 tyrosinase 저해제를 미생물을 대상으로 얻고자 tyrosinase 저해제를 강하게 분비하는 방선균 F-97을 토양으로부터 분리하였다. 이 균의 기균사는 회색의 나뭇가지형 (tree type)이며, 포자의 끝부분이 유선형으로 회전되어 있고, 포자 표면은 구형의 smooth형으로 분류되었다. 세포벽의 DAP 성분은 LL-DAP이었으며, 세포균체의 당 성분은 검출되지 않았다. 기타 배양학적 및 생리적 특성을 종합한 결과 streptomyces aburaviensis 또는 근연균으로 추정된다.
