Maximizing Biomass Productivity and $CO_2$ Biofixation of Microalga, Scenedesmus sp. by Using Sodium Hydroxide

  • A series of experiments were carried out with three native strains of microalgae to measure growth rates, biomass, and lipid productivities. Scenedesmus sp. IMMTCC-6 had better biomass growth rate and higher lipid production. The growth, lipid accumulation, and carbon dioxide ($CO_2$) consumption rate of Scenedesmus sp. IMMTCC-6 were tested under different NaOH concentrations in modified BBM. The algal strain showed the maximum specific growth rate (0.474/day), biomass productivity (110.9 mg $l^{-1}d^{-1}$), and $CO_2$ consumption rate (208.4 mg $l^{-1}d^{-1}$) with an NaOH concentration of 0.005 M on the $8^{th}$ day of cultivation. These values were 2.03-, 6.89-, and 6.88-fold more than the algal cultures grown in control conditions (having no NaOH and $CO_2$). The $CO_2$ fixing efficiency of the microalga with other alternative carbon sources like $Na_2CO_3$ and $NaHCO_3$ was also investigated and compared. The optimized experimental parameters at shake-flask scale were implemented for scaling up the process in a self-engineered photobioreactor. A significant increase in lipid accumulation (14.23% to 31.74%) by the algal strain from the logarithmic to stationary phases was obtained. The algal lipids were mainly composed of $C_{16}/C_{18}$ fatty acids, and are desirable for biodiesel production. The study suggests that microalga Scenedesmus sp. IMMTCC-6 is an efficient strain for biodiesel production and $CO_2$ biofixation using stripping solution of NaOH in a cyclic process.

Organic Matter Degradation and Nitrification Characteristics in Aerobic Digestion of Sewage Sludge (슬러지 호기성 소화공정에서의 유기물 분해 및 질산화 특성)

  • Continuous operation of aerobic sludge digestion reactor was attempted for 279 days. Anaerobic digester sludge, the target material of the experiment, was pretreated by sodium hydroxide at $40^{\circ}C$ for 120 minutes, and the pretreated sludge was fed to 5 L CSTR (continuous stirred tank reactor). Reactor performance was affected by properties of input sludge and HRT (hydraulic retention time). 6 days of HRT showed best and stable performance, and under this condition, removal rates of $NH_3$-N, SCOD, TKN, TCOD, SS, and VSS were 97.4%, 81.7%, 68.7%, 61.4%, 50.6%, and 47.0%, respectively. 73.9% of SS in anaerobic digester sludge was reduced by pretreatment and aerobic digestion. Effluent sludge had low soluble COD of 350 mg/L. This implied the sludge was stabilized and suitable for use as liquid fertilizer. Nitrification took place when HRT was higher than 4 days. $NO_3$-N concentration was as high as 658 mg/L while $NH_3$-N was as low as 20 mg/L.

  • In the present study, kenaf fibers were treated with sodium hydroxide using soaking and ultrasonic methods prior to biocomposite processing, respectively. The effect of alkali treatment on the kenaf-PLA interfacial adhesion and mechanical and thermal characteristics of kenaf/poly(lactic acid) biocomposites was investigated in terms of their interfacial shear strength, flexural properties, dynamic mechanical properties and thermal stability and also microscopic observations of kenaf fibers and the composite fracture surfaces. As a result, use of both soaking and ultrasonic methods for treating kenaf fiber surfaces played a role in increasing the fiber-matrix adhesion and the mechanical properties of the biocomposites. Their characteristics depended not only on the fiber surface treatment method but also on the treatment condition like alkali concentration and treatment time.

  • A study on the solvent extraction of hexavalent chromium from aqueous solutions has been investigated. The results showed that the extraction efficiency of chromium increased in proportion to extractant concentration. Aliquat 336 of quarternary amine effectively extracted hexavalent chromium and was superior to Alamine 336. And the stabilizers like polyhydric alcohols of octanol, decanol and dodecanol showed the similar effect fur prevention of third phase and a little promotion of extraction efficiency in comparison with non-use of polyhydric alcohols. On the other hand, in solvent extraction of hexavalent chromium from aqueous solutions, the hexavalent chromium was completely extracted at pH range lower than 7 with 1% Aliquat 336 as extractant and 5% decanol as stabilizer and the hexavalent chromium in extractant was completely stripped with 1M sodium hydroxide solution in the stripping step.

Effect of Dilute Alkali on Structural Features and Enzymatic Hydrolysis of Barley Straw (Hordeum vulgare) at Boiling Temperature with Low Residence Time

  • This work was conducted to evaluate the effect of dilute sodium hydroxide (NaOH) on barley straw at boiling temperature and fractionation of its biomass components into lignin, hemicellulose, and reducing sugars. To this end, various concentrations of NaOH (0.5% to 2%) were applied for pretreatment of barley straw at $105^{\circ}C$ for 10 min. Scanning electron microscopy (SEM), atomic force microscopy (AFM), and Fourier transform infrared (FTIR) spectroscopy studies revealed that 2% NaOH-pretreated barley straw exposed cellulose fibers on which surface granules were abolished due to comprehensive removal of lignin and hemicellulose. The X-ray diffractometer (XRD) result showed that the crystalline index was increased with increased concentration of NaOH and found a maximum 71.5% for 2% NaOH-pretreated sample. The maximum removal of lignin and hemicellulose was 84.8% and 79.5% from 2% NaOH-pretreated liquor, respectively. Reducing sugar yield was 86.5% from 2% NaOH-pretreated sample using an enzyme dose containing 20 FPU of cellulase, 40 IU of ${\beta}$-glucosidase, and 4 FXU of xylanase/g substrate. The results of this study suggest that it is possible to produce the bioethanol precursor from barley straw using 2% NaOH at boiling temperature.

  • A study on the behavior of boron extraction by TMPD(2,2,4-trimethyl-1,3-pentanediol) was carried out to find the optimum conditions for the boron extraction from brine. In case of boron extraction from 0.736 g/L boron-containing brine, typical optimum extractive conditions would be confirmed to be 0.75 mol/L of extractant concentration at 3.0 pH of brine with 1 of phase ratio and 20 min. of shaking time at 298 K respectively. And 1 mol/L sodium hydroxide of stripping agent was shown above 99 % of boron extraction and stripping efficiencies. Also, extraction equilibrium equation were obtained through experiments as follows : log D = 1.7 log $[TMPD]_O$ + constant at pH < 6.

A Simple and Effective Purification Method for Removal of U(VI) from Soil-Flushing Effluent Using Precipitation: Distillation Process for Clearance

  • Background: The purpose of this study is to purify uranium (U[VI])-contaminated soil-flushing effluent using the precipitation-distillation process for clearance. Precipitation and distillation are commonly used techniques for water treatment. We propose using a combination of these methods for the simple and effective removal of U(VI) ions from soil-flushing effluents. In addition, the U concentration (Bq/g) of solid waste generated in the proposed treatment process was analyzed to confirm whether it satisfies the clearance level. Materials and Methods: Uranium-contaminated soil was decontaminated by soil-flushing using 0.5 M sulfuric acid. The soil-flushing effluent was treated with sodium hydroxide powder to precipitate U(VI) ions, and the remaining U(VI) ions were removed by phosphate addition. The effluent from which U(VI) ions were removed was distilled for reuse as a soil-flushing eluent. Results and Discussion: The purification method using the precipitation-distillation process proposed in this study effectively removes U(VI) ions from U-contaminated soil-flushing effluent. In addition, most of the solid waste generated in the purification process satisfied the clearance level. Conclusion: The proposed purification process is considered to have potential as a soil-flushing effluent treatment method to reduce the amount of radioactive waste generated.

Preparation of Conjugated Linolenic Acid from Urea Fractionated Perilla Seed Oil Hydrolysate (우레아 분별된 들기름 가수 분해물을 이용한 Conjugated Linolenic Acid(CLnA)의 합성)

  • Conjugated linolenic acid (CLnA) and conjugated linoleic acid (CLA) are positional geometric isomers with three and two double bonds, respectively. In this study, perilla seed oil containing 60% ${\alpha}$-linolenic acid (C18:3) and 30% linoleic acid (C18:2) was used as a reaction substrate. After the perilla seed oil was hydrolyzed, conjugated fatty acids were synthesized using different reaction parameters, such as reaction time and concentration of sodium hydroxide. As a result, CLnA, CLA, and other newly synthesized conjugated isomers were present at levels of 14.5%, 14%, and 42.2%, respectively, when the reaction was performed with 20% NaOH, at $180^{\circ}C$, and for 1 hr. The results of GC-MS and fourier transform infrared spectroscopy (FT-IR) showed that CLnA isomer of cis-9, trans-11, and trans-13 octadecatrienoate, CLA isomer of cis-9, trans-11, and trans-10, cis-12 octadecadienoate, and other conjugated isomers were produced. Using urea, ${\alpha}$-linolenic acid could be concentrated from perilla seed oil hydrolysate. After concentration by urea, the concentration of ${\alpha}$-linolenic acid reached about 70%. After alkaline-isomerization was performed on the urea fraction containing 70% ${\alpha}$-linolenic acid, the content of CLnA increased up to 16.6%.

Studies on the Production of Artificial Zeolite from Coal Fly Ash and Its Utilization in Agro-Environment

  • 1. Production of the artificial zeolite from coal ash Coal fly ash is mainly composed of several oxides including $SiO_2$ and $Al_2O_3$ derived from inorganic compounds remained after burning. As minor components, $Fe_2O_3$ and oxides of Mg, Ca, P, Ti (trace) are also contained in the ash. These components are presented as glass form resulting from fusion in the process of the combustion of coal. In other word, coal ash may refer to a kind of aluminosilicate glass that is known to easily change to zeolite-like materials by hydrothermal reaction. Lots of hot seawater is disposing near thermal power plants after cooling turbine generator periodically. Using seawater in the hydrothermal reaction caused to produce low price artificial zeolite by reduction of sodium hydroxide consumption, heating energy and water cost. As coal ash were reacted hydrothermally, peaks of quartz and mullite in the ash were weakened and disappeared, and new Na-Pl peaks were appeared strengthily. Si-O-Si bonding of the bituminous coal ash was changed to Si-O-Al (and $Fe^{3+}$) bonding by the reaction. Therefore the produced Na-Pl type zeolite had high CEC of 276.7 $cmol^+{\cdot}kg^{-1}$ and well developed molecular sieve structure with low concentration of heavy metals. 2. Utilization of the artificial zeolite in agro-environment The artificial zeolite(1g) could remove 123.5 mg of zinc, 164.7 mg copper, 184.4 mg cadmium and 350.6 mg lead in the synthetic wastewater. The removability is higher 2.8 times in zinc, 3.3 times in copper, 4.7 times in cadmium and 4.8 times in lead than natural zeolite and charcoal powder. When the heavy metals were treated at the ratio of 150 $kg{\cdot}ha^{-1}$ to the rice plant, various growth inhibition were observed; brownish discoloration and death of leaf sheath, growth inhibition in culm length, number of panicles and grains, grain ripening and rice yield. But these growth inhibition was greatly alleviated by the application of artificial zeolite, therefore, rice yield increased $1.1{\sim}3.2$ times according to the metal kind. In addition, the concentration of heavy metals in the brown rice also lowered by $27{\sim}75%$. Artificial Granular Zeolites (AGZ) was developed for the purification of wastewater. Canon exchange capacity was 126.8 $cmol^+{\cdot}kg^{-1}$. AGZ had Na-Pl peaks mainly with some minor $C_3S$ peaks in X-ray diffractogram. In addition, AGZs had various pore structure that may be adhere the suspended solid and offer microbiological niche to decompose organic pollutants. AGZ could remove ammonium, orthophosphate and heavy metals simultaneously. Mixing ratio of artificial zeolite in AGZs was related positively with removal efficiency of $NH_4\;^+$ and negatively with that of $PO_4\;^{3-}$. Root growth of rice seedling was inhibited severely in the mine wastewater because of strong acidity and high concentration of heavy metals. As AGZ(1 kg) stayed in the wastewater(100L) for 4days, water quality turned into safely for agricultural usage and rice seedlings grew normally.

Ion Exchange of Glutamic Acid Coupled with Crystallization (결정화 반응이 결합된 글루탐산의 이온교환)

  • A specific ammino auid in a mixture can be crystallized inside an ion exchange column when displacer concentration is high enough to concentrate the amino acid in a pure band beyond its solubility limit. Glutamic acid formpd a discrete crystal layer in a cation exchanger column by operating displacement development mode and using a high concentration of displacer NaOH. The glutamic acid crystal formed was eluded from the column with the effluent stream and collected in a fraction collector. When 1.0 M of NaOH was used as a displacer, more than 60% of the loaded glutamic acid was recovered as crystal. The continuous crystallization and dissolution of crystal occurred, resulting in apparent movement of the crystal along the column without clogging or pressure increase. NaOH was proved a better displacer than NaCl because hydroxide ions neutralized hydrogen ions released from the resin and thus reduced the number of hydrogen ion competing with sodium ion for re-adsorption. The displacement development process coupled with crystallization provided higher concentration and recovery of glutamic acrid than conventional chromatography.
