This study was performed to assess the sodium intakes of Korean adults using a 24-hr urine analysis and dish frequency questionnaire (DFQ) according to each dish group and the regional area. The subjects of this study were comprised of 522 adults (male : 267, female : 285), aged 20-59yr residing in the metropolitan area (N=200), Chungcheng-Do (N=117), Jeolla-Do(N=117), and Gueongsang-Do provinces (N=118). The subjects were recruited from the residents who once participated or are participating in the various health programs offered by the public health center. The number of subjects who completed the 24-hr urine collection was 205 (male : 110, female : 95). The mean age and BMI of the subjects were $39.0{\pm}$11.7y and $23.1{\pm}2.9 kg/m^2$, respectively. The mean systolic and diastolic blood pressure was $119.5{\pm}15.4 mmHg$, and $77.1{\pm}11.1 mmHg$, respectively. Eighteen percent of the subjects responded that they are currently smoking, 36% drinking and 50.4% exercising. Twenty point six percent of the subjects were assessed as having hypertension according to their systolic or diastolic blood pressure($SBP{\ge}140mmHg$ or $DBP{\ge}90mmHg$) measurements in the present study. Salt intake of the subjects estimated with 24-hr sodium excretion was 12.7g/d (male : 13.4g/d, female : 12.1g/d) based on the sodium excretion rate as 82%. Salt intake estimated with DFQ was 14.7g/d (male : 16.2g/d, female : 13.4g/d), 2 g more than the salt intake estimated with 24-hr urine analysis. The four dish groups that contributed most to the sodium intake in order were kimchi (11571.4mg), soup and stew (1260.5mg), fish and shellfish(706.3mg) and noodle and ramyeon(644.3mg). Salt intake estimated with DFQ was the highest in the subjects of Gueongsang-Do(17.0g/d), second highest Chungcheong-Do (16.4g/d) and the lowest in the metropolitan area (13.0g/d). Subjects of Gueongsang-Do showed the highest sodium intakes in most of the dish group, whereas subjects of the metropolitan area showed the lowest. Residents of Chungcheong-Do revealed the highest sodium intake with kimchi and of Jeolla-Do the higher sodium intake with main dish (meat, fish and beans). The highest salt percentage of kimchi ($3.0{\pm}0.8%$) and soybean paste ($14.5{\pm}5.1%$) were observed in Gueongsang-Do, whereas individuals of the metropolitan area were observed as having kimchi ($1.6{\pm}0.5%$) and soybean paste ($7.4{\pm}1.6%$) with the lowest salt percenage. Men were observed as having more salty kimchi ($2.4{\pm}0.1%$) than women ($2.1{\pm}0.1%$).
This study was conducted to investigate the hypocholesterolemic effects of porphyran and insoluble dietary fiber isolated from laver in rats fed high fat diet containing 1% cholesterol, 0.25% sodium cholate and 12% lard. Rats were fed, ad libitum, diets containing 5% diet fiber as cellulose(normal control or high fat control), porphyran or insoluble dietary fiber for 4 weeks. Among the groups fed high fat diet, liver weight was significantly lower in high fat porphyran group than high fat control. Plasma GOT, GPT, total cholesterol, cholesteryl ester, LDL-cholesterol and liver total cholesterol concentration were significantly lower in high fat porphyran group than high fat control. The feeding of porphyran significantly increased fecal cholesterol and bile acid excretion. The feeding of insoluble dietary fiber had no significant effect on either plasma or liver cholesterol levels, although fecal cholesterol level in the insoluble dietary fiber group was significantly higher than that in the high fat control. The results indicate that porphyran isolated from the laver may exert their hypocholesterolemic effect by increasing excretion of fecal bile acid and cholesterol.
Renal pathways for excretion of sulfadiazine has been studied by standard clearance technique in the rabbit. 1. Large part of sulfadiazine filtered in the glomeruli is reabsorbed in the tubules, as visualized from the fact that Csd (clearance of sulfadiazine) amounts only a fraction of simultaneously measured Ccr (GFR). 2. Csd changed linearly with the rate of urine flow, whether it is increased by the duir etics or decreased by clamping u reter. 3. Csd remained unchanged until the plasma level of the Csdremained unchanged drug reached 10.0 mg%, and the amount transported in the tubules increased linearly with the increase in the load, exhibiting No maximum capacity for transport. 4. Csd was increased by 2,4-dinitrophenol which is an uncoupling agent of oxidative phosphorylation and decreased by probenecid which is on uricosuric agent. 5. During sodium bicarbonate infusion net secretion of sulfadiazine by tubules observed. All the evidences obtained in the rabbit indicated that sulfadiazine was reabsorbed by active, energy-requiring, or passive, simple diffusion, process, and secreted simultaneously by a probenecid-sensitive, active procss.
The mechanism of urinary excretion of sulfadiazine was investigated in the dogs by means of stop-flow technique Results of experiments were summarized as follows; 1. The ratios of U/P sulfadiazine to U/P creatinine $(U/P_{SD}:U/P_{cr})$ were always lower than 1 in all nephrons, showed a minimum in the proximal area. 2. $U/P_{SD}:U/P_{cr}$ were not affected by Probenecid or 2.4-DNP, whereas increased significantly by administration of sodium bicarbonate. 3. Probenecid did not alter the stop-flow patterns in alkalotic dog too. 4. $C_{SD}$ (clearance of sulfadiazine ) was appreciably influenced by change in urinary PH, or flow rate. All evidences lead to the conclusion that sulfadiazine reabsorption is passively transported by proximal tubules. No clue for active process, either secretory or reabsorptive, was obtained.
General pharmacological properties of recombinant human parathyroid hormone (hPTH) were examined. The administration of hPTH (7, 35 and 175 lU/kg sc) in mice had no effects in general behavior and central nervous system, and no influences on normal body temperature, writhing syndromes induced by 0.7% acetic acid solution and chemoshock produced by strychnine and pentetrazol solution. hPTH (9 and 44 lU/kg, sc) given to anesthetized rabbits showed no effect on blood pressure of carotid artery and respiration, and it did not influence the responses produced by injection of acetylcholine or epinephrine. It showed no direct effect at 4.4$\times$10$_{-2}$IU/ml in isolated stomach fundus and uterus of rats and ileum of guinea-pig, and it also showed no inhibition of contraction produced by acetylcholine, oxytocin, serotonin and histamine in the above-mentioned preparations. It did not influence intestinal propulsion at the doses of 7,35 and 175 lU/kg sc in mice. This drug exhibited no effect on urinary excretion at the doses of 7 and 35 lU/kg sc in rats. However, at a dose of 175 lU/kg sc, it showed a decreased urination along with decreased excretion of potassium, sodium and chloride ion. These results indicate that hPTH does not exert any of serious pharmacological effects except the inhibition of urination at a highest dose used.
In order to evaluate the effect of habitual Na and Ca intake on blood pressure regulation, we measured the habitual dietary intakes of Na and Ca, urinary excretion of Ca, Na and K, and plasma level of renin activity, aldosterone, and indices of Ca metabolism in 27 untreated hypertensive women and 30 age-matched normal women on a free diet. Hypertensive and total subjects were divided into four groups according to habitual dietary intakes of Na and Ca as low Na-low Ca(LNLC), low Na-high Ca(LNHC), high Na-low Ca(HNLC), and high Na-high Ca(HNHC). HNLC hypertensive group showed the lowest level of plasma renin activity, 25-(OH) Vit D$_3$, calcitonin and serum total Ca, and presented the highest level of PTH and urinary excretions of Na/K and Ca/Cr. There were no significant difference in plasma level of aldosterone and urinary excretion of Na and K among four hypertensive groups. When all subjects were divided into four groups according to the same method, HNLC group showed the highest level of blood pressure with no statistical significance and the lowest level of calcitonin and total serum Ca. The above results indicated that renin-aldosterone system and Ca regulating hormone has a mutual relationship in hypertension. Na and Ca may interact each other, rather than affecting independently blood pressure control. As a result, considering the fact that daily balance of Na and Ca intakes affects Na and Ca regulating hormones and urinary excretion of Na and Ca, it may be involved in blood pressure control. These results suggest that maintaining an adequate intake of Ca with less intake of Na may prevent from the risk of hypertension. (Korean J Nutrition 34(4) : 409~416, 2001)
산업체 근로자의 potassium 섭취상태와 배설상태를 평가하기 위하여 서울시내 H타이어 공장에 근무하는 건강한 성인남자 40명을 대상으로 3일간 식사 분석과 뇨 분석을 행하였다. 대상자들의 3일동안 평균 potassium 섭취량은 $54.5\pm16.7mEq/day(2.13\pm0.64g)$이었고, 24시간동안 뇨 중의 potassium 배설량은 $45.9\pm10.5mEq/day(1.77\pm0.41g)$로 소변중 배설은 83%정도였다. Na와 K의 섭취비율은 $4.15\pm0.58$인 반면에, 뇨 중 배설비율은 $5.20\pm1.11$이었다. 1일 섭취하는 potassium의 주요 급원은 밥 중에서 콩 밥, 국 종류에서는 감자된장국, 반찬류에서는 삼치무조림과 묵야채무침이었다. 단백질 섭취량과 K 섭취랑, N 배설량과 K 배설량, N 배설량과 Na 배설량, K 섭취량과 Na 섭취량, 그리고 K 배설량과 Na 배설량간에 모두 유의적인 상관을 나타내었다.
The present study examined pharmacokinetic profiles of KBP31705-Cl27 and KBP30603-901, new platinum coordination complexes synthesized as anticancer candidates, in comparison with two well-known platinum-containing anticancer agents, cisplatin and carboplatin in rats. Under sodium pentobarbital anesthesia of male Sprague-Dawley rats, urinary bladder, and femoral artery and vein were catheterized for urine collection, blood sampling and drug injection, respectively Following i.v. administration of cisplatin (2 mg/kg), KBP31705-C127 (2 mg/kg), carboplatin (20 mg/kg) or KBP30603-901 (20 mg/kg), blood samples were collected at 2, 4, 6, 8, 10, 15, 20, 30, 45, 60 and 120 minutes. Urine samples were collected at 1-hr interval for 4 hr. Platinum concentrations in plasma and urine were measured using an inductively coupled plasmamass spectrometer. The plasma concentration-time curves were biphasic for all drugs during the time period studied. Compared with cisplatin, KBP31705-C127 showed similar decay patters in the alpha- and betaphases with slightly lower plasma concentrations. Urinary platinum excretion for cisplatin and KBP31705-C 127 was 56 and 52% of the administered dose in 4 hr, respectively. With regard to carboplatin and KBP 30603-901, a similar decay pattern was also observed in the alpha-phase. The half life of KBP30603-901 in the beta-phase, however, was much longer than that of carboplatin, which was consistent with the urinary excretion results that 46 and 59% of the administered dose were excreted in the urine in 4hr, respectively. The results suggest that platinum coordination complexes are primarily excreted via the renal route and KBP30603-901 can elicit longer duration of action due to slower renal excretion compared to carboplatin.
The aim of this study was to investigate the effect of supplemental recombinant bovine somatotropin (rbST) and cooling with misters and fans on renal function in relation to regulation of body fluids in different stages of lactation in crossbred Holstein cattle. Ten, 87.5% crossbred Holstein cattle were divided into two groups of 5 animals each, housing in a normal shaded barn (NS) and in a shaded barn with a mister-fans cooling system (MF). The experiment in each group was divided into 3 phases, early- (Day 75 postpartum), mid- (Day 135 postpartum), and late stage of lactation (Day 195 postpartum). The pre-treatment study was conducted on the starting day of each stage of lactation and the treatment study was performed after the end of the pre-treatment, during which the animal was injected with 500 mg of rbST (POSILAC) every 14 days for three times. During the study, ambient temperature at the hottest period daily in the MF barn was significantly lower, while relative humidity was higher than that of the NS barn. The temperature humidity index (THI) in both barns ranged from 79-85 throughout the periods of study. Cows in the MF barn showed a lower rectal temperature and respiration rate as compared with cows in the NS barn. The effect of rbST administration increased both rectal temperature and respiration rates of cows housed in either the NS or MF barn. Milk yield significantly increased in cows treated with rbST in all stages of lactation. Increases in mammary blood flow, accompanied by increases of total body water (TBW), extracellular fluid (ECF), blood volume (BV) and plasma volume (PV), were observed in both groups of cows receiving rbST in all stages of lactation. No alterations of renal blood flow and glomerular filtration rate were observed in cows receiving rbST, but decreases in urinary excretion and fractional excretion of sodium, potassium and chloride ions appeared to correlate with reduction in the rate of urine flow and osmolar clearance during rbST administration. These results suggest that the effect of rbST supplementation to cows housed either in NS or MF barns on body fluid volume expansion is attributable to changes in the rate of electrolyte excretion by the kidney. The increased availability of renal tubular reabsorption of sodium, potassium and chloride ions during rbST treatment was a major factor in retaining body water through its colligative properties in exerting formation of an osmotic force mechanism.
본 연구는 해조류가 정상적인 Na, Ca, K 흡수대사와 지질대사에 미치는 영향을 살펴보기 위하여 시행되었다. 실험대상자들은 영양적으로 조절된 일상식이로 5일간 적응시킨 후 미역, 다시마, 파래, 김 등 해조류가 첨가된 식이를 5일간 섭취하였다. 이때 전 실험기간 동안 섭취된 Na, Ca, K 배설량을 측정하였고 또한 혈청중 total cholesterol, triglyceride, free fatty acid 함량을 분석함으로써 해조류와 Na, Ca, K 배설량과 지질대사와의 유의성을 규명하였다. 결과는 다음과 같다. 1) 실험기간동안 섭취된 Na와 K는 각각 56%, 64%의 비율로 소변으로 배설되었지만, 반대로 Ca는 주로 대변으로 많이 배설되었다. 2) 식이로 섭취된 양과 대변중으로 배설된 양과의 비교에서 Na와 K는 유의적인 차이를 나타내었다. 3) 반면에 식이로 섭취된 Na, Ca, K는 혈청 농도에 아무런 영향을 미치지 않았다. 4) 혈청 total cholesterol, triglyceride 농도는 유의적(P<0.05) 수준에서 현저히 감소함으로써 해조류가 정상인의 지질대사에 영향을 미친다는 것을 알 수 있었다. 결과적으로 본 연구에서 선택한 미역, 다시마, 파래, 김 등의 해조류는 체내 무기질 이온의 대변으로서의 배설을 촉진하여 정상인의 Na, Ca 및 K 대사조절에 영향을 미치며 혈청 지질 강하 효과가 있음을 알 수 있었다.
