Stress Responses of the Escherichia coli groE Promoter

  • GroEL is well known as a molecular chaperone. In order to determine the dynamic stress response of the Escherichia coli groE promoter, a groE-lacZ operon fusion in the chromosome was constructed. Stress leading to ${\sigma}^{32}$ synthesis induces transcription from E. coli groE promoter, since the promoter is ${\sigma}^{32}-regulated$. When the strain was stressed with ethanol, phenol, and sodium chloride, clear inductions of ${\beta}-galactosidase$ were observed. Two types of simultaneous stresses of sodium chloride and phenol induced the enze much more than either of the two alone, suggesting that stress was an additive. The combined stress resulted in the highest induction of the enzyme in this system. The groE-lacZ fusion strain developed in this study can conveniently be used to detect other harmful pollutants in the environment. Stress treatment of cells containing recombinant proteins, which need GroEl, by ethanol, phenol, or sodium chloride, might have a tendency to increase their biological activities.

Current Status of Eco-Friendly Deicing Material Development (친환경 제설제 개발 현황)

  • Recently, due to the climate change by global warming, the amount of snow has increased. The situations were demanded to use many snow removal materials. The snow removal materials that used in Korea were solid chloride deicing material. That is calcium chloride($CaCl_2$) and sodium chloride(NaCl). Solid chloride baesd snow removal materials have various property, for example good freezing point depression, deliquescence and economical solvents. However, there are problems such as water pollution, high corrosiveness and ecocide. For such reason, development of eco-friendly deicer was required obligatorily. This study investigated development situation of eco-friendly deicer materials.

Kinetic Evidence for the Interactive Inhibition of Laccase from Trametes versicolor by pH and Chloride

  • The interactive inhibitory effects of pH and chloride on the catalysis of laccase from Trametes versicolor were investigated by studying the alteration of inhibition characteristics of sodium chloride at different pHs for the oxidation of 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid). At pH 3.0, the addition of sodium chloride (50 mM) brought about a 40-fold increase in $K{_m}^{app}$ and a 4-fold decrease in $V_{max}{^{app}}$. As the pH increased to 7.0, the inhibitory effects of sodium chloride became significantly weakened. The mixed-inhibition mechanism was successfully used to quantitatively estimate the competitive and uncompetitive inhibition strengths by chloride at two different pHs (pH 3.0 and 6.0). At pH 3.0, the competitive inhibition constant, $K_i$, was 0.35 mM, whereas the uncompetitive inhibition constant, $K{_i}^{\prime}$, was 18.1 mM, indicating that the major cause of the laccase inhibition by chloride is due to the competitive inhibition step. At a higher pH of 6.0, where the inhibition of the laccase by hydroxide ions takes effect, the inhibition of the laccase by chloride diminished to a great extent, showing increased values of both the competitive inhibition constant ($K_i=23.7mM$) and uncompetitive inhibition constant ($K{_i}^{\prime}=324mM$). These kinetic results evidenced that the hydroxide anion and chloride share a common mechanism to inhibit the laccase activity.

Effects of Heat, Salt and Hydrocolloid Treatments on Flying Fish Cypselurus agoo Roe Analogs Prepared Using Calcium Alginate Hydrogels

  • The present study was conducted to elucidate effects of heat, salt and hydrocolloid treatments on flying fish Cypselurus agoo roe analogs prepared using calcium alginate gel. The changes in size, sphericity and rupture strength of the analogs as affected by treatments of heat, sodium chloride and hydrocolloids were investigated. The size (mm), sphericity (%), and rupture strength (kPa) of the analogs were $2.2{\pm}0.1$, $98.2{\pm}0.2$, and $74.7{\pm}1.7$, respectively. When the analogs were heated at $95^{\circ}C$ in water, the size was slightly decreased. The rupture strength by curing with 2% sodium chloride was slightly increased. Sphericity didn't show significant differences by sodium chloride and heat treatment. The rupture strength of the analogs was slightly decreased by heat treatment, whereas remarkably decreased by curing with sodium chloride. In order to prevent a remarkable decrease in rupture strength of the analogs by curing with sodium chloride, the analogs were treated with hydrocolloids such as xanthan gum, gum guar, glucomannan, pectin and gelatin. The hydrocolloids treated analogs showed an increment in size and no significant changes in sphericity. On the other hand, the rupture strength of the hydrocolloids treated analogs exhibited remarkable increase than that of untreated ones.

Effect of High Concentrations of Sodium or Chloride Salts in Soil on the Growth of and Mineral Uptake by Tomatoes (토양에의 고농도 Na 및 Cl 염류가 토마토의 생육 및 무기성분 흡수에 미치는 영향)

  • This study was conducted to investigate the effect of high concentration of sodium salts and chlorides in soil on the growth of tomato and the uptake of minerals. The growth inhibition rates of plant height and dry weight were different depending on salts, but they were not related to the electric conductivities (EC) and acidities (pH) in the soil solution. The orders of growth inhibition were Cl, SO$_4$, CO$_3$, PO$_4$>NO$_3$ in the sodium salts series, and Na, K, Mg, NH$_4$>Ca in the chlorides. The growth inhibition rates of the sodium salts series tended to be larger than those of the chloride series. Yield was lower 30%~10% in the sodium salt and chloride series than in the control. Chlorophyll content, photosynthetic rate and stomatal conductance were lower in the sodium salts and chloride series than in the control. Mineral concentration was lower in sodium salts and chlorides than in control. The nitrate absorption was inhibited in all salts except for NaNO$_3$ and NH$_4$Cl, and specially in NaCl and Na$_2$SO$_4$ treatments of the sodium salts and in KCl treatment of chloride series. K concentration was reduced NaCl and Na$_2$SO$_4$ treatments compared with the other salts. In the sodium salt series, calcium and magnesium concentration were decreased antagonistically when sodium concentration was increased.

A Study on the Contents of Some Minerals in Korean Daily Foods (한국상용식품중(韓國常用食品中)의 무기물함량(無機物含量)에 관(關)한 연구(硏究) (특(特)히 Lithium, Sodium 및 Potassium함량(含量)))

  • In order to observe the contents of some minerals in Korean daily food, the lithium, sodium and potassium were determined by E.E.L flame photometry. The results obtained are summarized as follows: 1) The lithium content was most higher (13. 8 to 16.8mg/100g) in sodium chloride, the contents of the lithium End sodium in each food were increased according to supplement of sodium chloride. 2) The potassium contents in garlic and bean paste were 137.6 mg and 186.3 mg per 100g sample, respectively, therefore these values were higher than the values in other samples. 3) The contents of lithium, sodium and potassium in rice, barley pressed and boiled rice lower than other samples.

Sodium Chloride and Nitrogen Metabolism of Korean Females (한국여자의 소금 및 질소대사에 관하여)

  • In order to study the dally metabolism of sodium chloride and of nitrogen, 24-hour urine samples were collected from 1,593 normal Korean females whose ages varied from 2 to 80 years old. The volume, the concentration of chloride and the osmolality of the urine, add the total nitrogen were determined, along with the resting pulse rate and the blood pressure. The daily urine volume was maintained at $1,000{\sim}1,300\;ml/m^2/day$ in all age groups while the chloride concentration and osmolality of the urine samples were approximately 200 mEq/liter and 600 milliosmoles, respectively, in most of age groups. Hence the daily urinary output of sodium chloride was estimated to be approximately $15g/m^2$/day in adult groups. On the other hand, the daily excretion of total nitrogen amouted to approximately $5{\sim}6g/m^2/day$. These findings indicated that the average Korean females live on low-protein and high-salt diets throughout their life. Despite a known correlation between the incidence of hypertension and the high salt intake, none of the subjects employed in this work showed any sign of hypertension.

Studies on Sodium Chloride and Nitrogen Metabolism of the Korean (한국인(韓國人)의 식염(食鹽) 및 질소대사(窒素代謝)에 관(關)하여)

  • In order to study the daily metabolism of sodium chloride and of nitrogen, 24 hour urine samples were collected from 298 normal Korean males whose ages varied Iron 26 to 80 years old. The volume and the concentration of chloride and the total nitrogen were determine, along with the resting pulse and the blood pressure. The daily urine volume was maintained at approximately $1,000\;ml/m^2$ in all age groups while the chloride concentration was at approximately 230 mEq/l. Hence the daily urinary output of sodium chloride was estimated to be approximately 21 gm. On the other hand, the daily urinary output of total nitrogen amounted to approximately 10 gm. Theses findings are similar to those obtained earlier by Yi et al. (1966) in Korean subjects younger than 25 fears old, indicating that average Koreans live on low protein and high salt diets throughout their life. Despite a known correlation between the incidence of hypertension and the high salt intake, none of the these subjects employed in the present investigation showed any sign of hypertension.

Effect of Sodium Chloride Supplementation on Bone Metablism in Rats Consuming a Low Calcium Diet (저칼슘 식이 섭취시 식염첨가가 흰쥐의 골격대사에 미치는 영향)

  • This study was performed to evaluate the effect of sodium cholride supplementation on bone metabolism in female rats consuming a low calcium diet. Twenty five female rats were divided into three dietary groups (control Na : 0.1038%, 1% Na : 1.036%, 2% Na : 2.072%). All experimental diets contained 0.27% Ca and were fed to rats with deionized water for 7 weeks. Bone mineral density(BMD) and bone mineral content(BMC) of total body, spine and femur were measured using energy x-ray absorptiometry(DEXA) by small animal software. Then Ca efficiency was calculated from BMD and BMC. Serum Ca, P, Na and urine Ca, P, Na were determined. Urinary pyridinoline, serum ALP were measured to monitor bone resorption. Following 7 weeks, sodium cholride supplemented groups had higher urinary Ca excreteion, urinary pyridinoline, crosslinks value and serum ALP. There was no significant difference in case of serum Ca among all groups. Sodium chloride supplemnted groups had lower Ca effciency of total, spine and femur BMD and BMC than that of control group. In conclusion high salt intake not only increases urinary Ca excretion as urinary Na excretion does but also increase bone resorption and decrease Ca efficiency of each bone. It is been suggested that high salt intake may be harmful for bone maintenance. Therfore, the decrease of salt intake to the level of recommendation would be desirable.

Determination of Some Inorganic Anions in Saline Water by Ion Chromatography with UV Detection (이온크로마토그래피를 이용한 소금물중의 무기음이온들의 분리정량)

  • A stepwise gradient elution with two wavelengths detection was performed for the separation and determination of some anions in saline water. The eight anions such as iodate, bromate, nitrite, bromide, nitrate, chromate, iodide and thiocyanate were successfully separated using AS-7 column and sodium chloride/sodium phosphate buffer solution as an eluant within 40 min. The separation behaviors of anions were studied at various sodium chloride concentrations. The peak shapes of anions of bromate, nitrite, bromide and nitrate gradually broadened as the concentration of sodium chloride increased until 1.0 M in the sample solutions. However, no effect was observed in the peak shapes of chromate, iodide and thiocynate. A good linearity was obtained at the range of ppm(mg/L). The detection limit was proved to be $10-720{\mu}g/L$ for the eight anions with $50{\mu}L$ injection volume. This method was applied to the determination of $Br^-$, ${NO_3}^-$ and $I^-$ in sea water.
