Assessment of Organic Compound and Bioassay in Soil Using Pharmaceutical Byproduct and Cosmetic Industry Wastewater Sludge as Raw Materials of Compost (제약업종 부산물 및 화장품 제조업 폐수처리오니 처리토양에 대한 유기화합물 및 Bioassay 분석 평가)

  • This study was conducted to assessment organic compound and bioassay (density of inhabited animal, fluctuation of predominant fungi, and survival ratio of earthworm) for finding damage on red pepper by heavily amount application of sludges in soil, which was treated with 3 pharmaceutical byproducts and a cosmetic industry wastewater sludge as raw materials of compost, and for establishing estimation method. HEM contents in the soil treated with pharmaceutical byproducts sludge2 (PS2) and cosmetic sludge (CS) were 0.51, 1.10 mg/kg respectively. PAHs content of PS2 treatment in the soil was 3406.8 ug/kg on July 8. In abundance of soil faunas, the pharmaceutical byproducts sludge2 treatment was the most highest. The next was decreased in the order of pig manure (PM) and the cosmetic sludge treatment. However the other pharmaceutical sludge treatments were remarkably reduced populations of soil inhabited animals. In upland soil treated with organic sludges, the numbers of bacteria and fungi of the pharmaceutical sludge treatment were 736, 909 cfu/g and those of the cosmetic sludge treatment were 440, 236 cfu/g, respectively. The pharmaceutical sludge treatments and the cosmetic sludge treatment in identification of predominant bacteria were not any tendency to compare with non fertilizer and pig manure treatments, but they had diverse bacteria than NPK treatment. In microcosm tests, the survival of the tiger earthworm in five soil samples was hardly affected against the soil of PSI (20%) after three months treated in the upland But after six months, survival of PS1 was 80%. At present, raw material of compost was authorized by contents of organic matter, heavy metal (8 elements), and product processing according to 'The specified gist on possible materials of using after analysis and investigation among raw materials of compost', however, for preparing to change regulation of raw material of compost and for considering to possibility of application, this study was conducted to investigate toxic organic compound and bioassay methods using inhabited animal, fungi, and earthworm without current regulation.

Characteristics of Thermophilic Methane Fermentation Using the Organic Wastes (유기성 폐기물을 이용한 고온 메탄 발효의 특성)

  • In this work, it was investigated that various aspects of process, application situation, merits and short-coming results of the thermophilic methane fermentation with highly concentrated organic waste substances such as sewage sludges, food wastes and excretions. The merits of this methane fermentation were that it had a very fast reaction rate and was possible to proceed in high loads. It was also high in mortality for pathogenic microorganism and the digested sludge was more hygienic. However, the short-comings were that more energy was required for heating in the fermentation facility, no surplus energy could be gained from low concentration of organic waste, the fermentation treatment dropped level of water quality, thus burdens discharging process of water. Especially, the high concentration of methane fermentation could possibly lack nutritious salt and could face the disturbance by ${NH_4}^+-N$, a proper alternative was required. In general, thermophilic methane fermentation was considered as a better mean in disposing of cow excretion and food waste which were highly concentrated organic wastes. On the other hand, under the condition where the concentration of waste material was low and the high concentrate waste material became higher than 3,000 mg/L in ${NH_4}^+-N$, thermophilic methane fermentation resulted less desirable outcome.

An Assessment on the Behavior of Nitrogenous Materials during the First High-rate Phase in Composting Process (퇴비화 공정의 1차 발효단계에서 질소성 물질의 거동 평가)

  • Composting of N-rich wastes such as food waste and wastewater sludges can be associated loss of with substantial gaseous N, which means loss of an essential plant nutrient but may also lead to environmental pollution. We investigated the behavior of nitrogenous materials during the first high-rate phase in composting of food waste. Air dried food waste was mixed with shredded waste paper or wood chip and reacted in a bench scale composting reactor. Samples were analyzed for pH, ammonia, oxidized nitrogen and organic nitrogen. The volatilized ammonia nitrogen was also analyzed using sulfuric acid as an absorbent solution. Initial progress of composting reaction greatly influenced the ammonification of organic nitrogen. A well-balanced composting reaction with an addition of active compost as an inoculum resulted in the promoted mineralization of organic nitrogen and volatilization of ammonia. The prolongation of initial low pH period delayed the production of ammonia. It was also found that nitrogen loss was highly dependent on the air flow supplied. With an increase in input air flow, the loss of nitrogen as an ammonia also increased, resulted in substantial reduction of ammonia content in compost. The conversion ratio of initial nitrogen into ammonia was in the range of 28 to 38% and about 77~94% of the ammonia produced was escaped as a gas. Material balance on the nitrogenous materials was demonstrated to provide an information of importance on the behavior of nitrogen in composting reaction.

  • Organic waste, which is defined as wastes derived from various biological organisms that contain more than 40% of organic materials, is generated about 100 million tons per year in Korea. These organic wastes are now controlled by several governmental entities, under different rules and regulations, leading to the improper management and inefficient treatment. Therefore, integrated management system is primarily needed for the efficient recycling of organic waste. In this study, six kinds of organic wastes, which are food waste, sludges(sewage, waste water, night soil), animal excreta, animals and plants residues, and three kinds of recycling by-products(compost, feed, anaerobic digestion by-products) made of organic wastes, were analyzed for their physical and chemical characteristics. On the basis of this result, a possibility for the efficient recycling of organic waste was investigated.

Isolation and Characterization of Bacteria Capable of Degrading Bisphenol A (Bisphenol A 분해세균의 분리 및 특성)

  • Eighty-seven microbial strains capable of growing on bisphenol A (BPA) as a sole carbon source were isolated from soils, waste waters and sludges. Among them, three bacterial strains were finally selected as potential decomposers through measuring BPA-degradation efficiency by HPLC analysis. Two of these bacterial strains were identified as Serratia marcescens 1901 and S. marcescens 1902, and another was Pseudomonas putida 1401 by 16S rDNA partial sequences and based on morphological and physiological properties. They showed higher cell growth and BPA degradation in PAV (PAS medium containing vitamin mixtures) than in PAS medium. The degradation efficiencies of these bacterial strains were within a range of 20-40% in the PAV containing 500 mg/1 or 100 mg/l of BPA fur 3 days. S. marcescens 1901 showed higher degradation efficiency at 100 mg/1 of BPA than those of other selected strains, while S. marcescens 1902 and P. putida 1401 degraded a high concentration of BPA (500 mg/l) with a degradation efficiency of 40% for 3 days. The BPA degradation using a mixed culture of three selected strains showed the similar level of dog-radation efficiency with that using a pure culture.

A Study on the BOD Removal Characteristics of Aerobic Submerged Biofilter (Media를 충전(充塡)한 간접폭기식(間接曝氣式) 침지여상(浸漬濾床)에 의한 BOD제거 특성에 관한 연구)

  • The BOD removal characteristics of submerged biofilters filled with three kinds of filter media respectively were experimentally examined with constant temperature, pH value and variable BOD loading and recirculation ratio. Obtained results are as follows; 1. The BOD removal ratio decreases with the increasing $BOD_5$ volumetric loading rate, and the loading rate for the BOD removal over 90% is lower thean $1.6kg{\cdot}BOD_5/m^3{\cdot}d$ for the plastic media of Netring and cubic wire meshes. This is a much large value than $0.3{\sim}0.8kg{\cdot}BOD_5/m^3{\cdot}d$ for conventional activated sludge process. The required submerged biofilter volume is found to be much samller than that of conventional activated sludge process. 2. The decreasing order of BOD removal is Netring (random plastic media), cubic wire meshes (plastic module), and then gravel (stone media). This is mainly due to the media characteristics such as void ratio, specific surface area and media shapes. 3. The $BOD_5$ removal rate increases with the recircuration ration, but the rate of increases becomes samaller as the recirculation ratio increases over 20. When $BOD_5$ loading is $1.8kg{\cdot}BOD_5/m^3{\cdot}d$, the required recirculation rationto obtain 90% $BOD_5$ removal is about 20 for Netring and it was about 30 for cubic wire meshes. 4. Reynold's Number increases with recirculation ratio, and the Reynold's Numbers corresponding to the recircuration ratio of 10~50 are less than 52, showing laminar up flows in the filter. The settled and effluent sludges increase with increase of Reynol's Number, and there are the definite Reynold's Numbers at which the settling sludge concentrations become nearly constant respectively in each filters. 5. In this submerged biofilter system, small volume of sludge hopper can be substituted for a separated settling tank.

A Study on the Removal of Ammonia by Using Peat Biofilter (미생물 활성토탄을 이용한 암모니아 제거에 관한 연구)

  • Conventional deodorization filters using soil and compost reach the capacity limitation of deodorization in short period, because its removal mechanism primarily depends on adsorption. Therefore, in this study the experiment was performed on the removal of ammonia which is a strong inorganic malodor, frequently emitted from night soil treatment plants and sewage treatment plants, by seeding activated sludges on the bio-peat containing higher organic contents, water conservation capacity, permeability and lower pressure drop. As a result, in raw peat filter natural ammonia outlet was observed in consequence of pH increase resulted from ammonia ionizing in liquid phase. Ammonia removal mechanism primarily depended on the adsorption onto the anion colloidal substances in peat. In peat bio-filter, theoretical ammonium salts ratio was higher than that of raw peat, resulted from slight pH increase by microorganism activity, however, the experimetal value of ammonia-nitrogen accumulated in bio-peat was lower than that of raw peat because of nitrification by nitrifying bacteria. In the initial reaction period, adsorption was predominant in the ammonia removal mechanism, but nitrification was conspicuous after the middle period. Mass balance of nitrogen was established using experimental data of input $NH_3$ loading, output $NH_3$ loading, $NH_4{^+}$-N, $NO_x$-N, and Org-N. The critical time of unsteady state, which is the maximum activating point of microorganism in bio-filter, was determined using experimental data, and the ammonia adsorption curve was computed using regression analysis. On the basis of the results obtained by above analysis, the delay days for the saturation of adsoption capacity in peat bio-filter was calculated.

  • To investigate the effects of paper sludge, by-products of paper manufactory, on the chemical properties of paddy soil and growth of paddy rice, paper sludges were applied to the pots at the rates of 300, 600, 900 and 1,200㎏/l0a which were either pread-justed C/N ratio to 30 : 1 or not adjusted. The effects were compared with those of control. Seasonal variations of various forms of soil organic nitrogen, the mineralization of organic nitrogen and the contents of soil minerals were analyzed. The results were summarized as follows: 1) The contents of soil organic nitrogen, especially amino sugar-N and amino acid-N, organic matter, CEC and available $SiO_2$ were increased by application of paper sludge compared with that of control. 2) The mineralizations of organic nitrogen after 6 weeks of incubation at $30^{\circ}C$ were 12. 2, 12.6, 15.1, 9.7 and $15.2^{\circ}C$ in the control, 300㎏/l0a sludge treatment, 300㎏/l0a sludge treatment(C/N ratio adjusted), 600㎏/l0a sludge treatment and 600㎏/l0a sludge treatment(C/N ratio adjusted), respectively. 3) Cu and Pb contents in the soil were in the range of $0{\sim}0.2100$ and $0{\sim}0.0013$ ppm, respectively. Cr and Cu in the soil were not detected at all. There were no differences in the contents of all the heavy metals among all the treatments in the soil.

Isolation of Cadmium-Tolerant Bacteria and Characterization of Cadmium Accumulation into the Bacteria Cell (카드뮴 내성균(耐性菌)의 분리(分離), 동정(同定)및 균체내(菌體內) 카드뮴 축적(蓄積) 특성(特性))

  • Of the cadmium-tolerant 162 bacterial strains isolated from soils, river waters or active sludges of waste-water disposal plants in the Gyeongnam province a strain C1, which showed considerably higher growth rate in the agar plate containing 2000 ppm than any other strains isolated, was identified as a Pseudomonas putida or its similar strain when analyzed by taxonomical characteristics. Optimum pH and temperature for the growth of the P, putida were 7.0 and $30^{\circ}C$, respectively. This strain was resistant to antibiotics(ampicillin, chloramphenicol and streptomycin), and heavy metals(lithium, cupper, lead and zinc). This strain utilized salicylate, naphthalene or xylene as a sole carbon source. The rate of cadmium accumulation in P. putida cell was enhanced at low concentration of Cd in the growth media. The maximum cadmium absorption by this strain grown in 1 and l0ppm of Cd was respectively 78% and 60% 24 hrs after culture, but in 100 ppm Cd, 40% 48 hrs after culture. Addition of a non-ionic surfactant Triton X-100(0.1%) to the medium enhanced the accumulation of cadmium in the P. putida up to approximately 37%.

Effects of Sewage Sludge Composts on the Chemical Properties of Soil and Growth of Paddy Rice (하수슬러지 퇴비 시용이 토양의 화학성 및 벼 생육에 미치는 영향)

  • To investigate the effects of the composts made of sewage sludges on the physico-chemical properties of soil and growth of paddy rice, the variations of chemical properties in soil, the contents of inorganic matters and heavy metals in plant tissues, the contents of heavy metals in brown rice, the growth response of paddy rice and grain yield were investigated. The results were summarized as follows : The contents of organic matters, N, available $P_2O_5$ and exchangeable K in soil were increased by application of the composts. The contents of N and available $P_2O_5$ in soil were remarkably decreased at the harvesting time in comparing with the 20th day after application of the composts. The contents of heavy metals in soil were increased very slightly by application of the com-post. At the harvesting time, the contents of N and K were not varied in all plant tissues at the plot of the compost in comparing with the plot of the NPK. The content of $P_2O_5$ in plant was increased by application of the composts. The contents of heavy metals in plant tissues and brown rice were not changed among the treatments. When the composts were applied to the pot at the rates of 1,000, 2,000, 4,000 and 6,000kg/10a with NPK, the numbers of grain yield were increased with 3, 18, 44 and 54% in comparing with NPK plot, respectively. Dry weight, the numbers of tillers and spikelet of the rice plant were increased by application of the composts.
