The mixture of 5-chloro-2-methylisothiazol-3(2H)-one (CMIT) and 2-methylisothiazol-3(2H)-one (MIT), CMIT/MIT, is a preservative in cosmetics. CMIT/MIT is a highly effective preservative; however, it is also a commonly known skin sensitizer. Therefore, in the present study, a risk assessment for safety management of CMIT/MIT was conducted on products containing 0.0015% of CMIT/MIT, which is the maximum MIT level allowed in current products. The no observed adverse effect level (NOAEL) for CMIT/MIT was 2.8 mg/kg bw/day obtained from a two-generation reproductive toxicity test, and the skin sensitization toxicity standard value for CMIT/MIT, or the no expected sensitization induction level (NESIL), was $1.25{\mu}g/cm^2/day$ in humans. According to a calculation of body exposure to cosmetics use, the systemic exposure dosage (SED) was calculated as 0.00423 mg/kg bw/day when leave-on and rinse-off products were considered. Additionally, the consumer exposure level (CEL) amounted to $0.77512{\mu}g/cm^2/day$ for all representative cosmetics and $0.00584{\mu}g/cm^2/day$ for rinse-off products only. As a result, the non-cancer margin of safety (MOS) was calculated as 633, and CMIT/MIT was determined to be safe when all representative cosmetics were evaluated. In addition, the skin sensitization acceptable exposure level (AEL)/CEL was calculated as 0.00538 for all representative cosmetics and 2.14225 for rinse-off products; thus, CMIT/MIT was considered a skin sensitizer when all representative cosmetics were evaluated. Current regulations indicate that CMIT/MIT can only be used at concentrations 0.0015% or less and is prohibited from use in other cosmetics products. According to the results of this risk assessment, the CMIT/MIT regulatory values currently used in cosmetics are evaluated as appropriate.
Ninety-six specimens of nine puffer fishes landed at a fish market of Pusan, Korea were assayed for anatomical distribution of toxicity. The puffers, Lagocephalus gloveri ('geommilbog'), Fugu rubripes rubripes ('jajubog'), Fugu rubripes chinesis ('geomjajubog'), Lagocephalus wheeleri ('hinmilbog') and Arothron firmamentum ('byeolbog'), were shown to be nontoxic in muscle and skin. However, toxicities in skins of four specimens of Lagocephalus gloveri and a specimen of Arothron firmamentum were more than 10MU/g In the puffer, Fugu xanthopterus ('ggachibog'), the skin showed to be weakly toxic as far as nine specimens out of 27 ones are concerned, and the muscle was nontoxic in all specimens. Meanwhile, it was noted that the puffer, Fugu vermicularis radiatus was found to be a toxic species, differing from the toxicity known so far. Their highest toxicities were 3,880, 1,191, 1,115, 219, 289, 5,620 and 753MU/g for liver, intestine, skin, muscle, testis, ovary and bile, respectively.

Skin disease is one of the most common diseases and its incidence is increasing dramatically in modern society. Specially, many attempts have been made to treat chronic skin inflammation diseases, such as psoriasis and atopic dermatitis, but effective therapies for the immune cell-mediated skin diseases, including psoriasis and atopic dermatitis have not been developed. Until recently, several drug candidates which were claimed to be effective for skin diseases have been reported, but most of them are not used to treat chronic skin disease. Especially, Psoriasis is characterized by excessive growth and aberrant differentiation of keratinocytes, but is fully reversible with appropriate therapy. The trigger of the keratinocyte response is thought to be activation of the cellular immune system, with T cells and various immune-related cytokines. Formation of new blood vessels starts with early psoriatic changes and disappears with disease clearance. Several angiogenic mediators are up-regulated in psoriasis development. Contact- and mediator-dependent factors derived from keratinocytes, mast cells and immune cells may contribute to the strong blood vessel formation of psoriasis. New technologies and experimental models provide new insights into the role of angiogenesis in psoriasis pathogenesis. TMP and its derivatives themselves effectively inhibited in vitro cell migration, tube formation, and the expression of angiogenic factors. However, TMP and its derivatives induced side effects including hemolysis and local side effects. Therefore, in an attempt to reduce the toxicity and the undesirable side effects of TMP and derivatives, a liposomal formulation was prepared and tested for its effectiveness. TMP and derivatives liposomes retained the effectiveness of TMP in vitro while side effects were reduced. These results support the conclusion that TMP effectively inhibits in vitro angiogenesis, with the possibility that use as a psoriasis relief agent.

We tried to check the antioxidant activity and toxicity of trichosanthis cucumeroidis radix extracts in skin cells, and check the possibility of their use as a functional material that can be effectively used on the skin. Total polyphenol and total flavonoid content, which are indicators of antioxidant activity of trichosanthis cucumeroidis radix extracts, were confirmed, and cytotoxicity was confirmed using Neutral red assay in the skin. As a result of the study, the content of total polyphenols and total flavonoids increased concentration-dependent. High survival rates in fibroblast HDF cells were identified, and cell survival rates were significantly lowered from 5 ㎍/mL in melanocytes B16F10 melanoma cells and inflammation-related macrophages RAW 264.7 cells. He results of this study are believed to be available as basic data for antioxidant activity of trichosanthis cucumeroidis radix extracts and skin cells.

Arsenic is one of the most toxic environmental toxicants. More than 150 million people worldwide are exposed to arsenic through ground water contamination. It is an exclusive human carcinogen. Although the hallmarks of arsenic toxicity are skin lesions and skin cancers, arsenic can also induce cancers in the lung, liver, kidney, urinary bladder, and other internal organs. Arsenic is a non-mutagenic compound but can induce significant cytogenetic damage as measured by chromosomal aberrations, sister chromatid exchanges, and micronuclei formation in human systems. These genotoxic end points are extensively used to predict genotoxic potentials of different environmental chemicals, drugs, pesticides, and insecticides. These cytogenetic end points are also used for evaluating cancer risk. Here, by critically reviewing and analyzing the existing literature, we conclude that inorganic arsenic is a genotoxic carcinogen.
The purpose of this study is to evaluate the effectiveness of Scirpi rhizoma ethanol extract (SREE) on skin whitening using in vitro test. In the antioxidative activities, it was found that SREE contains 38.9 mg/g of polyphenol and 74.5 mg/g of flavonoid in total. In the electron donating ability, SREE showed a dose-dependent response, showing a high antioxidative capacity of 86.1% at 1000 ppm. It was found that the maximum permissible level of SREE to Melan-a cells was over 200 ppm, showing a quite low toxicity of SREE against Melan-a cells. Both in the inhibitory measurement for tyrosinase activity and melanogenesis using Melan-a cells, SREE presented a dose-dependent response with excellent efficacy.

Lophatheri Herba, Patriniae Radix, Alli tuberosi Herba, and Menthae Herba have long been used externally for treatment of injuries or inflammations in Korea. This study was carried out to examine the anti-dandruff effect of the mixture made with equal parts of 70%-ethanol extracts of Lophatheri Herba, Patriniae Radix, Allii tuberosi Herba, and Menthae Herba (LPAM) and of each one above, compared with 1%-zinc pyrithione (Standard) used commercially as anti-dandruff. The results showed the mixture was more effective than each crude drug. The 10%-LPAM produced no toxicity in skin reaction test and eye irritation test, and showed significant antimicrobial activity against Pityrosporum ovale, the major cause of dandruff, and also showed significant anti-dandruff and itch-improvement effects on head skin.

Objectives: Hyaluronic acid, high molecular glycosaminoglycan, exists in extracellular matrix of tissue, especially, in skin and has been known to be deeply involved in skin hydration. In this study, we investigated the effect of methanol extract of Hwang-gi, Astragalus membranaceus root, on hyaluronic acid production in human keratinocyte HaCaT cells. Methods: We determined hyaluronic acid synthase 2 gene expression and hyaluronic acid production in HaCaT cells by using RT-PCR and ELISA, respectively. Results: Hwang-gi extract didn't show the toxicity to HaCaT cells within the treated concentration and increased the hyaluronic acid synthase 2 gene expression and hyaluronic acid production. Conclusions: Hyaluronic acid production increased by Hwang-gi could be, partially, contribute to the moisturing effect in skin by it.
In the present study, the therapeutic potential of purified and well-characterized bacteriophages was evaluated in thermally injured mice infected with Klebsiella pneumoniae B5055. The efficacy of five Klebsiella phages (Kpn5, Kpn12, Kpn13, Kpn17, and Kpn22) was evaluated on the basis of survival rate, decrease in bacterial counts in different organs of phage-treated animals, and regeneration of skin cells as observed by histopathological examination of phage-treated skin. Toxicity studies performed with all the phages showed them to be non-toxic, as no signs of morbidity and mortality were observed in phage-treated mice. The results of the study indicate that a single dose of phages, intraperitoneally (i.p.) at an MOI of 1.0, resulted in significant decrease in mortality, and this dose was found to be sufficient to completely cure K. pneumoniae infection in the burn wound model. Maximum decrease in bacterial counts in different organs was observed at 72 h post infection. Histopathological examination of skin of phage-treated mice showed complete recovery of burn infection. Kpn5 phage was found to be highly effective among all the phages and equally effective when compared with a cocktail of all the phages. From these results, it can be concluded that phage therapy may have the potential to be used as stand-alone therapy for K. pneumoniae induced burn wound infection, especially in situations where multiple antibiotic-resistant organisms are encountered.
Reactive oxygen species (ROS) disrupt the cellular redox balance, exert cytotoxic effects, and consequently promote the development of various diseases in humans. Previous studies have reported that antioxidants counteract the adverse effects of ROS. Several studies examine the whitening effects of various agents based on their ability to inhibit tyrosinase activity. Tyrosinase is a critical enzyme involved in the synthesis of melanin, which protects the skin against radiation. Various agents exhibiting antioxidant and tyrosinase inhibitory activities have been synthesized. However, these synthetic drugs are associated with toxicity, decreased safety, and poor skin penetration in vivo, which has limited the clinical application of synthetic drugs. This study examined the antioxidant and tyrosinase inhibitory activities of some microalgae. The methanol, dichloromethane, and ethyl acetate extracts of four microalgal species (Tetraselmis tetrathele, Dunaliella tertiolecta, Platymonas sp., and Chaetoceros simplex) were prepared. The physiological and whitening effects of microalgal extracts were investigated by measuring the antioxidant and tyrosinase inhibitory activities. The ethyl acetate extract of D. tertiolecta exhibited the highest antioxidant and tyrosinase inhibitory activities. Future studies must focus on examining the whitening effects of microalgae on cell lines to facilitate the development of microalga-based therapeutics for skin diseases, functional health foods, and whitening agents. Thus, microalgae have potential applications in the pharmaceutical, food, and cosmetic industries.
