Many seismically vulnerable regions in India and worldwide are located on deep soil deposits which extend to several hundred meters of depth. It has been well recognized that the earthquake shaking is altered by geological conditions at the location of building. As seismic waves propagates through uppermost layers of soil and rock, these layers serve as filter and they can increase the duration and amplitude of earthquake motion within narrow frequency bands. The amplification of these waves is largely controlled by mechanical properties of these layers, which are function of their stiffness and damping. Stiffness and damping are further influenced by soil type and thickness. In the current study, an attempt has been made to study the seismic site response of deep soils. Three hypothetical homogeneous soil models (e.g., soft soil, medium soil and hard soil) lying on bedrock are considered. Depth of half space is varied from 30 m to 2,000 m in this study. Controlled synthetic motions are used as input base motion. One dimensional equivalent linear ground response analyses are carried out using a computer package DEEPSOIL. Conventional approach of analysing up to 30 m depth has been found to be inadequate for deep soil sites. PGA values are observed to be higher for deeper soil profiles as compared to shallow soil profiles indicating that deeper soil profiles are more prone to liquefaction and other related seismic hazards under earthquake ground shaking. The study recommends to deal the deeper soil sections more carefully for estimating the amplification factors for seismic hazard assessment at the surface.
Excision and amplification of pre-determined, large genomic segments (taken directly from the genome of a natural host, which provides an alternative to conventional cloning in foreign vectors and hosts) was explored in human cells. In this approach, we devised a procedure for excising a large segment of human genomic DNA, the iNOS gene, by using the Cre/loxP system of bacteriophage P1 and amplifying the excised circles with the EBNA-1/oriP system of the Epstein-Barr virus. Two loxP sequences, each of which serves as a recognition site for recombinase Cre, were integrated unidirectionally into the 5'-UTR and 3'-UTR regions of the iNOS gene, together with an oriP sequence for conditional replication. The traps-acting genes cre and EBNA-1, which were under the control of a tetracycline responsive $P_{hcmv^*-1}$ promoter, were also inserted into the 5'-UTR and 3'-UTR regions of the iNOS gene, respectively, by homologous recombination. The strain carrying the inserted elements was stably maintained until the excision and amplification functions were triggered by the induction of cre and EBNA-1. Upon induction by doxycycline, Cre excised the iNOS gene that was flanked by two ZoxP sites and circularized it. The circularized iNOS gene was then amplified by the EBNA-1/oriP-system. With this procedure, approximately a 45.8-kb iNOS genomic fragment of human chromosome 17 was excised and successfully amplified in human cells. Our procedure can be used effectively for the sequencing of unclonable genes, the functional analysis of unknown genes, and gene therapy.

The evaluation of the seismic hazard for a given site is to estimate the seismic ground motion at the surface. This is the result of the combination of the action of the seismic source, which generates seismic waves, the propagation of these waves between the source and the site, and site local conditions. The aim of this work is to evaluate the sensitivity of dynamic response of extended structures to spatial variable ground motions (SVGM). All factors of spatial variability of ground motion are considered, especially local site effect. In this paper, a method is presented to simulate spatially varying earthquake ground motions. The scheme for generating spatially varying ground motions is established for spatial locations on the ground surface with varying site conditions. In this proposed method, two steps are necessary. Firstly, the base rock motions are assumed to have the same intensity and are modelled with a filtered Tajimi-Kanai power spectral density function. An empirical coherency loss model is used to define spatial variable seismic ground motions at the base rock. In the second step, power spectral density function of ground motion on surface is derived by considering site amplification effect based on the one dimensional seismic wave propagation theory. Several dynamics analysis of a curved viaduct to various cases of spatially varying seismic ground motions are performed. For comparison, responses to uniform ground motion, to spatial ground motions without considering local site effect, to spatial ground motions with considering coherency loss, phase delay and local site effects are also calculated. The results showed that the generated seismic signals are strongly conditioned by the local site effect. In the same sense, the dynamic response of the viaduct is very sensitive of the variation of local geological conditions of the site. The effect of neglecting local site effect in dynamic analysis gives rise to a significant underestimation of the seismic demand of the structure.

The seismic slope stability is most often evaluated by the pseudo-static limit analysis, in which the earthquake loading is simplified as static inertial loads acting in horizontal and/or vertical directions. The transient loading is represented by constant acceleration via the pseudostatic coefficients. The result of a pseudostatic analysis is governed by the selection of the value of the pseudostatic coefficient. However, selection of the value is very difficult and often done in an ad hoc manner without a sound physical reasoning. In addition, the maximum acceleration is commonly estimated from the design guideline, which cannot accurately estimate the dynamic response of a slope. There is a need to perform a 2D dynamic analysis to properly define the dynamic response characteristics. This paper develops the modified one-dimensional seismic site response analysis. The modified site response analysis adjusts the density of the layers to simulate the change in mass and weight of the layers of the slope with depth. Multiple analyses are performed at various locations within the slope to estimate the change in seismic response of the slope. The calculated peak acceleration profiles with depth from the developed procedure are compared to those by the two-dimensional analyses. Comparisons show that the two methods result in remarkable match.
An accurate estimation of water level variation when thewaves propagate to the coastal regionis very important for the port and harbor development plan. This study describes the application of a hybrid element model to harbor oscillation problem due to the construction of shore structure and implementation of shore boundary. The site selected is Pusan Harbor area with the third development and the Artificial Island plan. The observed water level changes at the site are compared with the result of the numerical experiment. The model gives a very important prediction of water level changes for navigation and harbor design.
The development of performance-based design methodologies requires a reasonable definition of a displacement-response spectrum. Although ground motions are known to be significantly affected by the resonant-like amplification behavior caused by multiple wave reflections within the surface soil, such a soil-resonance effect is seldom explicitly considered in current-displacement spectral models. In this study, an analytical approach is developed for the construction of displacement-response spectra by considering the soil-resonance effect. For this purpose, a simple and rational equation is proposed for the response spectral ratio at the site fundamental period (SRTg) to represent the soil-resonance effect based on wave multiple reflection theory. In addition, a bilinear model is adopted to construct the soil displacement-response spectra. The proposed model is verified by comparing its results with those obtained from actual observations and SHAKE analyses. The results show that the proposed model can lead to very good estimations of SRTg for harmonic incident seismic waves and lead to reasonable estimations of SRTg and soil displacement-response spectra for earthquakes with a relatively large magnitude, which are generally considered for seismic design, particularly in high-seismicity regions.

The purpose of this study is to estimate seismic damage for harbor site considering dynamic amplification characteristics. First of all, a series of ground response analysis is performed and then correlation equations between rock outcrop accelerations and peak ground accelerations (PGAs) are determined. These equations are saved into DB and when an earthquake occurs, PGAs are determined by them as soon as possible. For earthquake events, seismic damage grades of harbor structures are determined by using the correlated PGAs and fragility curves of harbor structures in real time. In this study, seismic damage was estimated and classified into several grades by applying two hypothetical earthquakes.

In this study, a seismic behavior of electrical cabinet system in Nuclear Power Plants(NPPs) was evaluated by the shaking table test. A 480V Motor Control Centers(MCCs) was selected for the shaking table test, and a real MCC cabinet for the Korea Nuclear Power Plant site was rented by manufactured company. For the shaking table tests, three kinds of seismic input motions were used, which were a US NRC Reg. guide 1.60 design spectrum, a UHS spectrum and PAB 165' floor response spectrum(FRS). Especially, the UHS input motion was selected for an evaluation of structural seismic amplification effects, three directional accelerations were measured at three points outside on the cabinet system and also that of the incabinet response amplification, accelerations were measure at two points which were mounted in electrical equipment such as relay. Seismic amplification effect is determined at the outside and inside of a cabinet as input seismic motion, and compared to the results which are calculated by analytical method based on NUREG/CR-5203.

The thickness of permafrost in Eastern Siberia is from 200 to 500 meters. The seasonally frozen layer can vary from 0 to 4m depending on ground temperature and its location. The shear wave velocity varies from 80m/s in summer to 1500m/s in winter depending on soil type. When melted, large impedence will occur due to the difference between the shear wave velocity of seasonally frozen soil and that of permafrost layer. Large displacement may occur at the boundary of the melted and the frozen layer, and this phenomenon should be considered in a seismic design. In this research, one-dimensional equivalent linear analyses were performed to investigate the effects of the seasonally frozen layer on ground amplification characteristics. Soil profiles of Yakutsk and Chara in Eastern Siberia were selected from geotechnical reports. 20 recorded ground motions were used to evaluate the effect of input motions. As the thickness of seasonally frozen layer and the difference in the shear wave velocity increases, the amplification is shown to increase. Peat, very soft organic soil widely distributed throughout Eastern Siberia, is shown to cause significant ground motion amplification. It is therefore recommended to account for its influence on propagated motion.

Rice bacterial leaf blight (BLB) by Xanthomonas oryzae pv. oryzae (Xoo) is considered to be one of the major rice diseases steadily occurring around the rice-producing countries. In this study, we developed a recombinase polymerase amplification (RPA) assay for the rapid, convenient and specific diagnosis of Xoo by targeting Xoo-specific transposase A gene. As the target gene can be amplified in 10 min without DNA extraction process and special equipment for temperature control, RPA for BLB can be useful and practical component for on-site diagnosis.
