The objective of this study was to establish conditions for transfection of a foreign gene into somatic cells using cationic lipid reagents and to evaluate the effects of transfection on in vitro development of somatic cell nuclear transfer (SCNT) embryos. Green fluorescent protein (GFP) gene was used as a foreign gene and a non-transfected somatic cell was utilized as a control karyoplast. Monolayers of porcine cells were established and subsequently transfected with a GFP-expressing gene (pEGFP-N1) using three types of transfection reagents (LipofectAMINE PLUS, FuGENE 6 or ExGen500). Donor cells used for SCNT included transfected fetal or adult fibroblasts and oviduct epithelial cells, either serum-fed or serum-starved. Oocytes matured in vitro for 42 h were reconstructed with either transfected or non-transfected porcine somatic cells by electric fusion and activation using a single DC pulse of 1.8 kV/cm for $30{\mu}s$ in $Ca^{2+}$ and $Mg^{2+}-containing$ 0.26 M mannitol solution. Reconstructed oocytes were subsequently cultured in NCSU-23 medium for 168 h and the developmental competence and cell number in blastocyst were compared. There were no significant differences (P>0.05) in fusion, cleavage rates or development to the blastocyst stage between non-transfected, transfected, serum-fed and serum-starved cells. However, the rates of GFP-expressing blastocysts were higher in the FuGENE 6 group (71.4%) among transfection reagents and in the fetal fibroblasts group (70.4%) for donor cells. These results indicate that fetal fibroblasts transfected with FuGENE 6 can be used as donor cells for porcine SCNT and that GFP gene can be safely used as a marker of foreign genes in porcine transgenesis.

Microbial exopolysaccharide (EPS) is a biothickener that can be added to a wide variety of food products, where it serves as a viscosifying, stabilizing, emulsifying, and gelling agent. The objective of this study was to investigate the optimum conditions of pH, incubation temperature, and whey protein concentration (WPC) for EPS production by Lactobacillus rhamnosus ATCC 9595. We found that maximal EPS production was achieved at a pH of 5.5 and temperature of $37^{\circ}C$. At the same fermentation conditions, EPS production was affected by the addition of L. rhamnosus GG (a weak-EPS producer). After growth for 24 hr, total EPS production was $583{\pm}15.4mg/L$ in the single culture system, and $865{\pm}22.6\;mg/L$ in the co-culture system with L. rhamnosus GG. Based on the presence of WPC, EPS production dramatically increased from $583{\pm}15.4$ (under no WPC supplementation) to $1,011{\pm}14.7\;mg/L$ (under supplementation with 1.0% WPC). These results suggest that WPC supplementation and the co-culture systems coupled with small portions of weak-EPS producing strain can play an important role in the enhancement of EPS production.
This paper presents a mathematical model for a double-fleet operation in Korean high speed rail (HSR). KORAIL has a plan to launch new HSR units in 2010, which are composed of 10 railcars. The double-fleet operation assigns a single-unit or two-unit fleet to a segment, accommodating demand fluctuation. The proposed model assumes stochastic demand and uses chance-constrained constraints to assure a preset service level. It can be used in the tactical planning stage of the rail management as it includes several real-world conditions, such as the capacities of the infra-structures and operational procedures. In the solution approach, the expected revenue in the objective function is linearized by using expected marginal revenue, and the chance-constrained constraints are linearized by assuming that demands are normally distributed. Subsequently, the model can be solved by a mixed-integer linear programming solver fur small size problems. The test results of the model applied to Friday morning train schedules for one month sample data from KTX operation in 2004 shows that the proposed model could be utilized to determine the effectiveness of double-fleet operation, which could significantly increase the expected profit and seat utilization rates when properly maneuvered.

Single shortest path routing is known to perform poorly for Internet traffic engineering (TE) where the typical optimization objective is to minimize the maximum link load. Splitting traffic uniformly over equal cost multiple shortest paths in OSPF and IS-IS does not always minimize the maximum link load when multiple paths are not carefully selected for the global traffic demand matrix. However, among all the equal cost multiple shortest paths in the network, a set of TE-aware shortest paths, which reduces the maximum link load significantly, can be found and used by IP routers without any change of existing routing protocols and serious configuration overhead. While calculating TE-aware shortest paths. the destination-based forwarding constraint at a node should be satisfied, because an IP router will forward a packet to the next-hop toward the destination by looking up the destination prefix. In this paper, we present a problem formulation of finding a set of TE-aware shortest paths in ILP, and propose a simple heuristic for the problem. From the simulation results, it is shown that TE-aware shortest path routing performs better than default shortest path routing and ECMP in terms of the maximum link load with the marginal configuration overhead of changing the next-hops.

In this study, thermal performance of recuperators with plain and offset strip fins is investigated to enhance the thermal efficiency of a micro gas turbine. Thermal cycle analysis is conducted to determine major design parameters of a single-pass counterflow recuperator. In order to evaluate the performance of the recuperator, the effectiveness and the pressure drop in the recuperators are chosen as the objective function and the design constraint, respectively. The optimized geometries for internal structure of the recuperators with plain and offset strip fins are obtained with varying the fin spacing and height. From the result, the recuperator with offset strip fins has better thermal performance when the fin spacing, s, is smaller than 1.45mm and the thermal performance of the recuperator with plain rectangular fins is higher than that with offset strip fins in the region of $s{\geq}1.45mm$. In addition, it is found that the entrance region effect and the longitudinal wall heat conduction effect should be taken into account for accurately predicting the thermal performance of the recuperators with both plain and offset strip fins.

The evolving internet-based services demand high-speed data transmission in conjunction with scalability. The next generation optical network has to exploit artificial intelligence and cognitive techniques to cope with the emerging requirements. This work proposes a novel way to solve the dynamic provisioning problem in optical network. The provisioning in optical network involves the computation of routes and the reservation of wavelenghs (Routing and Wavelength assignment-RWA). This is an extensively studied multi-objective optimization problem and its complexity is known to be NP-Complete. As the exact algorithms incurs more running time, the heuristic based approaches have been widely preferred to solve this problem. Recently the software-defined networking has impacted the way the optical pipes are configured and monitored. This work proposes the dynamic selection of path computation algorithms in response to the changing service requirements and network scenarios. A software-defined controller mechanism with a novel packet matching feature was proposed to dynamically match the traffic demands with the appropriate algorithm. A software-defined controller with Path Computation Element-PCE was created in the ONOS tool. A simulation study was performed with the case study of dynamic path establishment in ONOS-Open Network Operating System based software defined controller environment. A java based NOX controller was configured with a parent path computation element. The child path computation elements were configured with different path computation algorithms under the control of the parent path computation element. The use case of dynamic bulk path creation was considered. The algorithm selection method is compared with the existing single algorithm based method and the results are analyzed.

Due to increasing awareness on the treatment of end-of-use/life products, disassembly has been a fast-growing research area of interest for many researchers over recent decades. This paper introduces a novel lot-sizing problem that has not been studied in the literature, which is the service-parts lot-sizing with disassembly option. The disassembly option implies that the demands of service parts can be fulfilled by newly manufactured parts, but also by disassembled parts. The disassembled parts are the ones recovered after the disassembly of end-of-use/life products. The objective of the considered problem is to maximize the total profit, i.e., the revenue of selling the service parts minus the total cost of the fixed setup, production, disassembly, inventory holding, and disposal over a planning horizon. This paper proves that the single-period version of the considered problem is NP-hard and suggests a heuristic by combining a simulated annealing algorithm and a linear-programming relaxation. Computational experiment results show that the heuristic generates near-optimal solutions within reasonable computation time, which implies that the heuristic is a viable optimization tool for the service parts inventory management. In addition, sensitivity analyses indicate that deciding an appropriate price of disassembled parts and an appropriate collection amount of EOLs are very important for sustainable service parts systems.

Purpose: The objective of this study was to propose a method for developing a clinical phantom to reproduce various diseases that are clinically prevalent in the field of dentistry. This could facilitate diverse clinical research without unnecessarily exposing patients to radiation. Materials and Methods: This study utilized a single dry skull, which was visually and radiographically examined to evaluate its condition. Existing lesions on the dry skull were preserved, and other relevant lesions were artificially created as necessary. These lesions were then documented using intraoral radiography and cone-beam computed tomography. Once all pre-existing and reproduced lesions were confirmed by the consensus of 2 oral and maxillofacial radiologists, the skull was embedded in a soft tissue substitute. To validate the process, cone-beam computed tomography scans and panoramic radiographs were obtained of the fabricated phantom. All acquired images were subsequently evaluated. Results: Most lesions could be identified on panoramic radiographs, although some sialoliths and cracked teeth were confirmed only through cone-beam computed tomographic images. A small gap was observed between the epoxy resin and the bone structures. However, 2 oral and maxillofacial radiologists agreed that this space did not meaningfully impact the interpretation process. Conclusion: The newly developed phantom has potential for use as a standardized phantom within the dental field. It may be utilized for a variety of imaging studies, not only for optimization purposes, but also for addressing other experimental issues related to both 2- and 3-dimensional diagnostic radiography.

This study developed a model to evaluate the co-benefits and trade-off effects between biodiversity and carbon storage capacity based on the implementation locations of nature-based solutions. The model aims to propose optimal implementation locations by using the conceptual idea of edge effects for carbon storage and connectivity for biodiversity. The co-benefits were considered by simultaneously taking into account two effects rather than a single effect. Trade-off effects were observed among optimal plans through a comparison of benefits. The NSGA-II multi-objective optimization algorithm was utilized, confirming the identification of Pareto-optimal solutions. The implementation patterns of Pareto-optimal solutions for green areas were examined. This study holds significance in proposing optimal locations by evaluating various co-benefits and trade-off effects of nature-based solutions. By advancing models based on this evaluation framework, it is anticipated that the assessment of co-benefits and trade-off effects among various benefits of nature-based solutions, such as climate change mitigation, enhancement of biodiversity, and provision of ecosystem services, can be accomplished.
Objective: This study was conducted to generate single stranded DNA oligonucleotides with selective affinity to bovine spermatozoa, assess its binding potential and explore its potential utility in trapping spermatozoa from suspensions. Methods: A combinatorial library of 94 mer long oligonucleotide was used for systematic evolution of ligands by exponential enrichment (SELEX) with bovine spermatozoa. The amplicons from sixth and seventh rounds of SELEX were sequenced, and the reads were clustered employing cluster database at high identity with tolerance (CD-HIT) and FASTAptamer. The enriched nucleotides were predicted for secondary structures by Mfold, motifs by Multiple Em for Motif Elicitation and 5' labelled with biotin/6-FAM to determine the binding potential and binding pattern. Results: We generated 14.1 and 17.7 million reads from sixth and seventh rounds of SELEX respectively to bovine spermatozoa. The CD-HIT clustered 78,098 and 21,196 reads in the top ten clusters and FASTAptamer identified 2,195 and 4,405 unique sequences in the top three clusters from the sixth and seventh rounds, respectively. The identified oligonucleotides formed secondary structures with delta G values between -1.17 to -26.18 kcal/mol indicating varied stability. Confocal imaging with the oligonucleotides from the seventh round revealed different patterns of binding to bovine spermatozoa (fluorescence of the whole head, spot of fluorescence in head and mid- piece and tail). Use of a 5'-biotin tagged oligonucleotide from the sixth round at 100 pmol with 4×106 spermatozoa could trap almost 80% from the suspension. Conclusion: The binding patterns and ability of the identified oligonucleotides confirms successful optimization of the SELEX process and generation of aptamers to bovine spermatozoa. These oligonucleotides provide a quick approach for selective capture of spermatozoa from complex samples. Future SELEX rounds with X- or Y- enriched sperm suspension will be used to generate oligonucleotides that bind to spermatozoa of a specific sex type.
