Bioconversion of Citron oil by Co-Culture of E. coli EC3, EC4, and EC6 (E. coli 형질전환주의 공동배양에 의한 유자정유의 생전환)

  • E. coli transformants EC3, EC4. and EC6. harboring citron oil degrading pathway genes, were co-cultured in M9 media with citron oil as a sole carbon source at 28$^{\circ}C$. Each co-culture(EC3+EC4, EC3+EC6, EC4+EC6 and EC3+EC4+EC6) showed three to four times higher cell growth than each transformant single culture. Microbial conversion products from the co-cultures were determined by GC-MS. Linalool. 4-terpineol and ${\alpha}$-terpineol were the major common products from co-cultures. Various minor products also were detected and important in flavor characteristics of cultures.

A Personal Reformer(PR) for your Fuel cell system (연료전지를 위한 개인용 개질기)

  • The present paper relates to an apparatus in which all carbonaceous material such as coal, oil, plastics and any substance having carbon atoms as part of its constituents are reformed(gasified) into syngas at temperature above $1,200^{\circ}C$(KR patent No.0391121, and PCT/KR2001/01717 and PCT/KR2004/001020). It comprises a single-stage reforming reactor without catalyst and a syngas burner as shown in Fig.2. syngas is combusted with $O_2$ gas in the syngas bunter to produce $M_2O$ and $CO_2$ gas with exothermic heat. Reaction products are introduced into the reforming reactor, reaction heat from syngas burner elevate the temperature of reactor above $1,200^{\circ}C$, and reaction products reduce carbonaceous material down to CO and $H_2$ gases. Reactants and heat necessary for the reaction are provided through the syngas burner only, Neither $O_2$ gas nor steam are injected into the reforming reactor. Reformer is made of ceramic inner lining and sst outer casing. Multiple syngas burners may be connected to the reforming reactor in order to increase the syngas output, and a portion of the product syngas is recycled into syngas burner. The present reformer as shown in Fig.2 is suitable to gasify carbonaceous wastes without secondary pollutants formed from oxidation. Further, it can be miniaturized to accompany a fuel cell system as shown in Fig.3 The output syngas may be used to drive a fuel cell and a portion of electrical power generated in a fuel cell is used to heat a compact reformer up to $1,200^{\circ}C$ so that gas/liquid fossil fuel can efficiently reformed into syngas. The fuel cell serves as syngas burner in Fig.2. The reformation reaction is sustained through recycling a portion of product syngas into a fuel cell and using a portion of electric power generated to heat the reformer for continuous operation. Such reforming reactor may be miniaturized into a size of PC, then you have a Personal Reformer(PR).

  • Although there are no oil wells in Korea, yet more than 900 strains of petroleum hydrocarbon utilizing microorganisms have been isolated from 357 soil and sewage samples collected from oil depots and other sources there. From these samples 7 strains of yeast were selected on the basis of their superior cell yields. Five of them were identified as Candida tropicalis, the other being Candida lipolitica and Torulopis sp. Of the selected strains the mass doubling time is $2.9{\sim}4.5$ hrs., the yield is $6.5{\sim}16.3\;g/l$; the conversion rate of crude petroleum substrate into the microbial mass is $7.8{\sim}19.3%$; and protein content of dried cells is $48.8{\sim}59.8%$.

  • The growth characteristics of Candida tropicalis KIST 359, isolated from soil samples collected at an oil depot in Korea, have been studied by cultivating batches under varying conditions. The conclusions of the study were: 1. The yeast easily assimilates hydrocarbons in a range of $C_{14}-C_{17}$, and the optimum cultivation temperature and pH are $30^{\circ}C$ and 5.5, respectively. 2. Using this strain of micro-organism, gas oil gives a higher cell yield than kerosine and with gas oil except urea all other nitrogen sources $(NH_4NO_3,\;NH_4Cl,\;(NH_4)_2SO_4\;and\;(NH_4)_2HPO_4)$ similarly support a satisfactory growth of the yeast. 3. The highest yield is obtained with a gas oil level of 10%(v/v), and concentrations of nitrogen source and $MgSO_4{\cdot}7H_2O$ of 0.5 and 0.05%(w/v), respectively. 4. The protein content of dried yeast cells is 59.8%. Its amino acid composition can be compared well with that of FAO provisional patterns, but with a low methionine and a high lysine content.

Therapeutic effect of oregano essential oil on subclinical bovine mastitis caused by Staphylococcus aureus and Escherichia coli

  • Oregano (Origanum vulgare) is an aromatic herb belonging to the Lamiaceae family. This study evaluated the therapeutic effect of oregano essential oil (OEO) on clinical bovine mastitis caused by Staphylococcus (S.) aureus and/or Escherichia (E.) coli. Thirty-two quarters from 18 lactating cows with subclinical mastitis were selected from a dairy farm and were divided into 4 groups (8 quarters/group): negative control (CON), positive control (GEN) and two OEO-treated groups (OEO-1 and OEO-2). Each group was intramammarily treated with saline, 10 g/tube of gentamicin ointment, and single and double doses of 0.9 mL OEO ointment twice a day for three days, respectively. After the treatments, physical udder conditions were greatly improved in GEN and OEO groups. In CON group, somatic cell counts (SCCs), number of white blood cells (WBC) and bacteria were increased. However, in OEO groups, SCCs and number of WBC were significantly decreased and S. aureus and E. coli were not detected in milk as compared with those before the treatments as well as GEN group. These results suggest that OEO may be a useful alternative to antibiotics for the control of subclinical bovine mastitis caused by S. aureus and/or E. coli.

Inhibitory Effect of BCG Cell-Wall Skeletons (BCG-CWS) Emulsified in Squalane on Tumor Growth and Metastasis in Mice

  • The antimetastatic effect of BCG-CWS, which was emulsified in an oil-in-water form with either Drakeol 6VR mineral oil (BCG-CWS/DK) or squalane (BCG-CWS/SQA), on lung metastasis produced by highly metastatic murine tumor cells, Colon26-M3.1 carcinoma cells and B16-BL6 melanoma cells, was investigated in syngeneic mice. An intravenous (i.v.) administration of BCG-CWS (100 mg/mouse) 1 day after tumor inoculation significantly inhibited tumor metastasis of both Colon26-M3.1 carcinoma and B16-BL6 melanoma cells in experimental lung metastasis models. No differences in the antitumor activity of the two oil-based formulations (BCG-CWS/DK and BCG-CWS/SQA) were obverved. However, BCG-CWS/SQA administered through subcutaneous (s.c.) route was shown to be effective only when it was consecutively injected (3 times) after tumor inoculation. An in vivo analysis for tumor-induced angiogenesis shwed that a single i.v. administration of BCG-CWS/SQA inhibited the number of tumor-induced blood vessels and suppressed tumor growth. Furthermore, the multiple administration of BCG-CWS/SQA given at on week intervals led to a significant reduction in spontaneous lung metastasis of B16-BL6 melanoma cells in a spontaneous metastasis model. These results suggest that BCG-CWS emulsified with squalane is a potent inhibitory agent of lung metastasis, and that the anti metastatic effect of BCG-CWS is related to the suppression of tumor growth and the inhibition of tumor-induced angiogenesis.

Measurements and Predictions of Rotodynamic Performance of a Motor-Driven Small Turbocompressor Supported on Oil-Free Foil Bearings (무급유 포일 베어링으로 지지되는 소형 전동 압축기의 회전체동역학 성능 측정 및 예측)

  • This study presents experimental measurements of the rotordynamic performance of a motor-driven small turbocompressor supported by gas beam foil journal bearings (GBFJBs) and compares the test results with the predictions of a computational model. The experiments confirmed that the rotational synchronous frequency component dominates the behavior of the overall rotor vibrations, whereas the nonsynchronous components are insignificant, indicating the rotor-bearing system remains stable up to 100 krpm. The undamped natural frequency and imbalanced response of the rotor-bearing system are predicted when integrating the finite element model of the rotor-bearing system with the predictions of the bearing dynamic coefficients. The results are in good agreement with the experimental results. In addition, base excitation test results show that the small turbocompressor can endure large external forces and demonstrate limited rotor amplitudes. A simple single degreeof-freedom rotor model using the nonlinear stiffness of the GBFJBs can effectively predict the test results.

The phytohormone abscisic acid increases triacylglycerol content in the green microalga Chlorella saccharophila (Chlorophyta)

  • Microalgae are currently a very promising source of biomass and triacylglycerol (TAG) for biofuels. In a previous study, we identified Chlorella saccharophila as a suitable source of oil for biodiesel production because it showed high biomass and lipid content with an appropriate fatty acid methyl esters profile. To improve the TAG accumulation in C. saccharophila, in this study we evaluated the effect of abscisic acid (ABA) addition on cell concentration, lipid content and TAG production in this microalga. First, we evaluated the effects of four ABA concentrations (1, 4, 10, and 20 μM) added at the beginning of a single-stage cultivation strategy, and found that all concentrations tested significantly increased cell concentration and TAG content in C. saccharophila. We then evaluated the addition of 1 μM ABA during the second stage of a two-stage cultivation strategy and compared it with a nitrogen deficiency treatment (ND) and a combination of ND and ABA (ND + ABA). Although ABA alone significantly increased lipid and TAG contents compared with the control, ND showed significantly higher TAG content, and ND + ABA showed the highest TAG content. When comparing the results of both strategies, we found a superior response in terms of TAG accumulation with the addition of 1 μM ABA at the beginning of a single-stage cultivation system. This strategy is a simple and effective way to improve the TAG content in C. saccharophila and probably other microalgae as a feedstock for biodiesel production.

  • Methods of separating yeast cells from oil-water-cell emulsion and subsequent purification of the recovered yeast have been studied. In addition, the results of preliminary feeding experiments in which a yeast grown on gas oil was incorporated into chick rations are reported. According to the present study, it appears that the recovery of the yeasts would be easier at pH 9, since the emulsion is relatively more unstable. A class of surface active agent at a concentration of 0.3% was found to facilitate the separation of the yeast from the emulsion. The use of electrolytes such as NaCl and KCl were found to be most effective in breaking the emulsion. Solvent treatment using iso-propyl alcohol and its azeotropic mixture with hexane at $58^{\circ}C$ are particularly suitable for purification of the yeast. In the feeding experiment it was found that 5 percent of the fishmeal in the control ration could be replaced by the yeast with no adverse effect on performance. However, when 8 percent of the fish meal in the control ration was replaced by the yeast, some effect on live-weight gain of the chicks was observed.

  • The present study was undertaken to evaluate rosette formation and NK activity of splenic lymphocytes in 3-methylcholanthrene (MCA) treated mice. Mice were sensitized iv with 0.1ml of 1% sheep red blood cell (SRBC) suspension were treated with a single ip injection of olive oil alone or with different doses of MCA in oil at various time before or after sensitization, and were challenged at 4 days after SRBC. Rosette formation and NK activity of splenic lymphocytes were measured at 24 hours after challenge. Erythrocyte(E) rosette formation of splenic lymphocytes was significantly depressed in mouse treated with large dose of MCA (5~50mg) regardless of injecting time. But, there was no difference in the response between the treated with small dose of MCA (0.5mg). Whereas erythrocyte-antibody(EA) rosette or erythrocyte-antibody-complement(EAC) rosette forming cells were significantly depressed by MCA. Under small dose of MCA (0.5mg), any difference of NK activity was not observed in all course of injecting time. But, under large dose of MCA, the activity was markedly inhibited to about half the values seen in control and this suppression was transient, resulting that the normal level was reached again 19 days after MCA. These results, which conform with the predictions of immunosuppression hypothesis, suggest that MCA inhibits immunological responses including NK activity and thereby allows the outgrowth of antigenic neoplastic cells.
