Disease suppression by ectomycorrhizal(ECM) fungi has been demonstrated on red pine seedlings. Culturing of pathogenic fungi on petri plates containing culture filtrates of ECM fungi showed that culture filtrates of the ECM fungus Hebeloma cylindrosporum may inhibit the mycelial growth of all tested soil-borne plant pathogenic(SBPP) fungi upto 60%, In order to examine the effects of ECM fungi on SBPP fungi and on red pine seedlings, both symbiotic and pathogenic fungi were inoculated into the soil with red pine seedlings by three inoculation methods; pre-inoculation of SBPP fungi 10 days before inoculation of ECM fungi, simultaneous inoculation of both fungi, post-inoculation of SBPP fungi 60 days after inoculation of ECM fungi. Seedling mortality, seedling growth, and ectomycorrhizal formation by the combined treatments were examined and compared. Pine seedlings were dead by the pre-inoculation of pathogenic fungi, except Rhizina undulate which required 9-12 days, within 6 days after inoculation. Among pathogenic fungi tested, Fusarium oxysporum was the most pathogenic with the mortality of 44%. However, no dead seedlings were shown by simultaneous inoculation of both fungi or pre-inoculation of ECM fungi. In addition, pine seedlings treated by simultaneous or post-inoculation of SBPP fungi were relatively higher than those treated by pre-inoculation in diameter at root crown and the number of ectomycorrhizal roots. There were no significant differences among inoculation methods in root length and dry weight of treated seedlings. It means that ECM fungi somehow play a role in protecting primary roots of red pine seedlings against invasion by the SBPP fungi.
The effects of three minor elements, S, Ce and Bi, on the inoculation ability and fading behavior of 1.0 wt%Ba-Fe-Si were investigated through thermal analysis. The performance of 1.0 wt%Ba-Fe-Si inoculant was better and more consistant at the high temperature range of $1,450{\sim}1,500^{\circ}C$ than that of low one of $1,300{\sim}1,400^{\circ}C$. That was improved with individual addition of three minor elements. The optimum amount of addition was 1.0% of the weight of inoculant added, respectively. That was improved also by the simultaneous addition of two or three kinds of minor elements. Even though worse with the addition of two kinds of them simultaneously than with individual addition, that was improved with the simultaneous addition of all three kinds over that with the individual one.
Four nonpathogenic isolates of Fusarium oxysporum isolated from spinach showed suppressive effect on the occurrence of the Fusarium wilt of spinach caused by F. oxysporum f. sp. sprinaciae, among which NF01 controlled the disease most effectively. And NF01 was not pathogenic to tomato, cucumber, radish and spinach. This isolate was further tested for the biological control of the disease. The isolate was not inhibitory to the growth of the pathogen on potato sucrose agar medium, however the Fusarium wilt disease occurred less by drenching spore suspension of the nonpathogenic isolate. The control effect of the isolate was higher at lower inoculum level of the pathogen than at the higher inoculum level, and in the pretreatments than the simultaneous treatment of the isolate with the pathogen inoculation. The nit mutants of the isolate were easily formed on chlorate containing media, and was reisolated selectively as nit mutant from infected soil and plants. The reisolation rate of the isolate as opposed to pathogen was high at preinoculated soil and plants relative to the simultaneous inoculation of the isolate with the pathogen.
We have previously identified genes for four different protease inhibitors (PIs) that were induced upon rice blast infection in a rice blast resistant mutant SHM-11. Our expression analysis of the PIs indicated that induction of the PIs was the highest 24 hr after rice blast inoculation in the rice mutant SHM-11. Three PIs in the group of serine PIs were highly expressed while a cystein PI was weakly expressed upon rice blast inoculation. Four PIs were weakly induced 48 hr after pathogen inoculation in rice blast susceptible wild type rice plant. The simultaneous expression of three serine PIs was apparent from SHM-11 and two of them were induced in rice blast resistant Taebaegbyeo. One of them was induced in rice blast resistant Hwayeongbyeo while none of them were expressed in rice blast susceptible Nagdongbyeo and rice blast resistant Dongjinbyeo. Our results suggest that the expression of PI gene is rice cultivar specific and may be linked with the rice blast resistance in a specific rice mutant by the simultaneous expression of the PI genes.

Detached chili pepper fruits were inoculated with the conidial suspension of Colletotrichum acutatum JC-24 simultaneously (simultaneous inoculation, SI) and at delayed time (delayed inoculation, DI) after wounding with (delayed wound inoculation, DWI) or without additional wounding (delayed non-wound inoculation, DNI) at the inoculation time. Disease severity was significantly lowered by DNI, compared to SI. By DNI, the disease reduction rates were proportional with the length of delayed time, and greater at the high temperature range (18, 23 and $28^{\circ}$) than at the low temperature ($13^{\circ}$) tested. DWI was also effective in reducing the disease severity especially at 18oC; however, its effectiveness was lower than for DNI. In light microscopy, parenchyma cells at the wounding sites were modified structurally, initially forming new cell walls crossing cytoplasm, enlarged with multiple periclinal cell divisions, and finally layered like wound periderms. In DWI, the above structural modifications occurred, showing the restriction of the fungal invasion by the cell walls in enlarged modified cells, while no definite cellular modifications were found with proliferation of fungal hyphae in SI. Sclerenchyma-like cells with thickened cell walls were proliferated around the wounding sites, which were partially dissolved by DWI, probably leading to some disease development. All of these results suggest that the decline of the anthracnose disease in pepper fruit by the delayed inoculations may be derived from the structural modifications related to the healing processes of the previous wound inflicted on the tissues.
Biological control of blue stain fungi, such as Ophiostoma and Leptographium spp., that reduce the quality of logs and cause economic losses in wood product industry, was carried out in laboratory and field trials by a colorless strain of Ophiostoma quercus, BSFcs-1. Inoculation of pine wood chips with the colorless strain 1 wk before inoculating wild-type strain demonstrated that BSFcs-1 colonized wood chips and excluded blue stain fungi from being established. Efficacy of BSFcs-1 was compared with colorless strain of O. piliferum, which is commercially available under the trade name of Cartapip. Inoculation of pine wood logs with the colorless strain 1 wk before inoculating wild-type strain of blue stain in isolated wood chips, while O. quercus and O. floccosum colonized 0% and 17%, respectively. Simultaneous inoculation of logs with the colorless and wild-type strains resulted in decreased colonization (28%) by BSFcs-1, but increased colonization by O. quercus (185) and O. floccosum (29%). On the other hand, BSFcs-1 and wild-type strain alone colonized 75% and 71%, respectively. Treatment of the surface of log ends with mycelial suspension of BSFcs-1 after cutting also showed good control of blue stain fungi in a pine forest stands.
The development of rapid, infallible, and sensitive methods of detecting foodborne pathogens has received much impetus in recent years owing to an increased public awareness of the health hazards. For the rapid and simultaneous detection of these foodborne pathogens, a multiplex PCR method was developed. Yersinia enterocolitica, Staphylococcus aureus, and Shigella spp. are bacteria of concern because of their specific growing condition that enables them to live at low temperatures. In order to detect each pathogenic bacterium, specific primers from Y. enterocolitica, St. aureus, and Sh. flexneri were selected and validated successfully. To apply this method to food stored at low temperature, Y. enterocolitica, St. aureus, and Sh. flexneri were artificially inoculated in lettuce and incubated for enrichment. The multiplex PCR assays were able to simultaneously detect three pathogens, and the presence of three bands was observed at initial inoculation levels of approximately 1$\times$10$^1$ CFU/g in lettuce. Therefore, this method could be used for simultaneous detection of Y. enterocolitica, St. aureus, and Shigella spp. contaminated in lettuce during cultivation, transportation, preservation, and storage.
The goal of this study was to improve the quality of cheonggukjang by the optimization of the inoculation methods of the Bacillus subtilis (B. subtilis) and Lactobacillus plantarum (L. plantarum) strains. In order to optimize the mixed cultivation of B. subtilis and L. plantarum, the B. subtilis strain was inoculated into steamed soybeans after cultivation of L. plantarum. Inoculation size of B. subtilis was changed to the simultaneous inoculation method in order to stimulate the growth of the L. plantarum in cheonggukjang. The viable cell count of L. plantarum increased from $2{\times}10^7$ CFU/g to $2-6{\times}10^8$ CFU/g and B. subtilis grew to $9{\times}10^8$ CFU/g. These results showed that 2 strains were successfully able to grow in the steamed soybean for good quality of cheonggukjang by optimization of the inoculation methods. The sensory evaluation indicated that a favorable aroma and overall acceptance of cheonggukjang by the optimized mixed cultivation of B. subtilis and L. plantarum, which was relatively higher than those of cheonggukjang by single strain inoculation of B. subtilis.
Ethanol productions were performed by separate hydrolysis and fermentation (SHF) and simultaneous saccharification and fermentation (SSF) processes using seaweed, Enteromorpha intestinalis (sea lettuce). Pretreatment conditions were optimized by the performing thermal acid hydrolysis and enzymatic hydrolysis for the increase of ethanol yield. The pretreatment by thermal acid hydrolysis was carried out with different sulfuric acid concentrations in the range of 25 mM to 75 mM $H_2SO_4$, pretreatment time from 30 to 90 minutes and solid contents of seaweed powder in the range of 10~16% (w/v). Optimal pretreatment conditions were determined as 75 mM $H_2SO_4$ and 13% (w/v) slurry at $121^{\circ}C$ for 60 min. For the further saccharification, enzymatic hydrolysis was performed by the addition of commercial enzymes, Celluclast 1.5 L and Viscozyme L, after the neutralization. A maximum reducing sugar concentration of 40.4 g/L was obtained with 73% of theoretical yield from total carbohydrate. The ethanol concentration of 8.6 g/L of SHF process and 7.6 g/L of SSF process were obtained by the yeast, Saccharomyces cerevisiae KCTC 1126, with the inoculation cell density of 0.2 g dcw/L.
Reproduction of the soybean cyst nematode (SCN), Heterodera glycines Ichinohe on the susceptible soybean cultivar, Lee 74, was significantly reduced by pre-, post- and simultaneous treatments of acetylsalicylic acid (ASA, aspirin). The control efficiencies were 60%, 64% and 87% for pre-, post- and simultaneous treatments, respectively. ASA had no significant effect on the survival of 2nd stage juveniles and their penetration into the soybean root tissues, but significantly inhibited the early stage nematode growth in the roots. Syncytia were formed 2∼3 days after inoculation in the susceptible soybean without ASA treatment, characterized by dense cytoplasm and increased cellular organelles such as mitochondria and endoplasmic reticulum. The nematode stylet was penetrated into the syncytial cell, and feeding tube was formed at the nematode stylet was penetrated into the syncytial cell, and feeding tube was formed at the nematode stylet entry. However, in the ASA treatments, syncytium was not formed or degenerated, depending on the root tissues. In the pre-treatments of ASA, nematode stylets did not penetrate into cells, showing callose-like cell wall thickening formed at the nematode probing sites, or retracted from the infected cells. The stylet penetration sites of syncytial cells appeared to be sealed off with fibrillar materials. With post-treatment of ASA, syncytia formed by the nematode were degenerated, characterized by degradation of syncytial cytoplasm.
