A Customized Cleaning Agent for the Maintenance of Electric Fume Collector Used for the Purification of Effluent Gas from the Textile Industry (섬유산업 배기가스 정화용 Electric Fume Collector 설비의 유지보수를 위한 맞춤형 세정제)

  • A customized cleaning agent was investigated for improving the performance decreased by the pollution of collecting plates in an electric fume collector (EFC) which was developed and applied for the purification of effluent gas including oil mist from the textile industry. The pollutants on the surface of collecting plates were blackened by the condensation of oil mist for a long time and difficult to remove by general cleaning agents. The composition of an optimized cleaning agent consisted of alkali, alcohol, glycol and non-ionic surfactant sources was determined by considering the pollutant properties and effect on the damage of the basic metal of collecting plate and so on. The developed cleaning agent solution diluted by 9.1% was applied to the field test, and also the pollutants strongly adhered on collecting plate surfaces were successfully removed by a simple spraying method. The effluent gas purification efficiency of EFC increased significantly by cleaning of collecting plates.

Production and Purification of Single Chain Human Insulin Precursors with Various Fusion Peptides

  • For the production and purification of a single chain human insulin precursor, four types of fusion peptides $\beta$-galactosidase (LacZ), maltose binding protein (MBP), glutathione-S-transferase (GST), and (His)(sub)6-tagged sequence (HTS) were investigated. Recombinant E. coli harboring hybrid genes was cultivated at 37$\^{C}$ for 1h, and gene induction occurred when 0.2mM of isopropyl-D-thiogalactoside (IPTG) was added to the culture broth, except for E. coli BL21 (DE3) pLysS harboring a pET-BA cultivation with 1.0mM IPTG, followed by a longer than 4h batch fermentation respectively. DEAE-Sphacel and Sephadex G-200 gel filtration chromatography, amylose affinity chromatography, glutathione-sepharose 4B affinity chromatography, and a nickel chelating affinity chromatography system as a kind of immobilized metal ion affinity chromatography (IMAC) were all employed for the purification of a single chain human insulin precursor. The recovery yields of the HTS-fused, GST-fused, MBP-fused, and LacZ-fused single chain human insulin precursors resulted in 47%, 20%, 20%, and 18% as the total protein amounts respectively. These results show that a higher recovery yield of the finally purified recombinant peptides was achieved when affinity column chromatography was employed and when the fused peptide had a smaller molecular weight. In addition the pET expression system gave the highest productivity of a fused insulin precursor due to a two-step regulation of the gene expression, and the HTS-fused system provided the highest recovery of a fused insulin precursor based on a simple and specific separation using the IMAC technique.

Application of simple and massive purification system of dsRNA in vivo for acute toxicity to Daphnia magna

  • The RNA interference (RNAi) has been considered as an important genetic tool and applied to develop a new living modified (LM) crop trait which is an improvement of nutrient quality or pest management. The RNAi of DvSnf7 has been used for resistance to LM maize and the Western Corn Rootworm which is a major agricultural pest for the US Corn Belt. Most of the environmental risk assessments (ERA) of double strand RNA (dsRNA) have been performed using in vitro transcript products, and not in vivo expressed product. A large amount of dsRNA was required for the acute toxicity assay of water fleas. Therefore development of massive dsRNA purification techniques is critical. Daphnia, a freshwater microcrustacean, is a model organism for studying cellular and molecular mechanism involved in life history traits and ecotoxicology. In this study, we established the massive dsRNA purification method using Escherichia coli and implemented acute toxicity assays to Daphnia magna. As a result, the present RNase A and DNase I, dsRNA was efficiently purified without any special techniques or equipment. Even though purified dsRNA existed during the acute toxicity test, lethality or abnormal behavior were not observed in D. magna. These results indicated that GFP and DvSnf7 dsRNA were not significantly affected to D. magna due to their lack of sequence matching in its genome. The purification method of dsRNA and the acute toxicity assay of water fleas using purified dsRNA would be suitable for the toxicological studies of LMOs to aquatic non-target organisms.

Determination of Chloramphenicol in Milk by High Performance Liquid Chromatography (HPLC를 이용한 우유중의 클로람페니콜의 정량 분석)

  • Seven different sorbents were evaluated for their adsorptivity and desorptivity of antibiotic, chloramphenicol. Among the sorbents studied, Carbopak B was found to be the most efficient in enriching the chloramphenicol from dilute aqueous solution. Interfering components in the milk matrix could be washed off by water and petroleum ether from Carbopak B column, while the chloramphenicol was retained on the surface of Carbopak B. The method of simple and efficient purification and enrichment of chloramphenicol using Carbopak B, followed by quantitative analysis employing $C_{18}$ reversed phase high performance liquid chromatography has been applied to the determination of chloramphenicol in milk.

Production of human leptin in Bacillus subtilis

  • Human leptin is a 16 kDa (146 amino acids) protein secreted from adipocytes and influences body weight homeostasis. In this report, active human leptin was successfully produced in the culture medium of Bacillus subtilis. After simple purification steps consisting of ammonium sulfate precipitation and anion-exchange column chromatography, 2.3 mg of leptin with a purity of greater than 95% was obtained from the 0.5 L culture with the recovery yield of 54.9%. The purified leptin showed the correct folding structure with one disulfide bond.

  • Techniques for protein refolding from inclusion body are discussed in view of its engineering application to large scale protein purification. Among the techniques, dilution and dialysis are mainly utilized due to simple operation. Membrane reactor, gel filtration chromatography, and continuous tank operation are emerging tools for their process-scale possibility in refolding. Reaction engineering approaches could be used to analyze the kinetic behaviour in the process scale refolding reactor. The kinetic analysis is helpful in the optimization of refolding yield in the refolding reactor.

  • Liquid-chromatography coupled with electrospray-ion trap mass spectrometry was applied to the analysis of the eleutherosides B and E in the Eleutherococcus senticosus cortexes. The optimum ESI/MS results were obtained in the positive ion mode using extracted ion chromatogram targeting Na-adduct molecular ion of each compound. This method allowed rapid and simple gradient separation of underivatized eleutherosides B and E without pre-purification steps at very low concentration.

  • A novel and simple purification method for microbial $poly-{\beta}-hydroxybutyrate$ (PHS) was developed. Sodium hydroxide was found to be efficient for digesting cell materials. Initial biomass concentration, NaOH concentation, digestion time, and incubation temperature were optimized. When 40 g/l of biomass was incubated in 0.1 N NaOH at $30^{\circ}C$ for 1 h, PHB purity of 88.4% with a weight average molecular weight ($M_w$) of 770,000 and a polydispersity index (PI) of 2.4 was recovered with a yield of 90.8% from the biomass which initially contained PHB of a $M_w$ of 780,000 and a PI of 2.3.

  • Chemistry laboratory class was produced for senior high school freshmen with the cooperation of university staffs, high school teachers and the university students. Although the students who will take the lab class are senior high school freshmen, we decided to prepare four experiments that are simple and have highly visible reactions or transformation. That is, 1) Water purification, 2) Surface modification, 3) Briggs-Rauscher reaction, and 4) Polymer synthesis and characterization. After the safety guideline and experiment instructions by the faculty staffs, two teaching assistants (TAs) supervised each experiment. Since taking a direct part in it will provide stronger impact than only being one who is just watching the experiment, all experiments contained some process that the high school students must handle the reagents, tools or the equipment, by themselves. Although, the operation performed by the students was limited to a series of simple actions, the various unique phenomena presented by the experiments impressed the students. The lab class was fruitful not only for high school students, but also good for university students. The lab class provided good opportunity for them to improve the abilities to teach and guide someone.
