Alkaline Protease of a Genetically-Engineered Aspergillus oryzae for the Use as a Silver Recovery Agent from Used X-Ray Film

  • Aspergillus oryzae U1521, which was a genetically engineered strain, produced 1,000,600 U per g . glucose of extracellular alkaline protease within 72 h in a submerged fermentation. However, the alkaline protease was not detected during the first 24 h. Northern blot analysis indicated that the enzyme synthesis was repressed at the transcriptional level during the lag period. Both catabolite repression and pH of the growth medium significantly affected the enzyme production. Use of this enzyme as a silver recovery agent from used X-ray film was confirmed by experiments in the shake-flask scale.

Improvement of Wrinkle Recovery and Functional Properties in Linen Fabrics (아마직물의 방추성과 복합기능성 향상을 위한 연구)

  • This study provides improved wrinkle recovery and UV protection capabilities as well as an antibacterial and deodorizing function to linen fabrics for summer shirts. The results obtained from this study are as follows. By setting catalyst concentration to 1.2% and DMDHEU concentration to 6% respectively and applying a heat treatment to them at $160^{\circ}C$ for 5 minutes, the decrease of fabric strength could be minimized and the crease resistance of linen fabrics improved. Compared to the treatment with DMDHEU only, the crease resistance of linen fabrics could be maintained and degradation of their properties could be more effectively prevented by applying the mixture of the UV absorber and the nano silver to the DMDHEU resin. The UV protection of fabrics could be improved by adding the UV absorber. Although the separate treatment of resin or the nano silver had no effect on the improvement of the UV protection properties for treated fabrics, they could increase the UV protection capability when they were combined with the UV absorber. Linen fabrics could possess an antibiosis and deodorizing capability by applying the mixture of the UV absorber, the nano silver, and the resin. The UV protection, crease resistance and flexibility of finished fabrics were maintained even after laundering. Washed treated fabrics maintained excellent antibiosis and odor free capabilities compared to untreated fabrics.

  • The removal characteristics of silver ion from waste photographic solution have been investigated by recovering silver electrochemically. Cyclic voltammetry for synthetic and actual wastewater which containing silver ion was investigated to understand its electrochemical behavior. For both synthetic and actual wastewater, the recovery of silver according to the electrowinning time was observed to be increased as the applied potential was raised. In addition, the applicable potential for the electrowinning of silver was found to be lower for synthetic wastewater compared with actual wastewater. As the temperature was increased, more silver was recovered for both wastewater, which indicated the electrowinning reaction was endothermic. The electrowinned silver was refined electrochemically to increase its purity and the variation of the purity of silver was examined according to the electrorefining potential.

  • To enhance the grade and recovery rate of the gold/silver ores which yield at Philippine Mankayan mine, we studied the characteristics which are the geologic and mineralogical features of gold and silver ore, the liberation by crushing and grinding, the separation by sieving and shaking table. Gold/silver ore is composed of the sulfide minerals like pyrite, sphalerite, galena; and the gangue minerals which is quartz, clay. Gold/silver element are mainly contained in a sulfide minerals like pyrite, sphalerite and galena. To increase the liberation rate of sulfide minerals containing gold/silver element, the gold/silver ore has to be grounded under $100{\mu}m$ very finely because the crystal size of sulfide minerals is distributed from $1{\mu}m$ to $100{\mu}m$. The liberation rate of gold/silver ore increases to 92% when the particle size ($d_{90}$) of ore is grounded below $100{\mu}m$ by jaw crusher $\to$ cone crusher $\to$ rod mill by steps. The grade and recovery of sulfide minerals could not be enhanced by sieving separation because those crystal size is distributed homogeneously below $100{\mu}m$. But, when we separated the sieved ore using shaking table, the gold and silver grade increased to 40 ppm and 140 ppm, respectively. Then the recovery rate of gold reach almost 100% but that of silver is no more that 50%.

Enzymatic Hydrolysis of Gelatin Layers of X-Ray Films and Release of Silver Particles Using Keratinolytic Serine Proteases from Purpureocillium lilacinum LPS # 876

  • Enzymatic decomposition of gelatin layers on used X-ray films and repeated utilization of the enzyme for potential application in silver recovery were investigated using keratinolytic serine proteases from Purpureocillium lilacinum LPS # 876. At pH 9.0, the enzymatic reaction was enhanced by the increase of enzyme concentration or by the increase of the temperature up to $60^{\circ}C$. Under the conditions of 6.9 U/ml, $60^{\circ}C$, and pH 9.0, hydrolysis of the gelatin layers and the resulting release of silver particles were achieved within 6 min. The protective effect of polyols against thermal denaturation was investigated. The presence of glycerol and propylene glycol increased enzyme stability. When the reusability of the enzyme for gelatin hydrolysis was tested, it could be seen that it could be effectively reused for more cycles when glycerol was added, compared with the enzyme without protective agents. The results of these repeated treatments suggested that a continuous process of recycling silver from used X-ray is feasible. Keeping in mind that recycling is (at the present time) needed and imperative, it can be remarked that, in this research, three wastes were successfully used: hair waste in order to produce serine proteases; glycerol in order to enhance enzyme thermal stability; and used X-ray films in order to recover silver and PET films.

Quantitative analysis of silver in chlorinated polymer (Chlorine을 함유한 고분자 물질 중 Ag의 정량분석)

  • Quantitative analysis of silver from the thermal decomposition of chlorinated polymer contained nano silver is described. The chlorine contained in the chlorinated polymer (e.g. PVC) is liberated as hydrochloric acid gas by heating and a lot of silver produces AgCl. $HNO_3$ and $NH_4OH$ were used for dissolving the Ag and the AgCl. The silver complex was formed by $NH_4OH$. Then the complex was decomposed to silver by heating at $500^{\circ}C$ and the Ag was dissolved by dilute $HNO_3$. Recovery of silver in PVC material was 99.0%.

  • The dissolution of gold and silver from concentrate was studied with acidic thiourea solution. The results showed that the gold and silver extraction was severely affected by concentration of thiourea and oxidant, pulp density, etc. Especially, oxidant such as ferric ions enhanced the leching rate of gold and silver. High concentration of ferric ions, however, dissolved the sulfide ore to form electrochemically passive layer on the surface of ore particles, which caused the precious metal not to be leached out from the ore. The use of $SO_2$could not effectively enhance the recovery of precious metal but reduce to some extent the consumption of thiourea. The leaching of gold and silver was achieved with recovery more than 90% and 80%, respectively, under the following conditions; Thiourea conc. :0.4M Oxidant : None $H_2SO_4$ conc. : 0.5M Pulp density : <10% Leaching time :4 Hrs Potential :250mV The Thiourea was consumed about 10% in comparison with its initial concentration.

  • In order to apply the silver cementation method using Fe powder from pregnant thiourea leaching solution. Parameters such as the amount of Fe powder addition, agitation speed, and temperature were investigated. The silver cementation rate was increased by the increasing of Fe powder addition, agitation speed, and temperature. The highest silver cementation rate was found when the addition of Fe powder was 50 g/L at the agitation speed of 500 rpm. The silver cementation rate increase with increasing temperature according to the Arrhenius equation and obeys $1^{st}$ order kinetics. The activation energy from the kinetics data was found to be between 13.73 KJ/mol and 17.02 KJ/mol. In the XRD analysis, goethite was detected in the precipitate of the thiourea leach solution. This indicates that an oxidation-reduction reaction had occurred in the thiourea solution due to the addition of the Fe powder.

  • A study on the recovery of silver and nitric acid in the liquid waste resulted from the mediated electrochemical oxidation(MEO) process was conducted. The removal of silver in the concentrated nitric acid solutions was carried out by the electrodeposition. The removal efficiency more than 98% could be obtained in nitric acid concentrations less than 3 M with the current efficiency of nearly 100%. The experimonts on the evaporation for the recovery of nitric acid were performed as well. At the evaporation factor of 25., the degree of nitric acid recovery in 3.5 M nitric acid solution containing 0.5 to 1.0 mol% NaNO, was 80~90% resulting in 2.8~3.1 M nitric acid. The design factors and operating conditions of the distillation tower were analyzed by using MEH model derived by Maphtali-Sandholm with the throughput of 4 kg/hr for the enrichment of dilute nitric acid solution recovered by evaporation to reuse in the MEO process. The distillation column composed of eleven theoretical stages having the overall tray efficiency of 70% are needed to obtain 1.03 kg/h of 12M nitric acid and 2.97 kg/h of water with feed being introduced to the column at tray 6 from the bottom at the reflux ratio of 0.25, the reboiler with the heat load of 2.7 kW, and the condenser with the cooling load of 0.5 kW.

Purification of Eicosapentaenoic Acid (EPA) and Docosahexaenoic Acid (DHA) Esters from Squid Oil by Silver Ion Chromatography (은 이온 크로마토그래피에 의한 오징어유로부터 eicosapentaenoic acid(EPA) 및 docosahexaenoic acid(DHA)의 분리농축)

  • EPA and DHA extracted from methyl esterified squid oil were purified by silver exchanged resin, silver nitrate-impregnated silica gel, silver exchanged zeolite, and silica gel column chromatography, among which column chromatography using mixture of silver exchanged resin and silica gel (10% by weight) showed the best result. By this simple purification method, EPA and DHA were concentrated from 12.5 to 27.9% (yield, 86,0%) and from 21.7 to 49.5% (yield, 87.3%), respectively. Silver exchanged resin had additional advantages of outstanding reusability and simple recovery of silver.
