액상 견 Fibroin 처리 직물의 표면가공에 관한 연구 (Studies on the Surface Modification of Fabrics Treated with Fibroin Solution)

  • The silk fibroin solution was prepared and applied to the surface of fabrics for the purpose of weighting as well as a surface modification. The water-soluble fibroin solution can be obtained by dissolving the cocoon fibroin in a boiling solution of 50% calcium chloride for 60 minutes. For the fixation of a water soluble fibroin onto the fabric surface, the various methods were investigated. The fixation can be achieved on a silk fabric by the after treatment with ethanol, stannous choride and methacrylamide. On the other hand, the epichlorhydrin compound is the most promising fixation agent for a cotton fabric. As a result of the examination of property changes, the softness and crease recovery were lessened for a silk crepe fabric by treating with 1-2% fibroin solution, while those properties were improved for a silk knit fabric.

Memory-Enhancing Effects of Silk Fibroin-Derived Peptides in Scopolamine-Treated Mice

  • Although enzyme-hydrolyzed silk fibroin has been reported to enhance cognitive function before, it has been still unknown which peptides can improve memory. Here we report that amino acid sequences of three novel peptides were identified from fibroin hydrolysate. Fibroin hydrolysate was obtained by hydrolysis with protease after partial hydrolysis with 5M $CaCl_2$. Synthesized peptides derived from these sequences improved scopolamine-induced memory impairments in mice. We confirmed this hydrolysate had effects that improved learning and memory abilities by performing the Rey-Kim test. From this hydrolysate of silk fibroin, amino acid sequences of eight peptides were identified by LC-MS/MS. Three peptides (GAGAGTGSSGFGPY, GAGAGSGAGSGAGAGSGAGAGY, and SGAGSGAGAGSGAGAGSGA) were synthesized to investigate whether they could improve memory. Passive avoidance test and Morris water maze test were performed, and all peptides showed memory-enhancing abilities on scopolamine-induced memory impairments in mice. In this study, we identified three novel peptides that could improve memory, and that silk fibroin hydrolysate was a mixture of various active peptides that could enhance memory.

견피브로인/히아론산 브렌드 필름의 적외선 분광 분석 및 시차열분석 (Infrared Spectroscopy and Differential Scanning Calorimetry of Silk Fibroin/Hyaluronic Acid Blend Film)

  • 견피브로인의 응용가능성을 높이기 위하여 대표적인 생체고분자인 히아론산을 이용하여 견피브로인/히아론산 브렌드 필름을 제조하였으며 구조 및 열 특성을 살펴보았다. 1. 적외선 분광분석 결과 견피브로인과 히아론산 두 고분자 사이에는 수소결합 등의 상호작용이 존재하지 않는 것으로 나타났다. 또한 브렌드 필름내에 존재하는 실크피브로인은 EDC 에탄올 용액처리에 의하여 $\beta$-sheet 구조로 전이되었다. 2. 견피브로인/히아론산 브렌드 필름에서 견피브로인과 히아론산 각각의 열분해 흡열피크와 발열피크 온도에는 큰 변화가 없었으나, 열분해 피크의 폭이 넓어지는 것으로 봐서 실크 피브로인과의 브렌드 또는 EDC 처리에 의하여 열분해 반응이 매우 복잡한 환경하에서 진행되고 있음을 알 수 있었다. 3 견피브로인/히아로산 브렌드 필름은 자기 중량 대비 평균 70배의 수분을 흡수하였다.

The Effect of Coagulant and Molecular Weight on the Wet Spinnability of Regenerated Silk Fibroin solution

  • The regenerated silk fibroin with various molecular weights (MW) was prepared by different dissolution condition and the effect of coagulant on the wet spinnability of the various MW silk fibroin solutions dissolved in formic acid was investigated by the observation of wet spun filament in coagulant and the measurement of maximum draw ratio. The observation on the wet spun filament in coagulation bath revealed that good fibers without bead were formed in a high MW and a very high MW silk fibroin samples. In contrast, beads were observed in the silk fibroin sample with medium MW. The maximum draw ratio of wet spun silk fibroin filament decreased with MW reduction. The decrease of maximum draw ratio in isopropanol, acetone, DMF and THF was remarkably higher than that in methanol and ethanol, indicating that the coagulant type strongly influenced the wet spinnability. The two simple evaluation methods used in this study showed complementary information for wet spinnability: (a) The observation of filament in coagulant was effective to check a continuous fiber formation and a bead formation, and (b) the maximum draw ratio measurement was useful to examine the post drawing ability related to molecular orientation.

가잠견사 Fibroin의 품종별 Amino산 분석 (Amino Acids Analysis of Silk Fibroins among Varieties)

  • 본 보고는 가잠사 fibroin의 amino산 성분을 품종별로 Paper chromatography에 의해 분석하여 품종간의 amino산 성분차이를 관찰하고 동 amino산이 잠체내에서의 유전성과 생합성에 어느정도의 관련성이 있나 고찰하기 위한 것으로 다음과 같은 결론을 얻었다. 1. Paper chromatography에 의한 각종 amino산의 전개상황은 품종간에 대체로 유의하였다. 2. 그러나 숫자적 분석치는 타인의 것과 유의하면서도 품종간에 다소의 차이를 보였다. 3. 각 교잡종 견사 fibroin의 amino산 성분비율은 모체의 유전인자와 어느 정도 관련성이 있는 것으로 보인다. 4. 영양성 amino산은 견사 fibroin의 성분으로서보다 잠아성장에 더욱 관련하는 것 같다. 5. 지금까지 문한상에 발견되지 않은 미지의 amino산이 특수영양종견사 fibroin에 나타났으며 Hydroxyprolin이 아닌가 추측된다. 6. 견사 fibroin의 견사선내에서의 생합성기구로서 Stepwise process나 Template 학설 어느측이나 일방적으로 지대할 수 없는 실험결과이었다.

전기분사법에 의한 다공성 실크 피브로인 미세입자의 제조 (Fabrication of Porous Silk Fibroin Microparticles by Electrohydrodynamic Spraying)

  • 실크 피브로인은 뛰어난 생체적합성 및 생분해성으로 인해 의료용 천연고분자소재로 각광받고 있으며 다양한 형태로 제조되어 이용되고 있다. 전기분사법은 고분자 용액에 고전압을 적용하여 미세입자를 제조하는 방법으로 진행과정이 간단하고 첨가제를 필요로 하지 않는다는 장점을 지닌다. 본 연구에서는 실크 피브로인 다공성 미세입자를 제조하기 위하여 폴리(에틸렌 글리콜)(PEG)을 첨가한 후 전기분사를 실시하였다. PEG를 첨가함으로써 분사원액의 전도도는 감소하였고 점도는 증가하였다. 제조된 미세입자의 크기는 PEG 첨가에 따라 감소하였는데 이는 분사용액의 전도도 및 점도보다는 피브로인과 PEG간의 상분리에 의한 효과인 것으로 보인다. 제조된 실크 피브로인 미세입자를 물에 침지한 결과 PEG가 제거되었으며 최종적으로 실크 피브로인 다공성 입자를 제조할 수 있었다. 제조된 다공성 실크 피브로인 미세입자는 약물전달체 및 조직공학용 세포전달체로 이용가능성이 높을 것으로 기대된다.

  • The intermolecular hydrogen bond between silk fibroin in aqueous solution facilitates gelation of silk fibroin. The dependence of gelation by the concentration of silk fibroin, pH and temperature was investigated. The gelation time of silk fibroin is decreased with increasing the concentration of glycerol and temperature within a limited range. The pH optimum for gelation found out to be 9. Hydrophobic additives were investigated to reduce gelation time.

  • Using gel filtration chromatography (GFC), pure separation of high molecular silk fibroin was obtained and silk fibroin microsphere particles (SFMP) could be simply made by spray dryer method. Also, some of its physicochemical properties and morphology were investigated. The average molecular weight (Mw) of pure silk fibroin protein dissolved in calcium chloride is about 61, 500 g/mol as measured by gel permeation chromatography(GPC). (omitted)

  • The permselectivity of alkali metal chlorides through silk fibroin membrane was investigated at $25^{\circ}C$: The Permeability coefficients were found to increase in a sequence of LiCl < NaCl < KCl < CsCl. This sequence was explained by considering the partition and the hydrophilic membrane. The dependence of the permeability on the salts concentration was interpreted by means of TMS theory. The ionic mobility ratio of alkali metal chlorides through this membrane decreased with the increase of the Stokes radius. The Effectiveness of the charged density was found to depend on the ionic species. The greater the Stokes radius the larger the effective charge density of membrane.

Antioxidation and anti-inflammatory effects of gamma-irradiated silk sericin and fibroin in H2O2-induced HaCaT Cell

  • Oxidative stress in skin cells can induce the formation of reactive oxygen species (ROS), which are critical for pathogenic processes such as immunosuppression, inflammation, and skin aging. In this study, we confirmed improvements from gamma-irradiated silk sericin (I-sericin) and gamma-irradiated silk fibroin (I-fibroin) to skin cells damaged by oxidative stress. We found that I-sericin and I-fibroin effectively attenuated oxidative stress-induced ROS generation and decreased oxidative stress-induced inflammatory factors COX-2, iNOS, tumor necrosis factor-α, and interleukin-1β compared to the use of non-irradiated sericin or fibroin. I-sericin and Ifibroin effects were balanced by competition with skin regenerative protein factors reacting to oxidative stress. Taken together, our results indicated that, compared to non-irradiated sericin or fibroin, I-sericin, and I-fibroin had anti-oxidation and antiinflammation activity and protective effects against skin cell damage from oxidative stress. Therefore, gamma-irradiation may be useful in the development of cosmetics to maintain skin health.
