Degumming with enzyme Degummase can be a viable alternative to the existing methods of degumming. Degumming, carried out with the conventional method of Marseille's soap 25%(owf) for 1.5h at boil gives a weight loss of about 24%. On Comparison, degumming with 10 enzyme at 5$0^{\circ}C$ for 60 min gives a weight loss of about 22%. Treatment with enzyme requires a much lower temperature and also the treatment time is shorter than the conventional process. Processing of silk at these conditions is likely to retain the lustre and softness of silk. To economise on the quantity of enzyme, conditions were optimised for a pad-store process. An enzyme concentration of 0.5% at 5$0^{\circ}C$ for a time period of 5~6h resulted in a weight loss of 22%. Though the concentration of the enzyme used was 20 times less than that used for long liquor method there was some strength loss. At 5$0^{\circ}C$ treatments carried out for 7~8 h resulted in some degradation. Strength loss occurs because enzyme in the absence of any stirring keeps acting at the place where it is deposited. Effect of temperature was more pronounced than time in degrading silk.
The purpose of this study is to investigate the change of the physical properties, bending and luster properties, which are related to the touch and appearance of silk fabrics modified by the degumming process. The silk has long been known as one of the most elegant and soft textile materials. The raw silk yarn, or cocoon bave, spun from the spinneret, is rather stiff due to the sericin covering the two fibroins together. The sericin can be removed during a degumming process. The removal of the sericin would result in remarkable change in the physical properties of the raw silk fabrics, including luster of the fabrics, which process parameters could possibly be utilized to adequately control the silk fabric properties. The KES(Kawabata Evaluation System) is a testing methodology that has been used with considerable success for predicting the hand and tailorability of apparel fabrics. This study uses one of the KES, bending tester, to measure the bending properties of the silk fabrics degummed for specified period to change the physical/mechanical properties of the fabric. The KES bending measurement revealed that the bending rigidity decreased for both the warp and weft direction of the silk fabrics with the increase of the degumming period. It has been shown in this study that the some of the hand-related physical properties, including the bending rigidity, drapability, and luster, could be modified with the change in the degumming period.
In this study we have evaluated the separation characteristics and concentration of sericin using tubular type ultrafiltration membrane in silk degumming solution that extracted from electrolytic reduction water process. Ultrafiltration membranes have used in sericin separation performance and the separation characteristics of membrane satisfied typical Hagen-Poiseuille equation. It had the increase of flux according to the increase of feed pressure and temperature in occasion of pure water flux. And also the flux and solute rejection had about $25{\sim}60{\ell}/m^2{\cdot}h$ and more than 95% in sericin feed solution with concentration 1.00~1.89% at feed pressure force of $3{\sim}8kgf/cm^2$ respectively. In addition, the separation performance of tubular type ultrafiltration membrane for silk degumming solution was very steady-state with long experiment time.
Silk was treated with calcium hydroxide for degumming at different treatment times, temperatures and Ca(OH)$_2$ concentration to optimize degumming conditions in this thesis. After degumming, soluble and insoluble sericin were seperated and then the soluble sericin was characterized by measuring the average degree of polymerization (D.P.), lysinoalanine (LAL) content, DSC, and by amino acid analysis. And degummed silk fibroin was characterized by measuring tenacity and SEM. Degumming loss was increased by increasing the treatment time and temperature until about 30 minutes. After then, a slight difference was found along with treatment times at the Ca(OH)$_2$ concentrations of 0.07% and 0.1% solutions. After degumming, insoluble sericin ratio on degumming solution was increased by increasing treatment temperature at Ca(OH)$_2$ 0.04% solution. At the concentration Ca(OH)$_2$ of 0.07%, a soluble ratio was almost 100% regardless of treatment time and temperature. At the beginning of treatment, insoluble ratio was high at Ca(OH)$_2$ 0.1% solution but it was decreased by increasing treatment time. At the Ca(OH)$_2$ concentration of 0.04%, D.P. of soluble sericin was maintained as a constant value of 10 at 100$^{\circ}C$ although treatment time was increased. However, at 80$^{\circ}C$ and 90$^{\circ}C$, it was hard to prepare a soluble sericin having a constant D.P. by increasing treatment time. At the Ca(OH)$_2$ concentration of 0.07%, D.P. was almost 10 irrespective of treatment temperature and time. Soluble sericins with high D.P. of 20∼30 were obtained at 0.1% and 100$^{\circ}C$. LAL was not detected in soluble sericin. As the results of amino acid analysis, it showed that Ca(OH)$_2$ degumming reduced the contents of hydroxy amino acids like Ser., Thr. and Tyr. In DSC analysis of soluble sericin, endothermic peak by thermal deformation and pyrolysis showed at 189$^{\circ}C$ and at 299$^{\circ}C$, respectively. The tenacities of degummed silk were 15∼30% lower than that of raw silk. And it was decreased with increasing treatment time. From the morphological study, the thickness of degummed silk fibroin became thinner by increasing degumming loss. The roughness of a silk fibroin surface was appeared as treatment concentration was increased.

Toll-like receptor (TLR) is responsible for the recognition of foreign protein. Accordingly, TLR is mainly expressed in the immune associated cells. When foreign protein such as silk sericin is considered for the graft, the response of TLR should be considered. TLR is not all or none responsive receptor. TLR can be activated differently by the intensity of the input. Silk sericin is easily fragmented. The protein conformation of silk sericin is different to the degumming method. TLR response to silk sericin may be different to the degumming method. Consequently, objective tailored extraction method should be investigated and developed.
This study was investigated on the effect of degumming of cocoon shell, raw silk yarn and grey silk crepe by use of Papain, comparing with soap, soda and soap-soda. The obtained results may be summarized as follows : 1. Degumming loss, when the pretreatment was done, was completed by Papain more than by alkali. 2. Lousiness result of Papain degummed silk yarn was apt to be improved more than that of alkali degummed one. 3. As a result of tensile property test, elasticity and resilience of Papain degummed crepe were good compared to those of alkali degummed one. 4. Not only bending rigidity of Papain degummed crepe was reduced more than that of alkali degummed one but also hysteresis of bending moment was decreased in papain degummed crepe. 5. Handle of Papain degummed crepe was superior to that of alkali degummed one.
To investigate the strong and effective sericin fixing agents and conditions, raw silk fibers were treated with the mixtures of urea-formalin, thiourea-formalin and melamine-formalin as sericin fixing agents, respectively. And the treated raw silk fibers were degummed by the degumming liquors of alkaline detergent and sodium carbonate to confirm the hardness of sericin fixation. The weight of raw silk fibers was decreased in the process of sericin fixation treating at 80$^{\circ}C$ for 60min. The effective sericin fixation was obtained by treating at $60{\sim}80^{\circ}C$ for 15min with the mixture of melamine and formalin. The mixture of melamine and formalin showed an outstanding ability of sericin fixation because the three amino groups of melamine were able to cross-link the hydroxy amino acids of sericin such as serine with the assistance of formalin.
The attaching treatment of sericin onto polyester fiber was attempted to improve its sanitary properties. Generally, sericin, a gummy material covering the outer layer of silk filament, is subjected to be removed during degumming process of silk textile process. For this study, sericin particle dissolved within the degumming waste water could be collected by sedimentation of polyaluminium chloride. It was revealed that sericin particle were attached onto the surface of polyester fiber evenly by treatment of glutaraldehyde, a crosslinking agent. A frictional static charge of the treated polyester fabric could be improved, while its hygroscopic property was little changed.
The sericin fixation of silk fabrics by epoxy resins was studied in the presence of aqueous salt solution in different solvents at the indicated temperature for the desired time. Heavy weight gains were obtained in the reaction with glycerol diglycidyl ether (EX-313) and ethylene glycol diglycidyl ether (EX-810) catalyzed by potassium thiocyanate in such solvents as carbon tetrachloride and p-chloroethylene. The obtained results were summarized as follows; 1) The tested resins were found similar in reaction behavior to silk fabric. The effect of fixation and weight gains was higher in EX-810 than EX-313. 2) The weight gains were increased with reaction time and temperature, and degumming ratio reached a constant value in 90 min at 70$^{\circ}C$. 3) The weight gains and the degumming ratio reached an equilibrium at 3-5% of epoxy concentration. 4) The weight gains were remarkably influenced and increased by the concentration of salt solution. The degumming ratio reached an equilibrium over 0.5N concentration of salt. 5) The weight gains were increased with the dipping time in gently-sloping. The degumming ratio reached an equilibrium over 15 min dipping. 6) The effect of sericin fixation of hydrophobic solvents, such as Carbon Tetrachloride, p-Chloroeth-ylene, Cyclohexane, Xylene and Toluene, was found suitable. 7) The effect of drying temperature was not remarkable on the weight gains and the degumming ratio. 8) There was a slight decrease in the moisture regain of sericin-fixed silk and it may be possible to maintain the moisture regain in the sericin-fixed silk by the epoxy resins. 9) The results on testing physical properties of sericin-fixed silk fabric were as follows; The crease recovery was almost not different from undegummed fabric, but inferior to degummed fabric. The tensile strength was improving in accordance with the effect of sericin fixation, either the elongation did.

The fixing behaviors of raw silk yarns treated with melamine and formaldehyde at a molar ratio of 1:3 for trimethylolmelamine were investigated. Sericin was fixed during the fixing process, but a part of sericin I was removed simultaneously by hot water. The weight losses by fixing and the degumming losses by degumming greatly decreased with increasing concentrations of melamine and formaldehyde. The silk yarns fixed with 0.011 M melamine and 0.033M formaldehyde were significantly degummed due to the insufficient fixation of sericin and the alkaline hydrolysis of sericin by sodium carbonate during the degumming process. On the other hand, the silk yarns fixed with 0.055M melamine and 0.165M formaldehyde were degummed slightly (the degumming losses of 3-8%) due to the strong fixation of sericin, which might result from the many cross-linkages between the sericin I molecules, which were formed by trimethylolmelamine. Those fixed with the fixing solution containing 15% owf softener showed the lowest weight and degumming losses because under the condition of 15% owf softener, the cation of the softener can effectively form ionic bonds with the negatively charged side chain of aspartic acid in sericin. In addition, van der Waals' forces may be also formed between the hydrophobic tail of the softener and the hydrophobic region of sericin, which may help inhibit the removal of sericin I.
