Five cytotoxic and antioxidative flavonoids were isolated from the stem bark of Cudrania tricuspidata by consecutive purification using HP-20, silicagel and prep-HPLC. They were identified as taxifolin, orobol, eriodictyol, dihydrokaempferol and steppogenin by means of spectral studies. The antioxidative activities $(IC_{50})$ assayed by TBA method of these compound $1{\sim}5$ to were 6, 3, 3, >50, and $10\;{\mu}g/ml$, respectively. The effect on the growth inhibition $(IC_{50})$ of these compounds against P388 cell line were found to be 0.18, 3.3, 15 and $6.2\;{\mu}g/ml$, respectively in the order of compound 2 to 5.
In order to isolate antimicrobial substances from Schizandra chinensis, the dried fruits were extracted with the methanol and the extract showed a strong antimicrobial activity. Also, the methanol exract was further fractionated with hexane, dichloromethane, ethylacetate and buthanol. The ethyl acetate-soluble fraction showed the strongest antimicrobial activity. These fraction were further separated by using various chromatographic methods including thin layer chromatography, silicagel open column chromatography and prep. HPLC. A major component S-EA-5-T1 and S-EA-5-T3 from the ethyl acetate fraction, which showed a strong antimicrobial activity was identified by Mass and NMR spectrometry. Two compounds were isolated and identified as trimethylcitrate and the essential oil of Schizandra chinensis and was estimated as gomisin C, respectively. The growth of S. typhimurium was also inhibited about 1.65 to 2.86 log cycle in minced pork by the addition 1% of Schizandra chinensis extract for 12 days at $4^{\circ}C$. These results suggested that these compounds have a strong potential as a natual food preservatives.
When the rice straw mulched, the inhibition of weed growth was observed in the paddy field. If we use these allelopathic effect of decreased weed establishment, we can save labor, and protect environment. Aqueous extract of rice straw were bioassayed using water foxtail seeds to investigate chair effects on the germination and the growth, and to identify allelopathic compounds. The major results obtained were summarized as follows : Aqueous extract was fractioned by solvent, among various fractions, II-3, II-4 fractions inhibited the coleoptile and the radical of water foxtail, by 100%, respectively. There were fumaric acid and 4 unknown organic acids. Seven organic acids including fumaric acid inhibited the growth and germination of water foxtail in $10^{-2}$M solution. One of the most effective allelopathic compounds in rice straw was identified the unknown organic acid, as molecular weight 253, by GC/MS.
To identify the volatiles of Platycodon roots, the three-year-old and the 24-year-old Platycodon roots cultivated in a local farm were extracted with ethyl ether by SDE method. 54.71mg% and 80.01mg% of volatile components were collected in the three-year-old and in 24-year-old Platycodon roots, respectively. As a result of GC/MS(TIC) analysis, 24 and 28 compounds were identified, respectively; The amounts of volatile components such as hexanol, trans-2-hexenol and nonanal in 24-year-old roots were detected above ten times than those in 3-year-old roots. The Volatiles of 24-year-old roots were fractionated by SCC and identified by GC/MS to be 10 hydrocarbons, 8 alcohols, 10 aldehydes, 5 esters, 6 acids and 1 unknown compound as the major components. The species and total amounts of volatile component in 24-year-old Platycodon roots were considered to be higher than those in 3-year-old Platycodon roots. *Abbreviations 1. Lipophilic brown pigments produced during the fermentation of domestic Meju and Doenjang were fractionated by column and thin -layer chromatography (TLC). Each of the fractions was tested for the antioxidant activity and then characterized by spectroscopic analysis. The lipophilic brown pigments were separated into chloroform -soluble and methanol-soluble parts in which Meju resulted the higher content of chloroform-soluble part than that of methanol-soluble part ; however, Doenjang exhibited the opposite result to that of Meju. More strong antioxidant activity was found in the methanol-soluble part than the chloroform-soluble part. Four and five fractions were separated from chloroform-soluble and methanol-soluble parts respectively. by silicagel TLC. The fraction that exhibited the high antioxidant activity showed a strong absorption at 260nm caused by amino compounds in UV spectrum The other fractions which did not have antioxidant activity absorbed at 240nm by carbonic acid and it ester,. IR spectrum of each fraction commonly showed absorption at 3400cm-1 , 2800cm-1 , 1700cm-1, 1600cm-1, 1400cm-1 , 1300cm-1 and 1100cm-1. Especially , the fraction which had a strong antioxidant activity showed absorption at 2800cm-1, 1400cm-1, 1600cm-1 suggesting that the fraction contain Schiff's base and primary amine structure.
One of yellowed rice toxins, luteoskyrin, was investigated with respect to its identification, quantitation and producibility by Penicillium islandicum isolated from deteriorated rice. 1) Luteoskyrin was best resolved by thin-layer chromatography with silica gel G plate impregnated with 0. 5 N oxalic acid and acetone : n-hexane : water (6 : 3 : 1.5, upper layer) solvent system. The isolated yellow spot showed maximum absorption bands at 426 and 448 nm and changed to purple color upon exposure to sunlight for $2{\sim}3$ hours. 2) Detection limit for luteoskyrin was 4 ppm in elution-colorimetry and 0.1 ppm in densitometry after TLC. Assuming that the tolerance for luteoskyrin in rice is set below 3.68 ppm, densitometry is usable for its screening in grain samples 3) Producibility of luteoskyrin by Pen. islandicum was shown to be 11 mg/g mycelial mat in liquid culture and 40 mg/g autoclaved rice.
Typical rose flowers, 'Red Corvette (red)', 'Nobless (pink)', 'Golden Metal (yellow)', and 'Rose Yumi (white)', were used as experimental materials to examine flower color changes as affected by pressing method and light treatment for four weeks and eight weeks by 2,000 lux fluorescent light at a 16 hours daylength. Pressing materials in silicagel mat resulted in the least change after pressing and light treatment for all kinds. On the contrary, an electric pressing device caused color to change to brown. Flower colors deteriorated the most by an electric pressing device made in Korea for pink rose; an electric pressing device made in Japan for white rose; and paper sheet made in Korea for red rose. The sequence of degree in severity of color change after pressing was pink, yellow, and white roses. Light treatment brought about the most extreme color change in yellow 'Nobless' rose, while other three cultivars were rather stable in change of color.
This study was undertaken to investigate the antioxidative substance and activity of ethyl acetate extracted from Rumex crispus. Sample extracted follow in proper course of a solvent. Material refinement was carried out using silicagel column and Sephadex LH-20 column chromatography. Material sorting was carried by Gas Chromatography(GC/MS). 1,1-Diphenyl-2-Picrylhydrazyl(DPPH) free radical scavenging and enzyme activity were measured for antioxidative activity. as result of testing by DPPH free radical scavenging activity, Antioxidative activity was shown as the highest in the root, then leaf and stem in order. Ethyl acetate extraction of root part were 50% inhibitory concentration (IC50) Rumex activty(6.1 ug/mL). Rumex nipponicus(9.8 ug/ml) and Rumex acetoceae(31.5 ug/mL) in leaf part. The highest antioxidative activity of sample refined through silicagel column chromatography of Rumex crispus was appealed Fraction 5(IC50;3.57 ug/mL) in root and Fraction 6(IC50;85.9 ug/mL) in leaf. Fraction 5 in roof & Fraction 6 in leaf were refined using Sephadex LH-20 column chromatography. The highest antioxidative activity were appeared Fraction 4 (IC50;3.57 ug/mL) and Fraction 4 (IC50;18.41 ug/mL)in leaf. As for main phenol compounds 2,6-Dichloro-4-nitropnenol and 2-Isopropyl-5-methyl Phenol were identified in root and leaf, While 4-Vinyl-2- methoxy-phenol and 2,3-Dihydro- benzofuran were identifica ted only in leaf. Enzyme activity was shown low both in peroxidase(PDD) Non-activate(IU/mg protein)and in Superoxide dismutase(SOD) non-activate(IU/mg protein). 2,6-Dichloro-4-nitrophenol, 2-Isopropyl-5-methyl phenol, 4-Vinyl-2-methoxy-phenol were obtained in this experiment and these compounds are phenolic compounds which have OH group in the structure. With the result of this study these phenolic compounds which are extracted from Rumex crispus have high antioxidative effect. This antioxidative effect of Rumex crispus can be applied for chromo-preventive and antioxidative supplements which can be used for anti-allegy, aging, anti-tumor, aging and other oxidative disease for health promotion.
Certain parts of 190 kinds of medicinal herbs and 171 kinds of original materials of food were extracted by methanol. The extracts were tested their microbial inhibition activities against several food spoilage microorganisms, Micrococcus luteus, Bacillus subtilis, Bacillus cereus, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhimurium, Escherichia coli, Saccharomyces cerevisiae, Candida albicans, Penicillium citrinum, Aspergillus flavus and Aspergillus niger. The methanol extracts of Cornus officinalis, Evodia officinalis, Glycyrrhiza glabra, Salvia miltiorrhiza. Schizandrae fructus, Coptidis rhizoma, aroma hop and bitter hop were shown inhibitory effect on certain species of gram(+) bacteria. Aroma hop and bitter hop were shown inhibitory effect on certain species of gram(-) bacteria. The methanol extract of Salvia miltiorrhiza exhibited a strong antibacterial activities. It was purified by solvent fractionation, silicagel column chromatography, prep. TLC, prep. HPLC. The purified active substance was identified as cryptotanshinone by EIMS, $1^H-NMR,\;{13}^C-NMR$ and DEPT. Cryptotanshinone showed a strong antibacterial activity against gram positive bacteria $(MIC\;:\;3.91{\sim}62.50\;{\mu}g/mL)$. Especially, this compound was the most strong activity against Bacillus subtilis $(MIC\;:\;3.91\;{\mu}g/mL)$.
This study was performed to describe analysis method of 209 PCB congeners in ambient air samples. The samples were collected by high volume air sampler in Chonju city. Extracted samples were cleaned by silicagel cleanup modified with sulfuric acid and activated carbon cleanup processing. The cleaned samples were analyzed by high resolution gas chromatography and high resolution mass spectrometry (HRGC/HRMS) with DB-5 column (60 m, 0.25 mm i.d., 0.25 m film thickness) to analyze the 209 kinds of PCB congeners. PCBs levels of air samples were detected to the range between 0.003 and $0.163pg-TEQ/m^3$. The PCBs congener of 162 kinds were detected in samples analysed using DB-5 column and 37 kinds peaks were overlapped with congeners more than one. It is difficult to isolate PCB 118/106 and PCB 105/127 in coplanar PCB, so it is likely to overestimate the concentration.. The distribution of coplanar-PCB congeners in origin source samples (Kanechlor and exhaust gas from incinerator) was compared with that in air samples, and PCB 81, PCB 77, PCB 126, and PCB 169 were higher in incinerator samples.
