Evaluation of interaction between organic solutes and a membrane polymer by an inverse HPLC method

  • Organic compounds are adsorbed on RO/NF membranes, and the adsorption may influence the rejection of organic compounds by the membranes. Because almost RO/NF membranes are composite membranes, the results obtained by adsorption experiment with using membrane pieces are unable to avoid the influence by the support membrane. In this work, the interaction between membrane polymer and organic solutes was examined by an inverse HPLC methodology. Poly (m-phenylenetrimesoylate), the constituent of skin layer of RO/NF membranes, was coated on silica gel particles and used as a stationary phase for HPLC. When water was used as a mobile phase, almost hydrophilic aliphatic compounds were not effectively adsorbed on the stationary phase, although hydrophobic compounds were slightly adsorbed. The results indicated that the hydrophilic aliphatic compounds are useful probe solutes to examine the molecular sieving effect of a membrane. When water was used as a mobile phase, the aromatic compounds were strongly retained, and therefore $CH_3CN/H_2O$ (30/70) was used as a mobile phase. It was revealed that the adsorption of aromatic compounds was controlled by stacking between solute and polymer and was hindered by non-planar structure and substituents.

Isolation and Characterization of Antitumor Agents from Dictamnus albus

  • This study was carried out to find new antitumor agents from plant resource. Three cytotoxic agents were isolated from the root of Dictamnus albus by hexane extraction, silica gel column chromatography and HPLC. They were identified to be dictamnine $(C_{12}H_9NO_2)$, preskimmianine $(C_{17}H_{21}NO_4)$ and fraxinellone $(C_{14}H_{16}O_3)$ on the basis of spectroscopic evidences. In this study, it was newly found that these compounds possess a cytotoxic activity against lung lymphoma L1210 cell line. Among them. Preskimmianine was the most potent against the lymphoma L1210 with a $IC_{50}$ of $3.125\;{\mu}g/ml\;(10.3\;{\mu}M)$. Toxicity of preskimmianine against normal Iymphocyte was observed at the concentration of $50\;{\mu}g/ml\;(165\;{mu}M)$. These results support the pharmacological role of D. albus, a herb known as Paeksun in Korea and used as an anticancer agent in folk medicine.

포물면 집광거울 방식의 태양광 집광채광시스템 개발 (Development of Solar Daylighting System Using Parabolic Mirrors)

  • We developed a parabolic reflector based daylighting system which can be used as an alternative indoor daytime lighting device such as for fluorescent lamps. The system comprises three main components : a daylight concentrator made of 4 pairs of parabolic reflectors and mirrors, a silica optical fiber bundle based light transmitter, and 4 light diffusers for the final indoor delivery of the collected daylight. We analyzed the performance of the system and revealed the system efficiency and daylighting factor. All test methods follow the rule, NR PV601 : 2007-daylighting system, governed by Korea Energy Management Corporation.

상업용 유리프릿의 소결 공정을 이용한 내수성을 갖는 CsPbBr3/Glass 세라믹 복합체의 제작 (Simple Fabrication of Green Emission and Water-Resistant CsPbBr3 Encapsulation Using Commercial Glass Frits)

  • In this work, narrow-band green-emitting CsPbBr3 particles are embedded in commercialized glass composites by a facile dry process. By optimizing the method through sintering in glass frit (GF) composites including CsBr and PbBr2, used as precursors, the encapsulation of CsPbBr3 particles made them waterproof with green fluorescence. To improve the fluorescent properties by reducing aggregation of CsPbBr3, fumed silica (FS) is additionally used to help particles avoid bulking up in the glass matrix. The CsPbBr3 perovskite/glass composites are characterized using scanning electron microscopy (SEM) images and energy-dispersive X-ray spectroscopy (EDS) maps, which support the existence of CsPbBr3 particles in the glass matrix. The photoluminescence (PL) properties demonstrate that the emission spectrum peak, full width at half maximum (FWHM), and photoluminescence quantum yield (PLQY) values are 519 nm, 17 nm, and 17.7 %. We also confirm the water-resistant properties. To enhance water/moisture stability, the composite sample is put directly into water, with its PLQY monitored periodically under UV light.

Estimation of lightweight aggregate concrete characteristics using a novel stacking ensemble approach

  • This study investigates the efficiency of ensemble machine learning for predicting the lightweight-aggregate concrete (LWC) characteristics. A stacking ensemble (STEN) approach was proposed to estimate the dry density (DD) and 28 days compressive strength (Fc-28) of LWC using two meta-models called random forest regressor (RFR) and extra tree regressor (ETR), and two novel ensemble models called STEN-RFR and STEN-ETR, were constructed. Four standalone machine learning models including artificial neural network, gradient boosting regression, K neighbor regression, and support vector regression were used to compare the performance of the proposed models. For this purpose, a sum of 140 LWC mixtures with 21 influencing parameters for producing LWC with a density less than 1000 kg/m3, were used. Based on the experimental results with multiple performance criteria, it can be concluded that the proposed STEN-ETR model can be used to estimate the DD and Fc-28 of LWC. Moreover, the STEN-ETR approach was found to be a significant technique in prediction DD and Fc-28 of LWC with minimal prediction error. In the validation phase, the accuracy of the proposed STEN-ETR model in predicting DD and Fc-28 was found to be 96.79% and 81.50%, respectively. In addition, the significance of cement, water-cement ratio, silica fume, and aggregate with expanded glass variables is efficient in modeling DD and Fc-28 of LWC.

아민-티타늄착체 촉매상에서 1,3-부타디엔의 삼량화반응에 의한 싸이클로도데카트리엔의 합성 (Synthesis of Cyclododecatriene from 1,3-Butadiene by Trimerization over Amine-Titanium Complex Catalyst)

  • 티타늄화합물과 티타늄부톡사이드를 각각 디아민과 결합시켜 새로운 구조의 중합촉매를 개발하였으며, 이들은 부타디엔의 삼중고리화를 통한 싸이클로도데카트리엔(CDT)의 합성반응에 대해 높은 촉매활성을 나타내었다. CDT합성반응은 고압식 액상반응기를 사용한 배취형 반응계에서 수행하였으며, 반응온도, 촉매의 종류, 촉매량, Al/Ti의 몰비 및 고정화방법 등이 생성물 CDT의 생성수율에 미치는 영향을 관찰하였다. 디아민과 4염화티타늄을 1:1로 결합시킨 촉매는 생성물 CDT에 대하여 90% 이상의 높은 선택성을 보였다. 생성된 CDT 중의 TTT/TTC 입체이성체비는 티타늄에 결합된 디아민의 종류와 Ti/디아민의 비율 등에 따라 달라졌다. 이들 균일계 착체는 담체상에 고정화시켜 사용할 수 있었으며, 티타늄 주촉매는 반응 중 추출되지 않고 활성을 유지하면서 여러 번 사용이 가능하였다. 실리카 담체보다는 탄소담체를 사용하여 티타늄화합물을 고정한 촉매가 보다 높은 활성을 보였으며, 특히 아미노실란 만을 중합시켜 제조한 담체에 티타늄을 결합시키면 BD의 전환율도 높고 CDT에 대한 선택도도 높게 나타났다.

Full validation of high-throughput bioanalytical method for the new drug in plasma by LC-MS/MS and its applicability to toxicokinetic analysis

  • Modem drug discovery requires rapid pharmacokinetic evaluation of chemically diverse compounds for early candidate selection. This demands the development of analytical methods that offer high-throughput of samples. Naturally, liquid chromatography / tandem mass spectrometry (LC-MS/MS) is choice of the analytical method because of its superior sensitivity and selectivity. As a result of the short analysis time(typically 3-5min) by LC-MS/MS, sample preparation has become the rate- determining step in the whole analytical cycle. Consequently tremendous efforts are being made to speed up and automate this step. In a typical automated 96-well SPE(solid-phase extraction) procedure, plasma samples are transferred to the 96-well SPE plate, internal standard and aqueous buffer solutions are added and then vacuum is applied using the robotic liquid handling system. It takes only 20-90 min to process 96 samples by automated SPE and the analyst is physically occupied for only approximately 10 min. Recently, the ultra-high flow rate liquid chromatography (turbulent-flow chromatography)has sparked a huge interest for rapid and direct quantitation of drugs in plasma. There is no sample preparation except for sample aliquotting, internal standard addition and centrifugation. This type of analysis is achieved by using a small diameter column with a large particle size(30-5O ${\mu}$m) and a high flow rate, typically between 3-5 ml/min. Silica-based monolithic HPLC columns contain a novel chromatographic support in which the traditional particulate packing has been replaced with a single, continuous network (monolith) of pcrous silica. The main advantage of such a network is decreased backpressure due to macropores (2 ${\mu}$m) throughout the network. This allows high flow rates, and hence fast analyses that are unattainable with traditional particulate columns. The reduction of particle diameter in HPLC results in increased column efficiency. use of small particles (<2 urn), however, requires p.essu.es beyond the traditional 6,000 psi of conventional pumping devices. Instrumental development in recent years has resulted in pumping devices capable of handling the requirements of columns packed with small particles. The staggered parallel HPLC system consists of four fully independent binary HPLC pumps, a modified auto sampler, and a series of switching and selector valves all controlled by a single computer program. The system improves sample throughput without sacrificing chromatographic separation or data quality. Sample throughput can be increased nearly four-fold without requiring significant changes in current analytical procedures. The process of Bioanalytical Method Validation is required by the FDA to assess and verify the performance of a chronlatographic method prior to its application in sample analysis. The validation should address the selectivity, linearity, accuracy, precision and stability of the method. This presentation will provide all overview of the work required to accomplish a full validation and show how a chromatographic method is suitable for toxirokinetic sample analysis. A liquid chromatography/tandem mass spectrometry (LC-MS/MS) method developed to quantitate drug levels in dog plasma will be used as an example of tile process.

  • 니켈을 알루미나와 실리카 담체에 담지 한후 CFC-113 ($CF_2Cl$-$CFCl_2$)을 수소 기류 중에서 3FCl ($CF_2$=CFCl, CFC-1113)로 하는 환원반응을 실시하였다. 실리카나 알루미나의 경우 모두 활성이 급격히 저하하였으며 니켈의 함량이 증가할수록 저하속도는 현저하였다. 그러나 선택성은 80% 수준을 유지하면서 변하지 않았다. 알루미나 상에 동과 리티움을 니켈과 동시에 담지 시켰을 시는 활성과 선택성이 크게 개선되었다. 그러나 과량의 리티움의 증가는 활성과 선택성을 모두 저하시켰다. 담체로 실리카를 사용한 경우에는 활성이 급격히 저하되었는데, 이는 반응중 생성 된 HF가 Silica와 반응, $SiF_4$로 증발됨으로서 표면적이 큰 실리카 담체의 손실 또는 알카리 금속의 첨가로 인한 활성점의 감소에 의한 것으로 보여 진다.

  • 광미를 이용하여 다공성 세라믹 비드를 제조한 후 NOx/SOx 제거용 촉매 변환기로 응용하였다. 변환기 표면에 코팅처리된 촉매 지지체는 합성한 메조포러스 실리카(SBA-15)를 사용하였다. 다공성 세라믹 변환기의 내부 구조는 기공과 기공이 서로 연결되어 있는 3차원 망상구조이며 기공율은 80%로 나타났다. 또한, 촉매 변환기의 비표면적은 SBA-15 코팅 전 0.8 $m^2/g$에서 코팅처리 후에는 55 $m^2/g$으로 크게 증가하였다. NOx/SOx 제거 실험은 다공성 세라믹 촉매 변환기 표면에 $V_2O_5$$V_2O_5$, CuO를 함께 담지한 것으로 실시하였다. NOx 전환율은 $V_2O_5$/CuO 변환기가 $V_2O_5$ 변환기에 비해 약 10% 정도 높게 나타났다. 또한, $V_2O_5$/CuO 변환기는 반응온도 $350^{\circ}C$, 공간속도 10000 $h^{-1}$, 산소농도 5%에서 NOx 95%, SOx 90% 이상의 전환율을 각각 나타냈다.

  • 실리카 지지 루테늄과 철의 합금($Ru/Fe-SiO_2$) 시료에 일산화탄소(CO)를 흡착시켰을 때 Ru/Fe의 몰비 9/1 이상에서 다섯 적외선 흡수띠($2138.7{\sim}2142.5cm^{-1}$, $2067.3{\sim}2073.1cm^{-1}$, $1976.7{\sim}2017.2cm^{-1}$, $1737.9{\sim}1799.3cm^{-1}$, $1625.7cm^{-1}$)를 관찰했고, Ru/Fe의 몰비 8/2 이하에서 두 적외선 흡수띠($1934.0{\sim}1990.2cm^{-1}$, $1625.7cm^{-1}$)를 관찰했다. $2138.7{\sim}2142.5cm^{-1}$ 흡수띠, $2067.3{\sim}2073.1cm^{-1}$ 흡수띠, $1976.7{\sim}2017.2cm^{-1}$흡수띠는 Ru/Fe 뭉치 표면의 Ru 원자에 선형결합한 CO의 신축진동에 의한 것으로, $1774.2{\sim}1799.3cm^{-1}$ 흡수띠는 Ru/Fe 뭉치 표면의 Ru 원자와 다리결합하거나 높은 지수 평면에 있는 Ru 원자에 결합한 CO의 신축진동에 의한 것으로, $1934.0{\sim}1990.2cm^{-1}$ 흡수띠는 Ru/Fe 뭉치 표면의 Fe 원자에 선형결합한 CO의 신축진동에 의한 것으로 제시할 수 있다. $Ru/Fe-SiO_2$에서 CO를 흡착시켰을 때 Ru/Fe 몰비가 8/2 이하에서 증가할 때 CO가 Fe에 선형결합하여 생긴 $1934.0{\sim}1990.2cm^{-1}$ 흡수띠의 흡광도가 증가하였는데, 이 현상은 Ru/Fe 몰비가 8/2 이하에서 증가할 때 Ru/Fe 뭉치 표면에서 시료의 함량과 비교하여 Fe 농도가 증가했고 $Fe-SiO_2$ 시료와 비교하여 표면적이 Ru/Fe 몰비의 증가에 따라 점차 증가했기 때문으로 설명할 수 있다.
