Isolation of Herbicidal Compounds from Pulsatilla koreana Roots (백두옹(Pulsatilia koreana Nakai) 뿌리로부터 제초활성물질의 분리)

  • To search herbicidal compounds in Pulsatilla koreana Nakai, methanol extract of P. koreana roots was purified by sequences of XAD-7 column chromatography, silica gel adsorption column chromatography, silica gel flash column chromatography, preparative layer chromatography and preparative high performance liquid chromatography(Prep, HPLC).The final Prep. HPLC gave two herbicidally-active fractions. These fractions treatment at 100ppm inhibited the root length of Echinochloa crus-galli seedlings by 48% and 60% as compared with the control, respectively. Components in the two active fractions were analyzed by GC-MS Spectrometry. These compounds, which were isolated from P. koreana roots, were identified as several fatty esters, hydrocarbons, squalene, evidonol, and a diazepin analogue.

  • In this study, an ethanol extract being obtained from Allium cepa var. cepa examins the inhibitory effects on the glutathione S-transferase and the separation had been done by silica-gel column chromatography using various eluents, such as ethyl acetate, methanol, and 50% methanol. A volume of column fraction was 50ml and evaporation has been performde by the rotary evaporator under reduced pressure. Each fraction is being examined by thin layer chromatography and the UV spectrum at 365 nm was used to investigate separation patterns of spots on thin layer chromatography. When the eluent was changed, the spot patterns showed another different pattern on thin layer chromatography, so on. Fractions showing similar pattern are combined and eventually, three fractions are obtained. Each fraction is testified to examine the inhibition effects on glutathione S-transferase. All of these showed inhibition effects on glutathione S-transferase. The GC-MS shows that each fraction contains more than 2 compounds.

A study on deoxynivalenol production by water-saturated silical gel chromatography (물포화 Silica gel chromatography에 의한 Deoxynivalenol 생산에 관한 연구)

  • Deoxynivalenol producing isolates of Fusarium Graminearum R 6576 was grown on rice for 25 days at 19,25 and $28^{\circ}C$. Maximum production of deoxynivalenol(DON) by Fusarium graminearum R 6575 occurred at $28^{\circ}C$ and 20 days. Maximum concentration of 940 ppm DON were obtained after 20 days at an initial moisture content of 40%. A DON derivative, 15-acetyl-DON (15-ADON), was also found at concentrations of 150~300ppm after 5~10 days. Crude culture extracts were purified by water-saturated silica gel column chromatography which selectivity extracted DON when methylene chloride was as the mobile phase. Purity of crystallized DON was verified by thin layer and high performance liquid chromatography. Also this method was advantage method or production of DON and require little organic sorbent than the other methods.

  • The stem barks of Fraxinus rhynchophylla was extracted with 95% EtOH, and the concentrated extract was successively partitioned with $CH_2Cl_2$, n-BuOH, and $H_2O$ in order to investigate the major phytochemicals. From the $CH_2Cl_2$ soluble fraction, a sterol (1) was isolated through the repeated silica gel column chromatographies. Three additional compounds (2-4) were isolated from the n-BuOH soluble fraction through silica gel, RP-18, and Sephadex LH-20 column chromatographies. Their chemical structures were elucidated as daucosterol $(1;{\beta}-sitosterol-3-O-{\beta}-D-glucopyranoside)$, caffeic acid (2), 6,8-dihydroxy-7-methoxycoumarin (3), and coniferaldehyde glucoside (4) by comparing their spectral data with those in the literatures. All isolates (1-4) were the first to be isolated from F. rhynchophylla.

  • The methanol soluble fraction of the hot water extracts from Hovenia dulcis Thunb showed antioxidative and antimicrobial activity. The methanol fraction was successively purified with solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, and octadecylsilane column chromatography. The purified active substances were isolated by high performance liquid chromatography. The isolated substances were identified as 3-methoxy-4-hydroxybenzoic acid (vanillic acid) and 3-methoxy- 4-hydroxycinnamic acid (ferulic acid) by LC-MS and GC-MS. Vanillic acid and ferulic acid showed antimicrobial activity against Gram-positive bacteria, Gram-negative bacteria and yeast. The DPPH-radical scavenging activity of ferulic acid appeared more active than that of vanillic acid. DPPH-radical scavenging concentration of ferulic acid and vanillic acid were $14\;{\mu}g/mL\;(SC_{50})$, $100\;{\mu}g/mL\;(SC_{10})$, respectively.

  • Areca catachu L. which was showed antimicrobial activity against intestinal pathogens through screening herbs related treatments of intestinal diseases, were extracted by methanol and fractionated by n-hexane, ethylether, ethylacetate, and water. Antimicrobial activities of the methanol extract and each fractionates were then investigated under the anaerobic broth system. All fractions of Areca catachu L. except of n-hexane showed antimicrobial activity against all tested pathogens. Especially, ethylacetate fraction of them had the most significant inhibition activity. There is no significant difference of antimicrobial activity among each fractionates. Fraction of Areca catachu L. ethylacetate fractionate, which were fractionated by Sephadex G-200 and Silica gel column chromatography revealed the strongest antimicrobial activity at 5 to 7 and 20 of fraction number, respectively.

Brassinosteroids in Rice Bran and Polished Rice (미강과 백미의 Brassinosteroid 활성물질)

  • To investigate the presence of brassinosteroids in rice bran and pㅐlished rice, they were extracted with MeOH. The extracts were purified through sequential procedure of solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 column chromatography and charcoal adsorption chromatography. The activity of brassinosteroids was monitored by the rice inclination test and its presence was confirmed in each purification step. The purified active components were further separated by TLC and HPLC. Brassinosteroids in active fractions of rice bran were identified as castasterone and teasterone by HPLC. We acknowledge that our work is probably the first report of brassinosteroids in mature seeds of rice The more amount of brassinosteroids was confirmed in rice bran than polished rice. The contents of castasterone and teasterone which were identified in rice bran were 0.15 ng/g and 0.37 ng/g, respectively.

  • Acanthopanax sessiliflorus Seeman fruits (Araliaceae) were extracted at room temperature with 70% aqueous ethanol (EtOH). The concentrated extract was partitioned with ethyl acetate (EtOAc), n-butyl alcohol, and $H_2O$, successively. From the EtOAc fraction, four compounds were isolated through the repeated silica gel and octadecyl silica gel (ODS) column chromatographies. According to the results of physico-chemical and spectroscopic data including NMR, IR, and gas chromatography/mass spectrometer, the chemical structures of the compounds were determined as stigmasterol (1), linoleic acid (2), ${\beta}$-sitosterol (3), and stigmast-5-en-$3{\beta}$,$7{\beta}$-diol (4).

  • The ethanol extract of leaf mustard(Brassica juncea) exhibiting high antimicrobial activities was fractionated in the order of hexane, chloroform, ethylacetate and butanol fractions to test antimicrobial activity. The highest antimicrobial activity for the bacteria tested was found in the ethylacetate fraction, but a lesser extent in the butanol fraction. In contrast to antimicrobial activity for the bacteria, both ethylacetate and butanol fractions showed weak antimicrobial activity for yeasts. Unknown compound A in the ethylacetate fraction which exhibited a strong antimicrobial activity was isolated by silica gel column chromatography and HPLC, and exhibited 9 times more antimicrobial activity than the ethylacetate fraction.

  • In the course of screening for the tipA promoter-inducing substances, we isolated an active compound, sulfomycin Ia, from the mycelium of a microorganism designated 50634. The producing organism was identified as Streptomyces sp. on the basis of taxonomic studies. Sulfomycin Ia was purified from mycelial extract by silica gel column chromatography, LH-20 column chromatography, silica gel TLC, and preparative HPLC. The molecular weight of sulfomycin Ia was determined to be m/z 1129 (M+Na)$^{+}$ by FAB mass measurement and $^{1}$H NMR spectroscopic analysis. The structure was assigned as a derivative of sulfomycin I with thiazole, methyloxazole, oxazole, and pyridine rings by $^{1}$H NMR spectral data.
