3'-methyl-4-diethylaminoazobenzene으로 유발된 랫트 hepatocellula carcinoma 모델에서 항암제의 항암효과에 대한 평가기법 개발 (Development of novel method for evaluation of antitumor effect of anticancer drugs on hepatocellular carcinoma induced using 3'-methyl-4-diethylaminoazobenzene in Sprague-Dawley rat)

  • This study was carried out for investigating antitumor effects of 5-fluorouracil(5-FU), methotrexate(MTX) and retinoic acid(RA) on hepatocellular carcinoma induced in Sprague-Dawley rat. Antitumor effects were examined a flow cytometric DNA distributions by flow cytometry and stuied ATP/Pi using nuclear magnetic resorance, and the enzymatic activity of thymidylate synthetase and dihydrofolate reductase as well as contents of total collagen and sialic acid were measured with spectrophotometer. In this study, S phase fraction, contents of sialic acid and total collagen were decreased in the induced hepatocellular carcinoma treated with 5-FU and MTX, and synergistic effects of anticancer drugs were exhibited in the hepatocellular carcinoma treated with 5-FU and MTX simultaneously, and the inhibition of thymidylate synthetic and dihydrofolate reductase activity were shown in the hepatocellular carcinoma treated with 5-FU, MTX, and 5-FU and MTX simultaneously. On the other hand, the ratio of ATP/Pi were increased in all groups except group treated with RA. The experimental results suggest that above method may be valuable for evaluating antitumor effect of anticancer drugs.

초고온 살균유의 저장 중 겔 형성 거동 (Gelation Behavior of Ultra High Temperature Pasteurized Milk during Storage)

  • In order to examine physicochemical gelation behavior of ultra high temperature(UHT) pasteurized milk during storage at 4$^{\circ}C$ and 25$^{\circ}C$, pH, electrophoresis, alcohol test, sialic acid contents and free amino groups contents were biweekly determined. The pH of UHT pasteurized milk decreased with increasing storage time. Gelation of the UHT milk occured faster at 25$^{\circ}C$ than at 4$^{\circ}C$ with larger decreasing rate of pH. The alcohol test showed positive results at lower pH than 6.5, which could indicate the casein instability and beginning of gelation. The electrophoretic patterns showed a decrease in the concentrations of all caseins. Degradation of k-casein was faster in all cases, while $\alpha$-casein and $\beta$-casein were also extensively degraded later. The sialic acid contents of the samples increased gradually during storage, and the increasing rate was higher before gel formation. The free amino groups of the samples increased gradually during storage. The increasing rate of free amino groups was faster at 25$^{\circ}C$ than at 4$^{\circ}C$. The samples stored at 25$^{\circ}C$ gelled earlier than those stored at 25$^{\circ}C$, with corresponding increase of free amino groups. The residual proteolytic enzymes, which survived during the UHT heat treatments and were reactivated during storage, could be responsible for UHT pasteurized milk gelation during storage. It is assumed that proteolytic degradation of caseins followed by aggregation would be attributable to complicated reaction mechanism.

Diagnosis Value of Membrane Glycolipids Biochemistry Index in Intracranial and Gastrointestinal Tumors

  • The diagnostic value of membrane glycolipid biochemistry index, the lipid-bound sialic acid (LSA) and total sialic acid (TSA) in cerebrospinal fluid (CSF) was evaluated in 30 intracranial and 65 gastrointestinal tumors. The plasma LSA, TSA and red cell membrane sialic acid (R-SA) in were determined according to the method of Sevenmerhulm. Our results showed that the levels of LSA and TSA in CSF of intracranial tumor patients was higher than that of normal group(p<0.01). The concentration of TSA and LSA in patients with malignant glioma was higher than that of benign meningioma patients(P<0.01). No significance was found between intracranial halmatoma patients and normal control group for levels of membrane glycolipids (p>0.05). Results also found that the plasma LSA, TSA and R-SA of gastric carcinoma were significantly higher than those of control group (p<0.05); while no significant difference was found in the plasma LSA, TSA and R-SA levels between chronic gastritis, gastrohelcoma and normal control group (p>0.05). Plasma LSA, TSA and R-SA levels of gastric carcinoma patient were significantly higher than those of chronic gastritis patients and gastrohelcoma patients(p<0.05). It was also found that plasma LSA, TSA and R-SA contents were significantly higher in large intestine carcinoma patients than in benign in stestine tumor patients (p<0.05) while no significant difference was found between intestine benign tumor and normal control group (p>0.05). The levels of LSA, TSA and R-SA were obviously higher in the patients with metastasis than in the ones without (p<0.05.) The membrane glycolipid biochemistry index LSA and TSA in CSF are sensive markers for diagnosing intracranial tumors. For gastrointestinal malignant tumors the plasma LSA TSA and red blood cell membrane SA may be considered as auxiliary indicators for diagnosis. They can be used for distinguishing benign from malignant tumors.

Increased α2-6 sialylation of endometrial cells contributes to the development of endometriosis

  • Endometriosis is a disease characterized by implants of endometrial tissue outside the uterine cavity and is strongly associated with infertility. Focal adhesion of endometrial tissue to the peritoneum is an indication of incipient endometriosis. In this study, we examined the effect of various cytokines that are known to be involved in the pathology of endometriosis on endometrial cell adhesion. Among the investigated cytokines, transforming growth factor-${\beta}1$ ($TGF-{\beta}1$) increased adhesion of endometrial cells to the mesothelium through induction of ${\alpha}2-6$ sialylation. The expression levels of ${\beta}$-galactoside ${\alpha}2-6$ sialyltransferase (ST6Gal) 1 and ST6Gal2 were increased through activation of $TGF-{\beta}RI/SMAD2/3$ signaling in endometrial cells. In addition, we discovered that terminal sialic acid glycan epitopes of endometrial cells engage with sialic acid-binding immunoglobulin-like lectin-9 expressed on mesothelial cell surfaces. Interestingly, in an in vivo mouse endometriosis model, inhibition of endogenous sialic acid binding by a $NeuAc{\alpha}2-6Gal{\beta}1$-4GlcNAc injection diminished $TGF-{\beta}1$-induced formation of endometriosis lesions. Based on these results, we suggest that increased sialylation of endometrial cells by $TGF-{\beta}1$ promotes the attachment of endometrium to the peritoneum, encouraging endometriosis outbreaks.

흰쥐 부정소 상피세포, 내강액 및 성숙 전후 정자에서의 생리화학적 변화 (The Physiochemical Changes of the Epitheliat Cells, Luminal Fluid and Spermatozoa in Rat Edpididymis)

  • 흰쥐 부정소 정자의 성숙 전후에 일어나는 변화를 몇가지 효소를 중심으로 관찰하였고 그 성숙과정중에 일어나는 상피세포, 내강 및 정자 사이의 상호관계를 알아보기 위하여 실험군 별로 몇가지 효소의 활성도와 탄수화물 잔기의 함량을 측정하였으며, 전기영동을 이용하여 각 군의 차이를 관찰하고 이에 대한 웅성호르몬의 관련성을 알아보았다. 1. 부정소 두 정자와 부정소미 정자에서 활성도 측정시 lactate dehydrogenase, glucose-6-phosphatase 및 Na+ -K+ -ATPase의 경우는 유의한 차이가 없었으며, $Mg^2$+-ATPase의 경우만이 부정소미 정자가 부정소두 정자보다 유의성 있게 높은 활성도를 가지는 것으로 나타났다. 또한 부정소두와 부정소미 상피세포 , 내강 및 정자의 세군으로 나누어 각각 탄수화물 잔기를 정량하였을 때 hexosamine은 상피세포에서, sialic acid는 상피세포와 내강액에서 부정소미의 경우가 더 높은 함량이 존재하였으며, 내강액과 정자의 crude membrance fraction을 SDS-PAGE 했을 때 분자량이 33-37 KD 사이에 존재하던 band가 부정소미 내강액과 부정소미 정자의 crude membrance fraction에서 관찰되었으므로 흰쥐에서 정자의 성숙과정과 관련된 부정소내의 여러 변화를 비교하는 자료가 될 수 있었다. 2. 부정소 내강액에서 $\beta$ -glucuronidase와 $\beta$ -glucosidase의 활성도 및 웅성호르몬에 대한 의존성을 측정하였을 때 거세 후 5일째부터 이 두 효소의 활성도가 모두 유의하게 감소하기 시작하였고, tentosterone을 투여하였을 때는 $\beta$ -glucuronidase는 투여 5일, $\beta$ -glucosidase는 투여 10일 후부터 유의하게 증가하였으며 웅성호르몬에 대한 내강액의 의존성을 알아보기 위하여 SDS-PAGE하였을 때 tentosterone투여군의 부정소두에서 분자량이 약 21 KD에 해당하는 band를 새로이 관찰하였다.

Erythropoietin in vivo 시험법의 in vitro 대체 시험법 확립 (Replacement of the in vivo Bioassay for Erythropoietin with the in vitro Bioassay)

  • 현재는 재조합 Erythropoietin의 생물학적 활성을 mouse를 이용한 in vivo bioassay로 실시하고 있으나, 이 방법의 여러가서 불편함으로 인하여, 이를 대체하기 위해 Ba/F3 세포주를 이용한 in vitro assay 방법을 확립하였으나, 위에 상술한 바와 같이 in vitro assay는 erythropoietin의 당분포에 의한 차이를 보이지 않게 때문에, 이를 보완하기 위해서는 in vivo bio-activity와 정량적인 상관관계가 있는 당단백질의 isoform 분석법인 Capillary zone electrophoresis (CZE) 와 sialic acid 함량 결과를 동시에 분석해야 했다. 위의 결과 sialic acid 함량은 erythropoietin 원액에서 10 mol/mol,EPO 이상의 sialic acid 함량을 가져야 되며, CZE 결과는 재조합 erythropoietin이 isoform 분포가 isoform 3의 경우 5.594~0.593%, isoform 4의 경우 31.598~11.704%, isoform 5의 경우 37.033~29.301%, isoform 6의 경우 27.837~18.807%, isoform 7의 경우 17.085~7.824%, isoform 8의 경우 7.642~1.964%을 보여줌과 동시에 isoform 4의 면적은 isoform 5의 면적보다 항상 작아야한다. 이상 두 가지 시험결과와 in vitro assay 결과를 combine해서 in vivo assay를 대체할 수 있다는 좋은 실험적 data를 얻었으므로 이상 위의 3가지 분석법을 활용한 combined in vitro bioassay 법의 기초를 확립하였다.

Structural analysis of sialyltransferase PM0188 from Pasteurella multocida complexed with donor analogue and acceptor sugar

  • PM0188 is a newly identified sialyltransferase from P. multocida which transfers sialic acid from cytidine 5'-monophosphonuraminic acid (CMP-NeuAc) to an acceptor sugar. Although sialyltransferases are involved in important biological functions like cell-cell recognition, cell differentiation and receptor-ligand interactions, little is known about their catalytic mechanism. Here, we report the X-ray crystal structures of PM0188 in the presence of an acceptor sugar and a donor sugar analogue, revealing the precise mechanism of sialic acid transfer. Site-directed mutagenesis, kinetic assays, and structural analysis show that Asp141, His311, Glu338, Ser355 and Ser356 are important catalytic residues; Asp141 is especially crucial as it acts as a general base. These complex structures provide insights into the mechanism of sialyltransferases and the structure-based design of specific inhibitors.

구내 재발성 아프타성 궤양 환자에서 타액내 바이러스 검출 및 단백질의 변화 (Detection of Viruses and Changes of Protein of Saliva in Patients with Recurrent Aphthous Ulcer)

  • This study was performed to discover the underlining influences of Herpes Simplex virus (HSV) and Varicella Zoster virus (VZV), to detect the changes of whole protein and mucin level and to observe protein profiles in the saliva when recurrent aphthous ulcer (RAU) was present. Unstimulated whole saliva was collected from 23 patients who for over three years had a clinical history of RAU, in a group of 10 women and 13 men, ranging from 11 to 72 years of age, and 20 healthy subjects, in a group of 8 women and 12 men, who did not have the symptoms nor a past history of RAU. Through the means of Polymerization Chain Reaction, genomic DNA from the HSV and VZV was purified from the saliva samples for identifying precisely the two types of viruses, and the level of whole protein and sialic acids in the saliva and the ratio of sialic acid to whole protein were measured, and SDS-polyacrylamide gel electrophoresis was performed. The results obtained were as follows ; 1. 39.13% of patients showed 224 bp bands of VZV DNA, those were appeared more in patients than in control group (p<0.01), but there was no significant difference between patients and control group in HSV DNA (p>0.05). 2. The concentration of whole protein in men patients was lower than in men control group (p<0.05), but there were no significant differences between patients and control in other groups (p>0.05). 3. The concentration of sialic acids from patients was lower than control group in all groups (p<0.05). 4. The concentration of sialic acids in proportion to that of whole protein was lower in patients than in control group (p<0.05), and in the two women groups (p<0.01), but no noticeable difference was found between the two men groups (p>0.05). 5. There were no consistent differences observed in the protein profiles of patients with control group except that certain protein bands near 50 kDa was lower in patients than in control group. These results suggest that viruses such as HSV and VZV and reduction of salivary whole protein and mucin levels are related to development of RAU.

Anti-bacterial effects of enzymatically-isolated sialic acid from glycomacropeptide in a Helicobacter pylori-infected murine model

  • BACKGROUND/OBJECTIVES: Helicobacter pylori (H. pylori) colonization of the stomach mucosa and duodenum is the major cause of acute and chronic gastroduodenal pathology in humans. Efforts to find effective anti-bacterial strategies against H. pylori for the non-antibiotic control of H. pylori infection are urgently required. In this study, we used whey to prepare glycomacropeptide (GMP), from which sialic acid (G-SA) was enzymatically isolated. We investigated the anti-bacterial effects of G-SA against H. pylori in vitro and in an H. pylori-infected murine model. MATERIALS/METHODS: The anti-bacterial activity of G-SA was measured in vitro using the macrodilution method, and interleukin-8 (IL-8) production was measured in H. pylori and AGS cell co-cultures by ELISA. For in vivo study, G-SA 5 g/kg body weight (bw)/day and H. pylori were administered to mice three times over one week. After one week, G-SA 5 g/kg bw/day alone was administered every day for one week. Tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), IL-$1{\beta}$, IL-6, and IL-10 levels were measured by ELISA to determine the anti-inflammatory effects of G-SA. In addition, real-time PCR was performed to measure the genetic expression of cytotoxin-associated gene A (cagA). RESULTS: G-SA inhibited the growth of H. pylori and suppressed IL-8 production in H. pylori and in AGS cell co-cultures in vitro. In the in vivo assay, administration of G-SA reduced levels of IL-$1{\beta}$ and IL-6 pro-inflammatory cytokines whereas IL-10 level increased. Also, G-SA suppressed the expression of cagA in the stomach of H. pylori-infected mice. CONCLUSION: G-SA possesses anti-H. pylori activity as well as an anti-H. pylori-induced gastric inflammatory effect in an experimental H. pylori-infected murine model. G-SA has potential as an alternative to antibiotics for the prevention of H. pylori infection and H. pylori-induced gastric disease prevention.
