The optimal conditions for production of Monascus sp. KM100l cell mass on submerged culture and production of monacolin K on rice solid culture were investigated. On optimal conditions, 4.1 g/l of the cell mass was obtained at 28$^{\circ}C$ for 3 days. The mycelium were inoculated on 500 g of steamed rice using vinyl bag ($30.6{\times}44$ cm) and incubated at $30^{\circ}C$, 85% humidity for 21 days. Lactone form monacolin K was rapidly increased for 2 days and reached highest concentration of monacolin K (2,930 mg/kg) for 15 days, and monacolin K was decreased after 15 days.
Strains degrading and decolorizing acid dyes, Nylosan red E-BL 150%. were isolated from natural system, was named as ARK3. The optimal culture conditions of temperature and pH were $35^\circ{C}$, 7.0, respectively. Growth rate of cells in conditions of aerobic shaking more than standing culture conspicuously increased, and optical density of those to strain ARK3 were found as 1.38 and 0.25 after 42 hrs. Decolorization efficiency in batch culture which used as immobilization media to natural zeolite was 15% after 6 hrs, while suspension culture was 5%, also its of immobilization and suspension culture were 90% and 85% after 48 hrs, respectively. Decolorization efficiency of air-lift bioreactor was more than 90% to a dilution rate of $0.038hr^{-1}$, but that was decreased as 70%, when the dilution rate was $0.05hr^{-1}$. Even though at maximum dilution rate of this study, there was not appeared "wash out" phenomienon of biomass. Decolorization efficiency was 97.7% at a dilution rate of $0.025hr^{-1}$, when influent dye concentration was $100mg/\ell$. But if influent dye concentration increased as $150mg/\ell$, even though MLVSS increased, that of treatment water decreased as 93%. Also, when influent dye concentration increased as $200mg/\ell$ and $300mg/\ell$, decolorization efficiencies of treatment water abruptly decreased as 85% and 63%, respectively. Decolorization efficiency was more than 92% to the limit volumetric loading rate of $3.75mg/\ell\cdot{hr}$hr, without regard to variation of influent dye concentration or hydraulic retention time. if volumetric loading rate was more than $3.80mg/\ell\cdot{hr}$, at same condition, decolorization efficiency was lower decrease of retention time than increase of influent dye concentration.entration.
We investigated the effect of 1-methylcyclopropene (1-MCP) on ethylene production induced by simulated transport stress in $Begonia$${\times}$$hiemalis$ 'Blitz' and 'Carnival' to improve the display life in potted plants. The simulated transportation conditions were imposed for 4 days in simulated export containers with darkness, vibration with continuous shaking ($150{\pm}20$ rpm) on a rotary lab shaker, and low temperature ($12^{\circ}C$). Plants were treated with 1-MCP at three concentrations (5, 25, or 125 $nL{\cdot}L^{-1}$) and for three different periods (0, 6, or 12 hours) before undergoing the simulated transport stress treatments. Treatment with 25 or 125 $nL{\cdot}L^{-1}$ 1-MCP inhibited the abscission of open flowers by more than 40% as compared to the untreated plants. One week after the treatments, the ethylene production decreased in the plants treated with 125 $nL{\cdot}L^{-1}$ 1-MCP for 'Blitz' and 25 $nL{\cdot}L^{-1}$ for 'Carnival'. Ethylene production was correlated with concentration and duration of 1-MCP treatment in 'Blitz', but not in 'Carnival'. To reduce flower abscission and ethylene production, thus improve the display life when plants are exposed to transportation stress, we recommend pre-treatment with 1-MCP before packaging, using concentrations and durations specific to each cultivar, 125 $nL{\cdot}L^{-1}$ for 6 h and 25 $nL{\cdot}L^{-1}$ for 12 hours for 'Blitz' and 'Carnival', respectively.

Even if odorous compounds remain very low concentration in water, it cause strong odor. Because Geosmin and most of odorous compound had very low vaporization, those were difficult to analyze with GC/MSD and Purge & Trap. So, we needed pre-treatment method for decreasing amounts of extracting solvents, improving recovery efficiencies and increasing analytical efficiencies. This study developed efficient technology for analyzing odorous compounds, using various adsorbents and extracting solvents. The optimum adsorbent was XAD resins. Especially, XAD-2, XAD-7 and XAD-2010 were superior, but XAD-2 of these and the optimum extraction solvent is MTBE. Other extraction solvents' efficiency is in order of MTBE>Dichloromethane>n-Hexane>Diethylether. The optimum NaCl dosage for increasing efficiency is 5 g in liquid-liquid extraction method. The shaking time(0~24hr) has no concern with adsorption efficiency. The optimum adsorbent is XAD-2 resin and extraction solvent is MTBE. Dosing NaCl, adsorption efficiency is increased in liquid-liquid extraction method, but NaCl has no effect on liquid-solid extraction method. In this experimental results, this algae toxins(Mycrocystin, Anatoxin etc.).
In order to investigate the combined effect of potassium sorbate and sodium benzoate and the synergistic effect of sodium chloride on bacterial cell growth inhibition, Bacillus subtilis was cultured with or without shaking in tryptone-glucose-yeast extract broth containing 0.1% potassium sorbate and/or 0.03% sodium benzoat, which are equivalent to half of the maximum permissible levels, respectively. The combined treatment of the two preservatives did not show any synergistic effect of tne growth inhibition of B. subtilis. Addition of 2% sodium chloride, however, showed remarkable synergistic effect on the growth inhibition of the bacterium by potassium sorbate.
A bacterial strain inhibiting the growth of Vibrio anguillarum, the causative agent of vibriosis, was isolated from fish intestines. The isolated strain HS36 was identified as Aerococcus urinaeequi based on the characteristics of the genus according to Bergey's Manual of Systematic Bacteriology and by 16S rRNA sequencing. The growth rate and antibacterial activity of strain HS36 in shaking culture were higher than those in static culture, while the optimal pH and temperature for antibacterial activity were 7.0 and 30℃, respectively. The active antibacterial substance was purified from a culture broth of A. urinaeequi HS36 by Sephadex G-75 gel chromatography, Sephadex G-25 gel chromatography, and reverse-phase high-performance liquid chromatography. Its molecular weight, as estimated by Tricine SDS-polyacrylamide gel electrophoresis, was approximately 1,000 Da. The antibacterial substance produced by strain HS36 was stable after incubation for 1 h at 100℃. Although its antibacterial activity was optimal at pH 6-8, activity was retained at a pH range from 2 to 11. The purified antibacterial substance was inactivated by proteinase K, papain, and β-amylase treatment. The newly purified antibacterial substance, classified as a class II bacteriocin, inhibited the growth of Klebsiella pneumoniae, Salmonella enterica, and Vibrio alginolyticus.
The surface modification of silica particles (SPs) was systemically conducted by the treatment of 0.1-10 wt % phenylsilanetriol (PST) on the basis of SPs used through two step processes: 1) the PST coating of SPs via evaporation under reduced pressure and 2) their thermal condensation leading to Si-O-Si bond formation via heating at $130^{\circ}C$. The evaluation of the modified SPs was conducted by the simple floating test on water and the measurement of the contact angle (CA) of water droplet on the 2-dimensional layer of modified SPs on slide glass. When PST was used about 2 wt % or above on the basis of SPs (about average size: 50 nm) used, the modified SPs were fully floated on the water and all dispersed into upper organic solvent layer after a shaking with the mixture of the water and benzene, indicating that the modified SPs have hydrophobic properties. The modified SPs were characterized by $^{29}Si$ MAS NMR and physicochemical properties including SEM, TEM, BET, adsorption/desorption isotherms, etc. were measured and compared each other in details. This research demonstrates that the organosilanetriol is a good modifier applicable for the surface modification of inorganic oxide particles using a low amount of modifier on the basis of oxide particles used.
Objectives : The aim of the study is to offer basic data that help to reduce dental fear by measuring adult dental fear level with DFS scale. Methods : The subjects were 300 persons including college students in W University and their parents in Jeollabuk-do Province. Data were collected by convenience sampling from May 1 to May 30, 2013. Results : 1. Dental clinic visit provoked pain in 99 people(47.4%). Oral examination and preventive treatment evoked pain in 13 people(6.2%). 2. Women tended to feel much pain than men. Both women and men felt the thrilling fear when a needle pricks the flesh. 3. Respondents having dental caries, gum bleeding, halitosis, shaking tooth, and painful tooth had a higher dental fear level. 4. The direct pain experience(p<0.001) had the greatest influence. The next influencing factor was the insufficient anesthesia(p<0.05). 5. The explanatory power that the pain experience has influence upon dental fear is $R^2$=0.151. Conclusions : The direct pain experience and the insufficient anesthesia experience have the great influence upon patients' dental fear level. Anesthetics and analgesics can be considered as one of the positive methods for pain control.
A novel bacterium isolated from Cheonggukjang was identified as a glutamate-dependent Bacillus subtilis HA with 98.3% similarity to Bacillus subtilis Z99104. Optimization of poly-$\gamma$-glutamic acid ($\gamma$-PGA) production by modulating fermentation factors including carbon sources, nitrogen sources, inorganic salts and fermentation time was investigated. Optimum culture broth for $\gamma$-PGA production consisted of 3% glutamate, 3% glucose and various salts, resulting in the PGA production of 22.5 g/L by shaking culture for 72 hr at $37^{\circ}C$. Average molecular weight of $\gamma$-PGA was determined to be 1,220 kDa through MALLS analysis. The $\gamma$-PGA solution showed a typical pseudoplastic flow behavior, and a great decrease in consistency below pH 6.0 regardless of the same molecular weight of $\gamma$-PGA. The molecular weights of isolated $\gamma$-PGA were drastically decreased by heat treatment in various acidic conditions, resulting in different hydrolysis of $\gamma$-PGA. The consistency of $\gamma$-PGA solution was greatly decreased with increase heating time in acidic conditions.
