Proliferation of Gladiolus 'Topaz' Callus by Liquid Shaking Culture (액체진탕배양에 의한 글라디올러스 'Topaz' 캘러스의 증식)

  • This study was performed to enhance the proliferation rate of Gladiolus 'Topaz' callus. The callus was induced from the cermet tissue explants on MS solid medium with 10 mg/L 2,4-D. In the case of liquid shaking culture, proliferation of the callus was effective in MS medium with 0.05 mg/L 2,4-D at 2$0^{\circ}C$ under 16 hours daylength and in a 100 mL Erlenmeyer flask containing 20 mL of the liquid medium and at 75 rpm in rotation speed of the horizontal shaking culture. Furthermore the callus was also able to be subcultured in the same liquid medium.

Effects of Environmental and Nutritional Conditions on Fibrinolytic enzyme Production from Bacillus subtilis BK-17 in Flask Culture (플라스크 배양에서 Bacillus subtilis BK-17의 혈전용해효소 생산에 대한 환경 및 영양 조건의 영향)

  • The production of fibrinolytic enzyme from Bacillus subtilis BK-17 was studied in the shake flask cultures. The important medium components studied include nitrogen source, carbon source and inorganic salts. The environmental conditions include initial pH, temperature, shaking speed and working volume. Among various N-sources, C-sources and inorganic salts tested, soybean flour, D-glucose and Na2HPO4 gave the best results, and their optimal concentrations were 1.5%, 0.5% and 0.05%, respectively. The optimal pH and temperature were 9.0 and 37$^{\circ}C$. With decreasing working volume in the range of 25∼100ml in the 250ml flask or increasing shaking speed in the range of 100∼300rpm, the enzyme production was greatly enhanced. The enzyme activity under the optimal conditions was about 1400I.U./ml with urokinase as a standard.

Improvement of Liquid Culture Methods of Paecilomyces japonica (눈꽃동충하초균의 액체배양법 개선)

  • This study was carried out to improve the liquid culture methods of Paecilomyces japonica. The results show that the size of granular mycelium is smaller when the shaking speed is increased. Especially, the granular mycelium is the smallest at the shaking speed of 150rpm under the photoperiod of 12L-12D. Dry weight of mycelium was averagely 1.216 g in the Sikworm larva (SL) medium, and the weight was 2 times heavier than in the Potato dextrose (PD) medium. By adding 6 g of 6 mmbeads in the SL medium, the dry weight is increased to 1.332 g. The optimal addition of silkworm larval powder to the culture medium for gest harvest was 1.360$\pm$0.67 g in dry weight.

Effect of Culture Conditions on Microbial Cellulose Production by Acetobacter sp. A9 in Shaking Cultures

  • Several culture conditions affecting cellulose production by a newly isolated Acetobacter sp. A9 were examined by cultivating cells under shaking cultures. The inoculum size in the range of 1-10% (v/v) did not influence cellulose production. Maximum cellulose production was obtained with 200 rpm of agitation speed. The cells grown in the 75 ml of medium in a 250-ml conical flask produced the highest level of cellulose. The strain was able to produce cellulose at 25-3$0^{\circ}C$ with a maximum at 3$0^{\circ}C$. Cellulose production occurred at pH 4.5-7.5 with a maximum at pH6.5.

  • A set of modal parameters of a stay-cable have been extracted from a moving picture captured by a digital camera supported by shaking hands. It is hard to identify the center of targets attached on the cable surface from the blurred cable motion image, because of the high speed motion of cable, low sampling frequency of camera, and the shaking effect of camera. This study proposes a multi-template matching algorithm to resolve such difficulties. In addition, a sensitivity-based system identification algorithm is introduced to extract the natural frequencies and damping ratios from the ambient cable vibration data. Three sets of vibration tests are conducted to examine the validity of the proposed algorithms. The results show that the proposed technique is pretty feasible for extracting modal parameters from the severely shaking motion pictures.

  • Inverse suspension polymerization of acrylamide (AM) in cyclohexane was carried out to study the effects of concentrations of sorbitan ester (Span) stabilizers and 2,2'-azobis (2-methyl propionamidine) dihydrochloride (AIBA) initiator, amount of monomer, shaking speed, and polymerization temperature on the particle size of the resulting poly (acrylamide) (PAM) beads and their molecular weights. It was found that the particle diameter. in general, decreased with increasing concentration of stabilizer, shaking speed, and water content in the aqueous phase, and with decreasing concentration of initiator and polymerization temperature. The average molecular weight of the resulting PAM beads was also found to increase with increasing concentrations of monomer and stabilizer, and also with decreasing concentration of initiator, polymerization temperature, and water content in the aqueous phase. In this study, PAM beads ranging 2 ~ 50 ${\mu}$m in diameter, with 8000000 ~ 12000000 in the weight average molecular weight were successfully prepared in almost 100% conversion.

  • This study was divided into experiment 1(191 g egg-albumen plus 150g sucrose) and experiment 2(222g egg-albumen plus 150g sucrose)which were subdivided into groups treated with 0, 8.71, 17.43 and 26.149 egg yolk. These experiments were incubated in a shaking water bath($50^{\circ}C$ ) with a speed of 92(turnaround) per minute (or a period of times. The pH and specific gravity were measured after 2, 4, 6, 8, 10 and 12 hours incubation. The results obtained were as follows : 1. The pH and specific gravity were steadily increased by shaking time, but decreased by the amount of egg yolk. 2. In the groups treated with egg-albumen, egg yolk, and sucrose of experiments 1 and 2, PH was increased by shaking time and reached highest value at 10 hours shaking time. However, after 12 hours shaking time pH slightly decreased though not significantly. 3. In the experiment 1 and 2, specific gravity had a higher correlation with pH and the regression equation between specific gravity and pH (X) were Y=0.1081x+0.272(r=0.899n) and Y=0.083X+0.476(r=0.825$^{* *}$), respectively.y.

Modeling the cumulative residual deformation of high-speed railway bridge pier subjected to multiple earthquakes

  • High-speed railway bridge piers in seismically active area may be subjected to multiple earthquakes and then produce cumulative residual deformation. To study the cumulative residual deformation of high-speed railway bridge piers under multiple earthquakes, a nonlinear numerical analytical model with multi-DOF (MDOF) system is presented and validated against two shaking table tests in this paper. Based on the presented model, a simple supported beam bridge pier model of high-speed railway is established and used to investigate the cumulative residual deformation of high-speed railway bridge pier under mainshock-aftershock sequences and swarm type seismic sequences. The results show that the cumulative residual deformation of the bridge pier increases with earthquake number, and the increasing rates are different under different earthquake number. The residual deformation of bridge pier subjected to multiple earthquakes is accumulated and may exceed the limit of code.

  • Now, Autonomous unmanned vehicle has become an issue in next generation technology. 2D Laser scanner as the distance measurement sensor is used. 2D Laser scanner detects the distance of 80m, measured angle is -5 to 185 degree. Laser scanner detects only the plane, but using motor swings. As a result, traffic signs detect and analyze patterns. Traffic signs when driving at low speed, shape of the detected pattern is very similar. By shaking the laser scanner, traffic signs and other obstacles became clear distinction.
