절간고구마를 이용한 RNA 생산에 관한 연구 (RNA Production from the Hydrolyzate of Sliced and Dried Sweet Potatoes.)

  • 절간고구마 당화액을 탄소원으로하여 RNA를 다량 축적하는 효모를 선정한 결과 Cryptococcus laurentii가 가장 높은 균체량 및 RNA량을 나타내었다. 이 선정 균주에 대한 배양조건을 검사한 결과 최적 PH 6.0, 3$0^{\circ}C$, 48시간 통기 진탕배양하였을 때 가장 좋은 결과를 얻었다. 이러한 조건 하에서 배지조성을 변화시켜 RNA 함량과 균체증식량을 검토한 결과 (N $H_4$)$_2$S $O_4$ 0.4%, Peptone 0.6%. yeast extract 0.4%가 질소원으로 좋은 결과를 보였으며 K $H_2$P $O_4$의 최적농도는 0.5 %, M $n^{++}$ 은 0.1 %, $Co^{++}$은 0.001 %를 나타내었다. 또 비타민 요구성 실험에서 Ca-pantothenate를 400$\mu\textrm{g}$/$\ell$ 첨가하였을 때 가장 좋은 효과를 얻었으며 기본배지의 배양조건을 검토하여 얻은 최종배지를 비교해 본 결과 효모 균체당 RNA 함량이 12.9%~16.8%로 증가하였다.

Blockade of Kv1.5 channels by the antidepressant drug sertraline

  • Sertraline, a selective serotonin reuptake inhibitor (SSRI), has been reported to lead to cardiac toxicity even at therapeutic doses including sudden cardiac death and ventricular arrhythmia. And in a SSRI-independent manner, sertraline has been known to inhibit various voltage-dependent channels, which play an important role in regulation of cardiovascular system. In the present study, we investigated the action of sertraline on Kv1.5, which is one of cardiac ion channels. The effect of sertraline on the cloned neuronal rat Kv1.5 channels stably expressed in Chinese hamster ovary cells was investigated using the whole-cell patch-clamp technique. Sertraline reduced Kv1.5 whole-cell currents in a reversible concentration-dependent manner, with an $IC_{50}$ value and a Hill coefficient of $0.71{\mu}M$ and 1.29, respectively. Sertraline accelerated the decay rate of inactivation of Kv1.5 currents without modifying the kinetics of current activation. The inhibition increased steeply between -20 and 0 mV, which corresponded with the voltage range for channel opening. In the voltage range positive to +10 mV, inhibition displayed a weak voltage dependence, consistent with an electrical distance ${\delta}$ of 0.16. Sertraline slowed the deactivation time course, resulting in a tail crossover phenomenon when the tail currents, recorded in the presence and absence of sertraline, were superimposed. Inhibition of Kv1.5 by sertraline was use-dependent. The present results suggest that sertraline acts on Kv1.5 currents as an open-channel blocker.

액침진탕 배양에 의한 Monascus sp..가 생산하는 적색색소에 관한 연구 제1보 균주의 분리 및 색소생산 배양 조건 (Studies on the Red Pigment Produced by Monascus sp. in Submerged Culture. Part I Isolation of Strain and Cultural Conditions of Pigment Produced)

  • 적색색소의 생산을 위하여 태국산 tapioca chips와 곡자, 공기, 식물의 잎으로부터 우수한 균주를 선별하였고 이의 형태적 특성을 조사 동정하였다. 배양조건으로 온도, pH, 탄소원, 질소원, 통기효과, amino산, vitamin, 무기염류 등이 색소생산에 미치는 영향을 조사하여 최적조건을 구하여 다음과 같은 결론을 얻었다. 1) 선별된 균주는 형태적 특성을 근거로 균사에 격벽과 균사체끝에 피자기를 가지고 자낭포자와 분생포자를 만들며 특수구조인 후피포자가 있는 Monascus sp. 로 동정되었다. 2) 최적 배양조건은 온도 32~33$^{\circ}C$, pH 6.5, 탄소원으로 Tapioca chips 분말 3.5%, 질소원으로 NaNO$_3$ 0.2%, 금속이온은 MnO$_2$ 0.001% 첨가가 가장 좋았고 통기효과는 100/500mι(v/v) 진탕배양 (180rpm)에서 4일 배양이 가장 효과적이었다.

Enhanced pectinase and β-glucosidase enzyme production by a Bacillus subtilis strain under blue light-emitting diodes

  • Bacillus subtilis B22, a chemotrophic and aerobic bacterial strain was isolated from homemade kimchi, identified by 16S rRNA gene sequencing. B22 was primarily screened by biochemical, carbon source utilization tests. B22 was used to produce pectinase and ${\beta}$-glucosidase by submerged fermentation under different light sources. B22 was incubated in pectin media and basal media (pH 7.0) under blue, green, red and white light-emitting diodes (LEDs), fluorescent white light, and in darkness at $37^{\circ}C$, orbital shaker 150 rpm for 24 hours. Fermentation under blue LEDs maximized pectinase production ($71.59{\pm}1.6U/mL$ at 24 h) and ${\beta}$-glucosidase production ($56.31{\pm}1.6U/mL$ at 24 h). Further, the production of enzyme increased to pectinase ($156{\pm}1.28U/mL$) and ${\beta}$-glucosidase ($172{\pm}1.28U/mL$) with 3% glucose as a carbon source. Activity and stability of the partially purified enzymes were higher at pH 6.0 to 8.0 and $25-55^{\circ}C$. The effect on the metal ions $Na^+$ and $K^+$ and (moderateactivity) $Mn^{2+}$ and $Ni^{2+}$ increased activity, while $Hg^{2+}$, $Cu^{2+}$, $Fe^{2+}$, and $Fe^{2+}$ inhibited activity. EDTA, phenylmethylsulfonyl fluoride and 5,5-dithiobis (2-nitrobenzoicacid) reduced activity, while tetrafluoroethylene and 1,10-phenanthroline inhibited activity. The amylase was highly tolerant of the surfactants TritonX-100, Tween-20, Tween-80 and compatible with organic solvents methanol, ethanol, isoamylalcohol, isopropanol, t-butylalcohol and the oxidizing agents hydrogen peroxide, sodium perborate and sodium hypochlorite, although potassium iodide and ammonium persulfate reduced activity. These properties suggest utility of pectinase and ${\beta}$-glucosidase produced by B. subtilis B22 under blue LED-mediated fermentation for industrial applications.

Rhodococcus sp. EL-GT에 의한 고농도 페놀의 분해 특성 (Characterization of Biodegradation of Highly Concentrated Phenol by Rhodococcus sp. EL-GT)

  • The isolated strain, Rhodococcus sp. EL-GT was able to degrade high phenol concentrations up to 10 mM within 24 hours in the medium consisting of 5.3 mM $KH_2PO_4$. 95 mM $Na_2HPO_4$, 18mM $NH_4NO_3$, 1 mM $MgSO_4{\cdot}7H_2O$,\;50{\mu}M CaCl_2$,\;0.5 {\mu}M FeCl_3$, initial pH 8.0, temperature $30^{\circ}C$ in rotary shaker at 200 rpm. This strain was good cell growth and phenol degradation in the alkaline pH range range, and the highest in the pH range of 7 to 9. The microorganism was able to grow at the various chlorinated phenols, benzene, toluene, and bunker-C oil. As Rhodococcus sp. EL-GT was good capable of attachment on the acryl media, it would be used as microorganism to consist of biofilm in wastewater treatment.

Low Serum Potassium Levels Associated with Disease Severity in Children with Nonalcoholic Fatty Liver Disease

  • Purpose: Recent studies have suggested that decreased serum potassium level may contribute to various metabolic disorders in adult patients including nonalcoholic fatty liver disease (NAFLD). We aimed to study the correlation between serum potassium levels and the histologic severity of NAFLD in children. Methods: Pediatric patients with biopsy-proven NAFLD were included in this study. Demographic, clinical, and histopathological data were obtained. Multivariable logistic regression analysis was used to assess whether potassium levels are associated with the presence of nonalcoholic steatohepatitis (NASH) or fibrosis after adjusting for possible confounders. A p-value <0.05 was considered statistically significant. Results: Among 125 biopsies, 49.6% (62) had evidence of NASH while 66.4% (83) had some degree of fibrosis (stage 1-3). Mean serum potassium was significantly lower in NASH group as compared to non-NASH group ($4.4{\pm}0.42mmoL/L$ vs. $4.8{\pm}0.21$, p<0.001). Higher potassium level had negative correlation with presence of steatosis, ballooning, lobular inflammation, fibrosis and NAFLD activity score (p<0.05). On multivariable analysis and after adjusting for the metabolic syndrome and insulin resistance, higher potassium level was significantly associated with lower likelihood of having a histological diagnosis of NASH on biopsy (odds ratio [OR], 0.12; 95% confidence interval [95% CI], 0.05-0.28; p<0.001). Similarly, the likelihood of having fibrosis decreases by 76% for every 0.5 mmoL/L increase in potassium (OR, 0.24; 95% CI, 0.11-0.54; p<0.001). Conclusion: Our study shows an inverse relationship between serum potassium levels and the presence of aggressive disease (NASH and fibrosis) in children with NAFLD.

Agrobacterium sp. KF-67에 의한 미생물 응집제 생산 (Production of Bioflocculant by Agrobacterium sp. KF-67)

  • 토양으로부터 분리한 120 여종의 집락 중 응집제를 생산하는 집락을 선발하고 가장 응집력이 우수한 K-67 균주를 최종 선발하여 Agrobacterium sp. KF-67로 동정하고 실험균주로 사용하였다. 응집제 생산시 최적 배지성분을 조사하기 위하여 응집활성을 kaolin과 활성탄 현탁액에서 응딥활성을 측정하였다. 최적 배지성분과 농도는 탄소원과 무기질소원은 glucose와 NH4NO3 각각 2%와 0.1%, 유기질소원은 yeast extract와 peptone 각 0.01%, 무기염으로 CaCO3을 0.04%를 첨가하였을 때 응집활성이 가장 우수하였다. 배지 중 0.03% NaCl 첨가로 약 19%의 응집활성을 향상시킬 수 있었다. 이상과 같은 결과로 볼 때 Agrobacterium sp.에 의한 응집제 생산시 최적배지의 성분 조성과 함량은 2% glucose, 0.1% NH4NO3, 0.01% yeast extract, 0.01% peptone, 0.04% CaCO3, 0.03% NaCl이었고, 이때 기초배지에 비해 약 76.1%의 응집활성력이 증가하여 응집제 생산에 효과적인 배지성분 조건으로 조사되었다.

반응표면법 및 다목적 최적화를 이용한 철근콘크리트 건물모델의 모델 개선 (Model Updating of a RC Frame Building using Response Surface Method and Multiobjective Optimization)

  • 본 논문에서는 반응표면법과 다목적최적화 기법을 이용한 유한요소모델개선기법의 절차를 제안하고 이를 저층의 철근콘크리트건물의 모델개선에 적용하였다. 대상건물은 전단벽 신설 및 댐퍼부착을 위한 부재의 강재보강을 통해 내진보강이 이루어진 건물로서 보강전후에 소형 가진기를 이용한 진동실험을 실시하여 동특성을 구하였다. 대상건물의 개선에 사용된 변수는 기존콘크리트, 신규타설된 콘크리트, 조적의 탄성계수, 신축줄눈부의 스프링계수, 강재보강된 부재의 유효강성비이다. 보강전후 건물의 초기모델을 구축한 후 중심합성법에 따라 개선변수의 값을 변화시키면서 얻은 해석결과를 통해 고유진동수의 오차와 모드형상의 오차를 나타내는 2개의 반응함수를 구하고, 이를 다목적최적화의 목적함수로 사용하였다.

가진기를 이용한 강제진동시험에 의한 전기 캐비닛의 실험적 모드특성 분석 (Analysis of Experimental Modal Properties of an Electric Cabinet via a Forced Vibration Test Using a Shaker)

  • 원자력발전소(이하, 원전)에 설치되는 안전관련 전기기기들의 합리적인 내진검증을 위해서는 사전에 정확한 동특성분석이 필요하다. 이 연구에서는 원전에 설치되는 전기기기 캐비닛 구조를 대상으로 입력 진동의 수준에 따른 모드특성의 변화를 평가하였다. 이를 위해, 실제 전기기기 캐비닛을 시편으로 선정하고 가진 시험기를 이용하여 입력진동에너지의 크기를 변화시켜 가면서 진동시험을 수행하였다. 시험은 캐비닛의 문짝을 부착한 경우와 탈거한 경우로 구분하여 수행하였다. 진동시험을 통하여 계측된 시편의 가속도응답신호와 입력운동신호로부터 진동의 크기에 따라 진동수응답함수를 작성하였다. 다항식회귀분석기법을 이용한 모드분석기법으로 시편의 진동수응답함수를 분석하여 모드특성을 추출하고, 진동수준에 따른 시편의 동특성 변화를 검토하였다. 연구결과, 대상 기기는 입력진동의 크기가 증가할수록 모드진동수와 모드감쇠비가 비선형적으로 변화하는 것을 확인하였다. 문짝이 부착된 경우에는 문짝이 탈거된 경우에 비하여 캐비닛의 모드감쇠가 증가한다.

PET섬유와 $\alpha-Fe_2O_3$ 입자의 표면전하간 상호작용이 고형오구의 세척성에 미치는 영향 (Influence of Interaction of Surface Charges of PET Fiber and $\alpha$-Fe2O3 Particle on Detergency of Particulate Soil)

  • The adhesion and removal of $\alpha$-Fe2O3 particles on the from PET fabric in NPE solution with different ionic strength were discussed in terms of interaction of surface charge of particle and substrate. The adhesion of $\alpha$-Fe2O3 particles to PET fabric and its removal from PET fabric were carried out by using water bath shaker and Terg-O-Tometer under various solution conditions. The ζ potential of PET fiber and $\alpha$-Fe2O3 particles in the detergent solution were measured by steaming potential and microelectrophoresis methods, respectively. The adhesion and removal amount of $\alpha$-Fe2O3 particles on the from PET fabric increased with increasing time of adhesion and removal, and the rates of adhesion and removal were high at the initial stage of adhesion and removal, and then the rates decreased with passing time. The adhesion and removal amount of $\alpha$-Fe2O3 particles on and from PET fabric increased with increasing pH of solution regardless ionic strength. The tendencies and degree of adhesion and removal were very similar regardless interaction of surface charge of particle and fiber. Therefore, in the presence of a surfactant and electrolyte, the influence of interaction of surface charge of particle and substrate on the detergency of particulate soil was small.
